Novel Splice Site Variant in ARFGEF2: Expanding the Phenotypic Spectrum of Periventricular Heterotopia.

Periventricular heterotopia (PH) is a rare developmental cause of intellectual disability (ID). One form is autosomal recessive PH with microcephaly (ARPHM), characterized by developmental delay and learning difficulties, with onset in infancy, plus recurrent seizures that onset in mid-adolescence. It is caused by biallelic variants in ARFGEF2. We report a consanguineous Pakistani family with three sibs afflicted with moderate/severe ID, developmental delay, muscle hypotonia, dystonia and contracture of distal limb joints, flat feet, and variable behavioral abnormalities. Single-nucleotide pol

TET loss of function selects for aneuploidies that confer a proliferative advantage.

Mutations in the Ten-Eleven Translocation (TET) family of DNA dioxygenases are among the most common epigenetic alterations in clonal hematopoiesis and hematologic malignancies, yet how TET loss of function reshapes genome evolution during cellular expansion remains to be fully understood. Here we show that loss of Tet2/3 in invariant natural killer T (iNKT) cells drives aggressive lymphoid expansion that is consistently associated with recurrent chromosome 17 trisomy, a dominant aneuploid state observed across more than ten independently derived tumors following transplantation into immunocom

Novel Strategy for Structural Variant Genotyping by Short-Read Genomic Sequencing From Restriction-Circles: Experimental and Bioinformatics Proof-of-Concept.

Structural variants (SVs) comprise copy-number variants (e.g., deletions and duplications) and copy-neutral SV (e.g., perfect inversions). Short-read high-throughput DNA-sequencing is the established technology for small-variant calling in clinical genomics, whereas Oxford nanopore or PacBio long-read NGS technologies improve SV identification but remain challenging to implement in clinical settings. We developed and validated a novel experimental protocol and bioinformatics pipeline for SV genotyping from short-read whole-genome sequencing (WGS) of restriction-circles of DNA. The experimental

Sister Chromatid Cohesion: A Multilayered Regulatory Network Beyond the Cohesin Ring.

Sister chromatid cohesion, mediated by the cohesin complex, ensures faithful chromosome segregation by providing the mechanical resistance required for bipolar spindle attachment. While the cohesin ring serves as the central executor, cohesion is regulated by a remarkably intricate multi-layered network extending far beyond the core complex. This review synthesizes current understanding of cohesion by integrating classical cohesin-centric regulation with emerging evidence implicating diverse non-cohesin factors, including DNA and RNA helicases, spliceosome components, microtubule-associated pr

Integrated Approaches in Drug Repositioning Highlight Ouabain and Helenalin as Potential Drug Candidates for Pulmonary Fibrosis.

BACKGROUND: Acute intermittent porphyria (AIP), an autosomal dominant inborn error, results from the half-normal activity of the heme biosynthetic enzyme hydroxymethylbilane synthase (EC 4.3.1.8; HMB-synthase). This disease is characterized by acute, life-threatening neurologic attacks that are precipitated by various drugs, hormones, and other factors. The enzymatic and/or biochemical diagnosis of AIP heterozygotes is problematic; therefore, efforts have focused on the identification of HMB-synthase mutations so that heterozygotes can be identified and educated to avoid the precipitating fact

All-optical microwave signal generation based on a DFB laser self-injection-locked to a fiber Fabry-Perot resonator.

Klein-Waardenburg syndrome or Waardenburg syndrome type 3 (WS-III; MIM 148820) is characterized by the presence of musculoskeletal abnormalities in association with clinical features of Waardenburg syndrome type 1 (WS-I). Since the description of the first patient in 1947 (D. Klein, Arch Klaus Stift Vererb Forsch 1947: 22: 336-342), a few cases have been reported. Only occasional families have demonstrated autosomal-dominant inheritance of WS-III. In a previous report, a missense mutation in the paired domain of the PAX3 gene has been described in a family with dominant segregation of WS-III.

Promoted Misrejoining of X-ray Radiation-induced DNA Double-Strand Breaks in Novel DNA Ligase IV-Deficient Mouse Cells.

DNA double-strand breaks (DSBs) are the most severe type of DNA damage in living organisms and are primarily repaired by two pathways: non-homologous end joining (NHEJ) and homologous recombination (HR). DNA ligase IV (LIG4) is essential for the final step of NHEJ, where it facilitates the rejoining of DSBs. Loss-of-function mutations in the LIG4 gene result in LIG4 syndrome, a condition characterized by combined immunodeficiency, developmental delay, microcephaly and radiosensitivity. In this study, we investigated cellular senescence, radiosensitivity, and X-ray radiation-induced chromosome

Biological effects of conventional and ultra high dose rate radiation in human cells.

FLASH radiotherapy (FLASH-RT) that uses an ultra-high dose rate (UHDR) radiation is emerging as an effective cancer treatment modality but the biological effects of UHDR are not fully understood. In this study, biological effects induced by conventional dose rate (CDR; 1 Gy/min) and UHDR (600 Gy/s) were evaluated in human peripheral blood lymphocytes of 10 donors at two different radiation doses (3 Gy and 8 Gy) of 9 MeV electrons. Cytogenetic analysis revealed that the unstable chromosome aberrations (dicentrics, rings and fragments) were reduced by 1.5–twofold after UHDR exposure (600 Gy/s) r

Occupational radiation exposure indicated by increased chromosomal damage in lymphocytes of orthopaedic surgeons in Japan.

