Liver sinusoidal endothelial cells (LSECs) regulate nutrient flux and immune surveillance within the hepatic niche, yet how they function as metabolic stress sensors that instruct adaptive immune remodeling during metabolic dysfunction-associated steatotic liver disease (MASLD) remains unclear. Here, single-nucleus transcriptomics of human MASLD revealed stage-dependent activation of cyclic GMP-AMP synthase/stimulator of interferon genes (cGAS/STING) signaling in LSEC comparable with that in macrophages, with endothelial activation showing greater responsiveness to metabolic stress. Schaaf-Yang syndrome and Prader-Willi syndrome are imprinting disorders that result from the disruption of paternally expressed genes within the 15q11-q13 region. Both conditions present with overlapping clinical features including developmental delay, hypotonia and endocrine abnormalities. Schaaf-Yang syndrome specifically results from heterozygous variants in the paternally expressed MAGEL2 gene. Because these variants are often de novo, determining that the variant is on the paternal allele is essential for a definitive diagnosis. Traditional methods, such as methylation-specific PCR, are l

Understanding how chromosome 21 gene dosage contributes to neurodevelopmental phenotypes in trisomy 21 (T21) remains a fundamental challenge. Here, we perform transcriptome-wide RNA-sequencing of fetal cortical and hippocampal tissues from T21 cases and euploid controls collected during mid-gestation, a critical window for human brain development. We identify widespread gene expression dysregulation with significant enrichment for chromosome 21 genes and perturbation of neurodevelopmental, synaptic, and immune-related pathways. Among the most strongly dysregulated genes is ADARB1, a chromosome

BACKGROUND: Most women experience hot flashes (hot flushes) during the menopause transition. Menopausal hot flashes typically persist for years, and for a sizeable minority of women, are substantially impairing. Genetic investigations can improve understanding of hot flash etiology.
METHODS: We conducted a trans-ancestry genome-wide association study (GWAS) of hot flashes (N = 149,560) among post-menopausal women age 35-88. The outcome variable was self-reported hot flashes in four samples (total n = 42,489) and menopausal hormone therapy as a proxy in one sample (n = 107,071). We estimated t

BACKGROUND: Fragile X Syndrome (FXS) is a rare X-linked genetic condition. Children with FXS exhibit a variety of challenging behaviors such as irritability, anxiety, and preference for solitary activities. The Aberrant Behavior Checklist-Community (ABC-C) is an observer-reported outcome (ObsRO) measure that captures many of the behavioral concerns (e.g., outbursts, social anxiety, difficulty connecting with others) salient to caregivers of individuals with FXS. An FXS-specific scoring algorithm of the ABC-C, considered more representative of the FXS phenotype (ABC-CFXS), has been included in

Fragile X syndrome (FXS) is characterized by sensory hypersensitivity and neural hyperexcitability linked to GABAergic dysfunction. BAER-101, formerly known as AZD7325, is a positive allosteric modulator of GABAA receptors with functional selectivity for α2/α3 subunits, designed to enhance inhibitory tone while minimizing sedation associated with α1 activation. This study evaluated the acute and chronic effects of BAER-101 on behavioral and electrophysiological outcomes in individuals with FXS. The study employed a within-subject 3 × 2 factorial design, with three dosing conditions (Low Dose:

PURPOSE: Prader-Willi syndrome (PWS) is a complex imprinting disorder associated with severe obesity and endocrine dysfunction, both contributing to increased cardiovascular morbidity. Emerging data suggest a disproportionately high incidence of thromboembolic events in PWS, potentially implicating an intrinsic hypercoagulable state.
METHODS: We conducted a cross-sectional, case-control study including 49 genetically confirmed PWS patients (22 pediatric and 27 adult) and 85 age-, sex-, and body-mass-index-matched controls. Participants underwent comprehensive hemostatic assessment including s

BACKGROUND: Fragile X syndrome (FXS) is a neurodevelopmental disorder caused by loss of fragile X messenger ribonucleoprotein (FMRP). In most cases, this results from a CGG expansion exceeding 200 repeats in the 5' untranslated region of the fragile X messenger ribonucleoprotein 1 (FMR1) gene, known as the "full mutation". While the trinucleotide expansion has long been thought to induce epigenetic silencing of this locus, studies have shown that many males with a full mutation still express FMR1 mRNA. However, these individuals produce little to no FMRP protein, due to mechanisms that remain

