Down syndrome, or Trisomy 21, is associated with excessive interferon (IFN) signaling and concomitant susceptibility to both autoimmunity and immunodeficiency. One of the many clinical phenotypes observed in people with Down syndrome is increased risk for oral candidiasis. Because oral candidiasis can be caused by excessive IFN-γ signaling, we asked whether IFN-gammopathy could be playing a role in susceptibility to oral candidiasis in Down syndrome. We used the Dp16 mouse model of Down syndrome, which displays mild systemic interferonopathy, to model oral candidiasis. We found that the Dp16 m

Currently, next-generation sequencing (NGS)-based non-invasive prenatal diagnosis testing (NIPT) of fetal T21 has been proven to be much more advantageous than traditional serum biochemical tests in terms of accuracy and sensitivity. Nevertheless, serum biochemical tests remain the first choice for the screening of trisomy 21 (T21) since NGS is too expensive and time-consuming. In this respect, this paper proposes a methylation-sensitive restriction endonuclease (MSRE)-digital PCR (dPCR) method for the screening of T21, which enriches the fraction of fetal-specific DNA by digesting maternal se

BACKGROUND: Interstitial deletions of the short arm of chromosome 12 are rare, and very little is known about the potential genetic basis of the most common phenotypic presentations to date described in the literature.
METHODS: In the present study, we present a new patient carrying a heterozygous de novo 12p deletion, identified by a-CGH.
RESULTS: Comparison between the new case with phenotypically similar 12p-deleted patients drawn from the literature and from the DECIPHER (the DatabasE of Chromosomal Imbalances and Phenotypes using Ensembl Resources) database allowed us to analyze 22 case

Genetic, hormonal, anatomical, and environmental factors underlie sex differences in the prevalence and progression of cardiovascular disease and responses to therapeutics. The presence of two X chromosomes in the female genome decreases susceptibility to X-linked recessive disorders but imposes the need for random X chromosome inactivation as an epigenetic mechanism controlling gene dosage. Long non-coding RNA XIST is essential for transcriptional repression of genes on inactive X chromosome but may also act as a miRNA sponge for post-transcriptional regulation of gene expression and a scaffo

BACKGROUND: Little is known about people with intellectual disabilities' views of facial attractiveness and and how desirable they feel they are (self-desirability).
METHOD: Twenty-four adults with intellectual disabilities and twenty-five adults without disabilities were recruited from further education colleges and voluntary community organisations. Participants were asked to rate the attractiveness of facial images of typically developing individuals and individuals with Down syndrome. This was followed by a semi-structured interview, exploring participants with intellectual disabilities'

CONTEXT: Down syndrome is a genetic condition caused by trisomy of chromosome 21, leading to intellectual and physical disabilities. Overexpression of the dual-specificity tyrosine-phosphorylation-regulated kinase 1A (DYRK1A) gene, located on chromosome 21, plays a critical role in neurodevelopmental abnormalities and synaptic dysfunction associated with the disorder. Identifying regulatory mechanisms capable of suppressing DYRK1A expression represents a promising therapeutic strategy.
METHOD: In this study, a consensus-based computational pipeline was employed to identify microRNAs (miRNAs)

BACKGROUND: Down syndrome (DS), which is caused by partial or complete triplication of chromosome 21, may cause a range of clinical features. Although most fetuses with DS exhibit typical characteristics, the molecular pathogenesis underlying DS remains unclear. Wnt signaling is known to play a crucial role in fetal growth and development. However, the link between Wnt signaling and the abnormal development of fetuses with DS remains poorly understood. In this study, our objective was to investigate the dysregulation of the Wnt/β-catenin pathway in the amniocytes of fetuses diagnosed with DS.

Angelman syndrome is a neurodevelopmental disorder caused by loss of the maternal UBE3A allele, the sole source of UBE3A in mature neurons owing to epigenetic silencing of the paternal allele. Although emerging therapies are being developed to restore UBE3A expression by activating the dormant paternal UBE3A allele, existing mouse models for such preclinical studies have limited throughput and utility, creating bottlenecks for both in vitro therapeutic screening and in vivo characterization. To address this, we developed the Ube3a-INSG dual-reporter knockin mouse, in which an IRES-Nanolucifera

BACKGROUND: Adults with Down syndrome (DS) are less physically active than those without DS. Among adults with DS, those with congenital heart disease (CHD) have lower physical activity than those without CHD. Many trials exclude people with CHD from exercise trials; thus, the safety and effectiveness of these interventions for increasing physical activity and fitness in adults with DS and CHD are not known. The purpose of this analysis was to evaluate the safety and preliminary effectiveness of an exercise intervention in adults with DS and CHD for increasing physical activity and fitness.

