Heterotaxy Syndrome Combining Univentricular Heart, Polysplenia and Truncated Pancreas: A Case Report.

BACKGROUND AND OBJECTIVES: Univentricular heart is a complex congenital heart disease with rare survival into adulthood. It could be part of a heterotaxy syndrome when, apart from the cardiac malformations, there are associated abnormal arrangements of extra-cardiac organs like the lungs, spleen and pancreas. The syndrome is said to be rarer than the univentricular heart and to have a worse clinical outcome. METHODS: After clinical assessment, the patient was investigated with Chest X-ray, Electrocardiography, Echocardiography, and contrast-enhanced CT scan of the chest and abdomen to make th

Multiple Basal Cell Carcinomas in an Adolescent with an MLPA-Detected PTCH1-Region Deletion and a Gorlin-Goltz Phenotype.

Gorlin-Goltz syndrome, also known as nevoid basal cell carcinoma syndrome or basal cell naevus syndrome, is a rare hereditary cancer-predisposition disorder most often caused by disruption of the Hedgehog signaling pathway, particularly alterations of PTCH1. Classical disease is characterized by multiple or early-onset basal cell carcinomas, odontogenic keratocysts, palmoplantar pits, skeletal anomalies and intracranial calcifications. Deletions involving chromosome region 9q22.3 may create an overlapping but broader phenotype because the deleted interval may include PTCH1 together with neighb

Clinical and Genetic Characterization of Russian Patients with von Hippel-Lindau Syndrome.

Von Hippel-Lindau syndrome (VHLS) is an autosomal dominant hereditary cancer disease caused by germline mutations in the VHL tumor suppressor gene and characterized by clinical heterogeneity and phenotype-genotype associations. We studied the VHL gene in 535 Russian patients referred for VHLS diagnostics using Sanger sequencing, multiplex ligase-dependent probe amplification, and, in some cases, high-throughput sequencing. A total of 177 causative VHL variants were identified, including 129 point mutations and 48 extended deletions. Missense variants were the most frequent (60.3%) and predomin

Types 1 diabetes mellitus and Graves' disease in Sanjad-Sakati syndrome: a case report in Saudi Arabia.

Sanjad-Sakati syndrome (SSS) is a rare autosomal recessive disorder caused by TBCE gene variants and characterized by congenital hypoparathyroidism, severe growth retardation, developmental delay, and dysmorphic features. We report a nine-year-old Saudi boy with genetically confirmed SSS due to a homozygous 12-bp deletion in exon 3 of the TBCE gene (c.155_166del12bp; p.S52_G55del), who presented with hypocalcemic convulsions at three weeks of age. At three years, he developed type 1 diabetes mellitus (T1DM) with profound insulin deficiency and positive anti-islet cell and anti-GAD-65 antibodie

The Role of the Immunoglobulin Superfamily of Cell Adhesion Molecules in Intellectual Disability.

Immunoglobulin superfamily (IgSF) cell adhesion molecules (CAMs) are cell surface glycoproteins that play critical roles during multiple stages of synapse formation, including axon and dendrite growth and guidance, target recognition, axo-dendritic contact formation and its maturation into a functional synapse. Intellectual disability is a condition characterized by significant limitations in intellectual functioning and adaptive behaviors. Growing evidence indicates that deficits in synapse formation underlie the pathogenesis of intellectual disability. In this review, we discuss studies show

Comparison of fecal continence and bowel function scoring systems following anorectal malformation repair: a systematic review.

PURPOSE: Multiple instruments are used to assess fecal continence and bowel function after anorectal malformation (ARM) repair, but their comparability is uncertain. This systematic review examined inter-system agreement and differences in the findings produced when two or more scoring systems were applied to the same ARM cohort. METHODS: MEDLINE, Embase, and Web of Science were searched from inception through 26 April 2026. Original studies applying at least two named fecal continence or bowel function scoring systems to the same ARM cohort were eligible. Risk of bias was assessed using an a

Changes in birth certificate-reported prevalence of congenital anomalies at live birth during the COVID-19 pandemic: a nationwide study of 26 million United States live births.

