[Duplication of the long arm of chromosome 3 (dup 3q) in a newborn infant whose the father is carrier of pericentric inversion of chromosome 9].

The authors report a case of a partial trisomy for the long arm of chromosome 3. The associated morphotypic anomalies are compared with the 31 cases already reported, and allow to further outline a well recognizable morphotype which has similarities with the Brachmann Cornelia de Lange Syndrome. The chromosomal mechanisms which may result in this unbalanced caryotype dup 3 q are discussed, and among them the possible role of a paternal chromosome 9 pericentric inversion, most commonly termed as a normal variant.

[Prenatal diagnoses in a family with pericentric inversion of chromosome no. 5].

A hereditary pericentric inversion of chromosome 5(p13 leads to q35) was detected in a family after the birth of a child with Cri-du-Chat-syndrome [46,XY,del(5)(p13)]. Prenatal diagnoses were carried out in three pregnancies in this family. The following results were found in the amniotic fluid cells: first pregnancy 46,XX; second 46,XY, inv(5)(p13 leads to q35) and the third 46,XX,der (5)(pter leads to q35::p13 leads to pter). The first two pregnancies ended with the birth of phenotypically normal children; the third one however was interrupted. Fetal kidney tissue cultures confirmed the resu

Familial paracentric inversion of 1p.

We report a paracentric inversion of 1p in a boy with mild mental retardation. The chromosome aberration was identified by high resolution chromosome banding, and was also present in his phenotypically normal mother and other relatives. The boy's karyotype was considered to be 46,XY,inv(1) (p31,2p36.22) ISCN (1981).

The origin of inverted tandem duplications, and phenotypic effects of tandem duplication of the X chromosome long arm.

Tandem repeats of chromosome material can arise as inverted or as direct duplications. Such duplications of the X chromosome are instructive regarding X-linked genetic determinants of phenotype. We describe a 40-year-old woman with a direct duplication Xq13.3 to Xq27.2, short stature, gonadal dysgenesis, and secondary amenorrhea. Comparison of her phenotype with that of two other women with a direct duplication of part of Xq confirms the existence of statural determinants within the region X13 to Xq21, determinants of ovarian function within X22 to X27, and the X inactivation center within or

Constitutive heterochromatin of chromosomes No. 1, 9, and 16 in 90 patients with malignant disease and 91 controls.

The variability of constitutive heterochromatin in chromosomes #1, #9, and #16 was investigated in 90 patients with malignant tumors (neoplasms of the head, neck, breast, and genital organs) and in 91 controls. The investigation was carried out on chromosome preparations from peripheral blood cultures. An analysis showed no significant difference in the frequency of localization variants, that is, inversions, in the patients and in the normal controls. Analysis of the variability of the C-segment size was made by measuring the length of heterochromatin blocks and showed that a correlation with

Duplication 9q34 syndrome.

Phenotypic, karyotypic, and developmental homology between affected children of carriers of an inverted insertion (9) (q22.1q34.3q34.1) led to recognition of a new chromosome syndrome: dup 9q34. Individuals with dup 9q34 have slight psychomotor retardation, understand simple directions, and acquire a limited vocabulary. In childhood, many are hyperactive. Clinical features include low birth weight, normal birth length, and initial poor feeding and thriving. Musculo-skeletal systems are affected: there are joint contractures, long thin limbs, and striking arachnodactyly. There is abnormal impla

Unusual chromosome aberrations in 3 children with Down syndrome.

In 3 children with Down syndrome extremely rare chromosome aberrations were found. In the first patient, the karyotype showed 46 chromosomes with a de novo duplication of the q22-qter segment. This finding supports that the 21q22-qter band was responsible for the characteristic mongoloid features. In the second case, trisomy 21 was present and out of 78 investigated cells, 60 contained a small, supernumerary marker chromosome in addition to trisomy 21. The parents were cytogenetically and clinically normal. In the third case trisomy 21 with inv(10) (p13q22) occurred. The inversion was inherite

Pericentric X inversion in dizygotic twins who differ in X chromosome inactivation and menstrual cycle function.

A 21-year-old female dizygotic twin was referred for cytogenetic evaluation because of mild mental retardation. Significant history, clinical, and physical findings included irregular menses, mildly coarse facies, and microcornea. Chromosome analysis revealed a pericentric inversion of the X chromosome, 46, X, inv (X)(p11;q22). Her twin who is phenotypically normal was also found to carry the same inversion. The twins differ significantly in X chromosome inactivation and menstrual cycle function.

Human chromosome 2 rod/ring mosaicism: probable origin by prezygotic breakage and intrachromosomal exchange.

