A register-based study of long-term health and social care costs among children with prenatal alcohol exposure.

Prenatal alcohol exposure (PAE) associated with fetal alcohol spectrum disorders (FASD) often remain underdiagnosed. They globally cause a wide range of health and social problems leading to high costs. To outline cumulative health and social care costs in children related with PAE with and without diagnosed FASD, we followed 427 children with PAE until the age of 20 years, and 1795 controls born 1992-2001 until the year 2016. All hospital care and out-of-home care episodes, including placements in foster or residential care, were analyzed, and their costs estimated. Age-dependent patterns of

Morphometric Analyses of Shape: The Analysis Software Toolbox for Craniofacial Shape Quantification in Zebrafish.

Fetal alcohol spectrum disorders (FASD) are characterized by a varying set of physical, cognitive, and behavioral disabilities caused by prenatal ethanol exposure, including those affecting the facial skeleton. Ethanol-sensitive genetic loci contribute to this high degree of variation in FASD, which complicates analyses of facial shape. We have previously shown that we can analyze these changes in facial shape from gene-ethanol interactions in zebrafish. Zebrafish are an ideal model to analyze this variation for several reasons; (i) 70% of genes are orthologs between humans and zebrafish; (ii)

Endothelial NMDA Receptor Involvement in Retinal Neurovascular Damage Following Prenatal Alcohol Exposure in a Mouse Model.

Prenatal alcohol exposure (PAE) induces neurodevelopmental damage leading to fetal alcohol spectrum disorders (FASD) by altering both brain and ocular development. Recent data showed that PAE impairs brain cortical and retinal vasculature leading to defective positioning of interneurons. In the retina, PAE disturbs vascular development and the association of calretinin neurons with vessels. The NMDA receptor (NMDAR) is a major target of alcohol in the brain, and both ligand binding to NMDARs and the expression of NMDAR subunits are altered in FASD. Given that NMDAR is also expressed in endothe

Families Moving Forward Connect mHealth Intervention for Caregivers of Children With Fetal Alcohol Spectrum Disorders: Randomized Controlled Trial.

BACKGROUND: Fetal alcohol spectrum disorders (FASD) affect 1.1% to 5% of the general population. Yet, most children with FASD and their families cannot access evidence-based interventions. Mobile health (mHealth) interventions have the potential to increase access to care on a broad scale. While numerous self-directed parenting apps exist, none have been tested for FASD. The FMF (Families Moving Forward) Connect app is a self-directed intervention derived from an empirically supported intervention for caregivers raising children with FASD. FMF Connect is the first self-directed parenting app f

Fluoxetine for Prenatal Alcohol-Exposed Mice: Addressing Impaired Enrichment-Mediated Neurogenesis.

BACKGROUND: Fetal alcohol spectrum disorder (FASD) encompasses a variety of disorders that occur after a fetus has been exposed to alcohol. Hippocampal related issues are a common neurological deficit found in FASD. The dentate gyrus within the hippocampus is a unique area of the brain that continues to generate new neurons into adulthood. This neurogenesis can be enhanced by an enriched environment (EE); however in prenatal alcohol exposed (PAE) mice this EE-mediated neurogenesis is impaired. In addition to EE, selective serotonin reuptake inhibitors (SSRIs), such as fluoxetine, also promote

Cardiovascular and Cardiometabolic Outcomes in Adults with Fetal Alcohol Spectrum Disorders: A Retrospective Cohort Study.

BACKGROUND: Fetal alcohol spectrum disorders (FASDs) impact up to 5% of U.S. school age children; however, the burden of cardiovascular disease (CVD) in adults with FASDs remains poorly defined. We investigated associations between FASDs, cardiometabolic abnormalities, and CVD using electronic health record (EHR) data. METHODS: We performed a retrospective cohort study of adults (≥18 years) receiving ambulatory care with a FASD diagnosis (n = 208, mean age 38.4±14.5, 50% female) and age- and sex-matched control patients without FASD (n = 824, mean age 41.6±14.5, 50% female). Disinhibited social engagement and reactive attachment behaviours in children assessed for Fetal Alcohol Spectrum Disorder (FASD): Distinguishing effects of maltreatment from inhibitory control difficulties.

The developmental origins of reactive attachment disorder (RAD) and disinhibited social engagement disorder (DSED) continue to be poorly understood. This cross-sectional study reports on the rates of RAD and DSED behaviours in a clinical sample of children presenting for Fetal Alcohol Spectrum Disorder (FASD) assessment (N = 123). It examines whether RAD and DSED behaviours, assessed by diagnostic interview with carers, are better explained by maltreatment (timing/type) or neurodevelopmental inhibitory-control deficits, assessed using the BRIEF-2, adjusting for age, sex and variables related t

Needs of professionals working with individuals with FASD in Spain.

Prenatal alcohol exposure is one of the main preventable causes of developmental disorders and health anomalies in children. Fetal Alcohol Spectrum Disorder (FASD) is a diagnostic term that encompasses a range of physical, cognitive, and behavioural impairments. Although this condition is gradually attracting greater attention among professionals, knowledge and awareness of its real impact - and of how to intervene effectively - remains limited. This lack of understanding shapes the work of professionals when they must support individuals affected by FASD. The aim of this study is to explore t

A Multiple Case Report of Fetal Alcohol Spectrum Disorders with Focus on Cognitive and Sensory Profiles.

