Ideal observer estimation for binary tasks with stochastic object models.

Objective. We propose a new formulation for ideal observers (IOs) that incorporate stochastic object models (SOMs) for data acquisition optimization.Approach. A data acquisition system is considered as a (possibly nonlinear) discrete-to-discrete mapping from a finite-dimensional object space,x∈Rnd, to a finite-dimensional measurement space,y∈Rm. For binary tasks, the two underlying SOMs,H0andH1, are specified by two probability density functions (PDFs)p0(x),p1(x). This leads to the notion of intrinsic likelihood ratio (LR)ΛI(x)=p1(x)/p0(x)and intrinsic class separability (ICS), the latter quant

Research progress of 3-n-butylphthalide and its derivatives in combating cerebral ischemia.

Ischemic stroke (IS) presents a major threat to human life and health due to its high disability and mortality rates. 3-n-Butylphthalide (NBP), derived from celery seeds of the Apiaceae family native to the Mediterranean region, was first introduced in China for acute IS treatment in 2004. NBP demonstrates multiple therapeutic actions, including reconstruction of microcirculation in the cerebral ischemia area, inhibition of platelet aggregation, reduction of cerebral infarction volume, maintenance of blood-brain barrier (BBB) integrity, and enhancement of cerebral blood perfusion. However, its

Effective and Robust Oxido-Molybdenum(V) Corrole Catalysts for Carbon Dioxide Utilization under Environmentally Benign Conditions.

Oxido-molybdenum(V) corrole complexes, Mo(O)DMPC (Mo-1) and Mo(O)DMPCBr14 (Mo-2), were synthesized and characterized using various spectroscopic techniques. The oxidation state of molybdenum(V) in Mo-1 and Mo-2 was confirmed by electron paramagnetic resonance (EPR) spectroscopy, revealing an axial compression with a dxy1 configuration. These complexes were utilized as catalysts for the cycloaddition of CO2 with various epoxides to produce the corresponding cyclic carbonates under solvent-less conditions in the presence of tetrabutylammonium bromide (TBAB) as a cocatalyst. These catalysts exhib

A wearable monitoring system for running gait analysis by diffusion transformer.

Conventional wearable monitoring devices often suffer from insufficient data accuracy and low posture recognition rates, making them inadequate for the demands of professional sports health monitoring. To address these issues, this study proposes a wearable monitoring system for running gait analysis based on the Diffusion Transformer (DiT). The system aims to achieve high-precision running posture recognition and real-time motion monitoring through multi-sensor data fusion and advanced deep learning architecture. First, a wearable system was developed using a nine-axis Micro-Electro-Mechanica

Chromosomal dynamics and evidence of differentiation of the proto-sex chromosomes XY in Farlowella oxyrryncha (Loricariidae; Loricariinae).

Farlowella is the second most species-rich genus within Loricariinae; however, cytogenetic data for its species remain scarce. In this study, we employed conventional and molecular cytogenetic procedures on Farlowella oxyrryncha, a species widely distributed across the Amazon and Orinoco River basins. Karyotypic analysis revealed a conserved diploid number (2n = 58), with 12m+22sm+18st+6a, in addition to notable structural variation compared to other species of the genus. The pattern of constitutive heterochromatin (CH) facilitated the characterization of chromosome pairs. Furthermore, a consp

Experimental investigation of high-density TeO2- and Sm2O3-modified ZnO-BaO-B2O3 glasses: Promising lead-free materials for nuclear radiation shielding application.

The replacement of lead (Pb) in radiation shielding materials is a critical priority for advancing safe medical diagnostics and environmental protection. However, viable alternatives that match Pb's attenuation performance remain limited. In this study, we present a new class of lead-free, high-density glasses: (30-x-y) B2O3-9ZnO-31BaO-(30+x) TeO2-ySm2O3, where x = 0, 2, 4, 6 mol% and y = 0, 0.5, 1, 1.5 mol%. These glasses were synthesised via the melt-quenching method. The shielding capabilities of the synthesised glasses were systematically investigated through experimental and Phy-X/PSD the

Generation of an isogenic human induced pluripotent stem cell line with a mutant propionyl-CoA carboxylase α subunit.

