Shared book reading in Down syndrome: parent-child interaction, language, and communication compared with typically developing peers.

PURPOSE: Shared book reading (SBR) is well-suited to children with Down syndrome (DS), yet evidence on parent-child communication during SBR remains limited. This exploratory observational study examined parental behaviours, interaction and communication, and child engagement and multimodal communication during SBR with children with DS (n = 12; 24-45 months), a typically developing (TD) group similar on chronological age (AM; n = 10; 25-47 months) and a TD group similar on receptive vocabulary (LM; n = 10; 12-29 months). METHOD: Parent-child dyads were video-recorded sharing an unfamiliar pi

Parental and grandparental age in relation to Down syndrome risk and congenital heart disease in affected children: a case-control study in Peru.

UNLABELLED: This study aimed to assess the association between grandparental age and the risk of Down syndrome and evaluate the relationship between paternal age and congenital heart disease (CHD) among affected children in Peru. A retrospective case-control study was conducted at a national pediatric referral center in Peru, including 290 participants (145 cases with free trisomy 21 and 145 controls without genetic disorders). Logistic regression models were used to estimate the odds ratios (ORs) and 95% confidence intervals (CIs). Parental ages were higher in cases than in controls, but ther

Unique coexistence of tetralogy of Fallot and congenital heart block linked to maternal autoantibodies in a Down syndrome fetus: a case report.

We report a rare fetal presentation of tetralogy of Fallot and complete atrioventricular block linked to maternal anti-Ro antibodies. However, trisomy 21 was confirmed postnatally. Despite pacemaker implantation, the neonate died on day 55 due to pulmonary complications. This complex case highlights diagnostic and management challenges involving tetralogy of Fallot, Down syndrome, and complete heart block associated with maternal autoantibodies.

Turkish adaptation and psychometric evaluation of the SATCo in infants and toddlers with Down syndrome.

BACKGROUND AND OBJECTIVE: Down syndrome (DS) is a genetic disorder characterized by hypotonia and laxity, with motor deficits that can lead to trunk control issues. This study aimed to examine the reliability and validity of the Turkish version of the Segmental Assessment of Trunk Control (SATCo) in children with DS. METHODS: In this prospective study, we included twenty-seven enrolled children with DS aged 6-18 months. We used Cronbach's alpha and the Intraclass Correlation Coefficient (ICC) to evaluate internal consistency and intra/inter-rater reliability. Spearman's correlation coefficien

An Uncommon Cause of Dyspnea: Parsonage-Turner Syndrome With Bilateral Phrenic Nerve Involvement.

Parsonage-Turner syndrome, also called neuralgic amyotrophy, typically affects the brachial plexus, while involvement of the phrenic nerve is far less commonly documented. When present, it can compromise diaphragmatic function, producing breathlessness with activity, orthopnea, or, in more advanced presentations, respiratory failure. These features are easily mistaken for other cardiac or pulmonary conditions, which often delays correct diagnosis. A 51-year-old woman presented with a five-month course of worsening breathlessness on exertion and inability to breathe comfortably while supine. Ca

NUDT21 Promotes Myelodysplastic Syndrome Progression by Regulating SRSF2 Expression via Alternative Polyadenylation.

Myelodysplastic syndrome (MDS) is a clonal hematopoietic neoplasm with a high risk of leukemic transformation. Alternative polyadenylation (APA) is an important post-transcriptional mechanism, and its dysregulation is closely linked to the pathogenesis of hematological malignancies. This study aimed to clarify the regulatory role of the NUDT21-APA-SRSF2 axis in the progression of myelodysplastic syndrome (MDS). To this end, we established MDS cell models overexpressing or knocking down NUDT21 and SRSF2. The regulatory mechanism and functional impacts of this axis were systematically evaluated

Left Stellate Ganglion Pulsed Radiofrequency Suppresses Ischemic Ventricular Arrhythmias: A Translational Study.

BACKGROUND: Cardiac sympathetic hyperactivity originating from the left stellate ganglion (LSG) drives ischemic ventricular arrhythmias (VAs). Stellate ganglion block provides only transient relief, whereas surgical denervation is invasive and irreversible. A minimally invasive approach with potentially sustained effects is needed. OBJECTIVES: This study sought to investigate whether pulsed radiofrequency (PRF) of the LSG-a nonthermal neuromodulatory technique-safely suppresses ischemic VAs, and to characterize its mechanism using integrated neural and cardiac transcriptomics. METHODS: Safet

Cytokine-driven recurrent pericarditis in Down syndrome: Role of pericardial fluid analysis and cardiac magnetic resonance imaging.

