Sheep fascioliasis causes considerable economic losses globally, yet risk factors for infection in Andes Highlands remain poorly characterized. We conducted a cross-sectional study to determine the risk factors of sheep fascioliasis in rural communities of the Southern highlands of Peru. Data on farm and animal characteristics, management practices, and water sources were collected. Stool samples were geographically tagged and examined microscopically for Fasciola and other parasites eggs. A total of 3,518 sheep from 268 farms were evaluated, of which 40.6% (1,429/3,518) were infected with F.

BACKGROUND: Fascioliasis is a zoonotic trematode infection caused by members of the genus Fasciola (F. hepatica and F. gigantica) that localize to the hepatic biliary ducts in humans. Parasites are acquired after ingestion of freshwater plants, and fascioliasis has been reported on every inhabited continent. Typically, fascioliasis is diagnosed by serology or stool examination. This case reports the unusual finding of adult Fasciola organisms during an endoscopic retrograde cholangiopancreatography (ERCP).
CASE SUMMARY: A 65-year-old woman who returned to the United States after traveling in

BACKGROUND: Iran is one of the most important foci of human fascioliasis. West Azerbaijan Province is known for its traditional rural livestock husbandry, supported by many rivers. This environment could facilitate the transmission of fascioliasis between animals and humans. Thus, the present study investigated the seroprevalence of fascioliasis in patients referred to the Reference Laboratory in Urmia, Northwest Iran.
MATERIALS AND METHODS: In this cross-sectional study, 919 sera were collected from patients referred to the Reference Laboratory of Urmia University of Medical Sciences from Ja

INTRODUCTION: Climate change impacts oceanic and atmospheric circulation patterns, precipitation, air and sea surface temperatures leading to altered intermediate host vector distribution, reproduction, and maturation. With temperatures predicted to rise by 1.5°C or more, understanding the influence of environmental characteristics and climatic conditions on distribution of intermediate host vectors, occurrence and habitat suitability is vital in vector borne diseases and their control policies. Hence, the study focused on the predicted changes in habitat suitability of Pseudosuccinea columell

Neorickettsia are obligate intracellular bacteria that persist as endosymbionts of digenean trematodes and include several vertebrate pathogens. However, many digenean-associated lineages remain poorly characterized, including Neorickettsia associated with the liver fluke Fasciola hepatica (nFh). Here, we present a large-scale population genomic analysis of nFh to assess its prevalence, genetic diversity, evolutionary history, and transmission dynamics. We screened whole-genome sequencing data from 502 F. hepatica samples collected across Peru, the UK, Uruguay, and the USA, detecting Neoricket

Fasciola gigantica, the predominant cause of fasciolosis in tropical regions, is a major cause of economic loss to the global farming community and is also recognised as a significant human pathogen. The surface tegument of the fluke, situated as it is at the host-parasite interface, is a metabolically active and highly dynamic secretory organ mainly concerned with ensuring the survival of the parasite in a hostile immunological and physiochemical environment. The period of migration of the juvenile fluke through the hepatic parenchyma of the host is critical in establishing a mutually sustain

BACKGROUND: Fascioliasis is a zoonosis and neglected tropical disease with worldwide distribution and significant economic impact. The search for anthelmintic compounds against Fasciola hepatica, one of the parasitic liver flukes responsible for this disease, has become important due to widespread resistances against existing drugs. To facilitate drug discovery, a useful strategy is the identification of proteins that are vital for the parasite. Protein kinases (PKs) emerged as potential targets in several parasites, given their critical role in many biological processes. To date, knowledge of

Fasciola hepatica, a food-borne trematode that parasitizes the liver and bile ducts of cattle, sheep, other ruminants, and humans, is a critical zoonotic disease responsible for substantial hepatic pathology. Kupffer cells (KCs), the resident macrophages of the liver, act as the first line of defense against liver damage. Investigating the gene transcription changes in KCs during F. hepatica infection is essential for identifying new therapeutic targets and improving disease intervention strategies. A sheep model was experimentally infected with F. hepatica to obtain adult flukes, from which e

We assessed the prevalence of three helminthic zoonoses-echinococcosis, fasciolosis and the taeniosis/cysticercosis complex-among residents of the Chucuito Health Network (Puno Health Region, Peru) over four years (2018-2021). Sera (n = 910) were analysed by ELISA to detect pathogen-specific antibodies, following national protocols. Echinococcosis predominated, whereas fasciolosis and taeniosis/cysticercosis occurred at comparatively low levels. Prevalence ranged from 4.4-9.2% for echinococcosis, 1.1-4.9% for fasciolosis, and 1.1-2.7% for taeniosis/cysticercosis across the four years. Prevalen

