Circulating BDNF in pediatric neurodevelopmental disorders: a meta-analysis of case-control differences and cognitive severity.

BACKGROUND: Brain-derived neurotrophic factor (BDNF) has been proposed as a neuroimmune biomarker of cognitive dysfunction in paediatric neurodevelopmental disorders other than autism spectrum disorder, but two related questions remain poorly characterised: whether circulating BDNF differs between affected and unaffected individuals, and whether it tracks the severity of cognitive impairment within affected populations. This systematic review and meta-analysis addressed both questions in children and adolescents with cerebral palsy (CP), intellectual disability (ID), or Down syndrome (DS). ME

Quantity down, quality preserved: a continuous HOMA-IR analysis of IVF/ICSI outcomes in PMOS patients.

OBJECTIVE: To evaluate the dose-response relationship between HOMA-IR as a continuous variable and embryo development outcomes in fresh IVF/ICSI cycles of PMOS patients (formerly PCOS), distinguishing IR effects on oocyte yield (ovarian responsiveness) from effects on embryo developmental efficiency (per-oocyte conversion potential), and to evaluate differential IR impacts on cumulative implantation versus pregnancy maintenance. DESIGN: Single-center retrospective cohort study of all fresh IVF/ICSI cycles from July 2022 to March 2026, comprising 3058 de-duplicated patients (2799 non-PMOS, 259

[What is the cost of neonatal respiratory distress for the Brazilian Unified National Health System? A national analysis of direct costs, regional distribution, and temporal variation].

Neonatal respiratory distress syndrome shows high clinical severity and economic burden for the Brazilian Unified National Health System. This study estimated the direct costs of hospitalizations from 2018 to 2021 and described their distribution by diagnosis, region, and period. A cost-of-disease study with a top-down approach and authorizations for hospitalization of newborns aged from 0 to 27 days was carried out. Values were updated to December 2025 and standardized per 1,000 live births. The period included 1,449,694 neonatal hospitalizations, of which 227,130 (15.7%) were due to respirat

Nerve growth factor metabolic dysfunction in Down's syndrome brains.

Basal forebrain cholinergic neurons play a key role in cognition. This neuronal system is highly dependent on nerve growth factor (NGF) for its synaptic integrity and the phenotypic maintenance of its cell bodies. Basal forebrain cholinergic neurons progressively degenerate in Alzheimer's disease and Down's syndrome, and their atrophy contributes to the manifestation of dementia. Paradoxically, in Alzheimer's disease brains, the synthesis of NGF is not affected and there is abundance of the NGF precursor (proNGF). We have shown that this phenomenon is due to a deficit in NGF's extracellular me

Dynamics of human cardiogenesis and its disruption in trisomy 21.

Developmental dynamics involve the specification of diverse cell types and their spatial organization into multicellular niches1. Here we combine single-cell and spatial multiomics to define 21 distinct tissue niches in the developing heart, which we use to develop a context-aware, resolution-agnostic niche classification tool (TissueTypist). Applying high-resolution spatial profiling to the developing sinoatrial node, we resolve three pacemaker cell subtypes arrayed along a linear axis. First trimester subpopulations, such as pacemaker cells in the sinus horn and sinoatrial node head region,

Evidence for functional convergence between LZTS2 and DYRK1A during craniofacial development.

BACKGROUND: Precise control of DYRK1A dosage is essential for embryonic development, including craniofacial morphogenesis. While LZTS2 is among the most consistently identified DYRK1A-interacting proteins, its roles in embryonic development remain incompletely understood, and its potential contribution to craniofacial development has not been examined. Xenopus laevis was used to test the role of LZTS2 in craniofacial development and its functional relationship with DYRK1A. RESULTS: Lzts2 and Dyrk1a showed overlapping expression during craniofacial development, with both proteins present in de

On markers of linguistic impairment in autism and their implementation.

Language abilities in autism show considerable variability across linguistic domains: vocabulary, grammar/structural language, and pragmatics (language in context) may be affected to differing degrees and not always simultaneously, as is the case in other neurodevelopmental conditions (NDCs), e.g. Down syndrome, Williams syndrome, or Developmental Language Disorder. This selective review synthesises empirical markers of language impairment across linguistic domains in autism, sometimes in comparison to other NDCs, while also evaluating theoretically driven assessment tools, both established an

Hyperfunction of PSD-95 leads to hyperexcitability and cognitive abnormalities in MEF2C haploinsufficiency.

