ImageNet pre-training and two-step transfer learning in chromosome image classification.

Chromosome image classification typically relies on ImageNet pre-training, yet the potential of leveraging intermediate domains from related staining techniques remains largely underexplored. Here, we evaluate two-step transfer learning-where classifiers are first fine-tuned on an intermediate domain before targeting the final classification task-across Q-band (BioImLab dataset) and G-band (CIR dataset) chromosome classification. Each dataset serves as intermediate domain for the other. Across 11 architecture families and three training approaches, models achieve improvements when domain simil

UBE3A isoform-selective and non-selective contributions to Angelman syndrome phenotypes.

Angelman syndrome (AS) is a neurodevelopmental disorder caused by UBE3A loss. In humans, UBE3A generates three isoforms that localize to distinct subcellular compartments-one mainly nuclear and two cytoplasmic. The nuclear and most highly expressed cytoplasmic UBE3A isoform are highly conserved in mice, whereas the cytoplasmic human isoform accounting for just ~1% of total UBE3A has no mouse counterpart. Loss of the nuclear-enriched UBE3A isoform causes AS and behavioral deficits in mice; no specific contribution of the predominant cytoplasmic UBE3A isoform has yet been identified. Because the

Growth patterns: Pathology vs. Normal variation.

Growth of children and adolescents follows a universal pattern with three distinct growth spurts. Common medical practice refers to normal child growth as a health and biological advantage, and considers "pathology in growth" as a deviation from the universal pattern. Growth can differ in amplitude and tempo with considerable variation both within and between populations over time, and depending on geographic region. Local growth charts are necessary to adapt the universal pattern of growth to characteristics of a specific historic or modern population. Local charts can visualize the distinct

Behavioral Phenotype Associations With Resting State EEG Signal Complexity and Power Spectral Density in Fragile X Syndrome.

Fragile X Syndrome (FXS), an X-linked genetic condition, is associated with a wide range of phenotypic manifestations, namely intellectual disability (ID), autism spectrum disorder (ASD), attention deficit hyperactivity disorder (ADHD), and atypical behaviors. Individuals with FXS present robust electrophysiological (EEG) alterations, notably on signal complexity and power spectral density (PSD) resting state measures. To which extent they are associated to specific behavioral phenotypes in the FXS population remains to be comprehensively addressed. This study aimed to investigate the relation

Exploring the pathogenicity and genotype-phenotype correlation of 16p13.11 microduplication: a report of two cases.

BACKGROUND: 16p13.11 microduplication syndrome is a rare genomic disorder caused by an extra copy of a small segment of DNA on the short arm of chromosome 16 at a specific location in band 13.11. The clinical presentation is highly variable. Some individuals are entirely asymptomatic, while others present with a spectrum of neurodevelopmental disorders. CASE DESCRIPTION: We report two cases with 16p13.11 microduplication to investigate the genotype-phenotype correlations. Peripheral blood was collected from the two patients and their respective parents and all participants underwent whole exo

Maternal mixed UPD3 and a homozygous PLXNA1 c.2497G>C variant in a fetus with severe anomalies.

BACKGROUND: Non-invasive prenatal testing (NIPT) is widely used for screening common fetal aneuploidies such as trisomy 21 (T21), trisomy 18 (T18), and trisomy 13 (T13). However, its utility in detecting trisomy 3 (T3) has been rarely reported. Furthermore, uniparental disomy (UPD) involving chromosome 3 is a rare genetic condition with potential phenotypic consequences. METHODS: NIPT indicated a high risk for fetal T3. This finding was further investigated using copy number variation (CNV) analysis via trio-based chromosomal microarray analysis (trio-CMA). Subsequent trio-based whole-genome

Angiotensin II type 1 receptor autoantibody (AT1-AA) is involved in ovulatory disorders: AT1-AA impairs spindle and chromosome organization.

Ovulatory disorders are a major cause of infertility. Studies have shown that the renin-angiotensin system (RAS) plays a vital role in the female reproductive system. The angiotensin II type 1 receptor (AT1R), an important mediator of the RAS, may induce anovulation when activated. Previous studies have shown that the levels of AT1R autoantibody (AT1-AA) are significantly higher in the serum of infertile women and are negatively correlated with the oocyte maturation rate. However, systematic understanding of the changes of AT1-AA associated with female ovulatory dysfunction remains limited. In

Condensin complexes: from chromatin organization to disease.

