Does multisomy of chromosome 1q confer a proliferative advantage in B-cell acute lymphoblastic leukemia?

Two patients fulfilled the clinical and hematologic criteria for B-cell acute lymphoblastic leukemia: the malignant cells had L3 morphology, bore B-cell markers, and carried the specific t(8;14) translocation. The leukemic cells of one patient were tetrasomic for 1q, and those of the other patient showed several separate cell lines with complete or partial trisomy of 1q. In the latter patient it appeared that a break close to the heterochromatin of 1q produced an unstable chromosome end which formed associations with the telomeres of at least seven other chromosomes. It is suggested that multi

[High frequency of subtotal deletion of the Y-chromosome hetero-chromatin in Khanty from the lower Ob River area].

High frequency of the Y chromosome heterochromatin subtotal deletion, del(Y) (q12) has been detected in the course of cytogenetic study of Khanty from the lower Ob river in West Siberia. This deletion was found in 32 out of 154 males examined, i.e. in 20.8%. Taking into account a small size and a long distance isolation of the population, it has been concluded that the high frequency of the del(Y) (q12) is caused by genetic drift of the marker chromosome.

Heterochromatin and nucleolus organizer regions in cells of patients with malignant and premalignant lymphatic diseases.

Peripheral blood lymphocytes (short-term cultures) and permanent lymphoid cell lines (long-term cultures) of patients with Hodgkin disease (5), lymphoma (1), plasma cell leukemia (1), angioimmunoblastic lymphadenopathy (1), and infectious mononucleosis (3) were investigated for C-band variants and nucleolus organizer region (NOR)-activities by C-banding and silver-staining, and compared to those of healthy adults (5) and newborn children (3). Heterochromatin polymorphisms were found in malignant diseases (33%) as well as in controls (29%). In the lymphoma patients, heterochromatin of class 3 (

Linkage and chromosome study of multiple endocrine neoplasia IIa.

A linkage and chromosome investigation of a large family with multiple endocrine neoplasia (MEN) IIa (medullary thyroid carcinoma, pheochromocytoma, and occasionally hyperparathyroidism) was undertaken. No significantly positive lodscores were obtained between MEN IIa and 25 different genetic markers. Conventional metaphase chromosome analysis showed normal karyotypes and no heterochromatin markers linked to the MEN IIa locus were found. High-resolution chromosome analysis in five MEN IIa carriers revealed no deletion within band 20p12.2. The present investigation could thus neither demonstrat

Familial supernumerary microchromosome mosaicism: phenotypic effects and an attempt at characterization.

A healthy normally intelligent man and his two daughters who had mild developmental delay and different malformations were determined to have an acrocentric supernumerary microchromosome (MC). The father and both daughters were noted to be mosaic for the MC, perhaps suggesting mitotic instability of the MC cell line with loss over time. The MC was negative staining on G-banding and showed absence of centromeric heterochromatin on C-banding. A centromere was identified with silver staining; no NORs were identified. The MC approximated the size of the short arm of the Y chromosome and showed cyt

An ovarian cancer family: clinical and cytogenetic data.

A family with five ovarian neoplasms in three subsequent generations was studied. Four women had ovarian cancer at age 38, 40, 47, and 53, and one had cystoma ovari at 24. There were other neoplasms and preneoplastic lesions in this family. Several developmental anomalies were revealed, and one of them (a tooth anomaly) may be associated with ovarian tumors. Cytogenetic studies have been carried out on six of the living relatives, including two treated for ovarian neoplasms. The incidence of spontaneous chromosome aberrations was not significantly increased in each of these cases. Polymorphism

46,XX/46,XY chimerism in a phenotypically normal man.

Some twenty cases of dispermic chimeras with the karyotype 46,XX/46,XY, discovered because of gonadal dysplasias or a true hermaphroditism, have been reported. This is a report of a phenotypically normal man with 46,XX/46,XY chimerism in whom a prepubertal finding of positive X-chromatin was interpreted as Klinefelter syndrome. The diagnosis was revised 11 years later when the family doctor, who doubted the earlier diagnosis because of the patient's normal-sized testes, sent him to an outpatient clinic. The young man was 23 years old, athletic (74kg, 180cm), with normal body proportions, norma

Stereologic analysis of the ultrastructure of isolated human T and non-T lymphoid cells. III. Studies in chronic lymphoid leukemias, hairy cell leukemia and some malignant lymphomas.

Malignant cells from 9 cases of chronic lymphocytic leukemia (CLL), 4 cases of prolymphocytic leukemia (PLL), 4 cases of hairy cell leukemia (HCL), 4 cases of malignant lymphoma of centrocytic type and 3 cases of malignant lymphoma of lymphoblastic type (Kiel classification), all examined for T and B markers, were analysed by electron microscopy. Stereological methods were applied to assess relative and absolute values concerning the whole cell and nuclear and cytoplasmic components of the average cell in each population, and comparisons were made between the morphologically and immunologicall

Absence of constitutive heterochromatin in a partially identified supernumerary marker chromosome.

