Analysis of survival in patients with sporadic hemangioblastoma versus that linked to von Hippel-Lindau. Experience of two reference centers.

OBJECTIVE: To establish the overall survival (OS) and disease-free survival (DFS) profiles regarding the patients diagnosed with hemangioblastoma in the context of a von Hippel-Lindau (VHL) disease and those of sporadic presentation. METHODS: An observational and analytical study was conducted using a retrospective historical cohort, followed prospectively. The inclusion criteria consisted of patients operated of hemangioblastoma in two reference hospitals, between 2000 and 2023. A total of 39 patients were included. The Kaplan-Meier estimator was used to summarize data, and the log-rank test

MEK inhibitors in NF1-associated high-grade glioma: partial responses and outcomes from a single-institution cohort and literature review.

PURPOSE: NF1-associated high-grade gliomas (HGGs) are rare, biologically heterogeneous tumors with poor outcomes. Although MEK inhibitors (MEKis) are established options for selected NF1-associated low-grade gliomas, their role in NF1-associated HGG remains undefined. We describe clinical and radiographic outcomes with MEKis, including mirdametinib. METHODS: We retrospectively reviewed adult patients with NF1-associated HGG treated at The University of Texas MD Anderson Cancer Center who initiated selumetinib, mirdametinib, or dabrafenib plus trametinib from December 1, 2019, to January 31, 2

High-Resolution Ultrasound in the Evaluation of Median and Superficial Radial Nerve Involvement in Neurofibromatosis Type 1.

This presentation highlights the value of high-resolution ultrasound for simultaneously characterizing superficial neurofibromas and detecting associated peripheral nerve involvement in young patients with neurofibromatosis type 1. In a 16-year-old female with hand pain and paresthesia, ultrasound demonstrated a hypervascular superficial neurofibroma together with marked epineural thickening of the median nerve, swelling of its recurrent motor branch, and edema of the superficial radial nerve. These findings provided an anatomic correlate for the patient's symptoms despite normal median and ul

[Newborn with neurofibromatosis type 1 and meningoencephalocele].

In this case report, a newborn with neurofibromatosis type 1 (NF1) and meningoencephalocele is presented. The foetus had inherited the same pathogenic defect in the NF1 gene as the father and was therefore closely monitored prenatally. The infant was born with a large frontonasal meningoencephalocele. This has not been described before and medical experience and knowledge is therefore sparse. In this case, early diagnostic imaging and scheduling of surgical intervention in collaboration with neuro- and plastic surgeons was performed.

Managing Plexiform Neurofibroma and Other Complications of Neurofibromatosis Type 1 in the Adult Neurology Clinic: A Review.

PURPOSE OF REVIEW: Loss-of-function variants in the NF1 gene result in the development of neurofibromatosis type 1 (NF1), a neurogenetic disease and multisystem disorder that includes a predilection for benign and malignant tumors of the central and peripheral nervous systems. Patients with NF1 experience a wide variety of comorbidities, such as chronic pain conditions, learning disabilities, and signs and symptoms that include migraines and NF1-associated plexiform neurofibromas (PNs), which can cause disfigurement and pain. All of these factors can severely affect health-related quality of l

Epilepsy in paediatric neurofibromatosis type 1: clinical features and associated factors in a single-centere cohort.

UNLABELLED: Neurofibromatosis type 1 (NF1) is associated with an increased risk of epilepsy, but the clinical spectrum and associated factors of epilepsy in paediatric NF1 remain incompletely characterized. We aimed to describe frequency and features of epilepsy in paediatric NF1 and to compare the characteristics of NF1 patients with and without epilepsy. We retrospectively reviewed 253 paediatric NF1 patients followed up between January 2020 and December 2025. Demographic, clinical, electroencephalographic, and radiological data were collected. Categorical comparisons between patients with a

Everyday executive functions in children with neurofibromatosis type 1: Data from nine institutions.

OBJECTIVE: Executive function (EF) difficulties are prevalent in neurogenetic syndromes such as Neurofibromatosis Type 1 (NF1), the most common autosomal dominant condition associated with nervous system tumors. However, age-related differences in EF problems in children with NF1 remain poorly understood. This study aimed to examine how EF problems in daily life vary across ages throughout childhood and adolescence and how these age patterns vary across demographic and NF1 disease-related characteristics. METHOD: This study integrated data of 1,294 observations from 1,025 children with NF1 (M

Clinical and Genetic Characterization of Russian Patients with von Hippel-Lindau Syndrome.

Von Hippel-Lindau syndrome (VHLS) is an autosomal dominant hereditary cancer disease caused by germline mutations in the VHL tumor suppressor gene and characterized by clinical heterogeneity and phenotype-genotype associations. We studied the VHL gene in 535 Russian patients referred for VHLS diagnostics using Sanger sequencing, multiplex ligase-dependent probe amplification, and, in some cases, high-throughput sequencing. A total of 177 causative VHL variants were identified, including 129 point mutations and 48 extended deletions. Missense variants were the most frequent (60.3%) and predomin

Prenatal Diagnosis of Neurofibromatosis Type 1: No Specific Prenatal Features Observed in Sporadic Fetal Presentations.

