Fatty acid profiles and tolerance to temperature extremes in Daphnia pulex.

Identification of physiological processes setting thermal tolerance limits is essential to describing adaptive response to temperature changes. We used the North American Daphnia pulex complex, which makes a remarkable model for comparative physiology as it is composed of clones differing in heat tolerance and ploidies, and with a wide geographic distribution. The fatty acid composition of 18 diploid and triploid D. pulex clones acclimated to 16°C and 24°C was measured and compared with their tolerance to extreme high and low temperatures (CTmax and CTmin, respectively). Eicosapentaenoic acid

Comparative Analysis of the Probiotic Features of Lysinibacillus and Enterobacter Strains Isolated from Gut Tract of Triploid Cyprinid Fish.

Gut mucosal immunity of teleost is mainly governed by mucosa-associated lymphoid tissues (MALT) and indigenous microbiota on mucosal surfaces of gut tract, which can confer protection against pathogenic invasion. However, the probiotic features of bacterial isolates from gut tract of triploid cyprinid fish (TCF) were largely unclear. In this study, Lysinibacillus and Enterobacter strains were isolated for probiotic identification. Whole genome sequencing (WGS) analysis indicated that Lysinibacillus and Enterobacter isolates possessed a variety of functional genes associated with probiotic feat

Natural repeated backcrosses lead to triploidy and tetraploidy in parthenogenetic butterfly lizards (Leiolepis: Agamidae).

Obligatory parthenogenesis in vertebrates is restricted to squamate reptiles and evolved through hybridisation. Parthenogens can hybridise with sexual species, resulting in individuals with increased ploidy levels. We describe two successive hybridisations of the parthenogenetic butterfly lizards (genus Leiolepis) in Vietnam with a parental sexual species. Contrary to previous proposals, we document that parthenogenetic L. guentherpetersi has mitochondrial DNA and two haploid sets from L. guttata and one from L. reevesii, suggesting that it is the result of a backcross of a parthenogenetic L.

Chromosomal rDNA Distribution Patterns in Clonal Cobitis Triploid Hybrids (Teleostei, Cobitidae): Insights into Parental Genomic Contributions.

Background: Interspecific hybridization between relative species Cobitis taenia (with a diploid genome designated as TT), Cobitis elongatoides (EE) and Cobitis tanaitica (NN) and the successive polyploidization with transitions from sexuality to asexuality experienced by triploid Cobitis hybrids likely influence their chromosomal rearrangements, including rearrangements of ribosomal DNA (rDNA) distribution patterns. Previously, we documented distinct karyotypic differences: C. elongatoides exhibited bi-armed chromosomes while C. taenia showed uni-armed chromosomes with rDNA-positive hybridizat

High-fidelity annotated triploid genome of the quarantine root-knot nematode, Meloidogyne enterolobii.

Root-knot nematodes (RKN) of the genus Meloidogyne are obligatory plant endoparasites that cause substantial economic losses to agricultural production and impact the global food supply. These plant parasitic nematodes belong to the most widespread and devastating genus worldwide, yet few measures of control are available. The most efficient way to control RKN is deployment of resistance genes in plants. However, current resistance genes that control other Meloidogyne species are mostly inefficient on Meloidogyne enterolobii. Consequently, M. enterolobii was listed as a European Union quaranti

Diploid aposporous sunflower forms triploid BIII progeny displaying increased apospory levels and non-random genetic mutations.

Apomixis (asexual reproduction via seeds) has the potential to revolutionize sunflower breeding. In previous studies, we identified a diploid sunflower line (Rf975) that naturally exhibits extra gametophytes resembling aposporous apomictic embryo sacs (AES). Here, we investigated the nature (reduced vs. unreduced) and viability of these AES-like gametophytes by examining the formation of triploid (3x) BIII hybrids (2n + n) in the progeny of Rf975. Flow cytometry analysis of immature seeds revealed that, on average, 42.8% of self-pollinated Rf975 progeny were triploids, although only 36.6% of t

The ecology of polyploid establishment and exclusion, with implications for polyploid biogeography.

