DNA repair-associated nucleases induce double-strand breaks following sequential exposure to UVA1 and UVB.

Solar UV comprises UVA and UVB; each exerts distinct biological effects, while their combined impact is not yet fully understood. We previously reported that human keratinocytes exposed to UVA1 followed by UVB irradiation exhibited severe cell death accompanied by DNA double-strand break (DSB) formation. In this study, we confirmed the DSB formation following sequential exposure to UVA1 and UVB and investigated the underlying mechanisms. The occurrence of DSBs was validated by biased sinusoidal field gel electrophoresis and the detection of phosphorylated histone H2AX and RPA. Notably, DSB ind

XAB2: a link between RNA metabolism, DNA damage repair, and human health.

Cells have evolved multiple mechanisms to preserve genome integrity, collectively known as DNA damage response (DDR). Rather than acting separately, the DDR often interacts with transcription and mRNA splicing; however, the underlying molecular mechanisms of this cross-talk are still poorly understood. Consistent with this, components of the splicing machinery are increasingly being recognized as factors with a direct role in sensing, signaling, and repairing DNA damage. Xeroderma pigmentosum group A-binding protein 2 (XAB2), which plays a well-characterized role in mRNA splicing, has also bee

Rare internal malignancies in xeroderma pigmentosum: A report of two cases from Tunisia and analysis of driver mutations.

Xeroderma pigmentosum (XP)-associated internal malignancies are characterized by their rarity, early onset, atypical histological presentations, and poorly characterized genomic landscape. In this study, we report the clinical, pathological, and molecular features of two rare XP-associated internal malignancies, focusing on their somatic mutation profiles. These tumors represented rare histological variants at their respective anatomical sites: an ovarian high-grade sex cord-stromal tumor (HG-SCST) with heterologous rhabdomyosarcomatous differentiation diagnosed in an 18-year-old female, harbo

Nucleotide excision repair and poly(ADP-ribose) polymerase 1 cooperate in HBV cccDNA synthesis: A potential viral therapeutic target.

BACKGROUND & AIMS: Blocking the synthesis of HBV covalently closed circular (ccc)DNA is crucial for achieving a viral cure. Previous work showed that inhibiting nucleotide excision repair (NER) reduced cccDNA levels in HBV-producing liver cells. Here, we aimed to clarify the role of NER endonucleases in addressing the flap structure on relaxed circular DNA (rcDNA) and to explore an antiviral strategy that disrupts viral replication reservoirs. METHODS: We investigated the need for NER endonucleases for cccDNA synthesis in liver cells by examining the interactions between the complex of excisi

A meta-analysis and expression profiling of DNA repair gene polymorphisms in leukemia.

BACKGROUND: Genetic susceptibility is believed to contribute to leukemia development. This study aimed to systematically evaluate the association between DNA repair gene polymorphisms, particularly xeroderma pigmentosum (XP) and excision repair cross-complementing (XRCC) genes, and leukemia risk. METHODS: A comprehensive search of PubMed, Web of Science, Scopus, and Cochrane Library was conducted until December 3, 2025, concerning the association between DNA repair gene polymorphisms and leukemia risk. Gene expressions were performed in Leukemia DB, GEPIA2, and BloodSpot datasets. RESULTS: F

Decoding Cancer-Associated Mutations in DNA Polymerase η through Atomistic Simulations.

DNA polymerases (Pols) are essential enzymes for DNA replication within the cell. However, DNA lesions, such as cyclobutane pyrimidine dimers (CPDs) induced by ultraviolet (UV) radiation, can impair Pols's function and DNA replication. Therefore, the presence of CPDs can, for example, lead to xeroderma pigmentosum variant (XP-V), a rare genetic disorder characterized by an increased risk of skin cancer. Nonetheless, in these situations, specific translesion synthesis (TLS) Pols, such as human DNA polymerase η (Polη), can overcome such lesions, enabling DNA polymerization. That is, Polη prevent

XPF mediates 3' flap processing for FEN1-independent Okazaki fragment maturation.

