A dynamic "hormonal tug-of-war" driven by a functioning ovarian remnant after testis-preserving partial gonadectomy in ovotesticular disorders of sex development: a case report.

The endocrine behavior of residual ovarian tissue following testis‑preserving surgery in ovotesticular disorders of sex development (DSD) during adolescence remains poorly characterized. We report an adolescent male who presented with Tanner stage B4 gynecomastia despite male‑typical external genitalia. Baseline evaluation revealed elevated gonadotropins, relative hyperestrogenism (estradiol 58.7 pg/mL), and SRY‑negative chromosome mosaicism (47,XX,+der(8)/46,XX). Pelvic imaging demonstrated a small prostate and bilateral intratesticular cysts, and laparoscopy and cystoscopy confirmed a male i

Mosaic Loss of Chromosome Y (LOY): From Genomic Instability to Disease and Single-Cell Detection.

Mosaic loss of chromosome Y (LOY) is the acquired absence of the Y chromosome in a fraction of cells, is most readily observed in the haematopoietic compartment, and represents the most common form of clonal mosaicism in older men. Once dismissed as a benign correlate of aging, LOY is now recognized as a clinically meaningful biomarker associated with reduced survival and an expanding range of age-related disorders, including multiple solid and haematological cancers, cardiovascular disease, Alzheimer's disease, and type 2 diabetes mellitus. Large-scale genome-wide analyses have shown that sus

Sex chromosomes and cardiovascular disease.

Sex differences in cardiovascular disease (CVD) are increasingly recognized. These are traditionally attributed to hormonal influences, but recent evidence underscores the potential role of sex chromosomes. This review describes the involvement of sex chromosomes in CVD, through loss of chromosomes, genetic variation, and altered expression. Mosaic loss of the Y chromosome (mLoY) is the most well-characterized mechanism linking sex chromosomes to CVD, with substantial evidence in heart failure. Also, the involvement of mLoY in CVD mechanisms, such as myocardial fibrosis and cardiac macrophage

Paternal genomic history and Y-chromosome phylogeny from the Chengdu Plain reveal multiple Indigenous East Asian farmer contributions to ancient Shu and their descendants.

BACKGROUND: The Chengdu Plain and surrounding regions (CPS) host a diverse mosaic of archaeological cultures and ethnolinguistic groups, yet the origins of these populations and the extent of Western Eurasian and East Asian influences remain unclear and underrepresented in genomic studies. As a molecular archaeological archive, Y-chromosome data provide vital insights into the paternal genetic history of ancient Shu populations and their descendants. METHODS: Here, we combine our newly generated genomic data with publicly available large-scale whole-Y-chromosome sequences from East Asia to re

Assessment of mosaic loss of chromosome Y in pulmonary fibrosis reveals limited association with susceptibility or disease severity.

BACKGROUND: Pulmonary fibrosis (PF) is a rare lung disease with diverse pathogenesis and biological mechanisms. Mosaic loss of chromosome Y (mLOY) has been reported to be associated with increased risk of fibrotic diseases. However, the exact role of mLOY in the development of PF remains to be elucidated. METHODS: Copy number on chromosome Y was used to estimate mLOY comparing patients in PROFILE and gnomAD cohorts and between cases and control patients from the GE100KGP cohort. Correlation of mLOY with demographic and clinical variables was tested using patients from the PROFILE cohort. Lung

Genetic regulation across germline and somatic variation on the Y chromosome contributes to type 2 diabetes.

Our understanding of the biological role of the Y chromosome remains limited. Here, we systematically profile germline Y haplogroups and somatic loss of the Y chromosome (LOY) in 122,683 East Asian males from BioBank Japan and 181,472 European males from the UK Biobank. A phenome-wide scan uncovers male-specific genetic regulation of complex traits, including pleiotropic effects of the Japanese-specific haplogroup D on height and type 2 diabetes (T2D). LOY increases T2D risk in East Asians but is associated with reduced T2D risk in Europeans. In East Asians, LOY contributes to T2D incidence pa

Genome assembly of the Neotropical marsh rat Holochilus nanus (Cricetidae: Sigmodontinae) brings insights on B and sex chromosome evolution.

The Neotropical region comprises approximately 27% of mammal diversity, and rodents of the tribe Oryzomyini represent a significant portion of that. This diversity is reflected in the karyotype variability of the tribe, with a huge number of chromosomal rearrangements involving autosomal, sex, and B chromosomes. Supernumerary B chromosomes were described for more than 10 species, four of them belonging to the genus Holochilus. Therefore, we sequenced the genome of two H. nanus specimens with different karyotypes: a female with (HNA-XXB) and a male without (HNA-XY) a B chromosome. We also seque

Optimizing the diagnostic approach for sex chromosome and autosomal mosaicism in amniotic fluid: integrating karyotyping with CNV-seq.

OBJECTIVE: Assessment of the clinical utility of combined karyotyping and CNV-seq for detecting sex chromosome and autosomal mosaicism in amniotic fluid. METHODS: A retrospective analysis was conducted on the data of 3307 pregnant women who underwent both amniotic fluid chromosome karyotyping and CNV-seq testing. RESULTS: The two techniques collectively detected 66 cases of mosaicism, with a detection rate of 2.00%. The concordance rate between the two techniques was 43.94% (29/66). The concordance rate for sex chromosome mosaicism was 77.78% (21/27), while it was 20.51% (8/39) for autosomal

X-Linked EGFP Reporter as a Tool to Examine X-Chromosome Inactivation in Mouse Embryos and Embryonic Fibroblasts.

