Executive functioning-related behavior and neuronal activation in the thalamic nucleus reuniens in a combined intubation and alcohol third-trimester binge exposure in rats.

Fetal Alcohol Spectrum Disorders (FASD) result from prenatal alcohol exposure (AE) and are linked to executive functioning (EF) deficits. The nucleus reuniens (Re) of the thalamus facilitates hippocampal-prefrontal synchrony, essential for EF. While AE alters Re structure and function, its role in EF impairments remains unclear. This study examined the effects of third-trimester equivalent AE on Re volume, neuron count and their activation following a Re-dependent object-in-place (OIP) task in adulthood. Using immunohistochemistry for NeuN (neuronal marker) and ΔFosB (sustained activation mark

Lived Experiences of Adoptive Parents Raising Children with Fetal Alcohol Spectrum Disorders in Israel: A Qualitative Study.

Background: Fetal alcohol spectrum disorders (FASDs) are lifelong neurodevelopmental conditions resulting from prenatal alcohol exposure. Despite high FASD rates in high-risk adoptive populations, little is known about the experiences of adoptive parents in Israel, where underdiagnosis and limited professional awareness persist. Drawing on ecological and family systems theory, this study explored the daily challenges, coping strategies, and service-related barriers encountered by adoptive parents raising children with FASD. Methods: Using a qualitative, exploratory design, semi-structured inte

From teratogens to trajectories: Integrating teratology and Developmental Origins of Health and Disease (DOHaD) frameworks for a new take on fetal alcohol spectrum disorders (FASD).

Research on the effects of prenatal alcohol exposure (PAE) developed within two research frameworks that progressed largely in parallel. Classical teratology was the dominant framework for many years, focusing on prenatal exposure to toxic agents that disrupted developmental processes, resulting in birth defects and neurobehavioral deficits. By contrast, recent studies have positioned PAE within the Developmental Origins of Health and Disease (DOHaD) framework, which suggests that low birth weight and other early life exposures reprogram developmental trajectories, increasing susceptibility to

Age- and sex- dependent effects of moderate gestational day 12 prenatal alcohol exposure on anxiety-like behaviors, ethanol intake, and mechanical sensitivity.

RATIONALE: Prenatal alcohol exposure (PAE) can result in Fetal Alcohol Spectrum Disorder (FASD), which consists of a group of diagnosable medical conditions that can include an increased risk for anxiety disorders and/or alcohol misuse, and sensory issues, such as increased mechanical sensitivity. OBJECTIVE: This study investigated how a single moderate PAE on gestational day 12 (G12) alters anxiety-like behavior, ethanol (EtOH) intake, and mechanical sensitivity across the lifespan of Sprague Dawley rats. METHODS: Pregnant dams were exposed to vaporized EtOH or room air (control) for 6 hour

Third Trimester-Equivalent Alcohol Exposure Reduces Neurons in Males and Increases Vglut2⁺ Neurons with Reduced Intrinsic Bursting in Females in The Murine Dorsal Subiculum.

UNLABELLED: Individuals with Fetal Alcohol Spectrum Disorders (FASDs) show reduced subicular volume, and preclinical studies compliment this by demonstrating that third-trimester-equivalent ethanol exposure induced apoptosis in corticolimbic regions, including the subiculum. The subiculum mediates hippocampal-cortical communication critical for long-term memory consolidation. Within the distal dorsal subiculum, a population of bursting neurons uniquely express VGLUT2 and they play a key role in memory processing. We hypothesized that third-trimester-equivalent ethanol exposure would reduce neu

Anterior middle cingulate cortex gamma-aminobutyric acid level is elevated in children with both familial and prenatal alcohol exposure-associated attention deficit hyperactivity disorder.

