Inherited epidermal differentiation disorders (EDD) in pediatric patients from a single Italian referral Center.

UNLABELLED: Inherited epidermal differentiation disorders (EDD) are a heterogeneous group of genetic skin diseases characterized by impaired epidermal differentiation and defective skin-barrier function. These conditions include congenital ichthyoses and palmoplantar keratoderma (PPK), which are particularly rare in the pediatric population and can present with evolving and, in mild forms, nonspecific or subtle clinical features, making early diagnosis challenging in children. A retrospective review was conducted on pediatric patients with a clinical and molecular diagnosis of EDD, who were ev

Burden of congenital birth defects in the ten countries with the highest numbers of births from 1990 to 2023, with projections to 2040: A retrospective population-level ecological study.

ObjectiveCongenital birth defects (CBDs) are major causes of neonatal mortality and lifelong disability worldwide. The ten countries with the highest numbers of births account for over half of global neonates, yet their CBD burden trends remain insufficiently studied. This study aimed to assess the CBD burden and temporal trends in these countries from 1990 to 2023 and project trends to 2040.MethodsWe conducted a retrospective population-level ecological study using publicly available data from the Global Burden of Disease Study 2023 on CBD burden. We analysed incidence, mortality, disability-

Clinical Outcomes After Recovered, Positive, and Negative Pulse Oximetry Screening Results Among Newborns.

IMPORTANCE: Pulse oximetry screening (POS) detects critical congenital heart defects (CCHDs) and other neonatal morbidities, but the clinical significance of initially abnormal results that normalize on repeat testing remains unclear. OBJECTIVE: To assess the prevalence and clinical implications of positive, recovered, and negative early POS results in asymptomatic newborns. DESIGN, SETTING, AND PARTICIPANTS: This population-based cohort study included asymptomatic neonates born at 35 weeks' gestation or later who underwent early POS at 2 to 6 hours of age at a tertiary-level university hosp

The kinase inhibitor dabrafenib halts progression of cerebral cavernous malformations in a BRAF-independent manner.

Cerebral cavernous malformations (CCMs) constitute the second most common type of intracranial vascular malformations. Cavernomas arise because of somatic or autosomal dominant inherited loss-of-function mutations in CCM genes, forming fragile mulberry-like malformed blood vessel structures that are prone to leak and break. Treatment is limited to surgical removal of solitary cavernomas, and because patients often present with multiple lesions, pharmacological alternatives to surgery are needed. We performed a high-throughput screen of 5200 bioactive molecules in programmed cell death 10 (PDCD

Poly(vinyl alcohol) dissolving microneedles for transdermal semaglutide delivery and anti-obesity therapy in rats.

Semaglutide is an effective anti-obesity peptide, but repeated subcutaneous injections may reduce its acceptability for long-term therapy. We prepared semaglutide-loaded poly(vinyl alcohol) (PVA) dissolving microneedles (Sema-MNs) by sequential vacuum-assisted molding and room-temperature drying. The 15 × 15 arrays concentrated Cy5-labelled semaglutide toward the needle tips, resisted axial compression, penetrated rat skin, and partially dissolved within 5 min; skin barrier function approached baseline within 4-6 h. Sema-MNs contained 29 ± 2 μg semaglutide per patch, with an apparent delivered

Changes Associated with Hyaluronidase Enzyme in Skin Connective Tissue: An Experimental and Qualitative Study in Rats.

BACKGROUND: Hyaluronidase enzyme is frequently used by plastic surgeons in aesthetic procedures to break down hyaluronic acid. The potential benefits of hyaluronidase in the clinical management of conditions associated with diseases and aging in the skin connective tissue are a subject of ongoing research. In this study, we investigated the changes associated with the hyaluronidase enzyme injected subcutaneously into the dorsal region of rats in the skin connective tissue. METHODS: This is an experimental and qualitative study. A total of 24 Wistar albino rats were divided into three groups o

Risk factors and outcomes of early postoperative complications following congenital heart surgery: a retrospective study at a tertiary care hospital in Nairobi, Kenya.

