Diffusion Tensor Imaging of Short T2 Tissues Using Quantitative Ultrashort Echo Time Double-Echo Steady-State: A Feasibility Study.

To evaluate the feasibility of diffusion tensor imaging (DTI) using quantitative ultrashort echo time double-echo steady-state (qUTE-DESS) MRI in short T2 musculoskeletal tissues, we validated it in phantoms, an ex vivo porcine hoof, and an in vivo human knee. The qUTE-DESS sequence was implemented on a clinical 3 T MRI system, enabling simultaneous estimation of T1, T2, and diffusivity in tissues with rapid signal decay. Data were acquired with six diffusion-weighting orientations (x, y, z, xy, yz, xz) to obtain mean diffusivity (MD) and fractional anisotropy (FA). Sucrose and agarose phantom

EXOSC10 haploinsufficiency causes primary microcephaly by derepression of Sonic hedgehog signalling.

The evolutionarily conserved RNA exosome complex modulates gene expression during development. Mutations in RNA exosome complex subunits have been implicated in various human brain disorders, suggesting that defects in RNA decay are linked to impaired neural development. In our study, we identified de novo variants of EXOSC10 in microcephalic individuals. The patient's phenotype can be replicated by heterozygous conditional knockout of Exosc10 in the developing mouse forebrain. The heterozygous loss of Exosc10 in the developing mouse cortex leads to increased cell cycle exit, with a premature

Aberrant Molecular Myelin Architecture in Charcot-Marie-Tooth Disease Type 1A and Hereditary Neuropathy With Liability to Pressure Palsies.

Charcot-Marie-Tooth Disease Type 1A (CMT1A) and Hereditary Neuropathy with Liability to Pressure Palsies (HNPP) are the most common inherited peripheral neuropathies and arise from copy number variation of the Peripheral Myelin Protein 22 (PMP22) gene. While secondary axon degeneration has been proposed as the primary driver of disability, our prior work demonstrated pronounced neuromuscular impairment in CMT1A model mice in the absence of overt axonal loss, prompting investigation into primary myelin dysfunction. Here, we reveal that altered PMP22 dosage profoundly disrupts molecular architec

Temporomandibular Disorders: Prevalence and Associated Biopsychosocial Factors. An Analysis of Data From the NAKO Health Study.

BACKGROUND: "Temporomandibular disorders" (TMD) is an umbrella term for symptoms in the area of the temporomandibular joints and muscles of mastication. TMD constitute one of the more common types of musculoskeletal pain. Previous estimates of the prevalence of TMD suffered from methodological limitations. The NAKO Health Study enables, for the first time, an examination-based estimate of the prevalence of painful TMD in a large population sample. METHODS: The baseline survey (2014-2019) was carried out in a partial population sample and involved standardized examinations of jaw mobility and

Gastrointestinal symptoms and quality of life in adults with Silver-Russell syndrome: a cross-sectional study.

BACKGROUND AND OBJECTIVE: Most children with the rare disorder Silver-Russell syndrome (SRS) have feeding difficulties and/or gastrointestinal symptoms such as vomiting and constipation, worsening their postnatal growth. Whether the complaints remain in adult life has not been evaluated by large specialised national centres. DESIGN/SETTING/PATIENTS: All 28 individuals who were referred to the national centre for SRS as children were approached at an adolescent or adult age. We used the validated Gastrointestinal Symptom Rating Scale (GSRS) questionnaire to assess gastrointestinal symptoms dur

Value and Use of Pediatric Postmortem Magnetic Resonance Imaging.

BACKGROUND: The gold standard for postmortem investigations of fetal and pediatric deaths involving neurological pathologies is a traditional autopsy; however, acquiring all diagnostic information from an autopsy alone can be challenging. In addition, there are substantial obstacles to performing traditional autopsies on the pediatric population such as parental opinion, religious beliefs, and cultural backgrounds. Postmortem computed tomography, sonography, and magnetic resonance (MR) imaging are less invasive alternative approaches for investigating central nervous system-related deaths. Alt

The E3 Ubiquitin Ligase DCAF13 Is Essential for Craniofacial Development.

Craniofacial morphogenesis requires precisely coordinated proliferation, migration, and differentiation of cranial neural crest (CNC) cells during development. Although DDB1- and CUL4-associated factor 13 (DCAF13) is known to play critical roles in early embryogenesis and tumorigenesis, its function in neural crest development remains unknown. Here, we identify a novel role for DCAF13 in craniofacial development. Conditional knockout of Dcaf13 in neural crest lineages resulted in severe craniofacial malformations characterized by impaired skeletal growth and differentiation. Mechanistically, D

HIF1α gates tendon response to overload and drives tendinopathy independently of vascular recruitment.

Tendons are sparsely vascularized connective tissues that link muscles to bones, withstanding some of the highest mechanical stresses in the body. Mechanical overloading and tissue hypervascularity are implicated in tendinopathy, a common musculoskeletal disorder, yet their mechanistic roles remain unclear. Here, we identify hypoxia-inducible factor 1α (HIF1α) as not only a marker but also a driver of tendinopathy. Histological and multiomics evaluation of human tendinopathic samples revealed extensive extracellular matrix remodeling, including pathological collagen cross-linking coinciding wi

Visual perioperative estimation of correction of the acetabular fragment in periacetabular osteotomies is sufficient but outliers exist.

