A qPCR-based algorithm for the diagnosis of classic and non-classic Turner syndrome.

Background and objectives Turner syndrome is an X-chromosome aneuploidy which include classic monosomy-X and variants like mosaicism, isochromosome-Xq, etc. It is diagnosed by karyotyping, which is time-staking, laborious and costly. Quantitative real-time PCR (qPCR) offers a faster and cheaper alternative testing strategy, but single- or dual-primer qPCR may miss some variants. This proof-of-concept study was conducted to evaluate multi-primer qPCR in detecting various karyotypes of Turner syndrome. Methods Genomic DNA was extracted from 50 cases with Turner syndrome (45,X=23; 45,X/46, XX=10;

Impaired Cardiorespiratory Fitness in Adolescents and Young Adults With 47,XXY/Klinefelter Syndrome.

BACKGROUND: 47,XXY/Klinefelter syndrome is associated with high rates of obesity, metabolic syndrome, type 2 diabetes, and cardiovascular disease. OBJECTIVE: This study compared cardiorespiratory fitness (CRF) in adolescents and young adults (AYA) with and without XXY and investigated the relationship between CRF and other biomarkers of cardiometabolic health. We hypothesized that CRF would be lower in XXY compared to healthy male controls. MATERIALS AND METHODS: This cross-sectional analysis included AYA with non-mosaic 47,XXY (n = 56) and 46, XY male controls (n = 37) without hypertension,

Molecular analysis of testicular germ cell tumor in a male patient with 45,X/46,XY mosaicism: case report with review of literature.

The 45,X/46,XY mosaicism is a rare condition with a wide range of phenotypes, including a nearly normal male or female phenotype and variable degrees of gonadal differentiation. Dysgenetic gonads containing Y chromosome material carry an increased risk of developing gonadal germ cell tumors. However, to date, molecular analyses of testicular germ cell tumors (TGCTs) in patients with 45,X/46,XY mosaicism have not been reported. We encountered a 17-year-old male patient with 45,X/46,XY mosaicism who presented with undervirilized external genitalia. Ultrasound examination revealed an abnormality

Co-occurrence of GATA2 deficiency and probable constitutional mosaic STAG2 alteration in a young adult with myelodysplastic syndrome: a case report.

Germline predisposition syndromes are increasingly recognized in young patients presenting with myelodysplastic syndromes (MDS) and bone marrow failure. Among these, GATA2 deficiency represents a well-established cause of hereditary susceptibility to myeloid malignancies, whereas constitutional STAG2 alterations are rare and mainly associated with cohesinopathies and neurodevelopmental disorders. We report the case of a 25-year-old male presenting with hypocellular MDS, recurrent severe infections, lymphedema, cytopenias, psoriasis, and multiple dysmorphic features. The patient had previously

Hematopoietic loss of Y chromosome and risk of symptomatic peripheral artery disease in men.

BACKGROUND: Mosaic loss of chromosome Y (mLOY) is the most common acquired somatic mutation in men and has been linked to multiple age-related diseases, including cardiovascular diseases. However, its association with symptomatic peripheral artery disease (PAD) remains poorly understood. We aimed to investigate the relationship between hematopoietic mLOY and incident symptomatic PAD in a large population-based male cohort. METHODS: We conducted a longitudinal cohort study including 171 698 male participants from the UK Biobank who were free of PAD history. mLOY was assessed using leukocyte-de

A Case of Disorder of Sex Development in Red Deer (Cervus elaphus).

INTRODUCTION: Disorders of sex development (DSDs) are rare in free-ranging cervids and remain poorly documented. CASE PRESENTATION: We describe a hunted red deer (Cervus elaphus) with well-developed antlers and male-typical morphology but female-like external genitalia, characterized by a hypertrophic clitoris/penile-like protrusion and absence of a scrotum. External genital tissues were available for histopathology, which revealed penile-type structures, including cavernous tissue and urethral-like epithelium. Genetic sex was assessed using PCR amplification of SRY and ZFX/ZFY markers from f

Case Report: Mixed gonadal dysgenesis with Müllerian remnants mimicking a prostatic utricle in a child with 45,X/46,XY/47,XYY mosaicism.