The relationship between polyploid formation, triploid fitness and plant reproduction has been studied for over a century, and uniparental reproduction has long been recognized to play a crucial role in polyploid establishment. Yet, we lack a synthesized framework of how polyploid establishment is expected to be influenced by different reproductive modes among angiosperms. Here, we provide new perspectives on how uniparental reproduction, pollination ecology, triploid fitness and assortative mating can impact minority cytotype exclusion (MCE) and, thereby, the likelihood of polyploid establish

A survey of triploid block inconsistency: potential mechanisms and evolutionary implications on polyploid speciation.

The triploid block, primarily caused by endosperm developmental issues, is known as a significant barrier to interploidy hybridization among flowering plants and, thereby, polyploid speciation. However, its strength varies across taxa, with some instances of leakiness, questioning its universal role as a barrier. We conducted a literature survey to explore the causes of the variation in the strength of the triploid block across 11 angiosperm families. We assessed the impact of interploidy cross direction, types of endosperm development, endosperm persistence at seed maturity, and divergence be

Optimization of Microsatellite Multiplex PCRs for Triploidy Verification and Genetic Diversity Assessment in the Pacific Oyster, Crassostrea gigas.

The ploidy detection is crucial for the oyster industry. The objective of this study was to develop a method that verifies ploidy of the triploid Pacific oyster Crassostrea gigas by analyzing the diversity of triploid through microsatellite multiplex PCRs using fluorescent universal primers. We developed four information-rich multiplex PCR panels, comprising a total of 12 genomic microsatellites located in the genome of the C. gigas, distributed across seven chromosomes with an average of 14 alleles per locus. Each panel used M13(-21) primers labeled with specific fluorochrome dyes, and the fo

Comprehensive analysis of lncRNA and mRNA expression in triploid rainbow trout (Oncorhynchus mykiss) liver in response to chronic hypoxia.

Long non-coding RNAs (lncRNAs) play a crucial role in response to environmental stresses (e.g. hypoxia) in triploid rainbow trout. However, the action mechanism of lncRNAs in triploid rainbow trout liver in chronic hypoxia is still not fully understood. Therefore, we investigated the mechanism of lncRNA and its target genes in response to chronic hypoxia in triploid rainbow trout using high-throughput RNA sequencing and bioinformatics analysis in this study. Results showed that 37 differentially expressed lncRNAs (DElncRNAs) and 780 differentially expressed mRNAs (DEmRNAs) were found in hypoxi

Overcoming Ploidy Barriers: The Role of Triploid Bridges in the Genetic Introgression of Cardamine amara.

Polyploidisation is a significant reproductive barrier, yet genetic evidence indicates that interploidy admixture is more common than previously thought. Theoretical models and controlled crosses support the 'triploid bridge' hypothesis, proposing that hybrids of intermediate ploidy facilitate gene flow. However, comprehensive evidence combining experimental and genetic data from natural mixed-ploidy species is missing. Here, we investigated the rates and directionality of gene flow within a diploid-autotetraploid contact zone of Cardamine amara, a species with abundant natural triploids. We c

Acclimation conditions and subsequent acute hypoxia challenges do not affect the frequency of erythrocyte nuclear segmentation in triploid brook charr Salvelinus fontinalis (Mitchill 1814).

Routine observations of blood smears have repeatedly shown that triploid fish have a greater proportion of erythrocytes with nuclear segmentation (ENS) than diploids; however, there is as yet no understanding of why this is the case and whether it affects erythrocyte function. In an attempt to address this, we examined blood smears from two previous experiments to determine whether ENS frequency is affected by rearing conditions (acclimation temperature or dissolved oxygen level) and subsequent acute aerobic distress in diploid and triploid brook charr Salvelinus fontinalis (Mitchill 1814). Genomic insights into evolution of parthenogenesis and triploidy in the flowerpot snake.

