TET loss of function selects for aneuploidies that confer a proliferative advantage.

Mutations in the Ten-Eleven Translocation (TET) family of DNA dioxygenases are among the most common epigenetic alterations in clonal hematopoiesis and hematologic malignancies, yet how TET loss of function reshapes genome evolution during cellular expansion remains to be fully understood. Here we show that loss of Tet2/3 in invariant natural killer T (iNKT) cells drives aggressive lymphoid expansion that is consistently associated with recurrent chromosome 17 trisomy, a dominant aneuploid state observed across more than ten independently derived tumors following transplantation into immunocom

Noncarrier mothers of hemophilia A patients with Intron 22 inversions often have other rearrangements.

BACKGROUND: Inversions involving intron 22 (Inv22) of the F8 gene are detected in approximately 45% of all severe hemophilia A (HA) patients. Digital droplet polymerase chain reaction using milepost assays and multiplex ligation-dependent probe amplification (MLPA) allows robust diagnosis of inversions, copy number variations, and more complex rearrangements in F8. OBJECTIVES: A total of 350 HA patients and 280 of their mothers from the Hemophilia Inhibitor Genetics Study cohort were analyzed to identify additional F8 mutations. METHODS: Digital droplet polymerase chain reaction and MLPA wer

Genetic landscape of male infertility: chromosomal abnormalities and Y chromosome microdeletions in a Turkish cohort (1 314 Cases).

Male infertility affects a significant portion of the global population and can arise from genetic and physiological abnormalities. This study assessed the clinical and genetic characteristics of 1 314 infertile males. Parameters included family history, age, marital duration, presence of physical conditions, clinical diagnoses (oligospermia and azoospermia), cytogenetic and Y-chromosome microdeletion analysis, and other laboratory data. The mean age was 31.37 years, with an average marital duration of 4.6 years. Oligospermia was observed in 601 patients, while 713 had azoospermia. Chromosomal

AURKB-driven dissolution of CIZ1-RNA assemblies from the inactive X chromosome in mitosis.

Cip1-interacting zinc-finger protein 1 (CIZ1) interacts with Xist lncRNA to form large RNA-protein assemblies at the inactive X-chromosome (Xi) in female mammalian nuclei, plus smaller assemblies in both sexes. CIZ1 assemblies influence underlying chromatin, and their disruption alters the expression of autosomal and X-linked gene clusters. Here, we explore the regulated dissolution of CIZ1-Xi assemblies during mitosis and show that, like Xist, CIZ1 is released in prometaphase under the regulation of Aurora Kinase B (AURKB). The part of human/mouse CIZ1 comprising 179/181 C-terminal amino acid

Quantifying rate-limiting genetic variation in breast and ovarian tumourigenesis.

BACKGROUND: The number and type of genetic alterations required to initiate breast and ovarian cancer remain unclear. While germline BRCA1/2 carriers show markedly elevated cancer risk, it is uncertain whether point mutations or copy number alterations constitute the rate-limiting events of tumourigenesis. METHODS: We developed a statistical framework extending prior incidence-mutation models to estimate the minimal number and type of driver events required for cancer initiation. Somatic mutation and copy-number data from >3000 breast and ovarian cancers in TCGA and METABRIC were compared bet

The Helicobacter pylori ribosomal silencing factor RsfS is required for low-growth states and chronic infection.

UNLABELLED: Helicobacter pylori is a prevalent bacterial pathogen that chronically colonizes the human gastric epithelium, but the bacterium's physiological mechanisms that promote this are understudied. Dormancy and low growth are known to facilitate other microbial chronic infections. A critical feature of low growth states is the down regulation of ribosome translational activity via regulation factors. The H. pylori genome is predicted to encode only one ribosome regulation factor, called RsfS (Ribosomal Silencing Factor S). In other bacterial species, RsfS prevents ribosome assembly by bi

Clinical diagnostic utility of MGMT promoter methylation.

