Erythropoietin improves spatial and nonspatial memory defects by suppressing oxidative damage, inflammation and apoptosis against ethanol neurotoxicity in the developing male rat hippocampus.

Excessive prenatal exposure to ethanol leads to a condition called fetal alcohol spectrum disorder (FASD). The neurotoxicity of alcohol causes changes in the hippocampus of animals during this time, resulting in impaired hippocampus-related functions, including memory/learning and cognition. The liver and kidneys produce erythropoietin (EPO). The synthesis of EPO by immature neurons also plays a decisive role in the embryonic stage. Also, exogenous EPO exerts its neurocognitive effects in the developing brain under pathophysiological conditions. The aim of this study was to investigate the prot

An updated systematic review of the literature on fetal alcohol spectrum disorder and the criminal legal system.

People with fetal alcohol spectrum disorder (FASD) can experience a range of individual, social, and systemic challenges that may increase the likelihood of life adversity, including contact with the criminal legal system (CLS). The purpose of this article was to update a 2018 systematic review of literature on this intersection of FASD and the CLS. We searched ten academic databases for studies with people with FASD involved in the CLS, as well as caregivers and service providers who support them. A total of 54 studies were identified, published between April 2017 and March 2024, which is mor

Fetal Alcohol Spectrum Disorders.

Prenatal alcohol exposure (PAE) can cause a wide range of physical, neurobehavioral, and neurocognitive impairments that impact developmental trajectories throughout the lifespan. Clinically, individuals who have been exposed to alcohol prenatally and who show physical or neurobehavioral difficulties may be classified as having a condition included in fetal alcohol spectrum disorders (FASDs). The aim of this chapter is to summarize the current knowledge of FASD including diagnostic criteria, neurobehavioral outcomes, lifespan considerations, and interventions. Individuals with PAE exhibit chal

The chick embryo model as an educational tool to explore the effect of alcohol on cardiovascular development.

In development, the interrelationship between physiology and anatomy is challenging as learners must appreciate how physiological processes and anatomical structures change over time. In addition, the dynamic relationships between structure and function are often concealed largely due to the inaccessibility of the embryo in higher-order organisms. This makes it difficult for students to appreciate normal intricate balances or interpret the physiological consequences of developmental disruptions to normal embryological development. In this paper, the applicability of the chick embryo model for

Prenatal Alcohol Exposure Disrupts CXCL16 Expression in Rat Hippocampus: Temporal and Sex Differences.

Prenatal alcohol exposure (PAE) affects around 40,000 newborns every year and poses a significant health risk. Although much is already known about the neurotoxic mechanisms of PAE, new findings continue to emerge. Studies with mouse models show that PAE leads to overexpression of proinflammatory cytokines and chemokines in the brain, which disrupts important neurodevelopmental processes such as cell migration, survival and proliferation of neurons. The chemokine CXCL16 is overexpressed in the brain following various impairments, including PAE. This study shows that CXCL16 expression varies by

Impact of Prenatal Dietary Soy on Cerebellar Neurodevelopment and Function in Experimental Fetal Alcohol Spectrum Disorder.

Background: Prenatal alcohol exposure (PAE) models can cause neurodevelopmental abnormalities like those observed in fetal alcohol spectrum disorder (FASD). Previous studies link experimental PAE effects in the brain to impaired signaling through insulin/IGF and Notch pathways that mediate neuronal survival, growth, migration, energy metabolism, and plasticity. Importantly, concurrent administration of peroxisome proliferator-activated receptor agonists or dietary soy prevented many aspects of FASD due to their insulin-sensitizing, anti-inflammatory, and antioxidant properties. Objective: To d

Prenatal Alcohol Exposure Inhibits Transient Expression of Autophagy and Synaptic Proteins in Developing Brain.

INTRODUCTION: Neuronal apoptosis and consequent inhibition of autophagy, with loss of synaptic connections are central events in the genesis of fetal alcohol spectrum disorders (FASD). However, studies of molecular mechanisms of autophagy in human fetal brain are limited. Recently, prenatal exposure to EtOH was associated with reduced miRNA-9 levels in fetal brain-derived exosomes (FB- Es) isolated from maternal plasma, which correlated with small eyes, an anatomical hallmark of fetal alcohol syndrome (FAS). Since miR-9 targets several genes that regulate synaptic plasticity, EtOH-induced inhi

Prenatal Ethanol Exposure Results in Cell Type, Age, and Sex-Dependent Differences in the Neonatal Striatum That Coincide with Early Motor Deficits.