A retarded child with multiple malformations was found to have a karyotype 47,XY,de1(11)(11 pter leads to q21:), +mar(11 qter leads to q21::?). The mitotically stable centric marker had no demonstrable C heterochromatin. Phenotype-karyotype correlation and the role of C heterchromatin in phenotypic effects are discussed.

Detection and isolation of fetal cells from maternal blood using the flourescence-activated cell sorter (FACS).

The presence of fetal cells in the maternal circulation during pregnancy has been suggested by repeated observations of small numbers of cells containing Y chromatin or a Y chromosome in the blood of pregnant women. With the fluorescence-activated cell sorter (FACS), we have used antibodies to a paternal cell surface (HLA) antigen, not present in the mother, to select fetal cells from the lymphocyte fractions of a series of maternal blood samples, collected as early as 15 weeks of gestation. These sorted cells have been examined for a second paternal genetic marker, Y chromatin. Y chromatin-co

Prenatal diagnosis in the presence of an extra small chromosome in amniotic cell culture.

The amniotic fluid of a 20 years old Caucasoid woman was brought in for analysis because of a previous child with anencephaly. Q-banded karyotypes disclosed 47,XY, +mar. Clinical significance of this karyotype could not be determined. The couple elected therapeutic termination but were denied because the pregnancy was over 23 weeks gestation. Her blood culture revealed also 47,XX, +mar. Silver staining revealed the marker chromosome bisatellited. C-banded karyotypes revealed the marker chromosome with one C-band. This inherited marker chromosome, composed of satellites, stalks, and constitutiv

A true hermaphrodite dispermic chimera with 46,XX and 46,XY karyotypes.

A 16-year-old male with hypospadias and gynaecomastia had a rudimentary uterus with a right Fallopian tube and ovary; the left gonad was a functioning testis. Cytogenetic studies showed cells with 46,XX and 46,XY sex chromosomes in cultured blood, skin and gonadal tissues. Cells with the 46,XX constitution predominated in all tissues. Extensive investigations failed to demonstrate blood cell and serum chimerism, but there was little genetic variation of these characters between family members. Cytogenetic studies demonstrated that the father had contributed different marker chromosomes to the

Chromosome 1 in 26 carcinomas of the cervix uteri: structural and numerical changes.

Cytogenetic studies on 26 carcinomas of the cervix showed that chromosome 1 was consistently involved in the changes: either one or more structurally abnormal chromosomes or a relative excess of normal chromosomes were present. Several types of structural change were repeatedly seen: short arm deletions (1p-, in seven tumors); long arm isochromosomes (i(1q), in six tumors); and translocations of unidentified chromosomal material onto one of the arms (possibly in eleven tumors; in four of these, there was an additional C-band on the long arm). In one tumor, there was a short arm isochromosome (

The origin and behavior of two isodicentric bisatellited chromosomes.

Karyotyping revealed three cell lines in a boy with mental retardation and few other abnormalities. Thirty cells exhibited a normal karyotype, and 54 had an extra acrocentric chromosome of E group size with satellites on the long and short arms. The remaining 20 cells each had, in addition to the first marker (M1), a second tiny bisatellited chromosome (M2). C-banding demonstrated that both markers were dicentric. G-, C-, and Q-banding and satellite association data were consistent with the markers having originated from chromosome 15 material. We propose that M1 was formed from a meiotic brea

Pitfalls in the use of chromosome variants for paternity dispute cases.

A huge 'satellite' (22s+) was observed in a mentally retarded and malformed girl. It consisted of constitutive heterochromatin based on the C-band technique (CBG). In neither of the parents was the variant observed. Genetic marker studies revealed that the father was, indeed, the father of the proposita (probability, 99.8%). It is emphasized that caution must be taken in the use of chromosome variants for paternity dispute cases.

The segregation of C-band polymorphisms on chromosomes 1, 9, and 16.

Eleven normal families with at least four children were studied cytogenetically using the C-band technique to identify polymorphisms in the constitutive heterochromatin of chromosomes 1, 9 and 16. Thirteen individuals showed one or more variants in such chromosomes. The analysis of the segregation ratios in the 35 offspring of these 13 individuals showed that these marker chromosomes generally segregated according to the expected 50:50. However, one of these variants, chromosome no. 9 with an increased heterochromatin block in the secondary constriction, has an apparently preferential segregat

X-chromosome inactivation and selection in somatic cells.

X-Chromosome inactivation leads to the formation of mosaic cell populations in the somatic cells of mammalian females. Cells have either a maternal or paternal X-chromosome active. If an individual is heterozygous for a cell autonomous X-linked trait, then a set of built-in cellular markers is provided for the investigation of various developmental phenomena, including selection. In the absence of somatic cell selection the tissues of an X-linked heterozygote should all be mosaic and should have a mosaic composition approaching 1:1. If somatic cell selection is occurring, it should be detectab
