Camp-based entertainment rehabilitation for young people with down syndrome in Saudi Arabia.

BACKGROUND: Down syndrome (DS) is associated with lifelong challenges across cognitive, physical, and social domains that can impact quality of life (QoL). Despite research evidence supporting the benefits of recreational and entertainment-based rehabilitation, such programmes remain underdeveloped in the Middle East. This study examined cultural appropriateness, and impact of a camp-based entertainment rehabilitation programme for young people with DS in Saudi Arabia. METHODS: This study was implemented with 11 participants (aged 17-23 years) attending an 11-day residential camp in Al-Baha.

Features of Biochemical and Hematological Parameters and Chromosomal Disorders in Lymphocytes of Aging Primates of the Kurchatovsky Complex of Medical Primatology.

Aging is a key challenge for modern society. In particular, brain aging is accompanied by chronic inflammation, depletion of energy potential, and an increased level of oxidative stress, with changes in blood composition playing a special role in this process. Recent studies also show that aging progresses non-linearly throughout life. Primates are genetically and anthropometrically the closest laboratory animals to humans, thus representing the most accurate model for research. This study establishes baseline values for biochemical parameters (including the state of the body's antioxidant sys

Oxytocin in infants with Prader-Willi syndrome to improve dysphagia and disease trajectory.

OBJECTIVE: Prader-Willi syndrome (PWS) is a genetic neurodevelopmental disorder with a characteristic trajectory. Infants display hypotonia, poor social and feeding skills, and high risk of choking, which have been shown to improve after oxytocin (OT) treatment. Our aim is to demonstrate the efficacy of intranasal OT treatment administered in the postnatal critical period on infant feeding skills and document its long-term effects. METHODS: We enrolled 52 infants with PWS (median age 2.2 months) in a European double-blind randomized placebo-controlled study. Infants were randomly assigned in a

Child Deictic Gesture Use and Maternal Labeling in Toddlers With Down Syndrome.

PURPOSE: The purpose of this study was to determine the association between the deictic gesture use of toddlers with Down syndrome (DS) and their mothers' use of labels. Relationships between child and maternal deictic gestures; maternal use of labels; and broader, traditional measures of maternal linguistic input were explored. METHOD: Participants were 25 toddlers with DS (14-31 months) and their mothers. Videos of a 15-min mother-child free-play were coded for child and maternal deictic gesture use. All maternal utterances were transcribed and coded to compute the number of complete and in

McIdas localizes to centrioles and controls centriole numbers through PLK4-dependent phosphorylation.

The centriole duplication cycle must be tightly controlled and coordinated with the chromosome cycle. Aberrations in centriole biogenesis can cause developmental disorders, ciliopathies and cancer, yet the molecular determinants controlling centriole numbers and the link between the two cycles remain poorly characterized. Here, we demonstrate that McIdas, previously implicated in cell cycle regulation and multiciliogenesis, plays a critical role in maintaining proper centriole numbers. McIdas localizes to centrioles, where it exhibits dynamic localization throughout the cell cycle, dependent u

Inflammatory signaling differentially changes chromatin accessibility and gene expression of the PD- associated kinase LRRK2 between human and mice.

The genomic locus that encodes the Leucine-rich repeat kinase 2 (LRRK2) gene is highly pleiotropic, being associated with both Parkinson's disease (PD) and Crohn's disease (CD). Coding variants associated with risk of PD and CD act as gain of function kinase mutations increasing phosphorylation of RAB substrates, and non-coding variants in the promoter region of LRRK2 increase expression of the gene, notably in immune cells. If regulation of LRRK2 expression is a causal contributor to age-related diseases, it is important to understand the mechanism(s) by which LRRK2 is regulated, particularly

Hippocampal glial alterations are associated with Lamin B1 dysregulation and abnormal nuclear morphology in a rat model of fragile X syndrome.

Fragile X syndrome (FXS) is the most common inherited intellectual disability and the leading monogenic cause of autism spectrum disorders (ASD). Although the pathological mechanisms underlying this neurodevelopmental disorder are challenging, recent studies have increasingly highlighted the involvement of glial cells in the pathogenesis of both ASD and FXS. Microglia and astrocytes are critical for brain development and homeostasis; thus, understanding glial dysfunction in both the developing and adult brain in these disorders may reveal novel therapeutic targets beyond the neuro-centric pers

ImageNet pre-training and two-step transfer learning in chromosome image classification.

Chromosome image classification typically relies on ImageNet pre-training, yet the potential of leveraging intermediate domains from related staining techniques remains largely underexplored. Here, we evaluate two-step transfer learning-where classifiers are first fine-tuned on an intermediate domain before targeting the final classification task-across Q-band (BioImLab dataset) and G-band (CIR dataset) chromosome classification. Each dataset serves as intermediate domain for the other. Across 11 architecture families and three training approaches, models achieve improvements when domain simil

UBE3A isoform-selective and non-selective contributions to Angelman syndrome phenotypes.

