Pragmatic weight estimation for dosing of a new paediatric treatment for schistosomiasis prevention and control in preschool-aged children: a feasibility study in Uganda and Côte d'Ivoire.

BACKGROUND: Schistosomiasis remains a major public health concern in endemic settings. The Pediatric Praziquantel Consortium has developed a new paediatric treatment, arpraziquantel, for preschool-aged children. Because the treatment is dosed according to body weight, dose determination by scales poses operational challenges in community-based mass treatment campaigns. This study assessed the performance of the Pediatric Advanced Weight Prediction in the Emergency Room Extra-Long Mid-Arm Circumference (PAWPER XL-MAC) tape for weight estimation during mass drug administration in Uganda and Côte

Cross-sectional study of prevalence and economic importance of bovine fasciolosis at Kombolcha Elfora Abattoir, Amhara Region, Northern Ethiopia.

The study was conducted from October 2024 to June 2025 at the Kombolcha ELFORA abattoir and Processing Plant to assess the prevalence and economic impact of bovine Fasciolosis in cattle sourced from three agroecological zones in northern Ethiopia. From the 382 examined bovine samples, 184 faecal samples (48.17%) and 195 livers (51.05%) tested positive for Fasciolosis. Infection rates were highest in cattle with poor body condition (51.55% in faecal samples and 54.64% in livers), followed by medium (47.62% and 50%) and good body condition groups (20% and 25%, respectively), with significant dif

Development of a novel organoid-based two-dimensional system to model the intestinal host-parasite interface in fasciolosis.

BACKGROUND: The earliest interactions between Fasciola hepatica newly excysted juveniles (FhNEJ) and the intestinal epithelium remain poorly understood, partly due to the limited availability of physiologically relevant in vitro models. Here, we establish a human duodenum organoid-derived epithelial monolayer (ODM) co-culture system to investigate the early host-parasite interface, providing a controlled experimental platform for analysing interactions between FhNEJ and the intestinal epithelium. METHODS: ODMs representing the human duodenum were co-cultured with either live FhNEJ or a tegume

Epidemiology of helminth infections in adults in rural rainforest communities of the Nkam-Wouri River Basin, Littoral Region of Cameroon: Implications for NTDs elimination using preventive chemotherapies.

BACKGROUND: Chronic helminth infections cause serious health problems; however, limited data on prevalence and distribution hinder effective control interventions. This study assessed the prevalence, distribution, and risk factors of helminth infections in adults across four health districts (HD) in the Littoral Region of Cameroon, while hypothesizing that helminth infection prevalences are significantly higher in densely forested and wetland forest plantation districts compared to degraded and mosaic plantation forest districts despite over more than two decades of preventive chemotherapy. Over-dispersion in malaria-schistosoma co-infection: insights from a meta-analytical approach and systematic review.

BACKGROUND: Malaria and schistosomiasis are two major parasitic diseases that are co-endemic in many regions of sub-Saharan Africa. Despite their frequent geographical overlap, the potential epidemiological patterns and potential interactions associated with Plasmodium--Schistosoma co-infection remain poorly characterized. This study aimed to systematically assess the epidemiology of malaria-schistosomiasis co-infection and to quantify whether co-infections occur more frequently than expected under an assumption of independence between the two infections. METHODS: We conducted a systematic re

Population Pharmacokinetics and Exposure-Response Analysis of Arpraziquantel Supporting Dose in Preschool-Aged Children Infected with Schistosoma.

Schistosomiasis is an acute and chronic parasitic disease that occurs mainly in sub-Saharan and African countries and for which no treatment is available in preschool-aged children (PSAC). An open-label, Phase III study of arpraziquantel pediatric formulation evaluated safety and efficacy in 280 Schistosoma-infected PSAC (NCT03845140). A population pharmacokinetics (PK) model for arpraziquantel and its active metabolite trans-4-hydroxy-arpraziquantel was developed from concentration measurements (from n = 74 and n = 30 children for arpraziquantel and its metabolite, respectively) after a singl

Morphology and molecular characterisation of two new species of Mymarothecioides (Monopisthocotyla: Dactylogyridae) parasitising the gills of anostomid fishes from Brazil.