Okazaki fragment maturation (OFM), the process that removes RNA-DNA primers, is a major source of DNA replication stress and mutations. It involves Polδ-mediated DNA strand displacement synthesis that produces 5' flaps, FEN1-mediated 5' flap cleavage, and LIG1-catalyzed nick ligation. Recently, we discovered that under hyperthermal stress conditions, yeast cells convert 5' flaps into 3' flaps, which are degraded by 3' flap nucleases to produce DNA nicks. However, little is known about this 3' flap-based OFM in human cells. Here, we report that 3' flaps frequently form in various human cancer c

Engineering quantum dot surfaces to preserve protein-DNA interactions for single-molecule visualization.

Quantum dots (QDs) are fluorescent nanoparticles widely used for single-molecule imaging because of their exceptional brightness and photostability. However, the impact of QD surface chemistry on biomolecular interactions has not been systematically investigated. Here, we report that commercial QDs unexpectedly destabilize protein-DNA complexes by inducing protein dissociation from DNA. Using the human nucleotide excision repair protein, xeroderma pigmentosum complementation group A (XPA) as a model system, we demonstrate that antibody-conjugated QDs promote dissociation of XPA from DNA substr

High cancer-associated mutational burden in normal blood of xeroderma pigmentosum group C, but not groups A, D, or F.

Xeroderma pigmentosum (XP) is a rare autosomal recessive congenital syndrome characterized by defective nucleotide excision repair (NER), leading to extreme photosensitivity and a strong predisposition to skin cancer. One of the 8 complementation groups, XP complementation group C (XP-C), is defective specifically in the global genome component of NER and presents not only with increased skin cancer but also with hematologic cancers. Using error-corrected single-molecule sequencing, we show a uniquely high spontaneous somatic mutational load in peripheral blood mononuclear cells (PBMCs) of pat

XPC and global genome nucleotide excision repair are essential for telomere stability after UVC-induced DNA damage in human cells.

Telomeric sequences are hotspots for ultraviolet light (UV) induced cyclobutane pyrimidine dimers (CPD) and pyrimidine(6-4)pyrimidone photoproducts (6-4 PP), due to pyrimidine runs on both the TTAGGG and CCCTAA containing strands. Photoproducts are repaired by global genome nucleotide excision repair (GG-NER) or by transcription-coupled (TC-NER) in regions of active transcription. Since telomeres are transcribed into long telomeric repeat-containing RNA (TERRA) molecules, here we tested roles for both TC-NER and GG-NER in telomere stability following UVC irradiation. XPC-deficient cells, incap

ATP Hydrolysis Gates an Ultra-Rapid Unwinding Mode of AfXPB Revealed by DNA Electrochemistry.

DNA helicases play a central role in DNA metabolism, enabling recombination, replication, transcription, translation, and repair. Recently, an unexpectedly rapid, exponential ATP-stimulated DNA unwinding rate was observed with an Archaeoglobus fulgidus helicase (AfXPB), markedly faster than that of conventional helicases, yet the mechanistic basis remains unresolved. Here, we distinguish the roles of ATP-helicase binding and ATP hydrolysis on the dynamic DNA unwinding activity of AfXPB via an electrochemical assay with redox-active DNA monolayers. To distinguish ATP-helicase binding from ATP h

Development of rutin-loaded cerosomes for topical photoprotection and skin barrier modulation.

Xeroderma pigmentosum (XP) is a rare DNA-repair disorder associated with extreme photosensitivity, high skin cancer risk, progressive xerosis and barrier dysfunction. Current management relies largely on strict photoprotection, which does not address impaired hydration and epidermal barrier integrity. This work developed cerosomes (ceramide-containing vesicular systems) as multifunctional topical carriers combining barrier-replenishing ceramides with rutin as an antioxidant/photoprotective agent. Box-Behnken design was applied to optimize rutin-loaded cerosomes by evaluating phospholipid conce