BACKGROUND: Propionic acidemia (PA) is a rare autosomal recessive metabolic disorder caused by defects in propionyl-CoA carboxylase (PCC), a mitochondrial enzyme composed of six alpha (PCCA) and six beta (PCCB) subunits. Mutations in PCCA/PCCB genes disrupt PCC function, leading to toxic metabolite accumulation and clinical manifestations. Current research is limited by inadequate patient-derived cellular models and ethical constraints in sample acquisition. METHOD: Using CRISPR/Cas9-mediated gene editing, we established an isogenic human induced pluripotent stem cell (iPSC) line carrying the

Novel stem cell therapy for cerebral palsy using stem cells from human exfoliated deciduous teeth.

BACKGROUND: Effective treatments for cerebral palsy caused by Hypoxic-ischemic encephalopathy are urgently needed. Current therapies primarily include prevention or acute intervention, leaving a major gap in the options for reversing established neurologic damage. Because of their ease of collection and unique trophic factor profile, stem cells from human exfoliated deciduous teeth (SHED) are promising candidates for cell-based therapy targeting neurological disorders. In this study, we examined the therapeutic potential of SHED in a rat model of cerebral palsy, focusing on neurogenic and func

Myeloid-lineage CAR knockin mice enable allogeneic immunotherapy for liver and lung fibrosis.

The preparation of chimeric antigen receptor immune cells (CAR-ICs) typically involves a costly and time-intensive process. Moreover, the transduction of CAR genes into ICs, particularly macrophages, presents inherent challenges. Here, we report a novel strategy to obtain CAR-modified macrophages (CAR-Ms) directly from living animals to treat pulmonary and hepatic fibrosis. We first validated adenovirus-transduced CAR-Ms targeting fibroblast activation protein (FAP) in vitro, as well as in mouse models of pulmonary and hepatic fibrosis. Using a Cre/LoxP strategy, we generated mice with myeloid

A Novel Systemic Inflammation-Based Model for Predicting Mortality in HBV-Related ACLF.

Excessive inflammation and immune activation are strongly associated with unfavorable outcomes and elevated mortality rates in hepatitis B virus (HBV)-related acute-on-chronic liver failure (ACLF). However, the role of the C-reactive protein to lymphocyte ratio (CLR) as a prognostic inflammatory biomarker in multiple diseases is rarely mentioned in HBV-ACLF. This investigation aimed to assess the utility of the CLR and additional parameters in identifying high-risk progression in HBV-ACLF patients. A cohort of 639 hospitalized patients diagnosed with HBV-ACLF based on the APASL criteria was di

Distorting Square-Planar Cu Using Adjacent La Atom Optimizes Electrocatalytic Nitrate Reduction.

Electrocatalytic conversion of nitrate to ammonia is a rapidly evolving field due to its potential applications toward nitrate pollution remediation and sustainable ammonia synthesis. As a group of well-established electrocatalysts, the activity of Cu-based materials is highly related to the coordination environment. Herein, a novel Cu-La-based dual-atom catalyst (Cu-La/DAC) was prepared using a Robson-type bimetallic Cu-La complex as precursor, where the square-planar Cu center of Cu-La/DAC effectively deviates due to the presence of adjacent La atom. As a result, the HOMO (dxy) energy of Cu

Two independent origins of XY sex chromosomes in Asparagus.

