Identifying genetic contributions of 6q21 loci and PPAR pathway to antipsychotic-induced metabolic syndrome: a Sex-Stratified Multi-Omics study.

Antipsychotics-induced metabolic syndrome (APs-induced MetS) is a common side-effect of antipsychotics, significantly increasing the risk of cardiovascular diseases and mortality. However, the genetic risk factors underlying APs-induced MetS remain poorly understood. Thus, we conducted a sex-stratified genome-wide association study (GWAS) in 3067 patients from Schizophrenia In Non-Occidental participants (SINO) trial, and significant results were validated in an independent cohort (all samples = 200) and proteomic data. Post-GWAS analyses were used to further explore the genetic mechanisms inv

Recurrent and Non-Recurrent Copy Number Variants in Native Americans and a Cosmopolitan Sample in Relation to Alcohol Use Disorder and Other Psychiatric Diseases.

Copy number variants (CNVs) can alter disease susceptibility by gene deletion, duplication, and other mechanisms, and have been implicated in neuropsychiatric diseases. However, their rarity or de novo nature impedes linkage analysis. Therefore, we identified recurrent CNVs (rCNVs) in Native Americans with low genetic admixture and high prevalence of alcohol use disorder (AUD) and other psychiatric disorders. Large (> 200 kb) rCNVs were abundant in Plains Indians (PI) and Southwest American Indians (SWI), almost all carrying at least one rCNV, with some CNVs found in both geographically and li

The insertion of an ATTTC repeat in an Alu element hyperactivates a neurodevelopmental enhancer in spinocerebellar ataxia type 37.

Alu elements are evolutionarily very old primate-specific interspersed repeat elements that constitute ∼11% of the human genome. They are a source of short tandem repeats (STRs), which often expand in size and cause inherited neuromuscular and neurodegenerative disorders. How expanded STR insertion mutations within Alu STRs culminate in disease remains unknown. Here, we report an Alu STR located in an intron of DAB1 that functions as a neurodevelopmental enhancer. We demonstrate that an ATTTC repeat insertion in this DAB1 Alu STR, known to cause spinocerebellar ataxia type 37 (SCA37), hyperact

Clinical Utility of Prenatal cfDNA Screening for Sex Chromosome Aneuploidies: A Single Center Experience.

INTRODUCTION: To assess the clinical efficacy of prenatal cfDNA screening for detecting sex chromosome aneuploidies (SCAs). MATERIAL AND METHODS: We conducted a retrospective study at a single center including women with singleton pregnancies who underwent prenatal cfDNA screening between January 2020 and December 2024. Cases with high-risk cfDNA screening results for SCAs were reviewed for confirmatory diagnostic results, prenatal ultrasound findings, and pregnancy outcomes. Positive predictive values (PPVs) were calculated for each SCA subtype, and associations between ultrasound findings a

The selective 5-HT1A receptor biased agonist, NLX-101, corrects anomalous behavioral phenotype in a mouse model of fragile X syndrome.

Fragile X syndrome (FXS) is the most prevalent X-linked dominant autism spectrum disorder, causing a range of developmental problems, notably characterized by mild-severe mood/cognitive dysfunctions. NLX-101 is a highly selective and fully efficacious biased agonist at post-synaptic 5-HT1A receptors, and has shown efficacy for reversal of sensory hypersensitivity and EEG anomalies in transgenic mouse models of FXS. Presently, we examined the ability of NLX-101 to "normalize" (i.e., restore to levels observed in wild-type mice) several aspects of behavioral anomalies displayed by adult male FMR

Interplay between DYRK1A dosage and aneuploidy-induced neuropathology in Down syndrome.

A reduction in the number of neurons and an increased susceptibility to apoptosis are one of the characteristic features observed in Down syndrome (DS). However, the multifaceted actions of chromosomal aneuploidy hinder the elucidation of the underlying mechanism. Here, using neurons and astrocytes differentiated from patient-derived induced pluripotent stem cells (iPSCs), we aimed to clarify the neuropathology of DS by focusing on aneuploidy-associated stress and the dosage effects of specific genes. Human chromosomal trisomies 13, 18, and 21 exert a stress response on neurons, resulting in t

Morphogen Gradients as Drivers of Mosaicism During Early Human Development.

