Klinefelter syndrome (KS), the most common sex chromosome aneuploidy in males, is often underdiagnosed until adolescence, delaying early intervention. We describe 11 pediatric patients with KS who were followed between 2005 and 2025 to identify early markers. Three were diagnosed prenatally; the remaining eight were diagnosed at a median age of 6.1 years, mainly due to neurodevelopmental problems. Of the total of 11 patients, 9 had delayed language acquisition, followed by 8 with psychomotor delay, 5 with behavioral disorders, 3 with sleep disorders, and 2 with epilepsy. Endocrinological comor

Sex differences in cardiovascular disease (CVD) are increasingly recognized. These are traditionally attributed to hormonal influences, but recent evidence underscores the potential role of sex chromosomes. This review describes the involvement of sex chromosomes in CVD, through loss of chromosomes, genetic variation, and altered expression. Mosaic loss of the Y chromosome (mLoY) is the most well-characterized mechanism linking sex chromosomes to CVD, with substantial evidence in heart failure. Also, the involvement of mLoY in CVD mechanisms, such as myocardial fibrosis and cardiac macrophage

The objective of this systematic review was to identify the evidence of testicular cancer risk for people with intersex conditions. This assessment is hoped to help refine risk stratification tools for assessing gonadal malignancy risk and guide the development of more robust evidence-based management strategies. The literature was searched in Ovid MEDLINE, Embase, and Cumulative Index of Nursing and Allied Health using a search string developed by a multidisciplinary team. The protocol was registered at Prospective Register of Systematic Reviews as CRD42021231313. A total of 3608 articles wer

The European Reference Network on Rare Endocrine Conditions (Endo-ERN) needs to take a cautious and evidence-based position regarding needs for genital surgery in individuals with differences in sex development (DSD). Given the multi-ERN coverage of DSD, the focus of our network is on rare endocrine care concerns in such surgeries and ensuring Endo-ERN's position aligns with the broader European emphasis on human rights, ethical medical practices, and patient-centered care. This discussion paper addresses the ethical, medical, and legal considerations surrounding genital surgery in individuals

OBJECTIVES: Differences of sex development (DSD) include rare diagnoses such as congenital adrenal hyperplasia, Turner syndrome, Klinefelter syndrome and other chromosomal, 46,XX- and 46,XY-DSD. Patient education is an essential aspect of an empowerment approach. This analysis evaluated a new training concept with regard to knowledge acquisition for individuals with DSD and their families to make self-determined decisions about treatment options.
METHODS: Two-day diagnosis-specific, and age-homogeneous group training sessions were evaluated in a five-centre, single-arm intervention study in a

Sex chromosome aneuploidies are associated with a wide range of neurodevelopmental outcomes, and notably, confer risk for cognitive impairments with varying degrees of severity. Yet, to date, few studies have compared multiple cognitive outcomes between different sex chromosome aneuploidy karyotypes, and it remains unclear how cognitive variation is linked to other elements of functioning. Leveraging high-dimensional data from a large battery of standardized neurocognitive tasks, we systematically characterize cognition across domains among 167 individuals with sex chromosome aneuploidy (n = 1

BACKGROUND: Klinefelter syndrome (KS) (47,XXY) is a genetic condition associated with infertility, hormonal imbalances, and a range of neurodevelopmental and psychosocial challenges, including language impairments, executive dysfunction, and difficulties in social functioning. Although these challenges can significantly affect daily life, no uniform terminology or standardized tool currently exists to systematically identify and assess them. Building upon research in tuberous sclerosis complex (TSC) and using the TSC-Associated Neuropsychiatric Disorders (TAND) Checklist as a foundation, this

INTRODUCTION: To develop a preliminary, decision-making protocol capable of predicting sperm retrieval success in non-mosaic Klinefelter's syndrome (KS) patients. Main outcome measure was to identify predictors of successful sperm retrieval and to build a decision-making protocol capable of predicting this success by assessing several variables including age, testicular volume, and preoperative hormonal responses.
METHODS: A retrospective cohort study included all men with non-mosaic KS who were treated with recombinant hCG and then underwent microdissection testicular sperm extraction (micro

Alcohol use disorder often causes patients to experience negative affect during withdrawal, and anxiety is positively correlated with relapse during abstinence. The neural adaptations that occur during the transition to dependence are not entirely understood, but they may include interactions between mesolimbic reward circuits and brain stress circuits. Previously, we demonstrated that chronic alcohol exposure leads to activation of ventral tegmental area (VTA) neurons projecting to the central amygdala (CeA) during acute withdrawal, raising the possibility that these cells may underlie some b

PURPOSE: Klinefelter syndrome (KS), the most common sex chromosome aneuploidy in males, is frequently associated with hypogonadism and metabolic dysfunction during adolescence and adulthood. However, data on the early endocrine and metabolic phenotype of prepubertal individuals with KS remain limited. This study aimed to descriptively characterize reproductive hormone profiles and metabolic parameters in prepubertal boys with 47,XXY KS and to explore early endocrine-metabolic associations across developmental stages.
METHODS: A retrospective cross-sectional analysis was conducted on 40 prepub

MOTIVATION: Chromosomal abnormalities, referred to as aneuploidies, occur in approximately 0.3% of live births. While the majority of aneuploidies in humans are incompatible with life, well-characterized exceptions include Down syndrome (47,+21), Patau syndrome (47,+13), Edwards syndrome (47,+18), Turner syndrome (45,X0), Klinefelter syndrome (47,XXY), and triple X syndrome (47,XXX). These chromosomal alterations disrupt gene expression and cellular function, leading to genetic and developmental disorders. With the increasing adoption of next generation sequencing (NGS) in clinical diagnostics

BACKGROUND: Klinefelter syndrome is associated with a markedly increased risk of venous thrombosis. While hypogonadism and metabolic dysfunction are known contributors, the role of the contact activation system in Klinefelter syndrome remains unexplored. contact activation system mediates crosstalk between coagulation, fibrinolysis, and inflammation and may contribute to thromboinflammatory risk.
OBJECTIVE: To investigate whether contact activation system is dysregulated in Klinefelter syndrome and assess the influence of hypogonadism, body composition, and testosterone replacement therapy on

BACKGROUND: Cell therapy, particularly those utilizing mesenchymal stem/stromal cells (MSCs), is gaining traction as a therapeutic option for regenerative treatment in patients with limited therapeutic options. Although the safety of MSC-based interventions is well established, uncertainties remain regarding how genetic abnormalities and culture conditions may influence their therapeutic efficacy. Klinefelter syndrome (KS; 47, XXY), the most common sex chromosome aneuploidy in males, is characterized by hypogonadism, infertility, and systemic comorbidities; however, its implications for autolo