Tumor suppressor genes and DNA repair genes can be inactivated not only by genetic lesions, like mutations or deletions, but also by aberrant DNA methylation of promoter regions. These DNA methylation alterations may serve as biomarkers for early detection, refinement of diagnosis, prognosis, or prediction of therapy response. Despite a long history of DNA methylation research in the field of oncology so far only very few aberrant DNA methylation events are of proven and generally accepted clinical value and are part of the routine diagnostic work‑up of cancer patient samples. Hypermethylation

Keeping up the beat of Kleefstra syndrome.

Long QT syndrome (LQTS) is a rare and potentially life-threatening arrhythmia characterized by delayed repolarization on electrocardiogram (EKG) evaluation. Although LQTS is primarily associated with defects in ion channels, in approximately 20% of cases the genetic cause remains unknown. Kleefstra syndrome (KS), a rare neurodevelopmental disorder caused by alterations in the EHMT1 gene, has been associated with various cardiac abnormalities, including structural defects and arrhythmias. Here we report the second case of LQT in a patient with KS, strengthening the association between these two

Live Birth Following Microdissection Testicular Sperm Extraction and Intracytoplasmic Sperm Injection: A Case Study of a Patient with Non-Obstructive Azoospermia and a Y-chromosome Microdeletion (Ym9; P3, Including AZFb, c Region).

The azoospermia factor (AZF) region on the long arm of the Y chromosome is instrumental in spermatogenesis. In particular, complete deletion of the AZFa, AZFb, and AZFb + c regions is not an indication for microdissection testicular sperm extraction (MD-TESE) as there is no chance of sperm retrieval. This report presents a rare case of a successful live birth, following intracytoplasmic sperm injection (ICSI) using sperm from a patient with a microdeletion of the Y chromosome, including part of the AZFb, c region, who underwent MD-TESE. A 3X-year-old Japanese man with azoospermia was admitted

Genomic spectrum of congenital heart disease combined with kidney and urinary tract anomalies uncovered by exome sequencing and array-CGH.

Congenital heart disease (CHD) combined with anomalies of the kidney and urinary tract (CAKUT) belongs to severe clinical conditions that represent isolated cases or can be part of complex inherited syndromes. To date, knowledge of the genetic basis of cardio-renal birth defects is very limited. Using chromosomal microarray analysis and exome sequencing, we investigated the spectrum of copy number variations (CNVs) and point genetic variants in a pediatric cohort of 30 patients presenting with combined CHD and CAKUT. Two patients possessed a pathogenic deletion in the 22q11.2 genomic region, w

Congenital Clitoromegaly With Concurrent Congenital Adrenal Hyperplasia and Turner Syndrome: A Case Report.

Clitoromegaly in a newborn with otherwise typically appearing female genitalia indicates exposure to excess androgens in fetal life. Congenital Adrenal Hyperplasia (CAH) is the most common cause of virilization at birth and empiric treatment is often pursued due to the risk of life-threatening adrenal crisis. Turner syndrome (TS) does not usually present with atypical genital appearance or other signs of hyperandrogenism, unless Y chromosome material is present. In this report, we describe a newborn who was noted to have clitoromegaly soon after birth with an otherwise normal exam and no syndr

Understanding histotripsy ablation safety: a systematic review of pre-clinical and clinical adverse events.

BACKGROUND: Facet joint tropism is asymmetry in orientation of the bilateral facets. Some studies have shown that tropism may increase the risk of disc degeneration and herniations, as well as degenerative spondylolisthesis (DS). It remains controversial whether tropism is a pre-existing developmental phenomena or secondary to progressive remodeling of the joint structure due to degenerative changes. As such, the following study addressed the occurrence of tropism of the lower lumbar spine (i.e. L3-S1) in a degenerative spondylolisthesis patient model. METHODS: An international, multi-center

Research progress of SPP1 in cancer regulation: Tumorigenesis, metastasis, tumor microenvironment, immunity and therapeutic targets (Review).