INTRODUCTION: Congenital Heart Disease (CHD) remains a major health challenge globally, with surgical interventions being the primary treatment modality for many defects. While advancements in cardiac surgery have improved survival rates, early postoperative complications continue to pose risks, particularly in settings with limited resources like Kenya. The study aimed to assess the rate of any early postoperative complications and identify factors associated with it in children after cardiac surgery at a tertiary care hospital in Nairobi, Kenya from 2019 to 2024. METHODS: This was an single

Pregnancy outcome after first-trimester exposure to JAK inhibitors - an evaluation of the German Embryotox cohort.

BACKGROUND: Janus kinase inhibitors (JAKi) are increasingly being used in the management of autoimmune diseases across rheumatology, gastroenterology, and dermatology, including in women of childbearing age. Consequently, pregnancies occurring during JAKi treatment are becoming more common. However, uncertainties regarding their safety during pregnancy remain, and current human data are insufficient to allow a comprehensive assessment of fetal risk. METHODS: Observational data of pregnancies with first trimester JAKi exposure, recorded in the German Embryotox database, was analyzed. Pregnanci

[Molecular diagnosis of non-syndromic craniosynostosis: new challenges for real clinical practice].

Craniosynostoses (CS) are a congenital malformation caused by absence or premature closure of skull sutures. This leads to deformation and impaired growth of skull. Traditionally, syndromic and non-syndromic forms are distinguished. For a long time, it was believed that molecular genetic mechanisms are significant exclusively for syndromic CS. However, clear data on involvement of somatic and germinal mutations in pathogenesis of non-syndromic forms were accumulated in recent years. Currently, there are no unified clinical guidelines defining the strategy for molecular genetic diagnosis for no

Addressing unilateral lambdoid craniosynostosis using endoscopic suturectomy and barrel-stave osteotomies: surgical technique and literature review.

PURPOSE: Unilateral lambdoid craniosynostosis (ULC) is a rare cranial deformity requiring intervention to correct asymmetry of the posterior skull. Conventional open remodeling and distraction osteogenesis offer robust expansion but carry risks of higher blood loss, hardware complications, or reoperation. We report a novel, minimally invasive technique combining endoscopic lambdoid suturectomy with barrel-stave osteotomies, supplemented by a literature review. CASE DESCRIPTION: A 2-month-old healthy male presented with classic left ULC features, including occipitoparietal flattening, ipsilate

Fraser Syndrome From Prenatal Diagnosis to Multidisciplinary Management and Long-Term Care: A Narrative Review.

Fraser syndrome (FS) is a rare autosomal recessive congenital malformation disorder characterised by cryptophthalmos, syndactyly, and variable respiratory, renal, genitourinary, craniofacial, and other systemic abnormalities. Considerable phenotypic heterogeneity creates challenges in prenatal recognition, prognostic counselling, neonatal management, and long-term care. Prenatal diagnosis may be particularly difficult because characteristic external abnormalities are not always detectable on ultrasonography, while oligohydramnios secondary to renal abnormalities may further limit fetal visuali

Central nervous system, cranial, and cerebrovascular conditions beyond the Kimonis criteria in patients with Gorlin syndrome: a retrospective cohort study.

BACKGROUND: Gorlin syndrome is a multisystem developmental and cancer-predisposing disorder commonly caused by germline PTCH1 variants. The Kimonis clinical criteria capture the canonical phenotype; however, central nervous system (CNS), cranial, and cerebrovascular conditions documented beyond these criteria remain poorly defined. METHODS: We conducted a retrospective descriptive cohort study of 32 patients who fulfilled the Kimonis clinical criteria and received care at either or both hospitals in Japan. Clinical information through March 31, 2026, including sufficiently documented care els