This study aims to assess chromosome aberrations in peripheral blood lymphocytes of orthopaedic surgeons in Japan, specifically focusing on potential occupational dose overexposure and its correlation with adverse health reactions. The main objective is to investigate the extent of chromosomal damage and evaluate the accuracy of estimating radiation dose with cytogenetic biodosimetry where no physical dosimetry exists. This study involved 18 male orthopaedic surgeons, with occupational experience spanning 15 to 33 years. Chromosome aberrations were analyzed in 32 573 and 45 674 cells with dice

Optimized dicentric chromosome dose-response curves with centromere probes improve low-dose radiation biodosimetry.

Accurate biological dose assessment is critical in radiation emergencies, particularly in cases of low-dose exposure. The dicentric chromosome assay (DCA) is the gold standard for biodosimetry, but conventional Giemsa staining often presents challenges in identifying centromeres, especially in morphologically distorted metaphase cells. In this study, we constructed dose-response calibration curves using both Giemsa and centromere-fluorescence in situ hybridization (FISH) staining across two dose ranges (0-5 and 0-1 Gy) according to ISO 19238:2023 guidelines. The centromere-FISH-based curves de

AI-Assisted Chromosome Aberration Analysis for Rapid Biological Dose Estimation of Ionizing Radiation.

Radiation dose assessment in exposed individuals relies on the dicentric assay, the gold-standard cytogenetic biodosimeter that quantifies radiation-induced chromosomal aberrations. Although fully automated methods have been proposed, their accuracy remains suboptimal for large-scale emergency response. Here, we present a hybrid, semi-automatic framework that couples a deep-learning-based dicentric classifier with expert review. Among 2 000 metaphase images analyzed with a semi-automatic method, the specificity was 99.8%, the accuracy 98.7%, and the area under the curve (AUC) 0.977. For radiat

Identification of a Novel Pathogenic ABCC6 Mutation Through Familial Genetic Analysis in Pseudoxanthoma Elasticum: A Case Report.

This study conducted genetic analysis on a patient clinically diagnosed with pseudoxanthoma elasticum (PXE) and their family to identify pathogenic mutations in the ABCC6 gene and analyze its inheritance pattern. We performed whole-exome sequencing of the ABCC6 gene on peripheral blood samples from the proband, his parents, and his sister, with validation via Sanger sequencing. Results revealed two heterozygous mutations in the proband: a previously reported missense mutation c.3412C>T (p. Arg1138Trp) and an unreported frameshift deletion mutation c.3160_3161del (p. Thr1054Glyfs*2). Both varia

Chromosome 3p deletion leads to extensive genomic alterations in diverse cancers and confers synthetic lethality in uveal melanoma.

Chromosome 3p (chr3p) is frequently deleted in multiple cancers, indicating the presence of shared tumor suppressors. Analysis of genomic alterations in 33 different cancer types implicates the deletion or deleterious mutations of SET-domain-containing 2 (SETD2) at chr3p21 in significantly facilitating the formation of isochromosomes, consisting of two identical mirror-imaged arms, thereby promoting genomic instability conducive to large-scale chromosomal rearrangements and rapid cancer genome evolution. Fracturing of dicentric isochromosomes during cell division is pervasive and follows the d

Clinical and molecular characteristics of constitutional mismatch repair deficiency syndrome: a case series of five children and appraisal of diagnostic guidelines.

DNA mismatch repair (MMR) is critical for maintaining genome integrity through correction of single-base mismatches and insertion-deletion loops arising from DNA replication. Heterozygous germline alteration of MMR genes (MSH2, MSH6, MLH1, PMS2) cause autosomal dominant Lynch syndrome (LS), most commonly manifesting as colonic or endometrial cancers, although brain, ovarian, and other organ systems may be involved. Neoplasia in LS usually arises after the age of 30 years. Constitutional mismatch repair deficiency (CMMRD) is inherited in an autosomal recessive manner due to biallelic germline a

A polymer physics model of the interphase cell nucleus for radiobiological simulations.

Advances in chromatin architectures achieved through super-resolution imaging and high-throughput chromosome conformation capture (Hi-C) techniques remain to be integrated into the modeling framework of radiobiology. In this study, the chromosomal and chromatin interactions in an interphase cell nucleus were described by polymer physics principles. To overcome the prohibitive computational cost, a multi-stage relaxation strategy was employed to decouple the relaxation processes of different structural levels. Cystinuria in an Australian Cattle Dog Family-A Seemingly Androgen-Associated Autosomal Dominant Trait.

In Australian Cattle Dogs (AUCDs), cystinuria was reported to be an autosomal dominant trait caused by a 6 bp deletion in the SLC3A1 gene (type II-A). Here we report an androgen association in this breed. A family of 11 adult AUCDs (five intact and one neutered male and five females) was genotyped for the SLC3A1 c.1095_1100del variant, and urine was examined for concentrations of cystine, ornithine, lysine, and arginine (COLA). Among this family, three males and five females tested heterozygous for the mutation, while all other AUCDs were homozygous for the wild-type allele. The two heterozygo