Recurrent pericarditis is uncommon in children with Down syndrome (DS) and may present diagnostic challenges. We report the case of a child with DS who developed massive pericardial effusion with minimal systemic inflammatory markers. Pericardial fluid analysis revealed markedly elevated interleukin-6 levels, suggesting localized cytokine-driven inflammation. Cardiac magnetic resonance (CMR) imaging revealed limited subepicardial enhancement with preserved ventricular function, findings consistent with pericarditis and secondary myocardial involvement rather than primary myocarditis. Exploring Perceived Barriers and Facilitators to Healthier and More Sustainable Dietary Behavior in 27 European Countries: Protocol for a Cross-Sectional Survey.

BACKGROUND: Food systems strongly affect human and environmental health. Across Europe, they often generate negative public health and environmental outcomes and contribute to inequalities in access to healthy, sustainable food. Addressing these challenges requires understanding the complex dynamics of the interaction of individual behavior within food systems, which involves a combination of internal mechanisms and external influences. OBJECTIVE: This study, part of the FEAST project (Food Systems That Support Transitions to Healthy and Sustainable Diets), aims to increase understanding of h

Efficacy and safety of risk-stratified chemotherapy with azacitidine for children with myeloid leukaemia with Down syndrome: protocol for a multicentre phase II trial (JCCG AML-D24).

INTRODUCTION: The prognosis of myeloid leukaemia associated with Down syndrome (ML-DS) is excellent with reduced-intensity chemotherapy; however, that of refractory or relapsed ML-DS remains dismal. These contrasting outcomes highlight the importance of risk-stratified therapy, based on reliable prognostic factors, to prevent treatment-related mortality and relapse. In addition, novel therapeutic strategies capable of overcoming the poor prognosis of high-risk patients are eagerly anticipated. Based on the results of molecular analyses, in vitro functional assays and anecdotal case reports, az

Domains of self-regulation in autism, Down syndrome, and Williams syndrome: Condition-specific or transdiagnostic?

BACKGROUND: This questionnaire study examined regulatory control in the conditions of autism, Down syndrome, and Williams syndrome by assessing effortful control and executive function across cognitive, emotional, social, and sensory domains. METHOD: A bespoke parent-report questionnaire was developed. Parents of children aged 5-13 with these conditions and neurotypical peers participated (n = 231). Factor analysis was used to identify regulatory structures across groups, and Bayesian analyses compared group factor scores. Cluster analysis explored whether profiles grouped by diagnosis or reg

Structured Evaluation of Model-Informed Precision Dosing of Mycophenolic Acid and Tacrolimus in Pediatric Patients with Kidney Disease.

BACKGROUND AND OBJECTIVE: Mycophenolic acid and tacrolimus are frequently used in kidney transplant recipients and patients with nephrotic syndrome. For both drugs, therapeutic drug monitoring is recommended. Model-informed precision dosing, not only uses population pharmacokinetic data but also increases flexibility. To optimize treatment with mycophenolic acid and tacrolimus using a limited number of samples, we evaluated model-informed precision dosing for both drugs. METHODS: We performed the following steps: (1) selection of population pharmacokinetic models; (2) retrospective evaluation

The Pinnaroo Project: a longitudinal 3-year cross-sectional study of the impact of a community-based arts-in-health program on the health and wellbeing of a remote Australian community.

INTRODUCTION: Rural and remote communities around the world face limited access to health and other services, often in the context of social, political, economic and environmental adversities. Evidence from WHO recognises art as a vital, non-invasive tool to enhance physical and mental health. The Pinnaroo Project was an arts-in-health project established by a remote rural community to address a dearth of locally available health services. METHODS: This study utilised a longitudinal 3-year cross-sectional study design from 2021 to 2023. The Pinnaroo Project team partnered with experts in crea

Redefining the Future of Skin Wound Healing with Modular Biomaterials.

The COVID-19 pandemic has presented a significant challenge to the world's public health and led to over 6.9 million deaths reported to date. A rapid, sensitive, and cost-effective point-of-care virus detection device is essential for the control and surveillance of the contagious severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2) pandemic. The study presented here aimed to demonstrate a solid-phase isothermal recombinase polymerase amplification coupled CRISPR-based (spRPA-CRISPR) assay for on-chip multiplexed, sensitive and visual COVID-19 DNA detection. The assay targets the SARS-

Continuity and Change of Delayed Sleep Phase From Adolescence to Early Adulthood.

BACKGROUND AND OBJECTIVES: The developmental trajectories and stability of delayed sleep phase (DSP) are poorly known. We investigated how behavioral and psychiatric characteristics, circadian thermoregulation, and the amount and intensity of physical activity (PA) were associated with the continuity of DSP from late adolescence to early adulthood. METHODS: We created a population-based longitudinal study in which 1374 adolescents (mean age, 16.8 years; SD, 0.6; 66% girls) self-reported their sleep and psychiatric and behavioral symptoms at timepoint 1 (T1). A DSP-weighted subsample at T2 (7