The relatively young and repeated evolutionary origins of dioecy (separate sexes) in flowering plants enables the investigation of molecular dynamics occurring at the earliest stages of sex chromosome evolution. With two independently young origins of dioecy, Asparagus is a model genus for studying the genetics of sex-determination and sex chromosome evolution. Dioecy first evolved in Asparagus ∼3 to 4 million years ago (Ma) in the ancestor of a now widespread Eurasian clade including garden asparagus (Asparagus officinalis). A second origin occurred in a smaller, geographically restricted, Me

ACTA2 Pathogenic Variants Activating Heat Shock Factor 1 and Increasing Cholesterol Biosynthesis in Smooth Muscle Cells Predispose to Early Onset Atherosclerosis.

BACKGROUND: ACTA2 pathogenic variants predispose to thoracic aortic disease, and a subset of variants lead to early onset atherosclerotic cardiovascular disease (ASCVD). The molecular pathway linking misfolded SMA (α-smooth muscle actin) monomers to augmented atherosclerosis-associated smooth muscle cell phenotypic modulation can be modeled in vitro by stably expressing the ACTA2 p.R149C variant in Acta2-/- smooth muscle cells. METHODS: The Montalcino Aortic Consortium patient registry was used to identify cases with ACTA2 pathogenic/likely pathogenic missense variants. These patients were su

Design and visual servo control of a three-axis piezo-flexural micro/nano-manipulator with suppressed coupling motions.

Compliant-mechanism-based precision positioning stages are widely employed in micro/nano manipulation applications. However, cross-coupling motions are major sources of positioning errors, particularly in multi-axis, multi-stage systems. Here, we develop a visual sensing-based compliant dual-stage nano-manipulator driven by piezoelectric actuators for decoupled motion control. It comprises a XY stage for lateral motion and a Z stage for axial motion, both designed using symmetrical four-bar flexure mechanisms. The load-displacement relationships of both stages were analyzed theoretically and t

Giant Specific Power Generation Capacity of Micro-Thermoelectric Generators Enabled by High-Entropy Cocktail Strategy.

Specific power generation capacity ΓP is a critical performance metric for micro-thermoelectric generators (μ-TEGs), yet the best reported values are constrained to a few hundred µW cm-2 K-2. Here, we report a giant ΓP of ∼5000 µW cm-2 K-2 in μ-TEGs based on the anomalous Nernst effect (ANE) in medium-entropy (FeCoNi)100- xPtx films. Leveraging the high-entropy cocktail strategy, we have simultaneously achieved a large anomalous Nernst thermopower Sxy (>1.4 µV K-1) and low resistivity ρxx (<85 µΩ cm), and suppressed the classical and quantum size effects on both Sxy and ρxx at the optimal comp

Multiemitting Lanthanide-Organic Complexes with Abundant Uncoordinated Lewis Base Active Sites as Luminescence Probes for Temperature Sensing, Ion Detection, and White Light Emission Films.

A series of isomorphic lanthanide-based metal organic complexes (LnMOCs), {[Ln(C14H10N4O4)0.5(C2O4)1.5(H2O)2]·3H2O}n (LnBTA, Ln = Eu, Tb, and Gd), and their doped complexes have been successfully synthesized using 2,5-di(1H-imidazol-1-yl)terephthalic acid (H2BTA) as the organic ligand. The as-synthesized LnMOCs were characterized by single-crystal X-ray diffraction (XRD), powder XRD, and thermogravimetric analysis (TGA), and their stability was systematically evaluated under a wide pH range, long-term storage conditions, and various organic solvents. In the range of 303-403 K, the doped Cu@LnB

Effects of an external electric field on the electronic properties of MSi2PxAsy monolayers and homojunctions: a first-principles study.

Using first-principles density functional theory (DFT), we systematically investigate the influence of a perpendicular external electric field (Eext) on the electronic properties of monolayer and homojunction structures of MSi2PxAsy (M = Mo, W; x + y = 4). Our results demonstrate that an external electric field of approximately 0.7 V Å-1 induces a direct-to-indirect bandgap transition, followed by a semiconductor-to-metal phase transition in the monolayer. The Rashba spin splitting around the Γ point, as captured by a derived k·p Hamiltonian, exhibits a tunable magnitude that depends linearly
