Cloning of human DING: Developmental expression and downregulation by EtOH in-utero.

INTRODUCTION: An estimated 15-20% of women consume alcohol (EtOH) during pregnancy. Women with alcohol use in early pregnancy are likely to have a child with fetal alcohol spectrum disorders (FASD). Recently, we reported neuroprotective effects of human DING (a member of the DING family of phosphatases) against EtOH-mediated toxicity in rats and in human fetal cortical neurons in vitro. Now, we report the sequencing and developmental expression patterns of endogenous DING in human fetal brain. METHODS: DING cDNA was cloned from human U87MG astrocytoma cells with primers specific to the plant

S-adenosyl-L-methionine reverses ethanol-induced developmental toxicity in FASD model of Danio rerio embryos via dual-modulation of oxidative stress and glutathione homeostasis.

Fetal Alcohol Spectrum Disorder (FASD) represents a major global public health concern, affecting approximately 7.7 per 1000 births worldwide and remains as the most common preventable cause of lifelong neurodevelopmental impairment. Despite its prevalence, current clinical interventions are largely symptom-supportive and fail to address the underlying developmental pathology, underscoring the need for targeted, mechanism-based therapeutic strategies. Given the central involvement of oxidative stress and inflammation in FASD pathogenesis, this study evaluated the protective efficacy of S-adeno

Paternal lineage effects of prenatal ethanol exposure: Offspring neuroanatomy and behavior in a multigenerational mouse model of FASD.

BACKGROUND: Fetal alcohol spectrum disorders (FASD) encompass a range of phenotypes in offspring exposed to ethanol via maternal consumption during pregnancy. In a series of studies, our laboratory identified deleterious effects of prenatal ethanol exposure (PrEE) in our FASD mouse model. The first filial generation (F1) of PrEE offspring exhibited abnormal neocortical gene expression, ectopic intraneocortical connectivity, altered neuroanatomy, and disrupted behavior. Our results suggest that PrEE can induce transgenerational transmission of the abnormal phenotypes, potentially via epigenetic

Developmental alcohol exposure: Impacts on medial prefrontal cortex morphology and the excitatory-inhibitory balance.

Alcohol is one of the most commonly used substances during pregnancy and adolescence, two of the most vulnerable periods of cognitive development. The prefrontal cortex (PFC) is a cortical region of the frontal lobe involved in various complex executive functions that are affected by alcohol exposure. Specifically, the medial prefrontal cortex (mPFC), a subregion of the PFC, performs critical roles in cognitive processing, such as conditioned fear, reward-seeking, impulse control, working memory, and pain. The mPFC begins developing in utero and continues to mature into adulthood. Thus, it is

Trends in Well-Being and Opportunities to Support Caregivers of Individuals With Fetal Alcohol Spectrum Disorder Across the Lifespan.

BACKGROUND: Caregivers of individuals with fetal alcohol spectrum disorder (FASD) experience unique challenges, stressors, and strengths, and there is growing understanding of the needs of these caregivers. However, less is known about how the broader caregiver context, including sociodemographic and ecological factors, may be associated with well-being. METHODS: Data for this study were collected between 2021 and 2025, gathered at baseline as part of an ongoing international longitudinal survey about caregiver experiences and perspectives of raising people with FASD (N = 234). Information on

Differentiating Fetal Alcohol Spectrum Disorders From Attention-Deficit Hyperactivity Disorder and Reviewing Diagnostic Guidelines.

Individuals with prenatal alcohol exposure (PAE) and fetal alcohol spectrum disorders (FASDs) present a range of neurodevelopmental deficits (e.g., inattention, hyperactivity, and executive dysfunction) which have shown marked overlap with attention-deficit/hyperactivity disorder (ADHD), making differential diagnosis challenging. While rates of comorbidity are high, evidence has suggested there are important distinctions between neurodevelopmental phenotypes. To understand these distinctions, we evaluated whether proposed neurobehavioral disorder associated with prenatal alcohol exposure (ND-P

Executive Functioning and Conduct Problems in Youth with Prenatal Alcohol Exposure: A Scoping Review.

Executive functioning impairment is a core neurobehavioral feature of Fetal Alcohol Spectrum Disorders (FASD) across all diagnostic frameworks and can result from prenatal alcohol exposure (PAE). Executive functioning (EF) is also associated with conduct problems, although the precise relationship between the variables in youth with PAE remains unclear. The primary aim of this scoping review is to clarify the relationship between executive functioning and conduct problems in youth with PAE based on existing literature. The secondary aim of the scoping review is to identify gaps in existing lit

Alcohol Consumption During Pregnancy and Conduct Problems: A Brief Overview of the Literature.