This study aimed to establish a model for investigating X chromosome inactivation using transgenic mouse strains expressing green fluorescent protein (GFP). The D4/XGFP-Tg (XGFP) strain carries the GFP transgene on the X chromosome; therefore, due to random X chromosome inactivation, female offspring from crosses between XGFP males and CD-1 females exhibit mosaic GFP expression. In contrast, the B5/EGFP-Tg (EGFP) strain harbours autosomal integration of the same reporter construct, resulting in uniform GFP expression in progenies. Analysis of CD-1 × XGFP attached blastocysts revealed strong GF

Network ontology transcript annotation identifies genetic signals underlying sex determination.

Cannabis sativa L. (marijuana, hemp, cannabis) is an angiosperm species currently evolving sex chromosomes. Genetic mechanisms, primarily an XY chromosome system, dictate cannabis sex expression in dioecious populations. However, sexual expression is also governed by the interplay of hormone regulatory gene networks, influenced by both genetic and environmental factors. Within the species, some populations exhibit dioecy, monoecy, or a gradient of both. Dioecious individuals produce exclusively male or female flowers, while monoecious plants bear both male and female flowers. Remarkably, throu

Loss of Chromosome Y Associates With Altered Immune Cell Trajectories and X-Inactivation Features.

The loss of chromosome Y (LOY) in leukocytes is the most prevalent form of clonal mosaicism observed in older men. Previous studies provided multiple pieces of evidence for the effect of LOY on the immune system and connected LOY to elevated risk of all major causes of mortality, including cardiovascular diseases and cancer. Despite these associations, the dynamic effects of LOY across the developmental trajectories of immune cell populations remain unclear. We utilized single-cell RNA-sequencing data from the peripheral blood mononuclear cells of 416 male donors (median age = 68) from the One

Establishment of an inducible knockout model for the chicken Z-chromosome-linked gene DMRT1.

Sex determination is a crucial process in animal development, regulated by complex genetic networks. In avian species, Doublesex and mab-3 related transcription factor 1 (DMRT1) plays a vital role in gonadal development and sex determination. To unravel the function of DMRT1 in chicken sex determination, establishing an inducible DMRT1 knockout model is essential. In this study, we constructed an inducible DMRT1 knockout system and verified its efficiency and effects on related genes and physiological indicators. To achieve precise genomic ablation, we screened multiple sgRNAs targeting the DM

Association of hematopoietic mosaic loss of chromosome Y with dermatitis and eczema risk: an analysis in a large-scale prospective cohort study.

BACKGROUND: Evidence regarding the role of hematopoietic mosaic loss of chromosome Y (mLOY) in incident dermatitis and eczema (DE) risk, and its contribution to male susceptibility remains limited. OBJECTIVE: To evaluate the associations of mLOY with incident DE and to assess whether mLOY may contribute to sex differences in DE susceptibility. METHODS: This prospective study included 189,572 UK Biobank men free of DE at baseline, recruited between 2006 and 2010. Hematopoietic mLOY was derived from genotyping intensity data and analysed as both categorical mLOY status and continuous mLOY perc

Computational analysis of bulk and single-cell RNAseq identifies prognostic signature genes in papillary renal cell carcinoma with Y chromosome loss.

Loss of Y chromosome (LOY) is the most commonly occurring somatic event in elderly males, and is associated with poor outcomes in most cancers. However, LOY is shown to be associated with improved survival in papillary renal cell carcinoma(pRCC). The study was designed to analyze the possible molecular mechanism behind this paradoxical event. For this purpose, the transcriptomics dataset of 214 male pRCC patients were retrieved from the TCGA database and the LOY score was calculated based on the expression of the 14 Y chromosome gene set. The LOY score was used to stratify the samples into two

Nanopore-based haplotype-resolved X chromosome inactivation for severity assessment in X-linked disorders: An AIFM1 study with a mosaic PDHA1 carrier.

X chromosome inactivation (XCI) modifies disease severity in females with X-linked variants, but clinically applicable high-resolution assessment remains limited. We report a family with an apoptosis-inducing factor, mitochondria-associated 1 (AIFM1) variant that shows marked intrafamilial phenotypic variability and evaluated whether haplotype-resolved nanopore sequencing can inform clinical interpretation. Targeted long-read sequencing was performed in a severely affected hemizygous male, his asymptomatic heterozygous mother, and a severely affected heterozygous sibling. In the hemizygous mal

Hematopoietic somatic mutations in atherosclerotic cardiovascular disease.

Age-related somatic mosaicism in the hematopoietic system is increasingly recognized as a risk factor for atherosclerotic cardiovascular disease. The most prominent example is clonal hematopoiesis of indeterminate potential (CHIP), defined as the expansion of hematopoietic clones carrying acquired mutations in genes recurrently mutated in myeloid hematological malignancies, typically at a variant allele fraction ≥2%. Across large human cohorts, CHIP is associated with increased risk of coronary artery disease and other atherosclerosis-related outcomes, although the magnitude of the association

Using SNP microarray to distinguish mosaicism from chimerism in a phenotypically normal male newborn with prenatal 46,XX/46,XY karyotype.

BACKGROUND: The coexistence of 46,XX/46,XY cells in cultured amniotic fluid sample is not uncommon. The vast majority of these cases are the result of contamination by maternal cells in an otherwise normal male fetus. However, this finding might be true chimera or mosaicism, which can be associated with sexual ambiguity. Conventional cytogenetic study could not exclude these abnormalities. Here, we present a case of 46,XX/46,XY at amniocentesis and ascertained a mosaicism by postnatal single nucleotide polymorphism (SNP) array in a phenotypic male newborn. CASE PRESENTATION: A fetus of a 37-y