WNT, BMP, FGF, and Nodal signalling gradients drive naive to primed epiblast transitions and primitive endoderm specification, as well as subsequent gastrulation of the implanted embryo. Recently, these pathways were shown to control a signalling rheostat that modulates chromosome replication and segregation fidelity in human pluripotent stem cells. In particular, WNT and BMP antagonists associated with embryo anteriorization during gastrulation (DKK1, Cerberus, LEFTY2, Noggin, and Chordin) induce DNA replication stress and damage in S-phase leading to ultra-fine-bridges and whole-chromosome m

Hyperphagia in craniopharyngioma- a real-world study from the international hypothalamic-pituitary brain tumors patient registry.

We previously showed that hyperphagia affects quality of life of craniopharyngioma (CP) survivors. Characteristics of hyperphagia are unknown. This study aimed to characterize and quantify hyperphagia in CP survivors with and without hypothalamic obesity (HO). Forty caregivers responded to online questionnaires including the Hyperphagia Questionnaire for Clinical Trials (HQ-CT) and Food Safe Zone (FSZ). Caregivers reported significantly more comorbidities in survivors with HO, with 82% presenting with hypothalamic syndrome. Survivors with HO had higher hunger, lower satiety, and higher food pr

Deletion in CACNA1F gene causes X-linked progressive retinal atrophy in English Cocker Spaniel dogs.

BACKGROUND: Progressive retinal atrophy (PRA) was diagnosed in four related male English Cocker Spaniels (ECS) between the ages of 3 and 4 years. All affected dogs were born to clinically unaffected parents that had tested negative for known PRA-causing variants through commercial vendors. Pedigree analysis revealed that the mothers of the affected dogs were paternal half-siblings, and their father had been reported as visually impaired, suggesting an X-linked recessive mode of inheritance. RESULTS: To identify the underlying genetic cause, we performed homozygosity mapping, identifying six ca

SOX9 gene anomalies and campomelic / acampomelic campomelic dysplasia: case report and literature review.

Campomelic dysplasia (CD) is a rare skeletal disorder characterized by the hallmark sign of bent femur or tibial bones or both. Subsequently, patients were identified as having features of CD but lacking the bent limbs. This constellation was later described as acampomelic campomelic dysplasia (ACD). Both CD and ACD are caused by anomalies in the SOX9 gene. Historically, CD has a high mortality rate, and ACD patients are often described similarly. Parents may be counseled to consider pregnancy termination or palliative care. This manuscript describes an index patient with ACD who exhibits prol

Female iPSC X-chromosome inactivation (XCI) erosion and its transcriptomic effects during CRISPR gene editing and neural differentiation.

Human induced pluripotent stem cells (hiPSC) and iPSC-differentiated neural cells, in combination with CRISPR editing, are commonly used for studying neurodevelopmental and other brain disorders. Female iPSCs undergo random X-chromosome inactivation (XCI) via epigenetic silencing by noncoding X inactive specific transcript (XIST). It is known that female iPSCs may lose XIST expression, leading to XCI erosion that affects both X-linked and autosomal gene expression. However, the effects of CRSIPR editing and neural differentiation on XCI erosion in iPSC-derived neurons and how this may confound

Characterization of Social and Repetitive Behaviors of Mllt11/Af1q/TcF7c Conditional Knockout Mice.

Mllt11 (myeloid/lymphoid or mixed-lineage leukemia translocated to chromosome 11; also known as Af1q/TcF7c) has been identified as a novel regulator of neural development, playing a role in the migration and outgrowth of cortical projection neurons. We previously reported that the conditional inactivation of the Mllt11 gene in the mouse superficial cortex resulted in reduced connectivity of the corpus callosum and white matter fiber tracts, resulting in reduced cortical thickness. However, the behavioral consequences of Mllt11 loss are unknown. Callosal abnormalities are thought to be present

Advancing genotype-phenotype analysis through 3D facial morphometry: insights from Cri-du-Chat syndrome.