OBJECTIVE: To present the indications for prenatal diagnosis (PND) of neurofibromatosis type 1 (NF1) in a Chinese referral medical center. METHOD: This study encompasses a 7-year experience in PND of NF1. The cohort consisted of women undergoing PND due to parental carrier status, as well as those whose fetuses were diagnosed with copy number variations (CNVs) involving NF1 gene or NF1 variants based on indications unrelated to parental carrier status. RESULTS: A total of 30 cases underwent PND related to NF1. Among these, 17 cases involved a parental carrier, with pathogenic NF1 variants id

Melanoma in an Unlikely Guise: Diagnostic Challenges of a Solid-cystic Popliteal Mass in Neurofibromatosis Type 1.

Malignant melanoma (MM) uncommonly presents as a solid-cystic soft tissue mass, posing a significant diagnostic challenge. We report a rare case in a 63-year-old woman with neurofibromatosis type 1 (NF-1) who presented with a painful, rapidly progressive swelling on the posterior aspect of her right lower thigh. Imaging revealed a large, well-defined solid-cystic lesion suggestive of a soft tissue sarcoma or malignant peripheral nerve sheath tumor. Initial fine-needle aspiration cytology was nondiagnostic. Definitive diagnosis was achieved following wide surgical excision, with histopathologic

Respiratory function in children and young adults affected by neurofibromatosis type 1 treated with selumetinib.

BACKGROUND: Plexiform neurofibromas (PNs) are benign tumors that can cause pain, disfigurement, and disability in patients with neurofibromatosis type 1. Selumetinib, a MEK1/2 inhibitor, is the first drug who showed efficacy in inducing PNs shrinkage, reducing the associated morbidities in pediatric patients. Overall, selumetinib showed a good safety profile. However, multiple adverse effects have been reported both in pediatric and adult patients, including respiratory ones. Serial spirometry is suggested in patients with thoracic neurofibromas to detect changes that may underly PN's modifica

Efficacy and safety of MEK inhibitors for NF1-associated symptomatic, inoperable plexiform neurofibromas: A systematic review and meta-analysis.

Plexiform neurofibromas arise in up to half of people with neurofibromatosis type 1 (NF1), causing pain, disfigurement and functional loss. Loss of neurofibromin releases RAS-MAPK signaling, making MEK1/2 a rational target. Two agents are now approved in the United States, the adult indication only since 2025. We evaluated the efficacy and safety of MEK inhibitor monotherapy in NF1-associated symptomatic, inoperable plexiform neurofibroma across children and adults. We searched six databases and registries from inception to 10 July 2026, reporting per PRISMA 2020. The primary outcome was objec

Ataxia-Telangiectasia, DNA Repair Deficits, Immune Dysfunction, and Cancer: A Review of Mechanisms, Clinical Presentations, and Emerging Therapies.

Ataxia-Telangiectasia (A-T) is a rare, autosomal recessive disorder caused by biallelic pathogenic variants in the ATM gene. Loss of ataxia-telangiectasia mutated (ATM) protein function impairs DNA damage responses, leading to progressive neurodegeneration, genomic instability, immunodeficiency, and an increased risk of malignancy. Some A-T patients develop cancers at a young age and display extreme sensitivity to ionizing radiation and certain chemotherapeutics. Therefore, standard treatments often cause severe, life-threatening toxicities in these patients. This review discusses the dual rol

Optic glioma in children: clinical presentation, diagnosis, and management.

INTRODUCTION: Optic gliomas are slow-growing astrocytic tumors that frequently occur in children. The general treatment approach for optic gliomas should be based on tumor location, patient age, and NF1 status. Optic gliomas respond well to chemotherapy; it is generally preferred over radiotherapy, especially for children under five years and those with NF1. Chemotherapy primarily results in stabilization of tumor volume, and this effect is often temporary. In some cases, a second line treatment may be required. Radiotherapy may be an option for treating patients with sporadic optic gliomas. T

Bibliometric mapping of the 100 most-cited publications on optic nerve and optic pathway glioma: A cross-sectional bibliometric analysis.

BACKGROUND: Optic pathway gliomas (OPGs), including optic nerve gliomas, are uncommon tumors of the anterior visual pathway that arise mainly in young children, often with neurofibromatosis type 1 (NF1). The literature is now substantial, but its structure is undescribed. We mapped the most-cited OPG publications to characterize citation patterns, contributors, collaboration, and thematic development. METHODS: Scopus was searched on April 30, 2025 using a title-abstract-keyword strategy combining 4 OPG-specific phrases, with no year or document-type restriction. Records were ranked by total c

Identification of CD74-Positive Antigen-Presenting Glioma Cells in Primary Human Tumors and Murine Models of NF1 High-Grade Glioma.

Compared with the general population, individuals with neurofibromatosis type 1 have a 50-fold higher risk of developing a high-grade glioma (HGG) in their lifetime. Despite improved understanding of the molecular and cellular drivers of these neoplasms, we have yet to translate this knowledge into therapies that improve overall survival. One limitation has been the paucity of in vivo models for drug testing within this population. We generated three distinct glioma stem cell lines from HGGs arising in mice with Nf1 and Trp53 mutations in cis (NPcis) and characterized the allografts resulting