OBJECTIVES: Psoriatic arthritis (PsA) has been defined as a systemic, chronic, inflammatory arthritis, usually seronegative for rheumatoid factor (RF), associated with cutaneous psoriasis. The exact prevalence of PsA is unknown and its estimation has been difficult, partly due to the lack of a widely accepted classification criteria. Agreed and validated criteria will facilitate comparison between centres and different countries in the areas of epidemiology, outcome studies and therapeutic trials. A number of classification criteria have been published by Moll & Wright (M & W), Bennett's, Vase

Exophytic left ventricular angiomyxoma.

BACKGROUND: Low muscle reserve combined with excess adiposity is clinically relevant in diabetes, but whether laboratory-based nutritional risk scores differ in their associations with this phenotype remains unclear. METHODS: This cross-sectional NHANES 2011-2018 analysis included U.S. adults aged 20-59 years with diabetes. The primary outcome was a DXA-defined low-lean-mass obesity phenotype, defined as low appendicular lean mass adjusted for body mass index combined with BMI-defined obesity. Genomic Characterization of Chordoma: Insights from the AACR Project GENIE Database.

Background: Chordoma is a rare primary tumor originating from embryonic notochord remnants, with limited systemic therapeutic options due to a poor understanding of its genomic landscape. This study aims to characterize the genetic alterations in chordoma using a large national patient-level genomic repository, the AACR Project GENIE, to identify potential therapeutic targets and improve disease modeling. Methods: A retrospective analysis of chordoma samples was conducted using the AACR Project GENIE database. Targeted sequencing data were analyzed for recurrent somatic mutations, tumor mutati

Shank3 modulates Rpl3 expression and protein synthesis via mGlu5: implications for Phelan McDermid syndrome.

Mutations or deletions in the SHANK3 gene have been identified in up to 1% of autism spectrum disorder cases and are considered the primary cause of neuropsychiatric symptoms in Phelan McDermid syndrome (PMS). While synaptic dysfunctions have been extensively documented in the absence of Shank3, other mechanisms through which Shank3 may regulate neuronal functions remain unclear. In this study, we report that the ribosomal protein Rpl3 and overall protein synthesis are downregulated in the cortex and striatum of Shank3 knockout (KO) mice and in neurons differentiated from human-induced pluripo

Factors that influence the caste ratio in a bacterial division of labour.

Colonies of the bacterim Streptomyces coelicolor divide labour between cells that specialize in growth and sporulation and cells that specialize in antibiotic production. This division of labour arises owing to costly chromosome deletions in the antibiotic overproducers. However, the spatial distribution and temporal emergence of these mutations in S. coelicolor colonies remain unknown, or whether mutation frequency-which we liken to the caste ratio in social insects-is phenotypically plastic. To elucidate changes in the proportions of specialized cells (measured as the mutation frequency), we

A review of antipsychotic therapy effectiveness and tolerability among individuals with copy number variants relevant to schizophrenia.

Although numerous copy number variants (CNVs) are considered pathogenic with respect to the development of schizophrenia, only eight loci have reached genome-wide significance. Reviews/studies characterizing antipsychotic use in this context exist for only three corresponding CNV syndromes. As these disorders also predispose to neurodevelopmental anomalies and various medical comorbidities, affected individuals may be particularly sensitive to the side effects of antipsychotic medications. As such, this review sought to identify and describe all reports of antipsychotic use among individuals w

Contrasting interferon-mediated antiviral responses in human lung adenocarcinoma cells.

UNLABELLED: Lung cancers develop from lung epithelial cells after a series of genetic and epigenetic changes, and these cells are major sites of influenza virus infection. Thus, we explored how changes found in patient-derived lung cancer cell lines impacted influenza virus replication and identified two lines with opposite responses to influenza A viral infection. We show that the NCI-H820 lung adenocarcinoma (LUAD) is resistant to influenza A virus and VSV infection, while LUAD line NCI-H322 is highly susceptible to infection by both viruses. H322 cells have a homozygous deletion in a region
