Klinefelter syndrome (KS) is the most common male sex chromosome aneuploidy, affecting approximately 1 in 450-600 male births. Characterised by a 47,XXY karyotype in the majority of cases, it is widely known for its association with hypergonadotropic hypogonadism, infertility, and impaired testicular function; however, the clinical consequences of the supernumerary X chromosome extend far beyond the reproductive axis. This review provides an updated and integrative overview of the multisystem involvement in KS, with particular emphasis on metabolic derangements, cardiovascular and thromboembol

BACKGROUND: The purpose of our study was to assess the effect of hypolipidemiant drugs on serum markers of vascular inflammation (E-Selectin, VCAM-1 and MCP-1) in dyslipidemic men without cardiovascular disease.
MATERIAL/METHODS: 84 dyslipidemic men were consecutively recruited from the Lipid Unit of a tertiary hospital. The patients were placed on statins (n=44) or fibrates (n=22), depending on the lipid profile, for 4 months. In the control group (n=18), a hypolipidemiant diet alone was indicated.
RESULTS: Baseline levels of VCAM-1 and MCP-1 were not correlated with the lipid profile. By c

46,XY disorders of sexual development (46,XY DSD) are conditions characterized by deviations from typical male sex development, which encompasses a broad spectrum of clinical features such as hypospadias, decreased sperm production, dysgenetic testes, bifid scrotum, and the presence of a mature uterus and fallopian tubes. The WWOX gene, which has been implicated in various cancers, is also thought to play a role in sex development, although its involvement in 46,XY DSD remains poorly understood. This study aimed to investigate the WWOX gene in a patient presenting with hypospadias. A 7-month-o

Background/Objectives: Contiguous Xq26.2 deletions involving FRMD7 and IGSF1 are rare and incompletely characterized. FRMD7 loss causes X-linked idiopathic infantile nystagmus (IIN), whereas IGSF1 loss causes central hypothyroidism and other pituitary-related abnormalities. We aimed to define the clinical and molecular spectrum of an Xq26.2 contiguous gene deletion in a large Bedouin kindred with infantile nystagmus. Methods: We clinically evaluated ten family members (3 males and 7 females); genomic analyses included chromosomal microarrays, whole-exome and whole-genome sequencing, breakpoint

Xp21 contiguous gene deletion syndrome is a rare X-linked disorder caused by interstitial deletions involving neighboring genes such as NR0B1, GK, DMD, and IL1RAPL1, and is clinically associated with primary adrenal insufficiency, glycerol kinase deficiency, Duchenne muscular dystrophy, and neurodevelopmental delay. The phenotype depends on deletion size and gene content. We report a male child with a 4.5 Mb hemizygous deletion at Xp21.3-p21.2 encompassing IL1RAPL1, NR0B1, GK, and exons 61 to 79 of DMD, confirmed by exome sequencing and shown to be maternally inherited. The presented patient w

INTRODUCTION: Disorders of sex development (DSDs) are rare in free-ranging cervids and remain poorly documented.
CASE PRESENTATION: We describe a hunted red deer (Cervus elaphus) with well-developed antlers and male-typical morphology but female-like external genitalia, characterized by a hypertrophic clitoris/penile-like protrusion and absence of a scrotum. External genital tissues were available for histopathology, which revealed penile-type structures, including cavernous tissue and urethral-like epithelium. Genetic sex was assessed using PCR amplification of SRY and ZFX/ZFY markers from f

Pathogenic rewiring of the three-dimensional (3D) genome architecture is increasingly being identified as the cause of genetic diseases, but recognizing the cis-regulatory effects of structural variation remains a challenge. The Xq27.1 region contains a quasi-palindrome identified as a pleiotropic hotspot for disease-causing interchromosomal insertions. In a large Danish family affected by X-linked recessive complex spastic paraplegia, we identified the segregation of a 149-kb interchromosomal insertion at Xq27.1 originating from 4q24. To understand the disease mechanism, we generated induced

BACKGROUND: Mosaicism for chromosomal structural rearrangements (Rea) is rare and the timing and mechanisms of mosaic Rea formation, maintenance, and clinical manifestation are poorly understood. To date, there are no published data on the cytogenetic profile of mosaic Reas. The question as to whether the proportion of abnormal cells in the carrier's cultured blood is clinically significant remains unanswered. A previous study showed a strong female preponderance among carriers of mosaicism for Rea with pericentromeric breaks, indicating female-specific instability in early embryos. However, t

Liposomal amphotericin B (AmBisome) is a lipid-associated formulation of the broad-spectrum polyene antifungal agent amphotericin B. It is active against clinically relevant yeasts and moulds, including Candida spp., Aspergillus spp. and filamentous moulds such as Zygomycetes, and is approved for the treatment of invasive fungal infections in many countries worldwide. It was developed to improve the tolerability profile of amphotericin B deoxycholate, which was for many decades considered the gold standard of antifungal treatment, despite being associated with infusion-related events and nephr

INTRODUCTION: Monorchidism is a rarely described condition in the horse and is not to be confused with cryptorchidism. The diagnosis is challenging and confirmed by surgery and histology in combination with hormonal assays. This report describes, to the best of the author's knowledge, the first case of monorchidism and abdominal cryptorchidism of the developed testicle in a horse.
METHODS: An Irish Cob underwent laparoscopic castration for removal of bilateral cryptorchid testicles. At surgery, the horse was diagnosed as a monorchid with the testicle retained intra-abdominally. Histopathologi