Using an integrative taxonomic approach, we describe two new species of Monopisthocotyla, Mymarothecioides leporini n. sp. and Mymarothecioides paranaensis n. sp., parasitising the gills of anostomid fishes from the Jaguaribe River and Paraná River basins, Brazil. These descriptions represent the first record of species of Mymarothecioides in hosts of the family Anostomidae. Mymarothecioides leporini n. sp. is distinguished by a male copulatory organ (MCO) with a spoon-shaped distal portion and a ventral bar with an ornamented anteromedial process. Mymarothecioides paranaensis n. sp. differs f

Novel environmental risk index reveals uneven impact of climate change on fascioliasis risk across Africa and Europe.

Fascioliasis, a zoonotic parasitic disease caused by the liver flukes Fasciola hepatica and Fasciola gigantica, poses a significant threat to livestock and human health globally. Climate change is altering the transmission dynamics of this disease; however, the direction and magnitude of this change remain uncertain due to a lack of mechanistically grounded predictions at continental scale. In this study, we developed a novel risk index to assess how climate change may affect the potential for fascioliasis transmission across Africa and Europe. The index integrates the following three componen

"We don't know schistosomiasis can affect the female reproductive system and increase the risk of HIV": A mixed-methods analysis reveals gaps in risk factor determinants and health consequences of female genital schistosomiasis in Southern Chad.

BACKGROUND: Urogenital schistosomiasis caused by Schistosoma haematobium infection is a public health problem in Chad. Despite reports of urinary S. haematobium prevalence in endemic communities, research on the genital aspect of the disease, which disproportionately affects women, remains neglected especially given the contemporary geographical overlap with HIV/AIDS. This study was undertaken to establish baseline data on the awareness and perception of female genital schistosomiasis (FGS) among community inhabitants and healthcare professionals in southern Chad to inform effective policy dec

Ontogenetic shifts in sound production and shared sonic mechanisms in two priacanthid fishes.

Sound production in teleost fishes relies on diverse anatomical adaptations, yet convergent mechanisms involving extrinsic sonic muscles acting on the swim bladder are widespread. This study investigates the acoustic and morphological features of two priacanthid species, Indo-Pacific glasseye Heteropriacanthus carolinus and moontail bullseye Priacanthus hamrur to explore interspecific similarities in sound production. Using recordings and anatomical analyses, we show that both species rely on a forced-response mechanism, where the contraction rate of fast sonic muscles determines the fundament

Revisiting the diversity of freshwater fish trematode metacercariae in cenotes (sinkholes) of Yucatán: molecular characterisation and comments on their phylogenetic interrelationships.

The limestone soil of the Yucatán Peninsula is highly permeable, allowing fluvial waters to filter into the aquifer, resulting in the absence of rivers on the surface and the formation of underground reservoirs that, on some occasions, open to the surface, forming the so-called cenotes (or sinkholes). Information about the freshwater fish parasite diversity of cenotes in the Yucatán Peninsula is scarce. Only 50 cenotes of the approximately 9,000 occurring in the peninsula have been sampled to assess the fish parasite fauna. Available information predates the molecular era and is based on morph

Occurrence of brachylaimid trematodes, alive and shell-trapped, in sympatric species of terrestrial gastropods in France.

Following the detection of Brachylaima spp., which are potentially pathogenic for domestic animals and humans, in Cornu aspersum and Cepaea nemoralis at a rural site in France, we investigated trematode distribution among sympatric gastropod species to assess their role in the parasite life cycle. Eight species were investigated (Cepaea nemoralis, Cernuella neglecta, Clausilia bidentata, Cochlicella acuta, Cornu aspersum, Lauria cylindracea, Pomatias elegans, and Theba pisana). Four species were parasitised by trematodes: C. aspersum (prevalence 56%), C. nemoralis (10%), and T. pisana (2%) as

Lamellodiscus louiseuzeti n. sp. (Monopisthocotyla: Diplectanidae: Lamellodiscinae): a parasite of Chaetodon hoefleri (Teleostei, Chaetodontidae) with distinct attachment organ morphology.

