Transverse Dental Arch Dimensions and Body Mass Index in Pediatric Patients with Marfan Syndrome: A Case-Control Observational Study.

Objectives: To describe BMI-for-age and transverse dental arch measurements in growing children with Marfan syndrome (MFS) and a non-Marfan pediatric comparison group. Materials and Methods: A retrospective case-control observational study included 41 children aged 6-11 years: 15 patients with MFS (7 males, 8 females; mean age 8.8 ± 1.5 years) and 26 non-Marfan controls (10 males, 16 females; mean age 8.9 ± 1.6 years). Anthropometric parameters and BMI-for-age z-scores were assessed using the WHO 2007 Growth Reference. Digital dental models were used to measure intermolar width (IMW), intercan

Vascular and Ventricular Responses Assessed by Exercise Stress Echocardiography in Pediatric Marfan Syndrome.

BACKGROUND: Pediatric Marfan syndrome (MFS) is associated with changes in aortic stiffness, endothelial function, and ventricular performance. Resting measurements may not fully capture cardiovascular functional reserve. Exercise stress echocardiography can reveal subclinical impairments not evident at rest. METHODS: This study included 31 pediatric patients with and 31 age-matched controls. Resting and exercise echocardiography were performed to assess systolic and diastolic function, aortic dimensions, and vascular parameters. RESULTS: Patients with MFS had larger baseline height and body

Increased Disarray of Extracellular Matrix Collagen-I Fiber Network and Compromised Biomechanics in Aortae From Marfan-Syndrome Mice Assessed Through Combined Opto-Biomechatronics.

Fibrillin‑1 (FBN1) mutations lead to extracellular matrix (ECM) defects with progressive aortic dilation in Marfan's syndrome (MFS). MFS thoracic aortas have increased stiffness, but how visco-elastic and microstructural abnormalities affect in vivo hemodynamics remains inconclusive. We applied ex vivo uniaxial visco-elasticity stress-relaxation testing and simultaneous 3D collagen Second Harmonic Generation (SHG) imaging to MFS and wt littermate linearized aortic strips with in vivo vascular function. MFS mouse (Fbn1mgR/mgR) aortas were stiffer and more viscous. SHG imaging revealed MFS ECM d

TRPV4 is Associated with a Marfan Syndrome-Related Mechanosensitive Gene Program in Aortic Smooth Muscle Cells.

Marfan syndrome (MFS) aortopathy involves extracellular matrix disruption, altered mechanical cues, and maladaptive vascular smooth muscle cell (VSMC) remodeling. However, the mechanosensitive regulators associated with MFS-related VSMC dysfunction remain unclear. This study aimed to identify and validate mechanosensitive candidate genes in MFS aortopathy. Public VSMC transcriptomic data from GSE128101 were re-analyzed to identify differentially expressed genes (DEGs). Protein-protein interaction analysis, functional enrichment, and minimum redundancy maximum relevance analysis were used to pr

Unexpected Coronary Malperfusion Requiring Bailout Coronary Artery Bypass Grafting after Valve-Sparing Aortic Root Replacement in a Patient with Anomalous Aortic Origin of a Coronary Artery: A Case Report.

Valve-sparing aortic root replacement (VSRR) requires coronary button reimplantation, and anomalous coronary anatomy may increase the risk of unexpected coronary malperfusion. A 38-year-old man with Marfan syndrome underwent VSRR for aortic root aneurysm and severe aortic regurgitation. The right coronary artery arose anomalously from the left sinus of Valsalva. Despite apparently satisfactory reconstruction, severe myocardial ischemia developed after coronary reimplantation during weaning from cardiopulmonary bypass. Intraoperative reassessment revealed no obvious kinking or obstruction of th

Molecular heterogeneity and diagnostic performance of phenotype-driven multigene panel testing in patients with marfanoid habitus.

Marfanoid habitus represents a clinically heterogeneous phenotype observed in Marfan syndrome (MFS) and other hereditary connective tissue disorders; however, the diagnostic utility of multigene panel testing in this group remains incompletely defined. Marfanoid features encountered in clinical genetics practice are often partial, age-dependent, or overlapping and may not meet diagnostic criteria for a single disorder, limiting phenotype-based classification and supporting multigene testing. We retrospectively evaluated the diagnostic yield of a custom-designed NGS panel targeting OMIM-listed

People With Marfan Syndrome Utilize an Ankle Dominant Strategy to Perform the Sit-to-Stand Task.