INTRODUCTION: This study investigated the impact of the carrier on transferable blastocyst rate and live birth outcomes in couples with structural chromosomal abnormalities.
METHODS: Couples were grouped into reciprocal translocation, Robertsonian translocation, or inversions groups, and clinical data were retrospectively analyzed. Preimplantation genetic testing for chromosomal structural rearrangements (PGT-SR) was conducted, and pregnancy outcomes were compared.
RESULTS: Embryo euploidy rates between male and female carriers differed nonsignificantly in the reciprocal translocation and in

Pediatric solid tumors are a leading cause of childhood disease mortality. In this work, we examined germline structural variants (SVs) as risk factors for pediatric extracranial solid tumors using germline genome sequencing of 1765 affected children, their 943 unaffected parents, and 6665 adult controls. We discovered a sex-biased association between very large (>1 megabase) germline chromosomal abnormalities and increased risk of solid tumors in male children. The overall impact of germline SVs was greatest in neuroblastoma, where we uncovered burdens of ultrarare SVs that cause loss of func

Differences or disorders of sex development (DSD) represent a range of congenital conditions that lead to discrepancies among a person’s sex chromosomes, gonads, and anatomical sex. Variants in the SRD5A2 gene can lead to 5-alpha-reductase type 2 (5α-RD2) deficiency, a condition within the DSD spectrum. Here, we report a case of 5α-RD2 deficiency in a Chinese child, resulting from a newly identified compound heterozygous variant in SRD5A2. The proband, a 2-month-old child assigned female at birth, was initially observed to have bilateral hypertrophy of the labial folds during routine child hea

The crucial role of Y chromosome genes in male gonadal determination and reproductive fitness has been recognized for decades. Y chromosome microdeletions are the most common molecular genetic causes of azoospermia and severe spermatogenic impairment. Since the late 1990s, screening for these microdeletions has become a routine part of the diagnostic work-up of severe male factor infertility. In this article, we provide a concise overview of the Y chromosome's structure and gene content. We describe its clinically relevant alterations, detectable through karyotyping or molecular genetic tools,

Individuals with Turner syndrome (TS) phenotypes may exhibit short stature, ovarian dysfunction, and neurocognitive disorders. Their genomes can include ring chromosomes formed from the X chromosome (RCX). Here, we present cytogenomic and clinical findings from seventeen individuals with TS who bore mosaic forms of RCX and frequently presented with short stature and concern for TS. The subjects were retrospectively included and tested at a single-site cytogenetics laboratory for over 42 years. Here, we illustrate each subject's comprehensive cytogenetic workup and phenotypes. The cohort shows

Although a routine semen analysis and an endocrine evaluation (usually of the gonadotropins and androgen levels) are the cornerstone of clinical diagnostic evaluation of the male seeking evaluation and treatment of the couple with infertility, in recent years, the American Urological Association/American Society for Reproductive Medicine Practice Guidelines for Male Infertility stressed the importance of assessing chromosomal anomalies, such as numerical and structural anomalies in men with spermatogenic failure. This narrative review of chromosomal anomalies as a cause of male factor infertil

BACKGROUND: Disorders of Sex Development (DSD) are caused by congenital abnormalities in the chromosomes, and subsequent development of gonads or sexual anatomy. XX-DSD pigs exhibit a series of adverse symptoms such as sterility, genital infections, and decline in meat quality, leading to significant economic losses in the breeding industry. However, the understanding of the etiology and pathogenesis of XX-DSD in pigs remains limited. To investigate the molecular mechanisms underlying abnormal gonadal development in XX-DSD pigs, we analyzed the gonads of 1-month-old XX-DSD pigs, normal females

Prenatal cell-free DNA (cfDNA) screening and carrier screening (CS) may have incidental findings that have implications for maternal health outside the scope of the test. We investigated outcome information for individuals with both a DMD full gene deletion/duplication on CS and a suspected maternal X chromosome abnormality on SNP-based prenatal cfDNA screening. This retrospective analysis included de-identified data from pregnant individuals referred for CS and prenatal cfDNA screening at a single reference laboratory (9/2019-12/2021). Maternal karyotype and/or chromosomal microarray analysis

BACKGROUND: Chromosomal abnormalities are a significant cause of miscarriages. Carriers of balanced chromosome rearrangement are often at risk of recurrent pregnancy loss (RPL), as they are more likely to produce gametes with unbalanced chromosome rearrangements.
OBJECTIVE: This study evaluated the chromosomal abnormalities detected in couples with history of primary recurrent pregnancy loss.
DESIGN: Retrospective, cross-sectional study.
SETTING: Single center, tertiary healthcare center in Turkey.
PATIENTS AND METHODS: This study reviewed conventional cytogenetic/molecular cytogenetic ana

BACKGROUND: Psychiatric symptoms are typically highly inter-correlated at the group level. Collectively, these correlations define the architecture of psychopathology - informing taxonomic and mechanistic models in psychiatry. However, to date, it remains unclear if this architecture differs between etiologically distinct subgroups, despite the core relevance of this understanding for personalized medicine. Here, we introduce a new analytic pipeline to probe group differences in the psychopathology architecture - demonstrated through the comparison of two distinct neurogenetic disorders.