Myocyte enhancer factor 2C (MEF2C) is a transcription factor critical for brain circuit development and cognitive behavior. The loss of one copy of MEF2C leads to MEF2C haploinsufficiency syndrome (MCHS), a neurodevelopmental disorder characterized by intellectual disability, epilepsy, and behavioral phenotypes associated with autism spectrum disorders. Given that numerous genes are regulated by MEF2C, the neuronal and behavioral deficits in MCHS are complex, and there is currently no effective disease-modifying strategy for this disorder. Here, we show that MEF2C haploinsufficiency in forebra

Actin-Related Causes of Aneuploidy in Human Eggs.

Gametogenesis (oogenesis and spermatogenesis) relies on a specialized form of cell division known as meiosis. During this process, chromosomes in parental cells undergo two successive rounds of segregation, resulting in a halving of their number and the formation of haploid germ cells, which is essential for subsequent fertilization. However, in human female gametogenesis, errors in chromosome segregation occur at a notably high frequency, leading to the formation of oocytes with abnormal chromosome numbers. Once fertilized, such oocytes often develop into aneuploid embryos, which exhibit seve

A Novel FAM111A Frameshift Variant Associated With Osteoclast Necroptosis and KCS2-like Syndrome.

Kenny-Caffey syndrome type II (KCS2) is a rare genetic disorder characterized by skeletal abnormalities, impaired growth, and developmental delay. This study investigates a novel heterozygous FAM111A variant's role in a patient presenting with KCS2-like features. An 11-year-old patient with clinical features consistent with KCS2-like syndrome underwent whole exome sequencing, which identified a novel heterozygous variant in the FAM111A gene. In vitro experiments and protein structure analysis were performed to elucidate the contribution of this mutation to KCS2-like syndrome. To confirm the di

Senotherapeutic potential against xeroderma pigmentosum.

BACKGROUND: Xeroderma pigmentosum (XP) is an inherited photoaging syndrome caused by mutations in genes involved in the nucleotide excision repair (NER) pathway. XP patients exhibit hypersensitivity to ultraviolet (UV) radiation, leading to accelerated skin aging and requiring lifelong sun avoidance. In this study, we aimed to investigate UV-induced cellular senescence in XP melanocytes and explore potential senotherapeutic strategies. METHODS: Melanocytes differentiated from iPSCs derived from an XP-A patient and a healthy individual, respectively, were used to induce senescence. Senescence

Muir-Torre syndrome in a patient with advanced duodenal carcinoma.

Muir-Torre syndrome (MTS) is a rare autosomal dominant disorder characterized by sebaceous neoplasms and visceral malignancies and is regarded as a phenotypic variant of Lynch syndrome. Although colorectal and genitourinary cancers are the most common visceral malignancies in MTS, duodenal cancer is extremely rare. We report a case of advanced duodenal cancer associated with MTS treated with multidisciplinary therapy. A 62-year-old man with a history of multiple colorectal cancers and sebaceous carcinomas presented with vomiting and epigastric pain. Upper gastrointestinal endoscopy revealed a

Clinical Diagnostic Value of miR-142-3p in Pediatric Acute Respiratory Distress Syndromes and Its Effects on Human Lung Epithelial Cells.

Pediatric acute respiratory distress syndrome (PARDS) is a major cause of morbidity and mortality in critically ill children. microRNAs (miRNAs) were discovered to be involved in regulating the progression of numerous disorders, including PARDS. We aim to explore the diagnostic value and relevant mechanisms of miR-142-3p in PARDS. Real-time quantitative PCR (RT-qPCR) was used to detect the levels of miR-142-3p and myristoylated-alanine rich C-kinase substrate (MARCKS) in the serum of PARDS patients or in A549 cells. Receiver operating characteristic (ROC) curve analysis predicted the diagnosti

Memory and Plasma pTau217 Refine Prognosis in Down Syndrome.

Individuals with Down syndrome (DS) are at high risk for Alzheimer's disease (AD), yet accessible tools for routine clinical screening of AD in individuals with DS remain limited. Using data from the Alzheimer Biomarker Consortium-Down Syndrome (ABC-DS), we show that integrating plasma pTau217 with performance on the modified cued recall task identifies a subgroup of cognitively stable individuals at markedly elevated risk of developing clinical impairment in the next 7 years. This low-burden, multimodal framework improves prognosis, supports more informed clinical management and efficient tri