BACKGROUND: Activities of daily living at home (ADL) are a fundamental aspect of adaptive behavior in children, youth, and adults with Down syndrome (DS). However, they have received limited attention in relation to this topic. This highlights the lack of a validated instrument for the DS population in Chile.
OBJECTIVE: a) Verify validity and reliability of the Scale for Assessing of Daily Living at Home (SAADL) and b) Develop benchmarks for daily activities in children, young people, and adults with DS in central-southern Chile.
METHODS: A descriptive cross-sectional study was conducted in

Obstructive sleep apnoea poses a significant health challenge for children with Down s syndrome. Its high prevalence is attributed to unique anatomical, physiological, and comorbid factors in this population. Early diagnosis and effective treatment are crucial, as children with Down s syndrome are particularly vulnerable to the health consequences of obstructive sleep apnoea. While adenotonsillectomy is the first-line treatment, its efficacy in children with Down s syndrome can be lower. This often necessitates further airway evaluation and interventions. Alternative treatment options include

BACKGROUND: Down Syndrome Regression Disorder (DSRD) is an acute neurocognitive regression in individuals with Down syndrome (DS), causing a profound loss of acquired skills. DSRD increases demands on caregivers, to sleep disturbances, financial distress, and negative impacts on caregiver-reported social connections and perceived social support. The goal of this study was to characterize the caregiver-reported impacts of DSRD on social relationships by comparing their experiences to those of caregivers of individuals with DS and other neurological disorders (DSN).

BACKGROUND: Down syndrome (DS) is associated with lifelong challenges across cognitive, physical, and social domains that can impact quality of life (QoL). Despite research evidence supporting the benefits of recreational and entertainment-based rehabilitation, such programmes remain underdeveloped in the Middle East. This study examined cultural appropriateness, and impact of a camp-based entertainment rehabilitation programme for young people with DS in Saudi Arabia.
METHODS: This study was implemented with 11 participants (aged 17-23 years) attending an 11-day residential camp in Al-Baha.

Aging is a key challenge for modern society. In particular, brain aging is accompanied by chronic inflammation, depletion of energy potential, and an increased level of oxidative stress, with changes in blood composition playing a special role in this process. Recent studies also show that aging progresses non-linearly throughout life. Primates are genetically and anthropometrically the closest laboratory animals to humans, thus representing the most accurate model for research. This study establishes baseline values for biochemical parameters (including the state of the body's antioxidant sys

OBJECTIVE: Prader-Willi syndrome (PWS) is a genetic neurodevelopmental disorder with a characteristic trajectory. Infants display hypotonia, poor social and feeding skills, and high risk of choking, which have been shown to improve after oxytocin (OT) treatment. Our aim is to demonstrate the efficacy of intranasal OT treatment administered in the postnatal critical period on infant feeding skills and document its long-term effects. METHODS: We enrolled 52 infants with PWS (median age 2.2 months) in a European double-blind randomized placebo-controlled study. Infants were randomly assigned in a

The centriole duplication cycle must be tightly controlled and coordinated with the chromosome cycle. Aberrations in centriole biogenesis can cause developmental disorders, ciliopathies and cancer, yet the molecular determinants controlling centriole numbers and the link between the two cycles remain poorly characterized. Here, we demonstrate that McIdas, previously implicated in cell cycle regulation and multiciliogenesis, plays a critical role in maintaining proper centriole numbers. McIdas localizes to centrioles, where it exhibits dynamic localization throughout the cell cycle, dependent u

The genomic locus that encodes the Leucine-rich repeat kinase 2 (LRRK2) gene is highly pleiotropic, being associated with both Parkinson's disease (PD) and Crohn's disease (CD). Coding variants associated with risk of PD and CD act as gain of function kinase mutations increasing phosphorylation of RAB substrates, and non-coding variants in the promoter region of LRRK2 increase expression of the gene, notably in immune cells. If regulation of LRRK2 expression is a causal contributor to age-related diseases, it is important to understand the mechanism(s) by which LRRK2 is regulated, particularly

Fragile X syndrome (FXS) is the most common inherited intellectual disability and the leading monogenic cause of autism spectrum disorders (ASD). Although the pathological mechanisms underlying this neurodevelopmental disorder are challenging, recent studies have increasingly highlighted the involvement of glial cells in the pathogenesis of both ASD and FXS. Microglia and astrocytes are critical for brain development and homeostasis; thus, understanding glial dysfunction in both the developing and adult brain in these disorders may reveal novel therapeutic targets beyond the neuro-centric pers