AIMS: Prenatal alcohol exposure (PAE) is associated with Fetal Alcohol Spectrum Disorders (FASD), characterized by persistent cognitive, adaptive, and neurosensory impairments. Alcohol disrupts processes of cerebral development underlying multisensory integration and lateralization, which are essential for cognitive and behavioral functioning. This study used the Brief Multisensory Integration Battery (BMIB) and the Global Lateral Preference Inventory (GLPI) to detect FASD-related neurocognitive and sensory features. METHODS: Three adolescents with documented PAE were assessed with the BMIB a

Mechanical properties of the developing brain in a model of fetal alcohol spectrum disorders and relationships to perineuronal net integrity.

Neuroimaging is a useful tool for examining altered neurodevelopmental trajectories in fetal alcohol spectrum disorders (FASD). FASD affects 1 in 20 infants in the United States with higher prevalence in specific regions across the globe. Advanced neuroimaging methods, such as volumetric morphometry and diffusion-weighted imaging, are critical for determining the effectiveness of interventions that support neurodevelopment in FASD. In this study, we introduce the use of magnetic resonance elastography (MRE), a cutting-edge neuroimaging technique used to measure the mechanical properties of bra

Prenatal alcohol damages the subiculum, a key region for learning and memory: A narrative review.

As an integral part of the hippocampal network, the subiculum (SUB) plays a critical role in learning, memory, and emotional processing. This region contains diverse neuronal populations with distinct projection patterns, each contributing uniquely to information processing. The SUB has also been implicated in several neuropathological conditions, including mood disorders and epilepsy, which are highly prevalent among individuals with fetal alcohol spectrum disorders. The objective of this review is to examine the potential involvement of the SUB, a key component of the hippocampal formation,

A Single Dose of a Psychedelic Drug Repairs Prefrontal Cortex Synaptic Physiology in a Mouse Model of Prenatal Alcohol Exposure.

BACKGROUND: Prenatal alcohol exposure (PAE) can cause fetal alcohol spectrum disorders (FASDs), which are characterized by neural circuit and behavioral dysfunction due to impaired brain development. At the neural and behavioral levels, PAE is associated with disrupted cortical synaptic transmission and lifelong impairments in learning and cognitive control. Despite the prevalence of FASDs (affecting up to one in 20 school-aged children in the United States) and the associated personal, familial, and societal costs, there are currently no treatments to reverse neural circuit dysfunction. Seizures in fetal alcohol spectrum disorder (FASD): evaluation of clinical, electroencephalographic, and neuroradiologic features in a large sample of Norwegian children.

AIM: Fetal Alcohol Spectrum Disorder (FASD) is an umbrella term encompassing developmental, cognitive, and behavioural impairments resulting from prenatal alcohol exposure. An increased prevalence of epilepsy has been reported in individuals with FASD, yet clinical characteristics, neuroimaging findings, and electroencephalography (EEG) profiles remain insufficiently described. This study aimed to determine the frequency of EEG pathology and epilepsy in a large Norwegian FASD cohort and to explore whether the identified epilepsies may have an etiology unrelated to FASD. METHOD: We conducted a

Molecular Networks of Redox Dysregulation in Fetal Alcohol Spectrum Disorders: Mechanisms and Therapeutic Prospects.

Fetal alcohol spectrum disorders (FASD) encompass a continuum of developmental abnormalities caused by prenatal alcohol exposure, resulting in persistent neurodevelopmental and structural defects. Accumulating evidence indicates that redox dysregulation plays a central role in the pathogenesis of FASD. Ethanol disrupts cellular redox homeostasis by promoting excessive reactive oxygen species production and depleting endogenous antioxidants, thereby perturbing key redox-sensitive molecular networks. Dysregulation of these pathways leads to mitochondrial dysfunction, endoplasmic reticulum stress

Third Trimester-Equivalent Alcohol Exposure Induces Sex-Dependent Alterations in Locomotor Activity, Anxiety-Risky Behaviors, and Enhances Mechanical Allodynia in Adulthood.

Prenatal alcohol exposure (PAE) causes fetal alcohol spectrum disorders (FASDs), which are neurodevelopmental conditions characterized by behavioral dysregulation, learning deficits, and cognitive inflexibilities. Alcohol exposure is harmful at all stages of human gestation, including the third trimester. This developmental window, characterized by rapid brain growth, myelination, and neural circuit formation, may be particularly vulnerable, yet the long-lasting behavioral and sensory consequences of exposure during this period remain poorly understood. In this study, neonatal mouse pups were

Linking prenatal alcohol-induced transcriptomic alterations to hippocampal-associated structural and behavioral deficits.

Prenatal alcohol exposure can have a lasting and devastating impact on the hippocampus. Structural deficits including volumetric loss, shape changes, and microstructural abnormalities are observed in clinical patients with Fetal Alcohol Spectrum Disorders (FASD) and in preclinical models. In addition, in utero alcohol exposure is associated with cognitive and behavioral impairments related to hippocampal dysfunction, including spatial and contextual memory deficits. This review discusses the cellular, genetic, and epigenetic changes caused by alcohol during early hippocampal development that c