BACKGROUND: Although the majority of studies have indicated that maternal severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) infection and coronavirus disease 2019 (COVID-19) vaccination are not associated with neonatal birth defects, it is relevant to examine whether the disease burden changed during the COVID-19 pandemic, as other influencing factors may have also changed. We aimed to compare the birth certificate-reported prevalence of congenital anomalies at live birth before and during the pandemic. METHODS: Using public birth data, all live births from 2016 to 2022 in the Unit

Family-Centered Care and Sociodemographic Characteristics as Predictors of Caregiving Burden and Parental Self-Efficacy in Mothers of Infants With Congenital Heart Disease.

BACKGROUND: Congenital heart disease requires complex care, placing mothers at risk of high burden and low self-efficacy. Family-centered care (FCC), emphasizing collaboration with healthcare providers, may alleviate these challenges. However, its predictive role in the Turkish context remains underexplored. METHODS: This cross-sectional, descriptive correlational study included mothers of 131 children aged 0 to 2 years who were followed at a pediatric cardiac surgery unit. Data were collected using the FCC Scale, Zarit Caregiving Burden Scale, and Parental Self-Efficacy Scale. Analyses invol

Limb Body Wall Complex: Report of a Discordant Case in a Monozygotic Twin Pregnancy.

Limb Body Wall Complex (LBWC) is a rare, severe congenital malformation syndrome characterized by thoracoabdominal wall defects, craniofacial anomalies, and limb and spinal malformations. Its incidence ranges from 0.04 to 3.3 per 10,000 births. We present a case of LBWC diagnosed in a monochorionic diamniotic twin pregnancy at 17 weeks of gestation. The mother was 28-year-old, primigravida, and had no notable medical history. Ultrasound identified abnormal positioning of 1 embryo, suggestive of adherence to the amnion. First-trimester scans revealed increased nuchal translucency and foot malpo

BCMA CAR T-cell therapy in a patient with heavily pretreated relapsed/refractory POEMS Syndrome: a case report.

B-cell maturation antigen (BCMA) chimeric antigen receptor T-cell (CAR T-cell) therapy is an established treatment for relapsed/refractory multiple myeloma. However, its application in POEMS syndrome remains largely unreported, despite overlapping pathogenesis involving plasma cell dyscrasia. Herein, we present a woman in her 30s with heavily pretreated relapsed/refractory POEMS syndrome who received equecabtagene autoleucel, a fully human anti-BCMA CAR T-cell product. The patient achieved a complete vascular endothelial growth factor (VEGF) response by day 14 post-infusion, with sustained rem

Alcohol Exposure Alters Proximodistal Nephron Segmentation During Zebrafish Embryogenesis.

Fetal alcohol spectrum disorders (FASD) encompass a broad range of effects caused by prenatal alcohol exposure. In the United States, conservative estimates place the incidence of FASD at a rate of 5 in 100 births. Children with FASD can have morphological afflictions including congenital defects of the brain, spinal cord, heart, eyes, and skeleton, as well as poor formation of facial structures such as the nose and mouth. Cognitive and behavioral abnormalities such as learning disabilities, socio-developmental delays and emotional disorders are also highly prevalent. There is no known cure fo

Interplay between genomic architecture alterations and GDF6 regulation: A candidate mechanism in Nablus mask-like facial syndrome.

Nablus mask-like facial syndrome (NMLFS) is a very rare disorder associated with 8q22.1q22 microdeletions and characterized by a distinctive facial phenotype and variable neurological, cardiological, and genital anomalies. However, not all affected individuals with overlapping deletions present the mask-like phenotype, thus suggesting the involvement of an additional regulatory mechanism. Here, we perform a comparative analysis of previously reported affected individuals (n = 30) with deletions in the 8q21q22 region, stratified by the presence or absence of the NMLFS phenotype, and including a
