Dynamic allele usage of X-linked genes ameliorates neurodevelopmental disease phenotypes in brain organoids.

While random X-chromosome inactivation in female cells of placental mammals silences one allele of the majority of X-chromosomal genes, a considerable fraction is only incompletely and variably inactivated. Human model systems to study the dynamics of incomplete X-inactivation are limited mostly to postmortem tissue, thereby disregarding developmental trajectories. Here, we used clonal human female induced pluripotent stem cells to track allele-specific expression of X-chromosomal genes along neural differentiation. We discovered dynamic reactivation and late-silencing of gene expression from

Associations between mosaic loss and schizophrenia or bipolar disorder of young age.

Mosaic chromosomal alterations (mCAs) accumulate in the brain tissues and are associated with psychiatric disorders. The association between mCAs in circulating blood and schizophrenia (SCZ) and bipolar disorders (BD) has not been fully evaluated. We detected mCAs from blood samples in 2470 SCZ, 3732 BD, and 177,773 control subjects. The associations between mCAs and SCZ or BD were evaluated using age-adjusted logistic regression models, further evaluated in age subgroups. We analyzed the associations between high cell fraction (CF) mosaic events (CF-mCAs >5% or CF-mCAs >10%) and SCZ or BD in

Chromosomal abnormalities diagnosed at the chromosomal microarray in pregnancies with isolated high risk of trisomy 21.

INTRODUCTION: In most European countries, if the risk of trisomy 21 exceeds 1/50, Women are offered further investigation through an invasive test. Since the development of the non-invasive test based on circulating free DNA from the entire genome (cfDNA), the use of non-invasive tests in this high-risk population is now being debated. Our study aims to investigate a cohort of women at high risk of isolated trisomy 21 with no ultrasound abnormalities at the first trimester ultrasound scan or nuchal translucency greater than the 99th percentile who underwent invasive sampling with chromosomal m

Harnessing the Potential of Natural Nutraceuticals Against Huntington's Disease.

Neurodegenerative disorders (NDDs) are a major global health concern and the fifth leading cause of death worldwide. Huntington's disease (HD) is an NDD regarded as a rare, genetic, and advanced disease that occurs due to the duplication of cytosine, adenine, and guanine (CAG) trinucleotide repeats on chromosome 4p, located in the Huntingtin gene (HTT). There is no specific therapy available for HD. This review examines current evidence on various nutraceuticals as therapeutic or preventive agents in HD and their benefits in protecting against neuronal damage, oxidative stress, mitochondrial d

Single-beat method echocardiographic comparison of ventricular-arterial coupling in adults with and without Down syndrome.

Adults with Down syndrome (Ds) demonstrate unique cardiovascular features, including smaller left ventricular volumes and lower blood pressure. Ventricular-arterial coupling (VAC), a key measure of cardiovascular efficiency, has not been previously studied in this adult population. Understanding VAC in adults with Ds may reveal compensatory mechanisms that maintain cardiac output despite distinct cardiovascular characteristics. This study used echocardiographic parameters to compare the VAC ratio of adults with and without Ds. Baseline echocardiographic data were collected from 28 adults with

Metabolic reprogramming during human neuron differentiation indicates glutaminase as a key determinant in Fragile X syndrome.

Metabolic homeostasis gone awry is a contributor to, if not an underlying cause of, several neurologic disorders. Fragile X syndrome (FXS) is a neurodevelopmental disorder caused by a trinucleotide repeat expansion in FMR1 and consequent loss of the encoded protein FMRP, which results in downstream molecular, neurologic, and mitochondrial deficits that are linked to cognitive impairment. In the human postmortem brain, many metabolites and solute carrier proteins are coordinately dysregulated, which also occurs during the differentiation of human induced pluripotent stem cells (iPSCs) into exci

Toward the future management of patients with CML and Ph + ALL: real-world safety insights from dasatinib pharmacovigilance.

BACKGROUND: Dasatinib, a second-generation BCR-ABL1 tyrosine kinase inhibitor, has transformed treatment for chronic myeloid leukemia and Philadelphia chromosome-positive acute lymphoblastic leukemia. However, its broad kinase inhibition leads to distinct adverse events (AEs), requiring systematic post-marketing surveillance. OBJECTIVE: To evaluate dasatinib-associated AEs using the United States Food and Drug Administration Adverse Event Reporting System (FAERS) and identify potential safety signals. METHODS: We extracted FAERS reports (Q1 2004-Q4 2024) listing dasatinib as the primary susp

Genetic counselors' perspectives on the expanded use of non-invasive prenatal testing.

Non-invasive prenatal testing (NIPT) is well established for detecting common autosomal aneuploidies. Its application to sex chromosome aneuploidies (SCAs), microdeletion/microduplication syndromes (MMSs), and single-gene disorders (SGDs) is expanding, yet opinions of genetic counselors on these indications remain variable. A cross-sectional web-based survey was conducted between October and December 2024 among 51 practicing genetic counselors across 14 countries. The 18-item questionnaire assessed perceived utility, analytic accuracy, and clinical appropriateness of NIPT for SCAs, MMSs, and S

Evaluation of Loss of the Y Chromosome in Peripheral Blood as a Biomarker for Prostate Cancer.

BACKGROUND: Prostate cancer (PCa) is the most common malignant tumor in men, but the widely used prostate-specific antigen (PSA) test has limited diagnostic accuracy. Research proposes that the loss of the Y chromosome (LOY) may affect the occurrence and development of prostate cancer, aiming to assess its potential as a diagnostic biomarker to improve the accuracy of prostate cancer detection and clinical management. METHODS: LOY levels were measured using droplet digital polymerase chain reaction (ddPCR) method, integrating relevant clinical indicators to evaluate the potential clinical sig

Vowel space change as a predictor of speech intelligibility gain in Down syndrome.

Reduced intelligibility is a frequent characteristic of the speech of children with Down syndrome (DS). While it is documented that compressed vowel space is often associated with low intelligibility, few studies have examined how acoustic vowel space changes during speech enrichment interventions are related to functional intelligibility gains. This study investigated whether change in acoustic vowel space can predict intelligibility improvement in DS. Thirteen children with DS completed a naturalistic speech enrichment intervention and were included in the present analysis. Vowel productions

A Case of Familial CDKN1C-Related Beckwith-Wiedemann Syndrome.

We report a case of a male fetus born to an unrelated couple with a fetal phenotype of an omphalocele and inferior vermian hypoplasia. Prenatal trio exome sequencing identified a maternally inherited pathogenic CDKN1C variant consistent with Beckwith-Wiedemann syndrome (BWS). This finding prompted targeted testing of the proband's sibling, who was confirmed to carry the same variant. Posterior fossa abnormalities have been reported in cases with BWS, and specifically in children with the CDKN1C loss-of-function variant.

Examining associations between foundational and complex mathematics skills in people with Down syndrome and typically developing children.

Acquiring mathematical competence is essential to independent living. In this study, we investigated the mathematics profile in young people with Down syndrome (DS), and the relations between foundational and more complex mathematics skills. The final sample included 33 participants with DS (58% male, 10.2-31.9 years) and a typically developing (TD) group matched on non-verbal mental age (N = 33, 58% male, 4.5-6.5 years). Participants completed mathematics tasks assessing symbolic number, non-symbolic number, arithmetic, reasoning and geometry. We found that performance was similar on measures