BACKGROUND: Prior neuroimaging suggests brain differences between children with attention deficit hyperactivity disorder due to prenatal alcohol exposure (ADHD+PAE) and non-exposed children with ADHD due to other, e.g., familial, causes (ADHD-PAE). There has been interest in regional brain levels of gamma-aminobutyric acid (GABA) and glutamate (Glu) measured in vivo with magnetic resonance spectroscopy (MRS) as possible indicators of local inhibitory, respectively, excitatory activity in ADHD. For the first time, we report here a comparison of GABA and Glu in ADHD+PAE vs. ADHD-PAE. METHODS: A

Effects of ethanol exposure in neonatal mice on retinoic acid signaling in forebrain neurons and astrocytes.

Toxicity of prenatal ethanol leading to fetal alcohol spectrum disorders (FASDs) has been linked to disturbances in retinoic acid (RA) signaling necessary for embryonic development. While ethanol exposure in the postnatal day 7 (P7) mice, which induces immediate neurodegeneration and long-lasting GABAergic cell loss and behavioral deficits, has been used for the third trimester FASD model, involvement of RA signaling in the process has not been well explored. Using RARE-LacZ reporter mice that express β-galactosidase (β-Gal) under the control of retinoic acid response element (RARE), we examin

Alcohol Consumption During Pregnancy Among Women Aged 18-49 Years - United States, 2021-2024.

Alcohol consumption during pregnancy can cause adverse birth outcomes and fetal alcohol spectrum disorders. One U.S. study identified a slight increasing trend in alcohol consumption during pregnancy from 2011 to 2018. During 2018-2020, 13.5% of pregnant women reported current drinking; more recent estimates are unavailable. CDC analyzed 2021-2024 Behavioral Risk Factor Surveillance System data to estimate prevalence of self-reported current drinking (one or more alcoholic drinks during the past 30 days), binge drinking (four or more alcoholic drinks on at least one occasion during the past 30

Epileptiform Discharges Drive Unique High-frequency Oscillations Within the Retrosplenial Cortex of Mice with Third Trimester Alcohol Exposure.

Fetal Alcohol Spectrum Disorders (FASDs) are associated with alterations in learning and memory that persist throughout the lifespan. Thus, determining the neural mechanisms driving these alterations has the potential to identify novel therapeutic targets for improving memory in those with FASD. Given the newly realized role of the Retrosplenial cortex (RSC) for learning and memory, as well as the profound neural apoptosis that exposure to alcohol during development causes to this brain region, we recorded electrophysiological activity from mice exposed to alcohol during the third trimester-eq

Hesperidin mitigates cognitive and anxiety-like deficits by enhancing hippocampal antioxidant defenses, reducing neuroinflammation, and preventing neuronal apoptosis in a third-trimester-equivalent rat model of developmental ethanol neurotoxicity.

Ethanol exposure during brain development has been associated with hippocampal oxidative stress, neuroinflammation, and apoptosis, resulting in lasting cognitive and emotional deficits characteristic of fetal alcohol spectrum disorders (FASD). This study evaluated the neuroprotective potential of Hesperidin, a citrus flavanone with antioxidant and anti-inflammatory actions, against developmental ethanol neurotoxicity. Neonatal Wistar rats received ethanol (5.25 g/kg/day; 11.9% v/v) from postnatal days 2-10 and were subsequently treated intraperitoneally with Hesperidin (25, 50, or 100 mg/kg).

Multimodal optical imaging for the assessment of the teratogenic effects of ethanol on zebrafish development.

SIGNIFICANCE: Prenatal alcohol exposure is a major cause of neurodevelopmental and growth impairments collectively termed fetal alcohol spectrum disorders (FASD). However, the mechanisms by which ethanol disrupts early embryonic development remain incompletely understood; for example, the potential link between ethanol treatment and biomechanical properties of tissues that lead to defects in development is not understood. AIM: To address this gap in knowledge, we used zebrafish (Danio rerio), a powerful vertebrate model with optical transparency and rapid development, to investigate ethanol-i

Neurodiversity and Intellectual Disability: Opportunities and Challenges for Functioning and Participation Across the Life Course.

