Characterization of HER2 Expression Levels, Including HER2-Ultralow, in a Retrospective Male Breast Cancer Cohort.

PURPOSE: Male breast cancer (maleBC) is an orphan disease. Aside from age, risk factors include genetic mutations and conditions like Klinefelter syndrome or other reasons of hypogonadism. Treatment is based on standards in women, but biological differences may exist, with maleBC exhibiting higher rates of hormone-receptor expression. Limiting data exists regarding the rate of low/ultralow HER2 expression, a predictive biomarker for the antibody-drug conjugate (ADC) trastuzumab deruxtecan (T-DXd) in HER2-negative disease in women. MATERIALS AND METHODS: We conducted a retrospective single-cen

Trisomy 21 Variants Among 3,124 Mexican Patients with Down Syndrome: A Three-Decade Experience of a Referral Laboratory.

BACKGROUND: Down syndrome (DS) is the most common genetic cause of intellectual disability, results from trisomy 21 (T21). T21 can be regular or due to translocation, isochromosome, or mosaicism, among other chromosomal alterations. In Mexico, frequencies of these chromosomal variants reported internationally are used; therefore, national data are necessary. OBJECTIVE: To determine the frequencies of cytogenetic findings in patients with DS referred to a research center in western Mexico over the past 31 years (1993-2024). METHODS: Physical and digital records from the cytogenetics laborator

Feminizing Therapy in People With Klinefelter Syndrome and Gender Incongruence: Considerations for Treatment Without Testosterone Blockers.

BACKGROUND: Klinefelter syndrome (KS; 47, XXY) usually involves male sex development and gender identity. Small studies suggest gender incongruence may be more common in KS, but feminizing gender‑affirming hormone therapy in this group has been scarcely reported. OBJECTIVES: To evaluate current feminizing hormone therapy in individuals with KS and the number of individuals with KS attending our gender clinic. MATERIALS AND METHODS: We conducted a care evaluation of individuals with KS referred to the Center of Expertise on Gender Dysphoria (Amsterdam UMC) between February 1972 and May 2025,

Network ontology transcript annotation identifies genetic signals underlying sex determination.

Cannabis sativa L. (marijuana, hemp, cannabis) is an angiosperm species currently evolving sex chromosomes. Genetic mechanisms, primarily an XY chromosome system, dictate cannabis sex expression in dioecious populations. However, sexual expression is also governed by the interplay of hormone regulatory gene networks, influenced by both genetic and environmental factors. Within the species, some populations exhibit dioecy, monoecy, or a gradient of both. Dioecious individuals produce exclusively male or female flowers, while monoecious plants bear both male and female flowers. Remarkably, throu

Reproducible Bradycardia After Flumazenil Reversal of Midazolam-Propofol Intravenous Sedation in a Patient With Klinefelter Syndrome: A Case Report.

Klinefelter syndrome (KS) is the most common sex chromosome aneuploidy in males and is associated with metabolic, cardiovascular, and neurodevelopmental features. Some individuals with KS have also been reported to exhibit altered heart rate (HR) regulation, although cardiovascular responses associated with intravenous sedation (IV-S) have not been well documented. We report a 14-year-old boy with KS, intellectual disability, and autism spectrum disorder who underwent multiple dental treatments under IV-S with midazolam and propofol. During three consecutive sessions at approximately two-week

Loss of Chromosome Y Associates With Altered Immune Cell Trajectories and X-Inactivation Features.

The loss of chromosome Y (LOY) in leukocytes is the most prevalent form of clonal mosaicism observed in older men. Previous studies provided multiple pieces of evidence for the effect of LOY on the immune system and connected LOY to elevated risk of all major causes of mortality, including cardiovascular diseases and cancer. Despite these associations, the dynamic effects of LOY across the developmental trajectories of immune cell populations remain unclear. We utilized single-cell RNA-sequencing data from the peripheral blood mononuclear cells of 416 male donors (median age = 68) from the One

Factors associated to motor development in Down syndrome patients.

Beyond monogenic obesity: the unmet need for anti-obesity pharmacotherapy in rare genetic syndromes.

