Quantitative ultrasonographic assessment of the orbicularis oris muscle following cleft lip surgery: A case-control study.

BACKGROUND: This case-control study aimed to quantitatively assess the orbicularis oris muscle (OOM) in children with unilateral cleft lip and palate (UCLP) after primary lip repair using high-resolution ultrasonography, and to compare these measurements with children presenting isolated cleft palate (CP) without lip involvement. METHODS: For this, 12 infants with UCLP who had undergone primary lip repair at 4-5 months of age were examined during subsequent palatoplasty at 9-12 months. They were compared to eight age-matched controls with isolated CP. Using a 22 MHz "hockey stick" transducer

Perinatal genetic diagnostic yield in a population of fetuses with the phenotype arthrogryposis multiplex congenita: a cohort study 2007-2021.

Arthrogryposis multiplex congenita (AMC) presents challenges for prenatal detection due to its heterogeneous etiology, onset, and phenotypical manifestations. This study aims to describe the genetic diagnostic yield in a population of fetuses with detailed phenotypic description over a 15-year period (2007-2021) at the Fetal Medicine Unit of Amsterdam UMC, the Netherlands. The fetal and neonatal phenotypes were classified into three clinical AMC Groups, with the exception that Groups 1 and 2 were combined in the prenatal classification. Group 1 involves limb involvement primarily, Group 2 incl

Understanding the role of Shroom3 in the developing mouse myocardium.

Loss of actin cytoskeleton control can hinder integral developmental and physiological processes and can be the basis for a subset of developmental defects. SHROOM3 is an actin binding protein, best characterized as being essential for neural tube closure in vertebrates. Shroom3 expression has also been identified in the developing heart, with some associated congenital heart defects. Here we show that the expression pattern of Shroom3 in the developing and adult mouse heart is specific to the myocardium. Using a gene trap line, we show that embryos with homozygous full-body Shroom3 loss die a

Billing Practices in Pediatric Fronto-Orbital Advancement Surgeries: Review of a National Billing Database.

BACKGROUND: Fronto-orbital advancement (FOA) for craniosynostosis is the standard treatment for patients with anterior cranial vault differences. Despite its widespread use, national billing practices for FOA have not been described. This study analyzes billing codes from a national multicenter claims database to evaluate variations in billing for FOA, aiming to inform standardization efforts. METHODS: We performed a retrospective study using PearlDiver, a national deidentified aggregate claims database covering over 100 million patients in the United States. Patients aged 0 to 16 years who h

Assessment of hip joint mechanics during walking in people with Marfan syndrome.

BACKGROUND: Marfan syndrome (MFS) is an autosomal dominant connective tissue disorder and is associated with muscle dysfunction. Individuals with MFS exhibit quadriceps weakness, hip joint pain, and an increased incidence of hip osteoarthritis (OA) compared to the general population, yet the underlying biomechanical factors associated with these poor hip-related outcomes are not well understood. RESEARCH QUESTION: The purpose of this study was to assess the effects of MFS on gait mechanics and to determine the relationship of gait mechanics with hip-related outcomes within the MFS group. Evaluating the quality of life in mothers caring for children with congenital clubfoot: prospective sequential evaluation along the course of Ponseti management.

The post birth revelation of child with physical anomaly taxes maternal hopes of a healthy child. However, unlike many other congenital anomalies, the Ponseti method has enabled an early and effective treatment of the clubfoot deformity. Our study aimed to assess the quality of life (QoL) of mothers with child born with idiopathic clubfoot through various stages of its management compared to a healthy infant. The study was conducted on mothers of children born with untreated congenital idiopathic clubfoot presenting within first 3 months after birth and undergoing Ponseti treatment. The WHO Qu

A systematic review of wide alveolar cleft repair techniques.