The flowerpot snake (Indotyphlops braminus) is the only known parthenogenetic and triploid species within Serpentes. However, the genetic basis underlying this phenomenon remains unresolved. We investigated the genomic complexities of this rare all-female triploid reptile. On the basis of the newly assembled genome, we revealed 40 chromosomes grouped into three subgenomes (A, B, and C). Comparative genomic analysis with related diploid species revealed a chromosome fusion event in ancestral genomes. This event shaped the unique genetic landscape of the flowerpot snake. We examined gene express

Duckweed genomes and epigenomes underlie triploid hybridization and clonal reproduction.

The Lemnaceae (duckweeds) are the world's smallest but fastest-growing flowering plants. Prolific clonal propagation facilitates continuous micro-cropping for plant-based protein and starch production and holds tremendous promise for sequestration of atmospheric CO2. Here, we present chromosomal assemblies, annotations, and phylogenomic analysis of Lemna genomes that uncover candidate genes responsible for the unique metabolic and developmental traits of the family, such as anatomical reduction, adaxial stomata, lack of stomatal closure, and carbon sequestration via crystalline calcium oxalate

Comparative study on the flavor characteristics of diploid and triploid oysters (Crassostrea gigas) before and after breeding.

In this work, the nonvolatile and volatile compounds were analyzed to investigate the effects of breeding and ploidy on the flavor of oysters. The oysters after breeding (November) exhibited enhanced flavor complexity characterized by malty and mushroom aromas, with diploid oysters displaying nutty aroma profiles, whereas triploid oysters demonstrated significantly higher intensity of fruity notes. Among oysters of different periods and ploidy, the November diploid oyster (ND) exhibited the most desirable flavor. This was primarily attributed to its significantly higher contents of adenosine 5

Quantitative genetic analysis of late spring mortality in triploid Crassostrea virginica.

BACKGROUND: Triploid oysters, bred by crossing tetraploid and diploid oysters, are common worldwide in commercial oyster aquaculture and make up much of the hatchery-produced Crassostrea virginica farmed in the mid-Atlantic and southeast of the United States. Breeding diploid and tetraploid animals for genetic improvement of triploid progeny is unique to oysters and can proceed via several possible breeding strategies. Triploid oysters, along with their diploid or tetraploid relatives, have yet been subject to quantitative genetic analyses that could inform a breeding strategy of triploid impr

Toll-like Receptor Type 2 and 13 Gene Expression and Immune Cell Profiles in Diploid and Triploid Sterlets (Acipenser ruthenus): Insights into Immune Competence in Polyploid Fish.

Toll-like receptors (TLRs) are key components of the innate immune system in fish, responsible for recognizing pathogen-associated molecular patterns derived from bacteria, viruses, and fungi. The sterlet (Acipenser ruthenus), an endangered sturgeon species valued for its meat and caviar, is a promising model for studying the effects of polyploidy on immune gene regulation. This study examined the expression of Toll-like receptor type 2 (TLR2) and type 13 (TLR13) in the heart, liver, gills, spleen, and kidney of diploid and triploid healthy sterlets using real-time PCR. TLR2 and TLR13 were exp

Genetic composition and heterozygosity of gibberellin-induced 2n gametes evaluated by SSR markers in Populus.

BACKGROUND: The artificial induction of unreduced (2n) gametes, due to first division restitution (FDR) or second division restitution (SDR), is an important method to produce triploids in higher plants. It is crucial to evaluate the genetic composition and heterozygosity transmission of induced 2n gametes with different formation mechanisms. RESULTS: Here, we produced 110 triploids by pollinating the female inflorescence of Populus alba × P. glandulosa with gibberellin-induced 2n pollen of Populus bolleana. The genetic composition and heterozygosity of the induced 2n pollen were inferred fro

Combining ability of banana triploid hybrid progenitors and genomic prediction of cross performance for agro-morphological traits.

Breeding disease-resistant cultivars that meet commercial criteria is essential to sustain banana production threatened by major diseases. Edible bananas are seedless triploid hybrids that represent end-breeding products. Hence, the crucial step in banana breeding is to improve and combine the parents. Currently, little information is available on parental combining abilities and on the inheritance of major traits to effectively guide banana breeding strategies. In this study, a breeding population of 2,723 triploid individuals resulting from multiparental diploid-tetraploid crosses was charac