Marfan syndrome (MFS) is a connective tissue disorder caused by structural changes in fibrillin-1 and is associated with muscle weakness and joint pain. The understanding of lower extremity (LE) joint mechanics associated with joint pain in people with MFS is limited. The goal of this study was to assess LE joint mechanics during the sit-to-stand (STS) task in people with MFS compared with asymptomatic controls. Sixteen people with MFS and 16 sex- and body mass index-matched controls were tested in this study. All participants performed the STS task at a self-selected speed. Peak LE joint exte

Phenotypic spectrum of 11p15.5 duplications: parent-of-origin and copy number variant size shape outcomes from Silver-Russell to Beckwith-Wiedemann syndromes.

BACKGROUND: Genomic imprinting at the 11p15.5 region is critically involved in fetal growth regulation. Disturbances in this region are primarily associated with two opposing growth disorders: Silver-Russell syndrome (SRS, growth restriction) and Beckwith-Wiedemann syndrome (BWS, overgrowth). While epigenetic alterations are well-characterized in SRS and BWS, copy number variants (CNVs) rearrangements remain incompletely understood, particularly in terms of how duplication size and parental origin shape the phenotypic spectrum. METHODS: We present a case series of three prenatally identified f

Pathogenic variants in the cohesin loader subunit MAU2 underlie a distinct Cornelia de Lange Syndrome subtype.

The role of the cohesin complex depends on the cohesin loader proteins NIPBL and MAU2. While NIPBL variants are a major cause of Cornelia de Lange Syndrome (CdLS), the role of MAU2 in disease is unclear. We describe 18 individuals carrying 15 heterozygous MAU2 variants and demonstrate pathogenicity through functional analyses. In-frame MAU2 variants predominantly impair NIPBL-MAU2 interaction, whereas truncating variants cause MAU2 haploinsufficiency and lead to NIPBL reduction. Most individuals exhibit a DNA methylation profile compatible with the CdLS episignature. We also describe two MAU2-

Quality of Evidence for Prenatal Down Syndrome Screening: An Umbrella Review.

OBJECTIVE: Many systematic reviews (SRs) report the accuracy of prenatal Down syndrome screening, but their methodological quality has not been comprehensively evaluated. This umbrella review assessed the quality, compared non-invasive prenatal testing (NIPT) with traditional screening, and evaluated the findings' robustness. METHOD: We searched MEDLINE, Embase, and multiple other databases from inception to March 24, 2026, for SRs, including gray literature sources as specified in the methods. Methodological quality, risk of bias, and evidence certainty were assessed using AMSTAR 2, ROBIS, a

Klippel-Trenaunay Syndrome in a Pediatric Patient: A Case Report.

INTRODUCTION: Klippel-Trenaunay syndrome (KTS) is a rare congenital vascular disorder defined by the triad of capillary malformation, venous or lymphatic anomalies, and soft tissue or bony hypertrophy. Pediatric presentations pose distinct challenges because clinical manifestations evolve with growth, and early phenotypes may not reliably predict long-term morbidity. In early childhood, accurate flow phenotyping and anticipatory surveillance are therefore central to management. CASE PRESENTATION: A 4-year-old girl presented with an extensive port-wine stain involving nearly half of the right

Emergency Bentall procedure at 15 weeks' gestation for giant aortic root aneurysm in Marfan syndrome: a case report.

BACKGROUND: Pregnancy in women with Marfan syndrome markedly increases the risk of life-threatening aortic complications due to superimposed haemodynamic and hormonal stress on an intrinsically fragile aortic wall. Management becomes particularly challenging when a giant aortic root or ascending aortic aneurysm with severe aortic regurgitation is diagnosed during early pregnancy before fetal viability. CASE SUMMARY: A 19-year-old primigravid woman at 15 weeks and 2 days of gestation with known Marfan syndrome presented with persistent palpitations. Evaluation revealed a giant fusiform aneurys

Secondary Thoracoabdominal Aortic Replacement Following Total Arch Repair With a Sutureless Integrated Stented Graft in Marfan Syndrome: A Case Report.

BACKGROUND Secondary thoracoabdominal aortic replacement after a total arch repair is a complex procedure that has a risk of complications such as stroke and renal failure. Patients with Marfan syndrome are more at risk of distal aortic re-interventions. This report describes a 52-year-old woman with Marfan syndrome and a history of type A aortic dissection treated with total arch replacement and frozen elephant trunk implantation using a sutureless integrated stented graft (SISG), who presented 3 years later with a distal aortic dissection aneurysm requiring secondary thoracoabdominal aortic