During a study on representatives of Diplectanidae (Monopisthocotyla) infecting Chaetodon hoefleri, a marine teleost from Senegal, we collected individuals belonging to a species of Lamellodiscinae, which showed a unique arrangement of the haptoral lamellodiscs, so far unreported in the literature, with the first lamella of the ventral lamellodisc open anteriorly, unlike the dorsal lamellodisc, which is closed. Due to this arrangement, we describe the species Lamellodiscus louiseuzeti n. sp.. Apart from the morphology of the lamellodiscs, the new species differs from its congeners by having a

A community-based participatory research and community health workers-led health education strategy for schistosomiasis control among schoolchildren in Pemba Island, Zanzibar: a cluster-randomized trial.

BACKGROUND: Schistosomiasis remains an intractable public health challenge in sub-Saharan Africa. While mass drug administration (MDA) has been the cornerstone of control, its limited impact on sustained behavioral change and persistent transmission in high-intensity transmission areas underscores the urgent need for complementary strategies. We aimed to evaluate the effectiveness of a community-based participatory research (CBPR) approach led by community health workers (CHWs), compared to conventional health education, in achieving sustainable schistosomiasis control among schoolchildren in

A systems-level analysis of global gene expression in resistant and susceptible Biomphalaria glabrata in response to Schistosoma mansoni infection.

BACKGROUND: The freshwater gastropod Biomphalaria glabrata is the major intermediate host of Schistosoma mansoni, the causative agent of human schistosomiasis. Elucidating the molecular basis of snail resistance to schistosomes is essential for understanding invertebrate immunity, host-parasite interactions, and schistosomiasis epidemiology. METHODS: Two new B. glabrata-S. mansoni interaction models were examined: homozygous lines (HZLs) iBS90_R (resistant) and iM line_S (susceptible), and recombinant inbred lines (RILs) RIL18_R (resistant) and RIL108_S (susceptible). Snail transcriptomes wer

First Molecular Detection of Infectious Spleen and Kidney Necrosis Virus (ISKNV) and the Parasite Centrocestus formosanus in the Ornamental Fish Platy (Xiphophorus maculatus) in Chile.

Infectious Spleen and Kidney Necrosis Virus (ISKNV) is a significant fish pathogen historically reported in East and Southeast Asia and increasingly detected in other regions, often in association with the international movement of ornamental fish. Centrocestus formosanus is a digenean trematode of sanitary and economic relevance, with zoonotic potential, that uses snails and fish as intermediate hosts. In this study, ornamental fish representing 24 species were sampled from 13 quarantine facilities across Chile. ISKNV was detected in samples of platy fish (Xiphophorus maculatus) from two quar

Colon polyps with or without Schistosoma japonicum.

Schistosoma japonicum, which has been endemic for a long time, associated colon abnormality has also been sporadically reported. The majority of patients with Schistosomiasis japonicum infected may mainly presented as acute manifestation or become chronic complication, known as schistosomiasis liver change, intestinal tract lesions and other system involved. Interestingly, both states could be observed in this patient. Acute infection presented as systemic hypersensitivity reaction, characterized by elevated eosinophils and intestinal symptom.

Review of the clinical, humanistic, and economic burden of focal segmental glomerulosclerosis.

Focal segmental glomerulosclerosis (FSGS) is a progressive glomerular disease characterized by podocyte injury, proteinuria, and risk of kidney failure. Until recently, no medicines had been approved by the FDA or European Medicines Agency, with management focused on supportive care and proteinuria reduction. This supplement explores the burden of FSGS from a clinical, humanistic, and economic perspective, informed by the results of 3 systematic literature reviews. FSGS is associated with poorer quality of life and high health care costs, and these outcomes are correlated with disease severity
