Fasciola hepatica in donkeys from southern Texas, USA.

Fasciola hepatica is a trematode parasite with a global distribution found in various mammalian hosts, most notably ruminants. It is associated with hepatic lesions including necrosis and inflammation, as well as can increase the risk of secondary bacterial infections. There is a lack of reports of fasciolosis in equids in North America. This report documents the pre- and postmortem findings of two cases of chronic fasciolosis in donkeys from southern Texas, United States. Grossly, left hepatic lobe atrophy and concurrent right lobe hypertrophy was observed. Histologic examination of the liver

Distinct gut microbiome features characterize Fasciola hepatica infection and predict triclabendazole treatment outcomes in Peruvian patients.

BACKGROUND: Fasciola hepatica, a globally distributed helminth, causes fasciolosis, a disease with significant health and economic impacts. Variability in triclabendazole (TCBZ) efficacy and emerging resistance are remaining challenges. Evidence suggests that the gut microbiome influences host-helminth interactions and is associated with anthelmintic effects, but its association with human F. hepatica infection and TCBZ efficacy is not well understood. METHODS: In this study, we investigated the relationship between Fasciola hepatica infection and the gut microbiome through metagenomic shotgu

In vitro co-culture of Fasciola hepatica newly excysted juveniles (NEJs) with 3D HepG2 spheroids permits novel investigation of host-parasite interactions.

Fasciola hepatica, or liver fluke, causes fasciolosis in humans and livestock. Following ingestion of vegetation contaminated with encysted parasites, metacercariae, newly excysted juveniles (NEJ) excyst in the small intestine and cross the intestinal wall. After penetrating the liver, the parasite begins an intra-parenchymal migratory and feeding phase that not only drives their rapid growth and development but also causes extensive haemorrhaging and immune pathology. Studies on infection are hindered by the difficulty in accessing these microscopic juvenile parasites in vivo. Thus, a simple

Exploring the impacts of El Niño-Southern Oscillation (ENSO) and North Atlantic Oscillation (NAO) on the spatial distribution of Fasciola spp. in the Gilan province, Iran.

This study investigated the correlation between large-scale climate phenomena, particularly El Niño-Southern Oscillation (ENSO) and North Atlantic Oscillation (NAO), and the distribution of Fasciola species in Gilan province, Iran. Climate change significantly impacts animal health, particularly through vector-borne diseases. Fasciola spp. is highly sensitive to temperature and soil moisture conditions, making its relationship with climate complex and influenced by various meteorological variables. ENSO and NAO index data were obtained from National Oceanic and Atmospheric Administration (NOAA

Independent origins and non-parallel selection signatures of triclabendazole resistance in Fasciola hepatica.

Triclabendazole (TCBZ) is the primary treatment for fascioliasis, a global foodborne zoonosis caused by Fasciola hepatica. Widespread resistance to TCBZ (TCBZ-R) in livestock and a rapid rise in resistant human infections are significant concerns. To understand the genetic basis of TCBZ-R, we sequenced the genomes of 99 TCBZ-sensitive (TCBZ-S) and 210 TCBZ-R adult flukes from 146 bovine livers in Cusco, Peru. We identify genomic regions of high differentiation (FST outliers above the 99.9th percentile) that encod genes involved in the EGFR-PI3K-mTOR-S6K pathway and microtubule function. Transc

γδ T cell distribution in the adventitial layer of non-fertile cystic echinococcosis cysts from cattle livers.

Cystic Echinococcosis (CE) is a zoonotic disease caused by Echinococcus granulosus sensu lato, forming cysts in ruminants and humans with major health and economic impacts. The immune response to CE cysts is complex, with fertility linked to the host's inflammatory reaction. This study examines γδ T cell distribution and role within the adventitial layer of non-fertile CE cysts in cattle, including cases co-infected with the trematode Fasciola hepatica (FH), a known immune response modulator. Using immunohistochemistry and double immunofluorescence, we observed γδ T cells dispersed in the adve

Genetic diversity and population structure of Fasciola gigantica isolated from cattle in Malawi.

Fasciola gigantica is an important trematode that affects the health of animals and humans in tropical and subtropical countries, including Malawi. Information on the genetic diversity and population structure of F. gigantica is important to understanding the parasite`s transmission patterns/ and in monitoring the development of resistance to commonly used anthelmintic agents. This study aimed to analyze the genetic diversity and population structure of Fasciola species collected from cattle at slaughter slabs and abattoirs in selected districts of Malawi. A total of 27 adult liver flukes were

The effects of triclabendazole, combined tetramisole with Oxyclozanide, and albendazole against ovine fasciolosis.

Recent estimates suggest that liver condemnation due to liver fluke in Ethiopia causes an annual economic loss of about 48.4 million Ethiopian birr. For approximately ten decades, the treatment of fasciolosis has relied solely on triclabendazole (TCBZ). In many clinical practices, combined tetramisole with Oxyclozanide and albendazole drugs have been used as treatment options for ovine fasciolosis. Therefore, this study aimed to compare the effectiveness of triclabendazole (TCBZ), combined tetramisole with Oxyclozanide (TETRA), and albendazole (ALBE) for the treatment of ovine fasciolosis and

Factors contributing to the abundance and spatial distribution of the invasive intermediate host snail (Pseudosuccinea columella) in uMgungundlovu district, KwaZulu-Natal, South Africa.