Condensin complexes are indispensable for mitotic chromosome condensation. In this review, we summarize their structural and functional features, focusing on ATP-dependent loop extrusion and mitotic chromatin folding. We discuss current models of mitotic chromosome formation with an emphasis on the emerging role of condensin in suppressing interphase chromatin contact patterns during mitosis. Additionally, we outline regulatory mechanisms governing condensin activity, including phosphorylation-dependent and -independent regulation of chromatin loading and unloading. Finally, we connect condens

Bioisostere-Driven Discovery of SePP: A Selenium-Containing Polypharmacological Agent Relevant to Fragile X Syndrome.

Diphenidine is a prototypical 1,2-diarylethylamine that functions as an uncompetitive N-methyl-d-aspartate receptor (NMDAR) antagonist and monoamine reuptake inhibitor. To examine the effects of phenyl-ring bioisosteric replacement within this scaffold, a series of diphenidine analogs incorporating chalcogen heterocycles (2-furan, 2-thiophene, 3-thiophene, and 2-selenophene) was synthesized. Compounds were evaluated for in vitro binding to rat forebrain NMDARs and inhibition of human DAT, NET, and SERT in cell-based assays, enabling assessment of polypharmacology. In silico analyses (molecular

Genotoxicity and DNA Damage in Long-Term SSRI Therapy: A Review Across SSRIs With Citalopram as a Case Study.

Selective serotonin reuptake inhibitors (SSRIs) are first-line therapies for depressive and anxiety disorders and are often used for years, including in reproductive-age patients and during pregnancy. This has intensified concern about possible genotoxicity and DNA damage. Citalopram is emphasized because it has the most extensive and consistently positive in vivo genotoxicity evidence among SSRIs, particularly in rodent comet and micronucleus endpoints. This review summarizes genotoxicity data for SSRIs, focusing on citalopram, across in vitro assays, animal studies, human biomonitoring, and

RO4938581, a GABAA-α5 negative allosteric modulator rescued behavioral and EEG phenotypes of a mouse model of Dup15q syndrome.

We investigated the correlation between increased gene copy number of gamma aminobutyric acid type A (GABAA) receptor α5-containing subunits and electrophysiological and behavioral phenotypes in a mouse model of Dup15q syndrome (15q dup) and tested the hypothesis that selectively inhibiting the activity of GABAA-α5 receptors may have therapeutic effects. Dup15q syndrome is a rare neurodevelopmental disorder caused by copy number gains of the 15q11.2-q13.1 chromosomal region, which includes UBE3A and a cluster of three genes (GABRA5, GABRB3, and GABRG3) encoding GABAA receptor subunits, all of

Genomic Imprinting, Epigenetic Dysregulation, and Neuropsychiatric Mechanisms in Prader-Willi Syndrome: A Multi-Level Integrative Review.

Prader-Willi syndrome (PWS) is a rare imprinting-related neurodevelopmental disorder caused by loss of paternally expressed genes within the chromosome 15q11-q13 region, including SNORD116, MAGEL2, and NDN. It provides a natural model for examining how genomic imprinting disruptions shape neural development and psychiatric vulnerability. This review synthesizes current evidence to clarify the mechanistic pathways linking imprinting defects and epigenetic dysregulation to neuropsychiatric outcomes in PWS. Published studies-including patient-derived induced pluripotent stem cell (iPSC) models, a

Integrative modeling to improve bleeding risk prediction in adult female hemophilia A carriers.

BACKGROUND: Although bleeding phenotypes and factor (F)VIII activity (FVIII:C) in men with hemophilia A correlate highly with F8 mutations, FVIII:C in female HA carriers is more variable due to mosaicism arising from a second wild-type F8 allele and X-chromosome inactivation (XCI). Conventionally, only female HA carriers with low FVIII:C were considered at risk for bleeding. However, hemostatic FVIII:C levels in females remain unestablished. Moreover, HA carriers with normal FVIII:C levels can also have reproductive tract and other bleeding sequelae. OBJECTIVES: We sought to develop a statist

Spatial abilities in aging adults with Down syndrome.

INTRODUCTION: People with Down syndrome (DS) have a higher likelihood of developing Alzheimer's disease (AD). Deficits in spatial abilities could serve as early indicators of AD. We examined age effects on visuospatial construction and visuomotor integration in DS and whether spatial tasks could distinguish various extents of cognitive decline in DS. METHODS: We used the Alzheimer's Biomarkers Consortium-Down Syndrome project dataset, where 376 DS participants completed a series of cognitive measures. RESULTS: Age effects were found in visuomotor integration but not in visuospatial construct