Delayed motor development is an early clinical sign of fetal alcohol spectrum disorders. However, changes at the neural circuit level that underlie early motor differences are underexplored. The striatum, the principal input nucleus of the basal ganglia, plays an important role in motor learning in adult animals, and the maturation of the striatal circuit has been associated with the development of early motor behaviors. Here, we briefly exposed pregnant C57BL/6 dams to ethanol (5% w/w) in a liquid diet on embryonic days 13.5-16.5 and assessed the mouse progeny using a series of nine brief mot

Benzydamine rescues ethanol-induced teratogenesis in zebrafish FASD model.

Fetal alcohol spectrum disorders (FASD) are a group of physical, behavioral, and cognitive impairments caused by ethanol exposure during pregnancy. Zebrafish have emerged as a useful model for researching FASD and its variants in recent years. Oxidative stress has been identified as the primary damaging pathway, notwithstanding the possibility of other mechanisms at play. In this regard it's important to put an effort towards antioxidants which can exhibits and bare a potential to counteract the oxidative stress induced by ethanol during embryos development. Previosuly benzydamine has shown to

Use of the alternative test R-FETAX (Refined-Frog Embryo Teratogenicity Assay-Xenopus) to evaluate the Fetal Alcohol Spectrum Disorders (FASD).

Fetal Alcohol Spectrum Disorders (FASD) refer to a range of conditions in children caused by alcohol consumption during pregnancy, including morphological defects, developmental delays, and neurobehavioral impairments. Ethanol (EtOH) at high concentrations (1-3 % v/v) was shown to induce malformations and lethality in Xenopus laevis embryos exposed throughout the FETAX test (from the mid-blastula stage to the final pre-feeding larval stage). The aim of this work was to evaluate multiple morphological and neurobehavioral effects of EtOH exposure (0.1-3 % v/v) using the R-FETAX protocol. Embryos

Analysis of risk factors associated with suicidality in children and adolescents with fetal alcohol spectrum disorder in Western Australia.

BACKGROUND: Individuals with fetal alcohol spectrum disorder (FASD) are at an elevated suicide risk compared with those in the general population. This public health issue warrants further research attention to help inform the development of prevention and intervention efforts. Our study is the first to characterize suicidality (i.e., suicidal ideation/suicide attempt) and explore associated risk factors in young individuals with FASD within the Australian context. METHODS: Retrospective file reviews from a diagnostic clinic in Western Australia obtained data on demographic variables and risk

Neuroprotective Effects of Erythropoietin in Fetal Alcohol Spectrum Disorder: BDNF Upregulation, Reduced Astrogliosis, and Anxiety Relief.

OBJECTIVE: The term fetal alcohol spectrum disorder (FASD) refers to a variety of negative consequences that may develop in children born to women who have consumed alcohol during pregnancy. Anxiety disorders are common in FASD. Animals exposed to alcohol exhibit anxiety-like traits. Erythropoietin (EPO) is produced by the kidneys and liver. The synthesis of EPO by immature neurons also plays a crucial role in the embryonic stage. In addition, EPO inhibits astrogliosis and cell death while increasing the number of brain-derived neurotrophic factor (BDNF)-positive cells, which is linked to redu

Defects of Cortical Microcircuits Following Early Exposure to Alcohol.

The interplay between excitatory pyramidal neurons and GABAergic interneurons is the basic building block of neocortical microcircuits and plays a critical role in carrying out higher cognitive functions. Cortical circuits are deeply and permanently disrupted by exposure to alcohol during brain development, the main non-genetic cause of intellectual disability. Here, I review experimental studies of fetal alcohol spectrum disorders, dealing with permanent cellular and molecular alterations of neocortical neurons and their connections.

Ontogenetic Neuroimmune Changes Following Prenatal Alcohol Exposure: Implications for Neurobehavioral Function.

This chapter reviews the enduring effects of prenatal alcohol exposure (PAE) on neuroimmune function across the lifespan, including discussion of associated neurobehavioral alterations. Alcohol has potent teratogenic effects, with a large body of work linking PAE to perturbations in neuroimmune function. These PAE-related neuroimmune disturbances may have downstream effects on neurobehavioral function given the critical role of the neuroimmune system in central nervous system development. The neuroimmune system matures over time, playing distinct roles depending on the developmental processes