AIMS: To investigate potential resistance to anthelmintics containing triclabendazole in naturally acquired Fasciola hepatica (liver fluke) present on a sheep and beef farm in New Zealand.
MATERIALS AND METHODS: A necropsy study was conducted with three groups of mixed-age Romney ewes naturally infected with F. hepatica. Ewes were either left untreated as controls or treated with a single oral dose of either a combination of 0.2 mg/kg moxidectin and 10 mg/kg triclabendazole or 10 mg/kg triclabendazole alone, both at the manufacturer's recommended dose. At necropsy, 49 days later, livers were

Water buffalo are ideal for grazing in restored wetlands due to their anatomical and behavioural adaptations. Unlike cattle or sheep, the water buffalo thrive in such habitats, feeding on rushes and reeds while helping maintain open landscapes. However, these moist environments also harbor risks, particularly parasitic infections by liver flukes (Fasciola hepatica) and the recently widespread rumen flukes, which require aquatic snails as intermediate hosts. For the present study faecal samples were taken once a year from 2016 to 2024 and monthly in two 12-month periods (2019/20 and 2022/23) fr

Hepatic fasciolosis represents a major health and production concern in sheep farming. Although ultrasonography has been used to evaluate fasciolosis in ruminants, the feasibility of a rapid ultrasonographic protocol performed in standing sheep under field conditions has not been specifically described. This study aimed to provide a preliminary, descriptive evaluation of the feasibility of a fast on-farm ultrasonographic protocol for the assessment of hepatic alterations in sheep with suspected chronic fasciolosis. Sixteen Sarda sheep with Fasciola hepatica fecal eggs count > 100 eggs per gram

Fascioliasis is a parasitic liver disease of mammals induced by liver flukes, Fasciola hepatica and Fasciola gigantica. Fasciola spp. rely on their definitive hosts, ruminants, and intermediate hosts, snails, to survive and can incidentally infect humans as definitive hosts. Ruminant (goat, sheep, and cattle) liver and fecal samples were collected from the Kharaa River Basin (KRB) of Mongolia during 2018-2020. A total of 807 adult liver flukes were found in livers of 18 goats, 21 sheep, and 1 cattle, with morphological identification of F. hepatica species. A total of 350 fecal samples selecte

Parthenogenic Fasciola (Trematoda: Fasciolidae) flukes have been developed by the hybridization of Fasciola hepatica and Fasciola gigantica. They are aspermic (asF) but capable of clonal expansion through parthenogenesis and are spreading rapidly throughout the globe. Here, we unambiguously prove the occurrence of parthenogenic Fasciola in cattle in Bangladesh, along with their ex vivo culture protocol and anthelmintic efficacy. By employing multiple conventional and molecular tools, we confirmed the presence of both the spermic F. gigantica (sFg) (31.7%; 814/2575) and asF (68.3%; 1761/2575) i

BACKGROUND: Fascioliasis is a neglected infectious disease affecting agricultural communities worldwide, with the Peruvian Andes among the most severely affected regions. Identifying fine-scale environmental risk patterns could support targeted surveillance and control. We aimed to develop predictive models of Fasciola hepatica infection in humans and sheep using drone-derived environmental indices in a rural Andean community.
METHODS: We conducted a cross-sectional study in the Huayllapata community, Cusco, Peru. Demographic, socioeconomic, and georeferenced infection data were collected fro

Infection with Fasciola hepatica (liver flukes) causes fascioliasis in humans and fasciolosis in ruminants. The current strategy for controlling fascioliasis/fasciolosis is predominantly mediated by chemotherapy with triclabendazole (TCBZ). As this flukicide targets newly excysted juveniles (NEJs), immature flukes and mature liver flukes, it's use as the frontline chemotherapy for over four decades has led to widespread drug resistance. New chemotherapeutics are, therefore, urgently required. Here, continuing our studies of flukicidal phytochemicals derived from Hedera helix (common ivy), we i

BACKGROUND: Trematode parasites, or flukes, are a significant economic threat to ruminant production worldwide. Traditional diagnostic methods rely on egg sedimentation from faeces, a time-consuming methodology lacking sensitivity and specificity. This study aimed to develop and validate two detection methods: firstly, qPCR for accurate identification of Fasciola spp., and secondly, a deep amplicon sequencing technique for identifying fluke species using faecal sedimented egg DNA.
METHODOLOGY: To detect Fasciola spp., infection, primers targeting mitochondrial DNA were repurposed to develop a
