Osteogenesis imperfecta (OI) is a genetically heterogenous disorder characterized by brittle bone and recurrent fractures. The majority of OI is clinically categorized into mild, moderate, severe, and perinatal lethal forms. A clinical study on the use of intermittent parathyroid hormone (PTH) as an anabolic therapy for OI found PTH was effective in mild, but not in individuals with moderate/severe OI. However, this sub-group analysis was relatively small and considered preliminary by the authors. Since then, research on PTH therapy in OI has focused on mild OI and no studies of PTH use in mod

Our study is a retrospective multicenter observational cohort study to investigate effect of the use of antenatal corticosteroids (ACS) in preterm infants on the severity of necrotizing enterocolitis (NEC) and its associated complications. We collected clinical data from 443 preterm infants with gestational age (GA) of less than 37 weeks who were diagnosed with NEC in four hospitals across various provinces in China, covering the period from June 2020 to June 2024. According to whether they received a full course of prenatal corticosteroid treatment in the week before delivery, infants were di

Osteogenesis imperfecta (OI), characterized by bone fragility and low bone mass, is predominantly caused by mutations in type I collagen. High bone mass OI (HBM OI) is a rare form caused by heterozygous missense mutations at the type I procollagen C-propeptide cleavage site. Knock-in HBM OI mice were generated to elucidate the effect of this mutation on cells and bone. HBM OI murine femora contain increased monomeric pro-α1(I)C-propeptide and pC-collagen; their bone collagen fibrils have a "barbed-wire" appearance. Decreased C-propeptide cleavage diminishes bone strength. HBM OI femora are ext

Craniometaphyseal dysplasia is a rare sclerosing bone disorder that may be difficult to recognise in early childhood. We report a patient initially evaluated in the toddler period for macrocephaly and craniofacial dysmorphism, in whom early neuroimaging was unremarkable, supporting the diagnosis of macrocephaly of infancy. During early childhood, following head trauma, incidental radiological findings revealed progressive cranial sclerosis. Persistent macrocephaly and craniofacial features prompted further investigation, leading to the diagnosis of craniometaphyseal dysplasia, confirmed by ide

Osteogenesis imperfecta (OI) is a heterogeneous type 1 collagenopathy characterized by recurrent fractures, decreased bone mass, and short stature. Bisphosphonates reduce fracture incidence in children with OI, but do not improve growth velocity. C-type natriuretic peptide (CNP) is produced in the growth plate (also in the brain and heart), and it positively regulates linear bone growth; people with OI have been shown to have reduced serum levels of CNP. This pilot study evaluated whether a CNP analog combined with alendronate (ALN) improves growth and BMD in oim/oim (OIM) mice, a model of mod

SAPHO syndrome is a rare autoimmune disorder, and the occurrence of renal involvement in this condition is even more uncommon. We report the case of a 43-year-old male patient with SAPHO syndrome, who initially presented with palmoplantar pustulosis and pain in the chest wall and sternoclavicular joints, accompanied by proteinuria and microscopic hematuria. He was admitted to the hospital due to headache, lower limb edema, and elevated serum creatinine. Renal biopsy revealed stage II membranous nephropathy combined with analgesic nephropathy. Following treatment, his symptoms resolved, and at

SAPHO syndrome is a rare autoinflammatory disorder with osteoarticular and cutaneous involvement. Paradoxical skin lesions have been reported during treatment with tumor necrosis factor-α (TNF-α) inhibitors. We describe a 42-year-old man with a 6-year history of SAPHO syndrome presenting with chronic sternoclavicular and lumbosacral pain and sterile pustules on the lower extremities. Conventional therapy with etoricoxib and methotrexate improved musculoskeletal symptoms but failed to resolve skin lesions, while adalimumab treatment led to progressive worsening of cutaneous manifestations. OBJECTIVES: Successful transition from paediatric to adult healthcare is integral in the management of chronic conditions. Osteogenesis imperfecta (OI) is a rare genetic connective tissue disorder affecting the skeleton and several other organ systems, with lifelong implications for clinical management. Consensus guidelines for the transition of patients with OI do not currently exist. We aimed to evaluate our institution's transition program for patients with OI and identify strengths and weaknesses of the current process.
METHODS: Participants were recruited from April to August 2023 for th

BACKGROUND: Osteogenesis imperfecta (OI) is a heritable connective tissue disorder that presents significant physical and psychosocial challenges throughout life. In chronic conditions that may result in physical disability such as OI, resilience and the ability to adapt in the face of adversity may influence how individuals experience and manage these challenges.
OBJECTIVES: This study aimed to identify sociodemographic and clinical predictors of resilience among adults with OI.
METHODS: A cross-sectional study was conducted with 271 English-speaking adults in the United States with a self-

