Over one billion people worldwide suffer from obesity, and the number is continually rising. This epidemic is partly caused by the modern lifestyle. Animal models, especially mouse models, are crucial to identifying the genetic components of complex disorders and exploring the potential applications of these genetic findings. The body weight of the animals used in research is often measured regularly to monitor their health. Only endpoint measurements, such as ultimate body weight, are frequently examined in quantitative trait locus (QTL) studies; time series data, including weekly or biweekly

OBJECTIVE: This study aimed to evaluate the detection rates of aneuploidies and pathogenic/likely pathogenic copy number variations (pCNV) using low-pass genome sequencing (LP-GS), also known as CNV-seq, across different ultrasonographic anomalies and to systematically elucidate the associations between specific types of chromosomal aberrations and specific ultrasonographic anomalies.
METHOD: LP-GS was performed in 19,452 fetuses, including those with ultrasonographic anomalies (n = 13,312) and those with normal ultrasonography (n = 6140). The detection rates of pCNV among fetuses with variou

Recent progress in the development of genetic therapies promises that impactful treatments for single-gene neurodevelopmental disorders are imminent. But can derailed neurodevelopmental processes be mended after broken genes are replaced or otherwise restored? The results of ongoing clinical trials for Angelman syndrome will soon yield answers to this pressing question, yet the trials face significant obstacles. Here we identify insights needed to aid the quest for a disease-modifying Angelman syndrome therapy, which could serve as a roadmap for the expeditious development of genetic therapies

Down syndrome (DS) is the most common chromosomal abnormality associated with intellectual disabilities. Many metabolic issues begin in childhood, and children without DS are reported to have a high prevalence of metabolic syndrome (MS) according to various studies. However, our understanding of the risk of MS in children with DS is limited. A cross-sectional study assessed the prevalence of MS in DS children aged 10-18 during 2022-2024. Demographic details and anthropometric measurements were recorded for all participants. Fasting blood samples were collected for blood glucose, insulin, and l

Pigs (Sus scrofa domesticus) are used as biomedical and preclinical models for their comparable anatomy, physiology, and neurodevelopment to humans. Angelman syndrome is a rare genetic neurodevelopmental disorder, caused by the loss of the maternal Ubiquitin protein ligase E3A (UBE3A) allele, that was recently modeled and characterized in Yorkshire/Landrace pigs. The paternal UBE3A allele is intact but silenced in neurons of the central nervous system by the long non-coding RNA and polycistronic transcription unit Small Nuclear Host Gene 14 (SNHG14). Targeting SNHG14 to reactivate the paternal

OBJECTIVE: Cardiac soft markers are subtle mid-trimester sonographic findings that may indicate an increased risk of chromosomal abnormalities. While conventional karyotyping remains the standard for detecting aneuploidies and balanced rearrangements, chromosomal microarray analysis (CMA) offers higher resolution for submicroscopic copy number variations (CNVs). However, the incremental diagnostic value of CMA in fetuses with cardiac soft markers, particularly in Chinese populations, remains underexplored. This study aimed to compare the diagnostic yield of CMA and karyotyping in fetuses with

Sex-based differences in immune function influence susceptibility to infections and predisposition to autoimmunity, with women showing both stronger immune responses and a higher burden of autoimmune and chronic inflammatory diseases. While sex hormones contribute to these differences, accumulating evidence highlights a central role for the X chromosome, which is enriched in immune-related genes and subject to complex regulatory mechanisms such as X-chromosome inactivation, skewing, escape from inactivation, and imprinting. Within this context, X chromosome-encoded microRNAs (miRNAs) have emer

BACKGROUND: Neuropsychiatric regression in individuals with Down syndrome (DS), termed Down Syndrome Regression Disorder (DSRD), has been reported to occur in the 2nd and 3rd decade of life. Symptoms of DSRD include acute onset of encephalopathy, catatonia, hallucinations, obsessive compulsive tendencies and “new autistic” features. To determine whether abnormalities in folate transport and/or metabolism may play a role in the pathogenesis of this disorder, studies explored whether abnormalities in serum, red blood cell (RBC), or cerebrospinal fluid (CSF) 5-methyltetrahydrofolate (5-MTHF) are

Epilepsy disorders differ depending on sex, gender and hormonal status. This can change treatment efficacy and remains an understudied topic in epilepsy, despite several public health initiatives promoting the inclusion of female subjects in clinical and preclinical research over the last 40 years. Here, we summarize research presented at the 2025 American Epilepsy Meeting Scientific Symposium that described current knowledge in sex differences in epilepsy, including X chromosome-dependent and -independent genetic and epigenetic mechanisms, and discussed how these advances may impact epilepsy