Fascioliasis is a parasitic disease commonly affecting cattle, goats, and sheep globally. Lymnaeidae snail species are important in the epidemiology and dispersal of fascioliasis since they are intermediate hosts of the Fasciola spp. Our study mapped at micro-geographical scale, the distribution and abundance of Pseudosuccinea columella in uMgungundlovu district, which is in the northern part of the KwaZulu-Natal province, and measured physicochemical parameters at potential transmission sites. The study examined the impact of physicochemical parameters and presence of other snail species on P

The milk production impacts of liver fluke (Fasciola hepatica) infection in dairy cows on the West Coast of the South Island, New Zealand.

The impact of fasciolosis has been estimated on many occasions and can vary from mild to severe effects on animal productivity. The aim of the current study was to utilise observations on seroconversion of dairy cattle to Fasciola hepatica over the course of a lactation and compare their milk production with other cattle who remained serologically negative throughout the same lactation. Four spring-calving dairy herds on the West Coast of the South Island of New Zealand were selected based on prior knowledge of endemic liver fluke infection. Over the four herds, a total of 485 cows were blood

Transformer-based deep learning enables improved B-cell epitope prediction in parasitic pathogens: A proof-of-concept study on Fasciola hepatica.

BACKGROUND: The identification of B-cell epitopes (BCEs) is fundamental to advancing epitope-based vaccine design, therapeutic antibody development, and diagnostics, such as in neglected tropical diseases caused by parasitic pathogens. However, the structural complexity of parasite antigens and the high cost of experimental validation present certain challenges. Advances in Artificial Intelligence (AI)-driven protein engineering, particularly through machine learning and deep learning, offer efficient solutions to enhance prediction accuracy and reduce experimental costs. Infection mechanisms of Pochonia chlamydosporia: A breakthrough in Fasciola hepatica egg control.

Traditional parasite management has long been plagued by issues such as drug resistance and environmental pollution. Biological control using Pochonia chlamydosporia has emerged as a sustainable alternative, yet the underlying infection mechanisms remain elusive. This study aimed to comprehensively elucidate these mechanisms, with a particular focus on the role of gene editing. Employing optical, SEM, and TEM microscopy, we observed that P. chlamydosporia infects Fasciola hepatica eggs in three distinct stages. TEM analysis first visualized unique infection pegs crucial for initial eggshell pe

Evaluation of a novel vaccine candidate derived from newly excysted juveniles of Fasciola hepatica in sheep.

Vaccine approaches for controlling Fasciola hepatica present a promising avenue, particularly considering increasing resistance to anthelmintic treatments and concerns over chemical residues. Targeting vaccine candidates that are expressed and secreted during the early infective stage of F. hepatica could offer an effective alternative. This approach aims to inhibit the invasion and migration of juvenile parasites, which have not yet fully developed their immune evasion mechanisms, thereby preventing parasite establishment and development in the host. In this study, we evaluated the host immun

Construction, expression, and characterization of scFv fragment against Fasciola gigantica cathepsin L1H.

Fasciola spp. infection is a significant zoonotic disease. Fasciola gigantica cathepsin L1H (FgCathL1H) is expressed across the life stages of Fasciola gigantica: newly excysted juvenile (NEJ), juvenile, and adult. An emerging tool for diagnosing fasciolosis in humans and cattle involves single-chain variable fragments (scFv) antibodies. These antibodies, consisting of linked variable regions of heavy chains (VHs) and light chains (VLs), retain binding specificity and affinity. This study aims to construct, express, and characterize an scFv antibody for use in a diagnostic kit for fasciolosis.

Microinvertebrate consumption rates of Fasciola hepatica miracidia are not affected by alternate food.

The liver fluke (Fasciola hepatica) is a significant parasite of the global livestock industry, leading to negative economic and animal welfare impacts. Control of F. hepatica is becoming increasingly difficult as many liver fluke populations are developing resistance to commonly used anthelmintics. Additional or alternate control methods are, therefore, required. Microinvertebrates such as those of the order Cyclopoida and subclass Ostracoda are common organisms found in the same aquatic habitats as F. hepatica's intermediate snail host. We explore whether these microinvertebrates are effecti

Deciphering the Proteomic Profile of the Parasite Fasciola hepatica After Initial Vertebrate Host Infection Reveals New Insights Into Its Early Stages.

The migration of the trematode parasite Fasciola hepatica within its vertebrate host following infection by ingestion of the metacercariae represents a critical event in its establishment and survival. The early stages of infection, during which F. hepatica crosses the intestinal barrier and advances to the liver through the peritoneum, initiate changes in the parasite that drive its development from a free-living state on pasture to an obligate blood-feeding parasite. Using an in vivo mouse model, this study explores the proteomic changes in the parasite as it crosses the intestinal barrier a