BACKGROUND: Periacetabular osteotomy (PAO) is a well-established surgical treatment of symptomatic developmental dysplasia of the hip in young patients by improving the lateral centre-edge angle (LCEA) and the acetabular index (AI). Hence, the correction of the acetabular fragment is a hallmark of the PAO procedure. PURPOSE: To evaluate the efficacy of fluoroscopy-guided visual estimation of the correction of the fragment during PAO. METHODS: 49 patients undergoing PAO were enrolled. The surgeons (3) were asked to visual assess the perioperative LCEA and AI. Perioperative assessments were co

Domain-specific phenotypic profiles in RAF1-related Noonan syndrome.

Pathogenic variants in RAF1 are a common cause of Noonan syndrome (NS), accounting for approximately 5% of cases. Nonetheless, RAF1-related NS is often associated with severe clinical features, particularly hypertrophic cardiomyopathy (HCM). Although initial studies highlighted the occurrence of genotype-phenotype correlations, a comprehensive analysis specifically focused on RAF1 variants is still lacking. We conducted a retrospective observational study combining newly collected cases of RAF1-related NS from a national multicenter retrospective cohort with systematically reviewed cases from

Virus-induced endothelial senescence as a cause and driving factor for ME/CFS and long COVID: mediated by a dysfunctional immune system.

Myalgic encephalomyelitis/chronic fatigue syndrome (ME/CFS) and long COVID are two post-viral diseases, which share many common symptoms and pathophysiological alterations. Yet a mechanistic explanation of disease induction and maintenance is lacking. This hinders the discovery and implementation of biomarkers and treatment options, and ultimately the establishment of effective clinical resolution. Here, we propose that acute viral infection results in (in)direct endothelial dysfunction and senescence, which at the blood-brain barrier, cerebral arteries, gastrointestinal tract, and skeletal mu

Structure of SHOC2-KRAS-PP1C complex reveals RAS isoform-specific determinants and insights into targeting complex assembly by RAS inhibitors.

RAF activation is essential for MAPK signaling and is mediated by RAS binding and the dephosphorylation of a conserved phosphoserine by the SHOC2-RAS-PP1C complex. MRAS forms a high-affinity SHOC2-MRAS-PP1C (SMP) complex, while canonical RAS isoforms (KRAS, HRAS, NRAS) form analogous but lower-affinity assemblies. Yet, cancers driven by oncogenic KRAS, HRAS, or NRAS remain strongly SHOC2-dependent, suggesting that these weaker complexes contribute to tumorigenesis. To elucidate how canonical RAS proteins form lower-affinity ternary complexes, the cryo-EM structure of the SHOC2-KRAS-PP1C (SKP)

Analgesic effect of iliofascial compartment combined with quadratus lumborum block for pediatric developmental dysplasia of the hip surgery: a retrospective cohort study.

BACKGROUND: Effective postoperative analgesia for pediatric developmental dysplasia of the hip (DDH) surgery remains a challenge. This study aimed to compare the analgesic efficacy of a combined fascia iliaca compartment block (FICB) and quadratus lumborum block (QLB) versus patient-controlled intravenous analgesia (PCIA) in pediatric patients undergoing open reduction surgery for DDH under general anesthesia. METHODS: This retrospective cohort study enrolled children under 16 years old undergoing DDH surgery between January 2016 and January 2024. Patients were divided into two groups based o

Copy Number Variants in the 11p15.5 Associated Imprinting Disorders: An Attempt to Establish a Genotype-Phenotype Correlation.

Copy number variations (CNVs) affecting the imprinted regions in 11p15.5 (imprinting centre 1 and 2/IC1, IC2) account for more than 2% of the molecular disturbances in Beckwith-Wiedemann and Silver-Russell syndrome (BWS, SRS) and are associated with a recurrence probability of up to 50%. However, their clinical impact can be challenging to estimate, as it depends on the type of imbalance, the parental origin of the affected allele, its size and genomic content. As a result, a genotype-phenotype correlation of 11p15.5 alterations is still missing, at least for CNVs affecting only parts of the I

"You could get the best of both breeds or the worst of both": UK public attitudes towards crossbreeding in dogs - with a specific focus on brachycephalic dogs.

Extreme conformation and reduced genetic diversity are recognised to lead to severely reduced health, welfare and longevity in certain dog breeds. There is growing interest in applying strategic crossbreeding to promote more moderate conformations and greater genetic diversity within currently problematic breeds. Crossbreeding could therefore lead to more rapid and effective improvements in welfare compared to current practices of within-breed selection. Deliberate crossbreeding between distinct different dog breeds is not a new concept; it was historically commonly used to create the current

Digital planning of orthognathic surgery with aligners: ortho-facial-driven, a viable protocol for surgery-first treatment.

INTRODUCTION: Traditionally, orthodontic-surgical treatment is performed in three stages: presurgical orthodontics, orthognathic surgery and post-surgical orthodontics. Though predictable and stable, this protocol requires a long treatment time and can temporarily compromise the patient's aesthetics. Early benefit surgery eliminates or reduces the initial stage of orthodontic treatment, providing more immediate aesthetic and functional improvement. OBJECTIVE: To present a virtual diagnosis and planning protocol, combining cone beam computed tomography (CBCT) and digital models applied to earl