A 5-year-old male with mild mental retardation showed a chromosomal rearrangement involving duplication of part of 2q (2q33.3 leads to 2wter) in 70% of metaphases from peripheral blood; the remaining 30% of cells had a rearrangement of chromosome 2 in the form of a ring, viz., r(2)(2p25.2 leads to 2q33.2). Both configurations appeared to be missing a tiny portion of 2p (p25.3 leads to pter). All metaphases examined from cultured skin fibroblasts from the child had the abnormal rod configuration of chromosome 2; none had the ring. The pattern of the 2q duplication/2p deletion in the rod is that

Tiny interstitial duplication of proximal 7q in association with a maternal paracentric inversion.

Paracentric inversion of chromosome 7 was found in a female infant with multiple malformations and in her phenotypically normal mother. Examination of prometaphase chromosomes revealed an additional small dark band on the inverted chromosome 7 of the girl. It was assumed that an unequal crossing over at the base of a meiotic loop of chromosome 7 had occurred in the mother and resulted in a tiny interstitial duplication in the girl.

Recombinant inversion chromosomes in phenotypically normal chickens.

Some progeny resulting from interbreeding of individuals heterozygous for a pericentric inversion of chromosome 1 in the chicken have the two complementary types of recombinant chromosomes arising from a single crossing-over within the inverted segment. These individuals are capable of reproduction. Their progeny can have one or the other of the two recombinant chromosomes or, if crossing-over occurs, either a normal or an inversion chromosome.

Cytogenetic and histological studies of testicular biopsies from subfertile men with chromosome anomaly.

Testicular biopsies from eight men with abnormal karyotypes have been examined for histological and cytogenetic evidence of disturbances of meiosis. Quantitative analysis of this material showed one, with a 13;14 Robertsonian translocation, to have apparently normal spermatogenesis. Three patients, one with a 47,XYY and two with 45,XY, inv 9 karyotypes, had an overall depression of spermatogenesis. Four others, all with major chromosomal abnormalities, had apparently normal spermatogenesis until the primary spermatocyte stage. Two of these had sex autosomal translocations. One, 45,Y,t(X;21), h

The clinical significance of pericentric inversion of the human Y chromosome: a rare "third" type of heteromorphism.

A 28-year-old normal East Indian was found to have a pericentric inversion of the Y chromosome. After reviewing the literature, it was concluded that an inverted Y chromosome does not impede the production of normal sperm and does not predispose to non-disjunction of other chromosomes in the progeny. Thus, the earlier concept of nondisjunction was rejected, and it is suggested that aberrant cases with aneuploidy and an inverted Y are fortuitous. The pericentric inverted Y is inherited from generation to generation and has no clinical significance. The prevalence of males with pericentric Y inv

Familial pericentric inversion of chromosome 13 resulting in duplication 13q22 to qter.

A 10-year-old male with a recombinant (13)dup q chromosome is reported. The recombinant chromosome originated from a maternal pericentric inversion which was present in two of his four normal sibs. A segregation analysis of 60 pregnancies from which one of the parents was a carrier of inv(13) showed a significant predominance of males among the viable offspring.

Multiple karyotypic changes in retinoblastoma tumor cells: presence of normal chromosome No. 13 in most tumors.

There are conflicting reports on the frequency in retinoblastoma tumor cells of aberrations involving chromosome No. 13. To quantitate the frequency of various chromosome aberrations, we analyzed the karyotypes from the retinoblastoma tumors; all tumors contained chromosome abnormalities. Chromosome No. 13 was altered in only two tumors, but the aberrations in these two cases affected different portions of the chromosome. We have concluded that chromosome aberrations affecting chromosome No. 13 are relatively infrequent in retinoblastoma tumors. Chromosome No.1 was involved in rearrangements i

Chromosomal instability in ataxia telangiectasia.

We have examined various aspects of lymphocyte chromosomal instability in three families comprised of five individuals affected with ataxiatelangiectasia (AT), their obligate heterozygous parents, and their unaffected sibs. We found that neither baseline sister chromatid exchanges (SCEs) nor mitomycin-C-induced increments in SCEs showed any significant differences among family members or between AT heterozygotes or homozygotes. Chromosome breakage in first-division metaphases was found to be moderately elevated in three of the five AT homozygotes (range 1-12%); breakage in the six AT obligate

Cytogenetic and dermatoglyphic studies on severely handicapped patients in an institution.

Cytogenetic and dermatoglyphic studies were performed on a group of 197 institutionalized patients with severe mental and physical handicaps in order to evaluate the contribution of chromosomal aberrations on the etiology of the condition, and to determine whether any association exists between the dermatoglyphics and the severe handicaps. There were 4 patients with trisomy 21 and 2 patients with a de novo balanced reciprocal translocation. In addition, 9 patients were found to have a pericentric inversion of chromosome 9 (inv (9) (p11q13)). Other chromosome variations identified included inv
