Sex-Based Differences in Intraocular Pressure During Head-Down Tilt in a Spaceflight Analog Model.

INTRODUCTION: Spaceflight-associated neuro-ocular syndrome is a high-priority research gap, especially regarding its mechanisms and individual susceptibility, including sex-based differences. This study investigates sex-specific intraocular pressure (IOP) responses during head-down tilt (HDT) to improve understanding of differential risk profiles and inform the development of targeted countermeasures. METHODS: A total of 15 healthy subjects (8 males, 7 females) completed a 60-min -12° HDT protocol. IOP was measured in a seated baseline condition and at 0, 30, and 60 min during HDT using a reb

Global and Local Processing of Letters and Faces in Children and Adolescents with Typical and Atypical Development.

BACKGROUND/OBJECTIVES: this paper investigates the local vs. global visual processing preference in typically developing (TD) children, youth with Down syndrome (DS), and youth with Williams syndrome (WS). In particular, the global precedence effect (GPE) and the global interference effect (GI) have recently been described as two distinct and at least partially independent effects. METHODS: in this study, 50 participants (TD = 25, DS = 13, WS = 12) completed two experiments requiring the identification of either the global or local level of hierarchical stimuli, which consisted of letters and

Moving Beyond Somatic Alterations: Uncovering the Germline Basis of Myeloid Malignancies.

Myeloid neoplasms (MNs) with germline predisposition represent a distinct, increasingly recognized category in the WHO classification, encompassing myelodysplastic syndrome (MDS) and acute myeloid leukemia (AML) arising in the context of an inherited genetic alteration or mutation. While often presenting at a younger age or with characteristic cytopenias with or without organ dysfunction, some can manifest in adulthood, highlighting the need for vigilance regardless of age or family history. Key predisposing genes include transcription factors (e.g., RUNX1, CEBPA) and genes involved in RNA spl

Effects of 12-week aquatic HIIT on blood pressure lipid profile and BaPWV in postmenopausal women with different ACE genotypes.

The angiotensin-converting enzyme (ACE) insertion/deletion (I/D) polymorphism influences renin-angiotensin-aldosterone system activity and may modulate cardiovascular adaptations to exercise. Yet, evidence regarding genotype-dependent responses to aquatic high-intensity interval training (HIIT) in postmenopausal women is limited. We aimed to compare the effects of 12-week aquatic HIIT on blood pressure, lipid profile, and arterial stiffness between postmenopausal women with the ACE II genotype and those carrying at least one D allele (ID/DD genotype). Sixty postmenopausal women aged 45-75 year

The Effects of Tonic and Burst Spinal Cord Stimulation on Cortical Pain Processing and Their Interaction With Conditioned Pain Modulation.

INTRODUCTION: Spinal cord stimulation (SCS) is an effective treatment for certain chronic pain conditions. Proposed mechanisms of SCS include modulation of the ascending lateral, ascending medial, and descending pain pathways. Conditioned pain modulation (CPM) evaluates the descending pathway by measuring ways a first painful stimulus is affected by a second painful stimulus. OBJECTIVES: We aim to increase insight into SCS mechanisms by exploring cortical activity in response to painful stimuli under various SCS paradigms and assessing how these responses are influenced by CPM. MATERIALS AND

Intracerebral age-independent reduction of endothelium-dependent hyperpolarization and nitric oxide signalling in the Dp16 mouse model of Down syndrome.

Down syndrome (DS) is associated with a myriad of cardiovascular defects. Endothelial cells, which form the innermost layer of blood vessels, have been found to exhibit divergent morphology and functional impairments in the peripheral circulation of individuals with DS. DS is also a strong risk factor for Alzheimer's disease-related neuropathology. Recent evidence linking cerebral vasculature to Alzheimer's disease, along with established correlations between cardiovascular and cognitive functions, prompted us to investigate brain parenchymal microcirculation reactivity in a DS mouse model. We

Identification of RALY as a novel regulator of DSCR1 transcription.