Alcohol is widely consumed globally, with significant risks linked to both mental and physical health conditions. Alcohol exposure during pregnancy is prevalent, with nearly 10% of pregnant women worldwide consuming alcohol. There is no safe limit for alcohol intake during pregnancy, as even minimal exposure can cause severe developmental issues. This narrative review examines the impact of alcohol consumption during pregnancy on the development of specific conduct problems in the offspring, namely Oppositional Defiant Disorder (ODD) and Conduct Disorder (CD). A comprehensive review of existin

Decoding neurodevelopment risks of exposures to intoxication, infection, and trauma during pregnancy.

The maternal environment plays a pivotal role in fetal neurodevelopment, but adverse conditions during pregnancy may disrupt key developmental processes. Conditions such as intoxication, infection, or trauma during pregnancy have been associated with impaired fetal oxygenation, altered immune signaling, and disrupted placental function, which may contribute to long-term neurodevelopmental alterations. This review examines evidence suggesting how prenatal exposures to alcohol, cocaine, Zika virus, rubella virus, Toxoplasma gondii, influenza, and traumatic brain injury can impact fetal brain dev

Warfarin-Induced Developmental Toxicity: Insights into Embryogenesis, Teratogenicity, and Molecular Pathways.

Warfarin is a coumarin-derived oral anticoagulant widely used for the prevention and treatment of thromboembolic disorders, particularly in patients with mechanical heart valves. The drug exerts its anticoagulant effect by inhibiting vitamin K epoxide reductase, thereby impairing γ-carboxylation of vitamin K-dependent coagulation factors. Despite its clinical efficacy, warfarin therapy is associated with a narrow therapeutic index, substantial interindividual variability in dose response, numerous drug interactions, and significant hemorrhagic risk. Maternal warfarin therapy during pregnancy i

Stunting, underweight and thinness in internationally adopted children: prevalence and associated factors in a large cohort study.

Internationally adopted children (IAC) are at high risk of growth impairment due to early-life deprivation, institutional care, and prenatal exposures. Data on determinants of stunting and other forms of malnutrition in this population remain limited. To evaluate the prevalence of stunting, underweight, and thinness, and to identify factors associated with these conditions in a large cohort of internationally adopted children (IAC), we conducted a large cohort study including 1,955 IAC referred to a tertiary pediatric center in Italy between 2015 and 2025. The cohort mainly included children a

Developmental Trajectory of Auditory Brainstem Deficits in a Mouse Model of Fetal Alcohol Spectrum Disorders.

Individuals with fetal alcohol spectrum disorders exhibit impaired auditory processing and atypical auditory behaviors across the disorder's spectrum. Although low to moderate prenatal alcohol exposure (PAE) is a prevalent type of exposure, the developmental trajectory of auditory processing deficits resulting from this level of exposure remains unknown. To examine the effects of PAE in the auditory system, we utilized the maternal voluntary alcohol consumption paradigm to model chronic low to moderate PAE throughout gestation. We measured auditory brainstem responses (ABRs) in juvenile mice (

Beyond Procedural Universalism: Relational Ethics and the Limits of Global Public Health Codes.

BACKGROUND: Alcohol consumption during pregnancy is a significant public health problem and can result in a continuum of adverse outcomes to the fetus known as fetal alcohol spectrum disorders (FASD). Subjects with FASD show significant neurological deficits, ranging from microencephaly, neurobehavioral, and mental health problems to poor social adjustment and stress tolerance. Neurons are particularly sensitive to alcohol exposure. The neurotoxic action of alcohol, i.e., through ROS production, induces DNA damage and neuronal cell death by apoptosis. In addition, epigenetics, including DNA me

[Interest as Catalyzer: The Role of Interest in the Use of Incremental Scaffolds in Inquiry-Based Learning].

BACKGROUND: Use of human mesenchymal stromal cells (MSCs) is a promising strategy for cell therapy in injured tissues recovery. However, MSCs acquire genetic changes when cultivated in vitro that make them more susceptible to undergo neoplastic transformation. Therefore, genomic integrity of stem cells should be monitored carefully for the use in basic research and clinical trials, including karyotype analysis to confirm the absence of genetic instability. Here, we report a case of a male 67-year-old patient selected to join the study: "Autologous transplantation of mesenchymal cells for treat