Autosomal recessive osteopetrosis is a rare genetic disorder caused by impaired osteoclast function, leading to excessive bone mass, defective remodeling, and fragility fractures. While bone density alterations are well recognized, compositional parameters remain poorly characterized. Here, we applied Raman spectroscopy to characterize bone defects in two murine models, the RANKL-deficient Rankl-/- (mild form) and the TCIRG1-deficient oc/oc (severe form), compared with healthy controls. Raman analysis revealed a gradient in mineral-to-matrix ratio, progressively showing lower values from healt

BACKGROUND: Multiple osteochondromas (MO) is a rare, inherited disorder characterized by multiple benign bone tumors. Although pain and fatigue are commonly encountered in clinical practice, their impact on children with MO remains largely unaddressed in literature. This study aimed to (1) assess fatigue and pain levels in children with MO, (2) compare fatigue severity with healthy peers, and (3) identify variables associated with fatigue and pain.
METHODS: In this cross-sectional study, 230 children (4-18yrs.) with MO were invited to complete validated, age-specific online questionnaires add

BACKGROUND: Pyle disease is a rare metaphyseal dysplasia caused by loss-of-function variants in SFRP4, leading to abnormal cortical modeling with relative preservation of trabecular bone. The mechanisms involved in severe and persistent bone pain in adult patients remain incompletely understood.
CASE PRESENTATION: We report an adult woman with genetically confirmed homozygous SFRP4 p.Asn53Tyr-related skeletal disease presenting with marked and persistent bone pain. Dual-energy X-ray absorptiometry showed preserved lumbar bone mineral density but markedly reduced femoral neck bone mass, consis

Osteogenesis imperfecta (OI) is a genetically heterogeneous connective tissue disorder characterized by bone fragility, skeletal deformities, and wide clinical variability. While clinical manifestations of OI are well recognized, the microstructural relationships of specific genetic subtypes remain less systematically addressed. Therefore, we aimed to examine and review mutations in genes involved in collagen type I biosynthesis and its processing and relate these genetic alterations to mineralization patterns and distinct histological bone signatures. Common features across OI types include i

BACKGROUND: Osteogenesis imperfecta (OI) is a rare genetic bone disorder. Children with OI have a higher risk of intraoperative pain. The fascia iliaca compartment block (FICB) can reduce pain, improve intraoperative hemodynamic stability, and facilitate postoperative recovery. This study explores the feasibility and potential role of FICB in multimodal analgesia by describing its application experience in children with OI.
METHOD: This study involved children with OI who underwent surgery for femoral shaft fractures. The FICB group received ultrasound-guided FICB combined with general anesth

Monogenic diabetes covers single-variant inherited subtypes of diabetes, of which the most prevalent are HNF4A-MODY (MODY1), GCK-MODY (MODY2), HNF1A-MODY (MODY3), HNF1B-MODY (MODY5) and mitochondrial diabetes. Together, they account for about 1% of diabetes cases, but more than 70% of affected individuals are classified with other forms of diabetes. The classical phenotype is young-onset, non-autoimmune diabetes with partially preserved insulin secretion and a family history of diabetes. Correct genetic diagnosis is crucial for family counselling and optimal diabetes management, as reviewed in

We report a rare case of a 40-year-old woman with longstanding craniofacial fibrous dysplasia involving the right maxilla, orbital cavity, and fronto-parietal bones. The patient underwent partial maxillary debulking 7 years ago for cosmetic reasons and orbital decompression 3 years ago, and is currently on long-term antiresorptive therapy. Bone scintigraphy with 99mTc-MDP showed multiple sites of increased osteoblastic activity. 99mTc-FAPI SPECT/CT also revealed focal uptake in craniofacial and other lesions, indicating active fibroblast proliferation. This is the first reported case of 99mTc-

OBJECTIVES: Vosoritide, a C-type natriuretic peptide analog, is the first approved pharmacological treatment for achondroplasia that targets the overactive FGFR3 pathway and promotes longitudinal bone growth. As increased body height may improve biomechanical leverage and influence muscle function, this retrospective, monocentric, observational study aimed to evaluate changes in muscle performance parameters during long-term vosoritide treatment.
METHODS: Nineteen (19) children with achondroplasia treated with vosoritide for at least 12 months were included. Muscle function was assessed via t

Hereditary multiple exostoses (HME) represent a rare skeletal disorder characterized by multiple osteochondromas, often leading to angular deformities in the lower limbs as well as leg length discrepancy (LLD), managed with tension band plates (TBP) for deformity correction. However, the utility of both angular deformity and LLD in HME has not been comprehensively evaluated. In this study, we retrospectively reviewed 25 pediatric patients with HME who visited our institution and reached skeletal maturity between 2012 and 2024, assessing a total of 50 limbs. Surgical indications included patien