Down syndrome critical region 1 (DSCR1) is located on chromosome 21, whose N-terminal coding sequence lacks recognizable motifs, and its precise transcriptional and translational regulation remains incompletely understood. Utilizing a bicistronic reporter system, we identified the core promoter region of DSCR1, which enables investigation of its gene regulatory mechanisms. Proteomic analyses via LC-MS/MS revealed that RALY directly binds to the DSCR1 promoter, influencing its transcriptional activity. Notably, RALY overexpression markedly suppresses DSCR1 expression, whereas siRNA-mediated kno

BMAL1 downregulation exacerbates age-related nonalcoholic steatohepatitis by promoting NLRP3 inflammasome activation via HIF-1ɑ-mediated glycolysis.

INTRODUCTION: Epidemiological studies have demonstrated higher incidence and mortality rate of nonalcoholic steatohepatitis (NASH) in the elderly population than in younger groups. However, the mechanisms underlying this age-related exacerbation remain poorly understood. OBJECTIVE: This study aimed to elucidate the specific pathways through which aging exacerbates NASH progression, using an integrated in vivo and in vitro model. METHODS: Aged (18-month-old) and young (6-week-old) mice were fed a high-fat diet (HFD) for 16 weeks to induce NASH. A senescence-associated cellular model of NASH w

Acute low-dose benzo[a]pyrene exposure induces neurodegenerative syndrome in cyp1a low-expression zebrafish: A novel living Bioindicator for dioxin-like environmental pollutants.

Benzo[a]pyrene (BaP), a ubiquitous environmental polycyclic aromatic hydrocarbon (PAH), elicits toxicity that is closely linked to the activity of the key metabolic enzyme CYP1A. In this study, a cyp1a low-expression zebrafish strain, KI (cyp1a+/+-T2A-mCherry), was used to investigate the toxic effects and mechanisms of low-dose BaP exposure under conditions of cyp1a deficiency. The results revealed that cyp1a low expression significantly exacerbated BaP-induced mortality, growth inhibition, apoptosis, and oxidative stress. It also induced neurodegenerative lesions accompanied by reduced acety

Activating FcγRI inhibits RIG-I-mediated host antiviral innate immunity by FGR during PRRSV-ADE infection.

Currently, the antibody-dependent enhancement (ADE) effect has been a pivotal factor in the persistent infection of pigs with porcine reproductive and respiratory syndrome virus (PRRSV) and a major hurdle in developing safe and effective vaccines. Therefore, elucidating the molecular mechanisms of ADE in PRRSV infection is crucial. This study assessed the impact of Fc gamma receptor I (FcγRI), Fc epsilon receptor gamma chain (FCER1G), and FGR on the host's antiviral innate immune response during PRRSV-ADE infection in vitro. Our results revealed that the expressions of RIG-I, TBK-1, IRF-3, STA

Chromosomal abnormalities diagnosed at the chromosomal microarray in pregnancies with isolated high risk of trisomy 21.

INTRODUCTION: In most European countries, if the risk of trisomy 21 exceeds 1/50, Women are offered further investigation through an invasive test. Since the development of the non-invasive test based on circulating free DNA from the entire genome (cfDNA), the use of non-invasive tests in this high-risk population is now being debated. Our study aims to investigate a cohort of women at high risk of isolated trisomy 21 with no ultrasound abnormalities at the first trimester ultrasound scan or nuchal translucency greater than the 99th percentile who underwent invasive sampling with chromosomal m

Brain aging in neurodevelopmental disorders: a narrative review of oxidative, inflammatory, and mitochondrial mechanisms.

Neurodevelopmental disorders (NDDs), including autism spectrum disorder, attention deficit/hyperactivity disorder, intellectual disability, and Down syndrome, are increasingly examined through the lens of aging. Emerging evidence indicates that individuals with NDDs may exhibit accelerated or atypical brain aging, characterized by cognitive decline and increased vulnerability to neurodegenerative conditions such as Alzheimer's and Parkinson's disease. This narrative review synthesizes current findings on biological mechanisms implicated in altered aging trajectories, with emphasis on oxidative