BACKGROUND: Sex-chromosome mosaicism can cause discordant gonadal, ductal, and external genital development. Persistent Müllerian derivatives are classically associated with defects in anti-Müllerian hormone (AMH) production or AMH receptor type 2 (AMHR2) signaling in otherwise normally virilized 46,XY individuals, termed persistent Müllerian duct syndrome (PMDS). In complex mosaicism, however, Müllerian persistence may reflect mixed gonadal dysgenesis (MGD). High incidence of Y-chromosome mosaicism in male and female individuals with mild malformation of cortical development with oligodendroglial hyperplasia in epilepsy.

OBJECTIVE: Mild malformation of cortical development with oligodendroglial hyperplasia in epilepsy (MOGHE) is an underrecognized pediatric cortical lesion associated with somatic X-linked SLC35A2 variants in approximately 50% of individuals. The genetic etiology in individuals without detectable SLC35A2 mutations remains undefined, which limits our understanding of its pathomechanism and clinical characterization. The aim is to identify genetic correlates of MOGHE in individuals lacking pathogenic SLC35A2 variants and to investigate the clinicopathological and genomic correlations. METHODS: W

Elucidating the multitarget neuroprotective mechanisms of protocatechuic acid in neurological disorders (Review).

BACKGROUND: Mosaicism for chromosomal structural rearrangements (Rea) is rare and the timing and mechanisms of mosaic Rea formation, maintenance, and clinical manifestation are poorly understood. To date, there are no published data on the cytogenetic profile of mosaic Reas. The question as to whether the proportion of abnormal cells in the carrier's cultured blood is clinically significant remains unanswered. A previous study showed a strong female preponderance among carriers of mosaicism for Rea with pericentromeric breaks, indicating female-specific instability in early embryos. However, t

Phenotype-specific associations of mosaic chromosomal alterations in systemic sclerosis.

OBJECTIVES: Mosaic chromosomal alterations (mCAs) increase with age and are associated with many diseases, including autoimmune diseases. The associations between mCAs and systemic sclerosis (SSc) and its clinical subtypes have not been explored. METHODS: We recruited study subjects from 2 independent datasets (set 1: 635 SSc, 4401 controls; set 2: 347 SSc, 2170 controls) and detected mCAs (loss, loss of heterozygosity [LOH], gain, and mosaic loss of the X chromosome [mLOX]) from their peripheral blood samples. Logistic regression analyses were conducted with covariates in each cohort, and th

Plasma fractionation in Latin America: a path towards sovereignty and sustainability in the supply of plasma-derived medicines.

PURPOSE: Increased reactive oxygen species (ROS) released by NADPH oxidase and inflammation are associated with arterial hypertension and eye diseases associated with high blood pressure, including glaucoma, retinopathies (e.g., age-related macular degeneration), and choroidopathies affecting ocular function; however, the mechanisms underlying these adverse outcomes remain undefined. The present study was designed to highlight the importance of oxidative stress in severe hypertension-related eye damage. METHODS: Male Wistar rats (n = 7, unless otherwise specified for specific experiments) wer

Human Y chromosome pan-organ mapping reveals progressive mosaic loss from normal to cancer.

BACKGROUND: Loss of the Y chromosome (LOY) is a frequent event in male tumors and has been linked to cancer progression. However, the degree of mosaic LOY (mLOY) within normal tissues from men with or without cancer remains uncharacterized. METHODS: Here we used a FISH-based assay targeting X- and Y-chromosome centromeres to perform a pan-organ analysis of mLOY in 1,000 male tissue samples from 405 individuals representing 11 organs. Automated image processing generated a quantitative FISH-based mLOY score (YchrFISH) that we validated against a transcriptomic surrogate of Y-chromosome dosage

An Unusual Case of a Monorchid Horse with an Abdominally Retained Testicle.

