Treating pain and trauma: A systematic review of non-randomized studies of psychotherapeutic approaches.

The co-occurrence of chronic pain and trauma symptoms is prevalent; thus treatment approaches that address both are required. This systematic review of non-randomized studies aims to describe available psychotherapeutic interventions that concurrently target chronic pain and trauma-related symptoms for adults. Five databases (MEDLINE, Embase, the Cochrane Central Register of Controlled Trials, the Cochrane Database of Systematic Reviews, PsycINFO) were searched from date of inception until October 8th, 2025. Eligible studies featured a psychological intervention addressing chronic pain and tra

Psychoactive medication use and cardiac electrophysiology in hypermobile Ehlers-Danlos syndrome and hypermobility spectrum disorder: a retrospective analysis.

CONTEXT: Ehlers-Danlos syndrome (EDS) and hypermobility spectrum disorder (HSD) are connective tissue disorders associated with musculoskeletal manifestations and significant psychiatric comorbidities. Psychoactive medications commonly utilized in these populations raise concerns regarding potential cardiac effects. OBJECTIVES: This study explores associations between psychoactive medication utilization and electrocardiogram (ECG) parameters in patients with hypermobile Ehlers-Danlos syndrome (hEDS) and HSD to provide insight into potential cardiac considerations in this population. METHODS:

Pain, Sleep Latency, and Mental and Physical Health in Individuals with Self-Reported Hypermobile Ehlers-Danlos Syndrome.

Background: Individuals with hypermobile Ehlers-Danlos syndrome (hEDS) frequently report chronic pain, sleep-related complaints, and reduced mental and physical health. While sleep disturbance has been described in this population, less is known about how pain frequency relates to sleep initiation and broader health burden. This study aimed to examine the associations between pain frequency, sleep latency, and mental and physical health burden in individuals with hEDS. Methods: This analysis used cross-sectional survey data collected from adults with self-reported EDS. Pain frequency was treat

Mast cell score associates with wide-spread mast cell symptoms and comorbidities in patients with hEDS and HSD.

BACKGROUND: Wide-spread mast cell (MC)-associated symptoms and MC activation syndrome (MCAS) are often reported in patients with hypermobile Ehlers-Danlos syndrome (hEDS) and hypermobility spectrum disorders (HSD). The goal of this study was to develop a novel MC score based on 11 self-reported MC-related conditions with clinical and research utility to better understand MC symptoms in hEDS and HSD patients. METHODS: From November 1, 2019, to June 13, 2025, patients ( n =2,141) filled out an Intake Questionnaire at the Mayo Clinic Florida EDS Clinic that included 11 self-reported questions re

Zinc transporter ZIP13 in mesenchymal tissues: From intracellular metal distribution to systemic homeostasis.

BACKGROUND: Zinc is an essential trace element that plays pivotal roles in development, immunity, inflammation, and aging. Precise regulation of intracellular zinc distribution is mediated by zinc transporters of the ZIP (SLC39) and ZnT (SLC30) families, and their functions are increasingly recognized as tissue- and organelle-specific. Among these transporters, ZIP13 (SLC39A13) is a key regulator of mesenchymal biology. Genetic studies have demonstrated that loss of ZIP13 function causes connective tissue abnormalities, impaired bone and tooth development, and spondylodysplastic Ehlers-Danlos

Association between Ehlers-Danlos syndrome and complex regional pain syndrome: a large nationwide claims database study.

BACKGROUND: Ehlers-Danlos syndrome (EDS) is a genetic connective tissue disorder characterized by joint instability and recurrent injuries, which has been linked to chronic pain syndromes, including complex regional pain syndrome (CRPS). This study examines the co-occurrence of EDS and CRPS and evaluates demographic differences using a large national claims database, with findings interpreted as population-level signal detection based on diagnostic coding. METHODS: This retrospective cohort study used 2017-2022 Merative MarketScan data (IRB# 2017-0169), including commercially insured US patie

Distinct sensory and autonomic involvement in hypermobile Ehlers-Danlos syndrome compared with idiopathic small fiber neuropathy: a multimodal study.

Hypermobile Ehlers-Danlos syndrome (hEDS), frequently presents with pain and autonomic symptoms suggestive of small fiber neuropathy (SFN). However, systematic comparisons between hEDS and idiopathic SFN (iSFN) using combined clinical, functional, and morphological approaches are lacking. We prospectively studied a population of SFN patients who also fulfilled the 2017 criteria for hEDS (hEDS/SFN) and compared them with a group of iSFN patients of similar age. All underwent SFN-Symptoms Inventory Questionnaire (SFN-SIQ), Douleur Neuropathique 4 (DN4), and the Composite Autonomic Symptom Score-

Prevalence of Atopic Disease in Pediatric Patients with Hypermobile Ehlers-Danlos Syndrome.

Background:The Ehlers-Danlos syndromes (EDSs) are a group of connective tissue disorders characterized by skin laxity and hypermobile joints. EDS is associated with many comorbidities that often necessitate multidisciplinary care. Adults with EDS report more atopic conditions such as allergic rhinitis and asthma; however, gaps exist around the prevalence of atopic conditions in pediatric patients with EDS. The objective of this study was to determine the prevalence of atopic conditions (allergies, asthma, and atopic dermatitis) in pediatric patients with hypermobile EDS (hEDS) and compare it t

The hypermobility crisis: A review and call to action.

Hypermobile Ehlers-Danlos syndrome (hEDS) and hypermobility spectrum disorders (HSD) are common yet underrecognized connective tissue disorders characterized by joint hypermobility and multi-organ system involvement. Current diagnostic criteria inadequately capture the clinical heterogeneity of hEDS/HSD. Consequently, patients often face diagnostic delays and fragmented care, which exacerbate underlying comorbidities such as chronic pain and psychological distress. This narrative review explores the key limitations of existing diagnostic criteria, the burden of underrecognized comorbidities, a

Bone phenotype in a mouse model of Classical Ehlers Danlos syndrome with Col5a1 haploinsufficiency.