Wide alveolar clefts impair secondary alveolar bone grafting due to deficient mucoperiosteal tissue for grafted bone coverage. Consequently, preparatory or alternative techniques are often required for closure of such defects. We conducted a PRISMA-adherent systematic literature review on wide alveolar cleft repair to compare treatment efficacies and patient populations. With this information, we provide guidance on the relative advantages and disadvantages of each examined method. Forty-two studies published from 1987-2022 were included, containing 332 patients treated with distraction osteog

Myocardial reprogramming by HMGN1 underlies heart defects in trisomy 21.

Congenital heart defects (CHDs) are the most common developmental abnormalities, affecting around 1% of live births1. Aneuploidy causes around 15% of CHDs, with trisomy 21 (also known as Down syndrome) being the most frequent form2. CHDs occur in around 50% of cases of Down syndrome, with an approximately 1,000-fold enrichment of atrioventricular canal (AVC) defects that disrupt the junction between the atria and ventricles3,4. The AVC contains unique myocardial cells that are essential for valvuloseptal development; however, the specific combination of dosage-sensitive genes on chromosome 21

Craniofacial morphology of TcMAC21 and TcHSA21rat models of Down syndrome: An interspecific comparison.

Down syndrome (DS) is caused by an extra copy of human chromosome 21 (HSA21) and is associated with significant craniofacial anomalies among other systemic traits. While animal models have been pivotal in advancing DS research, most of the mouse models replicate only a portion of the human genetic condition. The recently developed transchromosomic rodent models of DS, TcMAC21 mice, and TcHSA21rat carry nearly complete copies of HSA21q and all of HSA21, respectively. While both TcMAC21 and TcHSA21rat express DS-like craniofacial malformations, no comprehensive interspecies comparisons have been

Remodelling of deformities and resolution of shortening in congenital posteromedial bowing of the tibia.

BACKGROUND: In congenital posteromedial bowing of the tibia, the remodeling of triplanar (medial, posterior and oblique) deformities, the resolution of tibial shortening and their correlation with each other have not been studied extensively. Additionally, the age at which tibial shortening at maturity can be accurately predicted under these conditions is not known. This study aimed to evaluate the natural history of tri-planar deformity remodeling, the resolution of tibial shortening and the difference between tibial shortening predicted by the multiplier method at first presentation and subs

Talar Morphology of Charcot-Marie-Tooth Patients With Cavovarus Feet.

BACKGROUND: Charcot-Marie-Tooth disease (CMT), a common inherited neurologic disorder, significantly impacts the morphology of foot bones, particularly the talus. The disease has been classified into types based on specific mutations, with the most common being CMT type 1 (CMT1; demyelinating) and CMT type 2 (CMT2; axonal). However, the specific osseous morphologic variations in CMT patients and their major genetic subgroups remain insufficiently understood, posing challenges in clinical management and surgical intervention. METHODS: This study analyzed talar morphology in individuals with CM

Cranial neural crest shortage leads to extensive craniofacial anomalies in mice mutant for the NR2F1/2 nuclear receptors.

Nuclear receptors are iteratively deployed during neural crest development, from pre-induction through differentiation stages. NR2F1 and NR2F2 in particular have been proposed as broad regulators of early neural crest gene expression in mammals, but the timing, extent, and redundancy of their developmental requirement has remained unclear, as Nr2f1 and Nr2f2 single mouse mutants present only minimal craniofacial phenotypes. Here we report the dynamic expression patterns of Nr2f1 and Nr2f2 in the mouse cranial neural crest from specification through post-migratory stages. Combined conditional k

Associated Coalitions of Tarsal Bones: Review of the Literature and Presentation of a Classification.

Associated coalitions of tarsal bones, either unilateral or bilateral, may be classified to developmental or syndromic types. There are no specific patterns for osseous or nonosseous configurations of associated tarsal coalitions. Associated developmental tarsal coalitions can be categorized into dual, threefold, massive, and total tarsal coalitions according to the number and sites of the involved joints. Dual coalitions are more common than other types. Among dual tarsal coalitions, the most frequent combination is talocalcaneal (TC) and calcaneonavicular coalitions, also referred to as doub