There is increasing recognition that intellectual developmental disabilities are best understood through strengths, everyday functioning, and participation rather than through deficits or predicted limitations. The "F-Words" framework (ie, Function, Family, Fitness, Fun, Friends, Future) translates the World Health Organization's International Classification of Functioning, Disability and Health into language that is meaningful for clinicians and families. Using three common pediatric scenarios (eg, Down syndrome, fetal alcohol spectrum disorder, prematurity), this review shows how early couns

A Label-Free Ultraviolet Photoacoustic Microscopy Enables Nuclear Imaging and Toxicity Assessment in an Intact Brain Organoid.

Fetal alcohol spectrum disorders (FASDs) are caused by prenatal exposure to ethanol (EtOH), leading to developmental brain abnormalities. Cortical organoids derived from human-induced pluripotent stem cells provide physiologically relevant models to study such neurotoxic effects. However, accurate imaging of nuclear morphology, a key indicator of cytotoxicity and developmental impairment, remains difficult. Traditional fluorescence-based techniques rely on staining and sectioning, limiting throughput and potentially altering native structures. Herein, we present an ultraviolet photoacoustic mi

Myelin Alteration in Neurodevelopmental Disorders: Integrating Clinical and Preclinical Evidence.

Myelination is a fundamental neurobiological process that enables rapid and efficient neural signal transmission, playing a key role in shaping brain connectivity and function-particularly during early development. Given the high energetic cost and tight developmental timing of myelination, disruptions in myelin formation or maintenance can increase vulnerability to genetic and environmental factors during critical periods. As such, impaired myelination and oligodendrocyte dysfunction may contribute to the pathogenesis of various neurodevelopmental disorders (NDDs). This review synthesizes cur

Maternal alcohol exposure and atopic dermatitis in offspring: A scoping review protocol.

Maternal alcohol use is a modifiable, yet significant public health concern, causing fetal alcohol syndrome and neurodevelopmental disorders. Research has predominantly focused on the neurodevelopmental consequences of maternal alcohol consumption, but growing evidence suggests that these effects may extend to the developing immune system, potentially increasing the risk of allergic and inflammatory conditions such as atopic dermatitis in offspring, which remains underexplored. This scoping review aims to map the available literature on the association between maternal alcohol exposure (precon

Linking Reductions in Alcohol and Birth Control Use Risk Behavior to Prevention of Alcohol-Exposed Pregnancy: A Population-Level Simulation of Preconceptual Prevention Programs.

BACKGROUND: The most popular alcohol-exposed pregnancy (AEP) prevention program (AEP-P) is a brief intervention that dually promotes reducing alcohol use and increasing birth control use. The primary outcome variable used to evaluate AEP-P involves aggregating behavioral change into a binary variable, despite differences in how AEP-P impacts each behavior. This study examined if all three possible AEP-P behavior-change outcomes (alcohol only (AO), birth control only (BCO), or both behaviors [BB]) are equally effective in preventing subsequent AEP itself. METHODS: We simulated implementing an

Developmental Neurotoxicity of Alcohol from Neuronal Basis to Behavioural Outcomes: A Comprehensive Review.

Prenatal alcohol exposure (PAE) is recognized as a major public health concern due to its profound and lasting effects on the central nervous system (CNS) and its ability to induce fetal alcohol spectrum disorders (FASD), which encompass a wide range of cognitive, behavioural, and neuropsychiatric disorders that persist throughout life. Experimental and clinical studies have identified several mechanisms underlying ethanol impairing brain development, including apoptosis, oxidative stress, disruption of morphogen and growth factor signalling pathways, impaired neuronal proliferation and migrat

Alcohol Metabolism into Acetaldehyde in Developing Cerebral Arteries.

Alcohol exposure during pregnancy leads to fetal alcohol spectrum disorders (FASD), yet the mechanisms through which alcohol disrupts the developing cerebrovasculature remain poorly defined. Acetaldehyde, the first oxidative metabolite of alcohol, can alter vascular function, but whether developing cerebral arteries possess intrinsic capacity to generate acetaldehyde is unknown. Alcohol is primarily oxidized by alcohol dehydrogenase (ADH), cytochrome P450 2E1 (CYP2E1), and catalase (CAT), and their local metabolic activity may contribute to cerebrovascular vulnerability. In this study, cerebra