PURPOSE: Facial dysmorphism is a feature of many monogenic disorders and is important in diagnostics, variant interpretation and nosology. Nevertheless, comprehensively assessing the complex facial shape changes associated with specific syndromes remains challenging. Here, we present three-dimensional (3D) morphometric approaches to overcome these limitations, using Cri-du-Chat syndrome (CdCS) as a model. METHODS: We analysed 3D facial images from 24 participants with CdCS, 4540 unaffected controls and five participants with rare 5p15.33-15.32 deletions, incorporating two methods to account f

Semaglutide Plus Low-Dose Metformin Combination Therapy for the Treatment of Obesity and Prediabetes in a Woman with Partial Deletion of the X Chromosome Long Arm.

Background and Clinical Significance: Over the last two decades, glucagon-like peptide-1 (GLP-1) receptor agonists have dramatically improved the management of type 2 diabetes mellitus and obesity. Currently, little is known about the use of semaglutide (a second-generation GLP-1 receptor agonist) in patients with X chromosome abnormalities. Herein, we describe the therapeutic use of semaglutide in a woman with a partial deletion of the X chromosome long arm (partial Xq deletion) and comorbid obesity. We also conducted a narrative mini-review on overweight, obesity and common metabolic derange

[Maternal serum markers profiles for Down syndrome: Review of comments and actionable recommendations].

In France, for the vast majority of patients, screening for Down syndrome using circulating cell-free fetal DNA is contingent upon prior assessment of maternal serum markers and calculation of a specific risk estimate for this aneuploidy. Certain marker profiles may also suggest other fetal or maternal pathologies. This article therefore aims: (i) to clarify and explain any comments that may appear in laboratory reports; (ii) to propose a clinical interpretation and a procedure to be followed for the monitoring of the considered pregnancy. It updates a first article published in 2014 and, like

Human Endogenous Retroviruses and Epigenetic Regulators Are Dysregulated in Beckwith-Wiedemann Syndrome.

Beckwith-Wiedemann syndrome (BWS) is an overgrowth disorder caused by genetic and epigenetic alterations at chromosome 11p15.5. Increasing evidence suggests that imprinting defects may be accompanied by broader epigenomic perturbations affecting repetitive elements such as human endogenous retroviruses (HERVs). We quantified the transcriptional levels of the HERV-H, HERV-K, and HERV-W-pol genes, the HERV-derived env genes, Syncytin-1 (SYN1) and Syncytin-2 (SYN2), and the epigenetic regulators, TRIM28 and SETDB1, in whole blood from children and adolescents with BWS, stratified by molecular sub

Down Syndrome and Obesity Management: Lifestyle Observations Made in the Framework of the GO-DS21 Project and Clinical Recommendations.

Introduction/background: Individuals with Down syndrome (DS) face a higher and earlier risk of overweight and/or obesity than the general population, influenced by factors like limited physical activity, suboptimal diet, stress, and sleep disorders. Despite the impact these lifestyle factors can have, there is a significant lack of robust, evidence-based guidelines for managing overweight and obesity in these individuals, particularly for the adult population. Results: Based on findings from an observational study across three European sites, within the EU Horizon-2020 GO-DS21 project, in this

The inactive X chromosome as a female protector in autism and beyond.

Female individuals require greater autosomal genetic risk than male individuals to manifest autism, yet the biological basis of this 'female protective effect' (FPE) remains unknown. Based on insights into the human sex chromosomes, we propose that the FPE arises from higher expression of a subset of X-linked genes in females due to transcription from both the active (Xa) and inactive (Xi) X chromosomes. These higher expression levels enable females to buffer mutations in autosomal genes and in pathways regulated by Xi-expressed genes. This framework unifies epidemiological, genetic and mechan