The advent of highly effective anti-obesity pharmacotherapy has transformed obesity management, yet patients with rare genetic syndromes associated with obesity remain largely excluded from evidence generation and clinical guidance. Conditions such as Turner syndrome, Klinefelter syndrome, Fragile X syndrome, and sex chromosome mosaicisms may represent relevant candidates for anti-obesity treatment, although dedicated studies are lacking. This evidence gap may contribute to therapeutic inertia and limit equitable access to therapeutic innovation. We highlight the need for dedicated studies, re

Establishment of an inducible knockout model for the chicken Z-chromosome-linked gene DMRT1.

Sex determination is a crucial process in animal development, regulated by complex genetic networks. In avian species, Doublesex and mab-3 related transcription factor 1 (DMRT1) plays a vital role in gonadal development and sex determination. To unravel the function of DMRT1 in chicken sex determination, establishing an inducible DMRT1 knockout model is essential. In this study, we constructed an inducible DMRT1 knockout system and verified its efficiency and effects on related genes and physiological indicators. To achieve precise genomic ablation, we screened multiple sgRNAs targeting the DM

Association of hematopoietic mosaic loss of chromosome Y with dermatitis and eczema risk: an analysis in a large-scale prospective cohort study.

BACKGROUND: Evidence regarding the role of hematopoietic mosaic loss of chromosome Y (mLOY) in incident dermatitis and eczema (DE) risk, and its contribution to male susceptibility remains limited. OBJECTIVE: To evaluate the associations of mLOY with incident DE and to assess whether mLOY may contribute to sex differences in DE susceptibility. METHODS: This prospective study included 189,572 UK Biobank men free of DE at baseline, recruited between 2006 and 2010. Hematopoietic mLOY was derived from genotyping intensity data and analysed as both categorical mLOY status and continuous mLOY perc

Intersex community perspectives on prenatal sex chromosome screening: "It silences intersex".

Intersex individuals and/or individuals with variations in sex characteristics (VSC)/differences in sex development (DSD), who will be referred to from this point forward as individuals with I/VSC, have sex characteristics that do not fit within the binary framework of sex. This community has historically been marginalized in healthcare due to the pathologization of intersex variations, including nonconsensual sex "normalizing" medical interventions. While previous research has explored the perspectives of individuals who are transgender and/or gender-diverse on counseling about fetal sex chro

Computational analysis of bulk and single-cell RNAseq identifies prognostic signature genes in papillary renal cell carcinoma with Y chromosome loss.

Loss of Y chromosome (LOY) is the most commonly occurring somatic event in elderly males, and is associated with poor outcomes in most cancers. However, LOY is shown to be associated with improved survival in papillary renal cell carcinoma(pRCC). The study was designed to analyze the possible molecular mechanism behind this paradoxical event. For this purpose, the transcriptomics dataset of 214 male pRCC patients were retrieved from the TCGA database and the LOY score was calculated based on the expression of the 14 Y chromosome gene set. The LOY score was used to stratify the samples into two

Neonatal Presentation of 49,XXXXY (Fraccaro) Syndrome with Ventriculomegaly: Expanding the Early Neuroimaging Phenotype.

49,XXXXY syndrome (Fraccaro syndrome) is a rare sex chromosome pentasomy, historically considered a severe variant within the Klinefelter spectrum. It is characterized by intellectual disability, craniofacial dysmorphism, skeletal anomalies, hypogonadism, and congenital cardiac defects. Although neuroimaging abnormalities have increasingly been recognized in 49,XXXXY syndrome, neonatal diagnosis prompted primarily by ventriculomegaly remains rare. We report a neonate with prenatally detected ventriculomegaly in whom postnatal evaluation revealed cleft palate, congenital cardiac defects, bilate

Nanopore-based haplotype-resolved X chromosome inactivation for severity assessment in X-linked disorders: An AIFM1 study with a mosaic PDHA1 carrier.

X chromosome inactivation (XCI) modifies disease severity in females with X-linked variants, but clinically applicable high-resolution assessment remains limited. We report a family with an apoptosis-inducing factor, mitochondria-associated 1 (AIFM1) variant that shows marked intrafamilial phenotypic variability and evaluated whether haplotype-resolved nanopore sequencing can inform clinical interpretation. Targeted long-read sequencing was performed in a severely affected hemizygous male, his asymptomatic heterozygous mother, and a severely affected heterozygous sibling. In the hemizygous mal