Angelman syndrome (AS) is a neurodevelopmental disorder caused by UBE3A loss. In humans, UBE3A generates three isoforms that localize to distinct subcellular compartments-one mainly nuclear and two cytoplasmic. The nuclear and most highly expressed cytoplasmic UBE3A isoform are highly conserved in mice, whereas the cytoplasmic human isoform accounting for just ~1% of total UBE3A has no mouse counterpart. Loss of the nuclear-enriched UBE3A isoform causes AS and behavioral deficits in mice; no specific contribution of the predominant cytoplasmic UBE3A isoform has yet been identified. Because the

Growth patterns: Pathology vs. Normal variation.

Growth of children and adolescents follows a universal pattern with three distinct growth spurts. Common medical practice refers to normal child growth as a health and biological advantage, and considers "pathology in growth" as a deviation from the universal pattern. Growth can differ in amplitude and tempo with considerable variation both within and between populations over time, and depending on geographic region. Local growth charts are necessary to adapt the universal pattern of growth to characteristics of a specific historic or modern population. Local charts can visualize the distinct

Behavioral Phenotype Associations With Resting State EEG Signal Complexity and Power Spectral Density in Fragile X Syndrome.

Fragile X Syndrome (FXS), an X-linked genetic condition, is associated with a wide range of phenotypic manifestations, namely intellectual disability (ID), autism spectrum disorder (ASD), attention deficit hyperactivity disorder (ADHD), and atypical behaviors. Individuals with FXS present robust electrophysiological (EEG) alterations, notably on signal complexity and power spectral density (PSD) resting state measures. To which extent they are associated to specific behavioral phenotypes in the FXS population remains to be comprehensively addressed. This study aimed to investigate the relation

Maternal mixed UPD3 and a homozygous PLXNA1 c.2497G>C variant in a fetus with severe anomalies.

BACKGROUND: Non-invasive prenatal testing (NIPT) is widely used for screening common fetal aneuploidies such as trisomy 21 (T21), trisomy 18 (T18), and trisomy 13 (T13). However, its utility in detecting trisomy 3 (T3) has been rarely reported. Furthermore, uniparental disomy (UPD) involving chromosome 3 is a rare genetic condition with potential phenotypic consequences. METHODS: NIPT indicated a high risk for fetal T3. This finding was further investigated using copy number variation (CNV) analysis via trio-based chromosomal microarray analysis (trio-CMA). Subsequent trio-based whole-genome

Angiotensin II type 1 receptor autoantibody (AT1-AA) is involved in ovulatory disorders: AT1-AA impairs spindle and chromosome organization.

Ovulatory disorders are a major cause of infertility. Studies have shown that the renin-angiotensin system (RAS) plays a vital role in the female reproductive system. The angiotensin II type 1 receptor (AT1R), an important mediator of the RAS, may induce anovulation when activated. Previous studies have shown that the levels of AT1R autoantibody (AT1-AA) are significantly higher in the serum of infertile women and are negatively correlated with the oocyte maturation rate. However, systematic understanding of the changes of AT1-AA associated with female ovulatory dysfunction remains limited. In

Condensin complexes: from chromatin organization to disease.

Condensin complexes are indispensable for mitotic chromosome condensation. In this review, we summarize their structural and functional features, focusing on ATP-dependent loop extrusion and mitotic chromatin folding. We discuss current models of mitotic chromosome formation with an emphasis on the emerging role of condensin in suppressing interphase chromatin contact patterns during mitosis. Additionally, we outline regulatory mechanisms governing condensin activity, including phosphorylation-dependent and -independent regulation of chromatin loading and unloading. Finally, we connect condens

Bioisostere-Driven Discovery of SePP: A Selenium-Containing Polypharmacological Agent Relevant to Fragile X Syndrome.

Diphenidine is a prototypical 1,2-diarylethylamine that functions as an uncompetitive N-methyl-d-aspartate receptor (NMDAR) antagonist and monoamine reuptake inhibitor. To examine the effects of phenyl-ring bioisosteric replacement within this scaffold, a series of diphenidine analogs incorporating chalcogen heterocycles (2-furan, 2-thiophene, 3-thiophene, and 2-selenophene) was synthesized. Compounds were evaluated for in vitro binding to rat forebrain NMDARs and inhibition of human DAT, NET, and SERT in cell-based assays, enabling assessment of polypharmacology. In silico analyses (molecular

Genotoxicity and DNA Damage in Long-Term SSRI Therapy: A Review Across SSRIs With Citalopram as a Case Study.

Selective serotonin reuptake inhibitors (SSRIs) are first-line therapies for depressive and anxiety disorders and are often used for years, including in reproductive-age patients and during pregnancy. This has intensified concern about possible genotoxicity and DNA damage. Citalopram is emphasized because it has the most extensive and consistently positive in vivo genotoxicity evidence among SSRIs, particularly in rodent comet and micronucleus endpoints. This review summarizes genotoxicity data for SSRIs, focusing on citalopram, across in vitro assays, animal studies, human biomonitoring, and
