BACKGROUND: Klinefelter syndrome (KS) is the most common genetic cause of male infertility and is associated with nonobstructive azoospermia. Advances in surgical sperm retrieval techniques have enabled biological fatherhood in a subset of men with KS. Limited data exist concerning the presence of spermatozoa in the ejaculate of adolescent patients with KS. The question remains, on how to discuss fertility preservation with adolescents with KS, and to what extent attempts to obtain ejaculated spermatozoa should be encouraged, knowing that the chances of sperm retrieval are limited.

PURPOSE: Varicocele occurs in up to 20% of adolescent boys and is associated with impaired testicular growth and potential infertility. Although surgical indications based on testicular asymmetry are established, the optimal age for intervention remains unclear. This study evaluated the long-term associations between varicocelectomy and testicular growth and semen parameters in children/adolescents, with special attention to preadolescent patients.
MATERIALS AND METHODS: We retrospectively analyzed 125 patients from a registry (2011-2022) who underwent annual follow-up until age 20. Inclusion

CONTEXT: Klinefelter syndrome and Kallmann syndrome are 2 rare genetic disorders characterized by reduced testosterone (T) levels but differing in their gonadotrophin profiles. To date, no studies have directly compared body composition in these 2 syndromes.
OBJECTIVE: This work aimed to assess the prevalence of altered body composition parameters in patients with Klinefelter and Kallmann syndromes and to compare body composition between the 2 groups. Secondary objectives included evaluating associations between body composition, bone mineral density (BMD), and serum follicle-stimulating horm

Male hypogonadism is associated with significant alterations in body composition, including reduced lean body mass (LBM), increased fat body mass (FBM), particularly visceral adiposity, and impaired muscle function, contributing to frailty and cardiometabolic risk. These changes reflect the disruption of a complex endocrine crosstalk among bone, muscle, and adipose tissue, mediated by cytokines such as osteokines, myokines, and adipokines. This dysregulation promotes the development of osteosarcopenic obesity, a condition characterized by the coexistence of low bone mass, sarcopenia, and exces

Primary germ cell tumors (GCTs) of the prostate are extremely rare in Klinefelter syndrome (KS), with only four cases having been reported. Here, we report a case of mixed GCT characterized by pagetoid spread into the epithelium of the vas deference in a patient with KS. A 34-year-old man diagnosed previously with KS presented with gross hematuria. Radiological examination revealed a tumor occupying the prostate and bilateral seminal vesicles. Histologically, the tumor predominantly exhibited the pattern of a yolk sac tumor and focally presented as a seminoma. Notably, the tumor cells exhibite

PURPOSE: Klinefelter syndrome (KS) is the most common sex chromosome numerical abnormality in males and exhibits substantial heterogeneity in clinical, hormonal, and reproductive features. This study aimed to characterize the comorbidity profile, hormonal status, and anthropometric features of a mixed-age KS cohort and to contextualize the findings within current literature. METHODS: Forty individuals diagnosed with cytogenetically confirmed KS between 2020 and 2025 were included in the study. Of these, 38 had classic KS and two had high-grade aneuploidy (HGA), which were analysed separately.

BACKGROUND: Aberrant expression patterns of nuclear lamins have been described in various types of cancer depending on the subtype of cancer, its aggressiveness, proliferative capacity and degree of differentiation. In general, the expression of A-type lamins (lamins A and C) has been correlated with a non-proliferating, differentiated state of cells and tissues.
OBJECTIVES: To establish and compare the expression patterns of lamins in normal human skin, actinic keratosis (AK), squamous cell carcinoma (SCC) and basal cell carcinoma (BCC).
METHODS: Expression patterns of the individual lamin

Using testicular sperm, men with azoospermia can father children. Sperm cryopreservation helps avoid the risk of cycle canceling because of the failure of testicular sperm retrieval. However, cryopreservation of testicular sperm can potentially impair sperm function. In addition, the different qualities of testicular sperm retrieved from men with various etiologies of azoospermia can lead to different intracytoplasmic sperm injection (ICSI) outcomes after sperm cryopreservation. A retrospective analysis was performed to elucidate the influence of frozen testicular sperm from men with different

Male infertility is increasing in prevalence due to cancer treatment, environmental exposures, and patient-specific syndromes. Traditional two-dimensional culture systems inadequately recapitulate the complex three-dimensional architecture, microenvironment, and endocrine milieu essential for spermatogenesis. Testicular organoid models aim to circumvent the challenges of two-dimensional culture, mimicking key aspects of the testicular microenvironment, while enabling controlled experimental manipulation and patient-specific disease modeling. Current testicular organoids demonstrate remarkable

INTRODUCTION: Klinefelter syndrome is most commonly associated with a 47,XXY karyotype, though rarer variants such as 48,XXXY, 48,XXYY, 49,XXXXY, and mosaic forms occur. The clinical spectrum includes intellectual disability, psychiatric disorders, endocrinopathies, and infertility, while epilepsy is rarely reported. The syndrome is also linked to an increased risk of malignancies, including breast cancer and mediastinal germ cell tumors. Multinodular and vacuolating neuronal tumors (MVNTs) are rare glioneuronal neoplasms.
CASE PRESENTATION: We report a 9-year-old male with a 2-year history o

BACKGROUND: Non-obstructive azoospermia (NOA) represents the most severe form of male infertility. Hypogonadism is common in NOA patients, and normal testosterone (T) levels are considered essential for spermatogenesis. Fertility-preserving hormonal therapy (FpHT) has been proposed to optimize hormonal milieu and improve sperm retrieval rates (SRR) at microdissection testicular sperm extraction (mTESE), although evidence remains controversial and meta-analyses have not shown consistent benefit.
AIMS: To determine whether FpHT improves SRR in hypogonadal men with NOA undergoing mTESE.