Congenital syndromes with subtle changes in maxillofacial morphology can pose significant diagnostic challenges, wherein artificial intelligence holds great promise in aiding diagnosis through shape analysis. The authors applied the recently proposed Swap Disentangled Variational Autoencoder (SD-VAE) in diagnosis of Beckwith-Wiedemann syndrome (BWS). The SD-VAE model was trained on a data set primarily comprised of surface 3D head scans [stereophotogrammetry (3D SPG)], gathered using a 3dMD Head System (3dMD LLC). It was also trained on CT scans when available. A total of 72 syndromic scans we

Cohesin is a fundamental genome-organizing complex that orchestrates three-dimensional chromosome folding and gene expression via DNA loop extrusion. Alterations to genes encoding cohesin subunits and cohesin loaders cause Mendelian disorders, including Cornelia de Lange syndrome (CdLS). By contrast, disruption of factors that remove cohesin from DNA, including WAPL and its binding partners PDS5A and PDS5B, have not yet been associated with human disease. Here, we explored the relevance of these cohesin release factors in Mendelian disease by establishing a rare disease cohort of deeply phenot

Sex-chromosome dosage and pubertal hormone exposure exert powerful but temporally distinct influences on human brain development, yet their interaction has rarely been formalized within a unified mechanistic framework. Variation in X-chromosome dosage provides a unique opportunity to dissociate early organizational constraints from later modulatory processes shaping cognitive circuits. Here, we synthesize genetic, neuroimaging, neuroendocrine, and neuropsychological evidence to propose a hierarchical gene-hormone-circuit framework of cognitive development. Using Turner syndrome as a biological

The pseudoautosomal region (PAR) of mammalian sex chromosomes is a small region of sequence identity shared by the X and Y chromosomes that allows pairing, crossover, recombination, and proper segregation of sex chromosomes. Although the mouse PAR sequence was largely a mystery, we have recently obtained the complete PAR sequences of the C57BL/6J and CAST/EiJ strains. Here, we report the complete PAR sequence of a Japanese wild mouse-derived strain, MSM/Ms, and compare the PARs of the three strains. There are considerable differences in the size of PARs between strains (MSM/Ms PAR is only abou

OBJECTIVE: We present a distinction between age-related X chromosome loss and true mosaicism in women over 50 years of age and the clinical course in adulthood due to the low frequency of X chromosome dual aneuploidy.
CASE REPORT: Clinical reports of adult women with the 45,X/46,XX/47,XXX karyotype are extremely rare. In a 51-year-old woman with short stature and schizophrenia with secondary amenorrhea shortly after the onset of her early menstrual period, the proportion of 45,X cell lines was 10 % by G-band and fluorescence in situ hybridization in the peripheral blood. The presence of 45,X

Ph-chromosome-negative myelo-proliferative neoplasms (MPNs) are characterized by over-production of mature blood cells driven by acquired somatic variants in JAK2, CALR and MPL in most instances. Ph-chromosome-negative MPNs are genetically and clinically heterogeneous. These variants are detected in blood cells of > 3 per cent of normal people but few develop Ph-chromosome-negative MPNs. People with Ph-chromosome-negative MPNs often have sub-clones which are important for disease evolution. Clonal architecture is shaped by hereditary factors, inflammation and environmental exposures such as so

Williams-Beuren Syndrome (WBS), a neurodevelopmental disorder caused by a heterozygous microdeletion at chromosome 7q11.23, is characterized by hypersociability and enhanced affective empathy. However, the specific genetic and neural mechanisms within the WBS locus underlying this elevated empathic response remain unknown. Here, we investigated empathy-related behaviors, including observational fear and allogrooming, in WBS mouse models harboring a deletion within the conserved syntenic region on mouse chromosome 5. We demonstrate that WBS mice exhibited emotional contagion and prosocial conso

BACKGROUND: Rearing a child with Angelman syndrome (AS) poses significant maternal burden. This study examines whether social support–including family, friends, and significant other dimensions–is associated with maternal caregiving burden among primary caregivers of children with AS in mainland China, and whether caregivers’ self-stigma mediates this relationship. METHODS: In the cross-sectional study, a total of 281 primary caregivers (263 mothers) of children with AS in China completed a questionnaire assessing sociodemographic factors, social support (MSPSS-12), self-stigma (ASS-22), and c

Fragile X syndrome (FXS) is a genetic neurodevelopmental disorder characterized by cognitive impairment, behavioral dysregulation, and emotional lability. While it is primarily known as a leading cause of inherited intellectual disability, patients with FXS may also present with complex neuropsychiatric symptoms including attention-deficit/hyperactivity disorder (ADHD), anxiety, autistic features, irritability, and self-injurious behaviors as they progress through childhood and adolescence. As such, these patients require close monitoring and routine adjustment of behavioral and pharmacologic