The integrated stress response (ISR) is essential for cellular homeostasis and cognitive function. We investigated how persistent ISR activation affects cognitive performance by studying the PPP1R15BR658C genetic variant associated with intellectual disability. To model this condition, we generated a mouse line with the pathogenic allele inserted. This variant destabilized the PPP1R15B•PP1 phosphatase complex, causing persistent ISR activation, impaired protein synthesis, and long-term memory deficits. We demonstrated that the cognitive and synaptic impairments in Ppp1r15bR658C mice arise dire

Alzheimer’s disease (AD) and Down syndrome (DS) are both characterized by early accumulation of amyloid-β (Aβ), but the underlying mechanisms differ. In DS, lifelong overproduction of Aβ due to triplication of the APP gene drives pathology, whereas in sporadic AD (sAD) impaired clearance and altered processing are considered to be major contributors to Aβ pathology. Despite these shared hallmarks, it remains unclear whether the molecular composition of plaques, such as Aβ isoform distribution and post-translational modifications, is truly comparable between the two conditions. The common marmoset is a New World monkey (NWM) commonly used as a model organism to investigate questions in primate evolution and human disease, including Alzheimer's and other neurodegenerative diseases, as well as neuropsychiatric disorders. Here we present the first telomere-to-telomere (T2T) reference genome for the common marmoset, adding over 88 Mb of sequence and resolving challenging genomic regions. An additional near-T2T assembly from a second unrelated individual yields a total of four high-quality haplotypes for analysis. The improved contiguity and accuracy of these assemblies e

The mortality rates attributed to cardiovascular diseases (CVD) are increasing within the United States. Atherosclerotic cardiovascular disease (ASCVD) and pulmonary arterial hypertension (PAH) are two severe, life-threatening subtypes of CVD. Although ASCVD and PAH are distinct vascular disorders, they share common mechanisms, including endothelial dysfunction, inflammation, smooth muscle proliferation, fibrosis, and vascular remodeling. It is noteworthy that patients diagnosed with PAH may have underlying atherosclerotic coronary artery disease at a rate of ∼28%, and conversely, patients wit

The mechanistic role of trisomy 8 in the development of myelodysplastic syndrome (MDS) remains poorly defined. Here, we generated a trisomy 8 mouse model by transferring a human chromosome 8 into murine embryonic stem cells and prospectively examined the effects on hematopoietic stem cells (HSC) by trisomy 8. The expression of inflammatory genes was enhanced, and hematopoietic programs mediated by transcription factors and polycomb repressive complex 2 (PRC2) were dysregulated in trisomy 8 HSC, which impaired their self-renewal and balanced differentiation. Trisomy 8 HSC altered the chromatin

BACKGROUND: Chronic myeloid leukemia (CML) is defined by the presence of the BCR::ABL1 fusion gene resulting from the t(9;22)(q34;q11.2) translocation. In addition to this primary rearrangement, secondary chromosomal abnormalities involving chromosomes 9 and 22 may arise during clonal evolution and influence disease progression and response to therapy. Pericentric inversion of chromosome 9 is usually regarded as a constitutional variant. However, when acquired in CML, particularly involving the derivative chromosome 9, it represents a rare secondary abnormality whose biological and clinical si

BACKGROUND: The World health organization (WHO) identifies Down syndrome (DS) as one among common congenital disorders, along with congenital heart defects. Approximately fifty percent of children diagnosed with DS are affected by congenital heart defects which significantly impact their survival. Nevertheless, in low and middle income countries, there is limited published data on congenital heart defects in children with DS and their treatment outcomes. Therefore, this study aimed to document the clinical characteristics and hospital outcomes of children with DS who were diagnosed with heart

BACKGROUND: Germ cell tumors are heterogeneous neoplasms arising from primordial germ cells. Although genome-wide association studies have identified numerous susceptibility loci for adult testicular germ cell tumors, the heritable basis of pediatric testicular germ cell tumors and germ cell tumors that arise outside the testes remain poorly understood.
METHODS: We conducted a multi-ancestry genome-wide association study of pediatric germ cell tumors, including 1927 cases from the Germ Cell Tumor Epidemiology Study and 10 601 controls. Cases were diagnosed with testicular (n = 678), ovarian (

BACKGROUND: Neurodevelopmental disorders have a strong male bias that is poorly understood. The placenta provides molecular information about environmental interactions with genetics (including biological sex) that shape developmental processes in the brain. We investigate placental-brain transcriptional responses in an established mouse model of prenatal exposure to a human-relevant mixture of polychlorinated biphenyls (PCBs).
RESULTS: To understand sex, tissue, and dosage effects in embryonic (E18) brain and placenta RNAseq data, we use weighted gene correlation network analysis (WGCNA) to