Type V collagen is a key regulator in the formation of type I collagen fibrils in connective tissues including skin, tendons, and bones. Abnormalities in type V collagen cause classical Ehlers Danlos syndrome (EDS), a connective tissue disorder with primarily skin and joint phenotype. There are several clinical reports suggesting increased prevalence of vertebral fractures in adult patients with EDS, among them patients with classical EDS. Yet, the literature lacks data about bone health specifically in the classical EDS patient population. The Col5a1 haploinsufficient mouse model (Col5a1+/-)

Family-oriented support in genetic counselling: a scoping review of clinical practice and psychotherapeutic interventions.

Families living with inherited genetic conditions (IGCs) face practical and emotional challenges that affect individual well-being and family relationships. Although genetic counselling recognises the familial nature of genomic risk, support is often limited to patient-led disclosure. This scoping review examined how family-oriented support is conceptualised, delivered, and evaluated within genetic counselling and related psychosocial services. We define family-oriented support as any practice that explicitly considers relatives and family relationships, and systemic family-oriented care as fa

Combined Mutations of LTBP3 and COL5A1A in Geleophysic Dysplasia.

Geleophysic dysplasias (GDs) are uncommon genetically predisposed abnormalities that interfere with skeletal growth and formation. Several GD subtypes have different clinical manifestations. The current report presents the case of a 7-year-old Syrian boy with a medical history of repeated bone fractures and noticeable facial characteristics. Initial laboratory examinations revealed normal results, except for low serum phosphate and ferritin levels. The X-ray images showed no abnormalities. The DEXA scan was like that of a 92.1-year-old. Karyotype analysis revealed 46 XY. Genetic testing result

Linking central sensitization to multisystemic manifestations in hypermobile Ehlers-Danlos syndrome.

Hypermobile Ehlers-Danlos Syndrome (hEDS) and Hypermobility Spectrum Disorders (HSD) are complex multisystemic conditions frequently associated with chronic pain. Central Sensitization (CS)-a state of neural amplification and hyperexcitability-is hypothesized to be a unifying mechanism underlying the heterogeneous symptoms in chronic pain patients. Our aim was to investigate the association between central sensitization and multisystemic symptom burden in patients with hEDS/HSD while identifying independent clinical predictors of CS. We prospectively enrolled 150 adults diagnosed with hEDS/HSD

Familial Chiari Malformation: Prevalence of Connective Tissue Disorders and Other Comorbidities.

BACKGROUND AND OBJECTIVES: Chiari malformation type 1 (CM-I) is frequently associated with connective tissue disorders (CTDs), particularly Ehlers-Danlos syndrome (EDS), which may increase surgical risk and complicate management. Familial clustering of CM-I and CTDs has been reported, but their prevalence within families remains underexplored. This study aimed to determine the prevalence of CTDs and comorbidities among family members of CM-I patients, comparing familial and sporadic cases. METHODS: We conducted a retrospective survey-based study of patients who underwent posterior fossa decom

Anterior Capsular Reconstruction With Dermal Allograft Augmentation for Multidirectional Shoulder Instability.

Multidirectional shoulder instability, particularly in patients with underlying connective tissue disorders poses a surgical challenge. After exhaustive nonoperative treatment, surgical intervention is focused on restoration of the anterior and inferior capsular restraints. Oftentimes, after longstanding recurrent instability, the anterior capsule is diminutive with diffuse scarring and adhesions, requiring meticulous surgical dissection and allograft augmentation. Although outcome data are sparse, dermal allograft augmentation offers enhanced structure to the glenoid and humerus, and variable

Recurrent Acute Coronary Syndrome in Spontaneous Coronary Artery Dissection.

BACKGROUND: Spontaneous coronary artery dissection (SCAD) is an important cause of acute coronary syndrome (ACS) in vascular Ehlers-Danlos syndrome (vEDS). CASE SUMMARY: A 35-year-old with vEDS, found to have SCAD on coronary computed tomography angiography, was admitted with recurrent episodes of ACS. After each event, a heart team recommended medical management, eventually leading to resolution of symptoms without the need for invasive revascularization. WHY BEYOND THE GUIDELINES?: There are few guidelines for the management of recurrent ACS in the setting of SCAD, and clinicians are left w

Arthroscopic Anterior and Posterior Glenohumeral Capsular Augmentation With Gracilis Allograft.

Multidirectional instability in patients with connective tissue disorders such as Ehlers-Danlos syndrome (EDS) is difficult to treat due to poor tissue quality and frequent failure of standard repairs. We describe an arthroscopic technique using gracilis tendon allografts to perform an anterior and posterior capsular augmentation in the revision setting with subscapularis deficiency. Allografts are anchored to the glenoid and humeral head in a sling configuration, restoring capsular restraint and stability. This method builds upon prior posterior-only augmentation by providing circumferential

Case Report: A hypothesis-generating case of gadolinium retention and persistent symptoms after MRI despite normal renal function in a patient with hypermobile Ehlers-Danlos syndrome.

Gadolinium-based contrast agents are commonly used to enhance magnetic resonance imaging. In 2006, Thomas Grobner identified a link between these agents and nephrogenic systemic fibrosis in patients with renal impairment. Gadolinium exposure has also been associated with acute kidney injury, skin plaques, and encephalopathy. Even in patients with normal kidney function, gadolinium may be retained in tissues for prolonged periods after contrast administration. Some patients develop persistent and varied symptoms following exposure. Although Richard Semelka and colleagues proposed the term gadol
