Research Priorities of Individuals and Families With Sex Chromosome Aneuploidies.

Sex chromosome aneuploidies (SCAs) are chromosomal variations that result from an atypical number of X and/or Y chromosomes. Combined, SCAs affect ~1/400 live births, including individuals with Klinefelter syndrome (47, XXY), Turner syndrome (45, X and variants), Double Y syndrome (47, XYY), Trisomy X (47, XXX), and rarer tetrasomies and pentasomies. Individuals with SCAs experience a wide variety of physical health, mental health, and healthcare experiences that differ from the standard population. To understand the priorities of the SCA community we surveyed participants in two large SCA reg

Beyond the Extra X and Y Chromosome: The Contribution of Familial Risk for Psychopathology to the Neurodevelopmental Phenotype of Children With Sex Chromosome Trisomy.

Individuals with an extra X or Y chromosome (sex chromosome trisomy or SCT) have an increased risk for symptoms of psychopathology and neurocognitive dysfunction. In this study, we evaluated the contribution of family history (FH) of neuropsychiatric or neurocognitive disorders to the phenotype of SCT. One hundred and six children with SCT and 102 nonclinical controls, all aged 1-7 years, and their primary caregiver (parent) participated. Rates of neuropsychiatric and neurocognitive disorders were collected for all first-degree family members of the children. Neurocognitive tests and parental

Cardiovascular risk and mortality in men receiving testosterone replacement therapy for Klinefelter syndrome in Denmark: a retrospective cohort study.

BACKGROUND: Men with Klinefelter syndrome (KS) have hypogonadism, increased morbidity, and excess mortality. Testosterone replacement therapy (TRT) has the potential to alleviate this burden. We assessed the risk of major cardiovascular events (MACE) and mortality in KS according to TRT exposure. METHODS: We performed a nationwide registry based matched cohort study. We compared incidences of MACE and mortality between TRT exposed (KS-TRT) or unexposed KS (KS-non-TRT), and a male background population comparison cohort. The study period was from 1 January 1994 to 31 December 2022. FINDINGS:

X-Linked Gene Dosage and SOX2 Act as Key Roadblocks for Human Germ Cell Specification in Klinefelter Syndrome.

Klinefelter syndrome (KS), characterized by the presence of at least one extra X-chromosome, is a common cause of male infertility. However, the mechanism underlying the failure of germline specification is not well studied. Intriguingly, the differentiation efficiency of female human pluripotent stem cells (hPSCs) is often lower than that of male. This study investigates how X-linked gene dosage affects human primordial germ cell-like cells (hPGCLCs) specification in both healthy and diseased conditions. This work reveals that X-linked genes play a multifaceted role against the fate competenc

Unexplained Loss of Consciousness in a Patient With Klinefelter Syndrome and Brugada Syndrome.

Managing patients with both Klinefelter syndrome (KS) and Brugada syndrome (BrS) is challenging because of the rarity of this unique interaction. A man diagnosed with KS during fertility treatment experienced chronic muscle pain, severe depression, and cognitive difficulties, managed with testosterone therapy and antidepressants. Increased testosterone doses led to recurrent episodes of loss of consciousness, initially considered epileptic. Hormonal assessments, cardiac imaging, Holter monitoring, electrophysiological studies, and genetic testing led to BrS diagnosis. A positive ventricular ta

[Fertility treatment of men with Klinefelter syndrome].

In this review is described, that men with Klinefelter syndrome (KS) usually have azoospermia, and only a few have obtained paternity with ejaculated sperm. In Denmark, we performed surgery (micro-TESE or subcapsular orchiectomy) in 93 men with KS and obtained testicular sperm in 42% (39). More than ten children are now born, and several couples still have sperm and embryos cryopreserved with the potential to increase the live birth rate further. Since sperm in men with KS originate from cell lines with a normal chromosome number, children born with a father with KS usually have a normal karyo

Impact of age at first visit on glycolipid metabolism, bone metabolism, and fertility potential in patients with Klinefelter syndrome.

BACKGROUND: Classic Klinefelter syndrome (KS) is characterized by one extra X chromosome (47, XXY), leading to hypergonadotropic hypogonadism and higher risk of alterations in glycolipid homeostasis, cardiovascular diseases, and low bone mineral density. Most frequently, KS is diagnosed in adulthood because of infertility. OBJECTIVES: To investigate the potential association between the age at first visit and the presence of comorbidities in patients with KS. MATERIALS AND METHODS: In this cross-sectional retrospective study, we analyzed the data from 445 patients affected by non-mosaic 47,

The selection of Y chromosome microdeletion detection methods based on seminal analysis results: a comparison of high-throughput sequencing and fluorescence quantitative polymerase chain reaction (qPCR) applications.

BACKGROUND: Infertility significantly impacts numerous couples worldwide, and male infertility is a common contributing factor. Y chromosome microdeletions are potential genetic causes of male infertility. However, due to the lack of comparative studies based on semen analysis results, we have difficulty selecting an appropriate method for detecting Y chromosome microdeletions. This study aims to compare the application of high-throughput sequencing and fluorescence quantitative polymerase chain reaction (qPCR) in different types of infertility patients. METHODS: This study used high-throughp

Epidemiology of disorders associated with tall stature in childhood: A 20-year birth cohort study.

Many primary and secondary disorders in childhood may cause tall stature (height of +2 standard deviations above the mean height for age and sex). Growth-monitoring programs are aimed at early detection of such disorders to avoid permanent health consequences and support children's wellbeing. However, age- and sex-specific data on the incidence of disorders associated with tall stature are scarce. This retrospective population-based cohort study aims to specify the epidemiological data that are needed to develop better diagnostic practices. The study population included 1 144 503 children (51%

Social communication skills profile in infants with sex chromosome trisomy at 12 months of age.

Sex Chromosome Trisomy (SCT) conditions are genetic disorders that affect approximately 1:500 children. Although there is considerable variability in phenotype, individuals with SCT have an elevated likelihood, compared to the general population, of developing social deficits and autism spectrum disorder (ASD). Studies report that 10-35 % of individuals with SCT meet criteria for ASD, depending on differences in ascertainment methods and diagnostic criteria. More specifically, ASD prevalence has been estimated at approximately 18 % (range 10-27 %) for XXY, 15 % (10.8-20 %) for XXX, and 30 % (1

scRNA-Seq-Based Transcriptome Profiling and Relevant Bioinformatics Approaches to Uncover Novel Insights in Studying Human Spermatogenesis.

This study employs single-cell RNA sequencing (scRNA-seq) to investigate human spermatogenesis across developmental stages and under pathological conditions, including non-obstructive azoospermia (NOA). The analysis highlights the critical role of Sertoli cells in supporting germ cell development by providing structural support, paracrine factors, and essential nutrients. Pathological conditions, such as NOA and Klinefelter syndrome, reveal significant disruptions in Sertoli cell function, including impaired cell signaling, mitochondrial activity, and transcriptional regulation. These changes

Update on Physical, Psychological, and Quality of Life Management in Klinefelter Syndrome.

CONTEXT: Klinefelter syndrome (KS), most commonly arises from a 47,XXY karyotype. While KS affects around 1 in 450 to 600 male births, an estimated 50% to 75% of cases go undiagnosed. Individuals with KS are at increased risk of metabolic, cardiovascular, and reproductive comorbidities, increasing mortality risk, psychological burden, and significantly diminished health-related quality of life (HR-QoL) compared to healthy controls. We provide an updated review of the recent literature on the clinical management for people with KS, associated comorbidities, and the implications on HR-QoL. EVID