Klinefelter syndrome (KS) is the most common male sex chromosome aneuploidy, affecting approximately 1 in 450-600 male births. Characterised by a 47,XXY karyotype in the majority of cases, it is widely known for its association with hypergonadotropic hypogonadism, infertility, and impaired testicular function; however, the clinical consequences of the supernumerary X chromosome extend far beyond the reproductive axis. This review provides an updated and integrative overview of the multisystem involvement in KS, with particular emphasis on metabolic derangements, cardiovascular and thromboembol

BACKGROUND: Klinefelter syndrome (KS) is a sex chromosome abnormality disease, with a reported incidence of 0.1%-0.2%, while the prevalence of KS in male infertility patients is about 3.1%. Patients with KS are prone to experience metabolic abnormalities over time, such as obesity, diabetes, and metabolic syndrome, along with sexual dysfunction.
CASE PRESENTATION: This report describes a case in which diabetic ketoacidosis (DKA) developed in an individual with KS. Specifically, the case report concerns a 29-year-old male who presented to our hospital with a 3-day history of headache and poor

BACKGROUND Gallbladder agenesis is a rare congenital anomaly resulting from failed cystic bud development or canalization during early embryogenesis. While frequently asymptomatic, up to half of patients develop postprandial right upper-quadrant pain resembling biliary colic. This report describes an unusual case of gallbladder agenesis in a 38-year-old man with Klinefelter syndrome who presented with hematemesis and right upper-quadrant pain. CASE REPORT A 38-year-old man with known Klinefelter syndrome (47, XXY) and no prior abdominal surgery presented with hematemesis, lightheadedness, sync

BACKGROUND: Klinefelter's syndrome (KS; 47, XXY) is associated with an altered risk profile for malignancies compared with non-KS males. In particular, several reports have noted a striking association between KS and extragonadal germ cell tumours (EGCTs), especially in the mediastinum, whereas the risk of testicular germ cell tumours (TGCTs) in KS remains unclear. KS patients also have a higher prevalence of cryptorchidism (undescended testes)-a known risk factor for TGCT in the general population-yet it is uncertain if cryptorchidism confers the same cancer risk in the KS population. This st

OBJECTIVE: To systematically evaluate and quantitatively synthesize the association between Klinefelter syndrome (47,XXY karyotype) and autoimmune susceptibility.
METHODS: PubMed/MEDLINE, Scopus, and Web of Science were systematically searched from database inception to April 30, 2026, for studies evaluating autoimmune diseases or autoimmune serological markers in individuals with KS. The protocol was registered in PROSPERO (CRD420261399097). Random-effects models were prespecified, summary data were extracted from published reports, and pooled effect estimates were expressed as odds ratios (

Klinefelter syndrome (KS) is the most common sex chromosome disorder in males and the leading genetic cause of hypogonadism, affecting approximately 1 in 450-600 male births. Despite its prevalence, many individuals remain undiagnosed. Although European cohort studies have demonstrated poorer health-related quality of life (HR-QoL) among individuals with KS, evidence from the United Kingdom remains limited. This cross-sectional online survey explored HR-QoL and perceived self-efficacy in clinical communication among adults with KS in the United Kingdom. Sixty adults with a confirmed diagnosis

BACKGROUND: 47,XXY/Klinefelter syndrome is associated with high rates of obesity, metabolic syndrome, type 2 diabetes, and cardiovascular disease.
OBJECTIVE: This study compared cardiorespiratory fitness (CRF) in adolescents and young adults (AYA) with and without XXY and investigated the relationship between CRF and other biomarkers of cardiometabolic health. We hypothesized that CRF would be lower in XXY compared to healthy male controls.
MATERIALS AND METHODS: This cross-sectional analysis included AYA with non-mosaic 47,XXY (n = 56) and 46, XY male controls (n = 37) without hypertension,

Klinefelter syndrome (KS) is the most prevalent sex chromosome disorder in men but often remains clinically silent until later life. We describe an elderly man whose evaluation for a rib fracture uncovered the diagnosis. A 76-year-old male presented after a low-impact fracture during a coughing episode. Clinical history revealed delayed sexual maturation, small testes, infertility, and mild gynecomastia. Hormonal testing demonstrated markedly reduced testosterone with elevated luteinizing and follicle-stimulating hormones, suggesting primary testicular failure. Karyotype analysis confirmed 47,