INTRODUCTION: Monorchidism is a rarely described condition in the horse and is not to be confused with cryptorchidism. The diagnosis is challenging and confirmed by surgery and histology in combination with hormonal assays. This report describes, to the best of the author's knowledge, the first case of monorchidism and abdominal cryptorchidism of the developed testicle in a horse. METHODS: An Irish Cob underwent laparoscopic castration for removal of bilateral cryptorchid testicles. At surgery, the horse was diagnosed as a monorchid with the testicle retained intra-abdominally. Histopathologi

The maternal X chromosome affects cognition and brain ageing in female mice.

Female mammalian cells have two X chromosomes, one of maternal origin and one of paternal origin. During development, one X chromosome randomly becomes inactivated1-4. This renders either the maternal X (Xm) chromosome or the paternal X (Xp) chromosome inactive, causing X mosaicism that varies between female individuals, with some showing considerable or complete skew of the X chromosome that remains active5-7. Parent-of-X origin can modify epigenetics through DNA methylation8,9 and possibly gene expression; thus, mosaicism could buffer dysregulated processes in ageing and disease. However, wh

Mosaic loss of Y chromosome and mortality after coronary angiography.

BACKGROUND AND AIMS: Acquired somatic mutations emerged as important drivers of adverse cardiovascular disease outcomes. Recently, mosaic loss of Y chromosome (LOY) in haematopoietic cells was identified to induce diffuse cardiac fibrosis in male mice. The aim of the present study was to determine the association between LOY and cardiovascular mortality in patients undergoing coronary angiography. METHODS: LOY was quantified in 1698 male participants of the LURIC study, who underwent coronary angiography, and its association with all-cause and cardiovascular mortality was determined. Furtherm

Estimation of mosaic loss of Y chromosome cell fraction with genotyping arrays lacking coverage in the pseudoautosomal region.

BACKGROUND: Mosaic loss of the Y chromosome (mLOY) in circulating leukocytes is the most frequently detected age-related chromosomal mosaic event in men. Current mLOY detection approaches use genotyping arrays and employ a phase-based approach that identifies B allele frequency (BAF) deviations in the pseudo-autosomal region (PAR) shared between the X and Y chromosome. As some widely used genotyping arrays lack sufficient probe coverage of the PAR, methods for accurately measuring mLOY utilizing the median log2 R ratio across the male-specific region of Y chromosome (mLRR_Y) are needed for det

Reliable detection of sex chromosome abnormalities by quantitative fluorescence polymerase chain reaction.

OBJECTIVES: Many patients with sex chromosome abnormalities (SCAs) are diagnosed late in life or remain undiagnosed, leading to delayed or inadequate medical intervention and care. This study aimed to develop a reliable, rapid and cost-effective test for identifying SCAs using a blood sample - an essential step toward establishing a neonatal screening program. METHODS: A total of 360 blood samples (180 SCA patients, and 180 controls) were obtained from four cross-sectional studies of adult patients with SCAs and age-matched controls. Informed consent was collected, and all procedures followed

Association of Hematopoietic Loss of Y Chromosome With Atrial Fibrillation Incidence and Sex Disparity.

Mosaic loss of chromosome Y (mLOY), the most common somatic mutation, prompts fibrotic heart diseases. We hypothesized that it accelerates atrial fibrillation (AF) and contributes to its sex disparity. A total of 214,405 male participants from the UK Biobank without prevalent AF were included, with a median follow-up of 14.31 years. mLOY was associated with a 16% higher AF risk, with 9.6% and 6.4% of the association mediated by neutrophils and monocytes. AF risk of males with mLOY was 98% higher than that of matched females. Thus, mLOY is associated with a slightly increased AF risk, and might

Hematopoietic mosaic loss of Y chromosome: from population cohorts to pathogenic mechanisms.

Mosaic loss of Y Chromosome (mLOY) refers to genetic mosaicism in males where some somatic cells have lost the Y chromosome (ChrY) while other cells remain their ChrY. mLOY is primarily found in the blood, not only because blood cells are easily accessible, but also because hematopoietic stem cells with LOY mutation gain competitive advantages, therefore producing a large number of LOY-positive blood cells via clonal hematopoiesis. Due to the specific structures, human ChrY is prone to be missegregated during mitosis, and driving by the germline variants, environmental insults and aging microe
