An oral M-cell targeted Lactococcus lactis vaccine against Echinococcus multilocularis infection.

BACKGROUND: Alveolar Echinococcosis (AE) is a serious infectious disease caused by Echinococcus multilocularis (E.multilocularis,Em) in the highlands of northwestern China and vaccination is currently the most effective means of preventing E. multilocularis infection. However, current vaccines are not sufficiently effective in preventing and controlling Alveolar Echinococcosis. METHODS: In this study, an oral M-cell targeted Lactococcus lactis (L. lactis) vaccine (LL-plSAM-GILE) was constructed by adding SAM gene sequence to the epitope vaccine GILE for E. multilocularis constructed in our pr

Osteopontin Promotes Liver Echinococcus multilocularis Growth and Invasion via p38MAPK Pathway.

This study aimed to elucidate how osteopontin (OPN) affects the growth, invasion, and extrahepatic metastasis of Echinococcus multilocularis (Em) through the p38MAPK signalling pathway of host hepatocytes. C57BL/6J mice were randomly divided into the control group (n = 10), the LV-OPN-0423 group (n = 10), the anti-p38MAPK group (n = 10), and the anti-p38MAPK + LV-OPN-0423 group (n = 10). Em infection models in mice were set up. The mice were fed for 6 weeks under suitable conditions. The mice in the anti-p38MAPK group and the anti-p38MAPK + LV-OPN-0423 group were then given SB202190 (this is a

In Vivo MicroRNA-Based Inhibition of Experimental Cestode Infection.

Cestodes (class Cestoda) include zoonotic parasites such as Echinococcus spp. and Taenia spp., which cause significant morbidity and mortality in endemic regions, particularly among pastoral and rural populations in low-, upper-, and middle-income countries. Their developmental plasticity reflects finely tuned regulatory mechanisms controlling gene expression throughout complex life cycles and infection stages. Despite expanding genomic resources, functional postgenomic studies remain scarce. MicroRNAs (miRNAs) are critical regulators of gene expression, influencing diverse developmental and p

Is a shortened postoperative albendazole duration after curative surgery for alveolar echinococcosis possible? Results from a prospective multicenter study.

In a prospective study of 15 patients with liver alveolar echinococcosis who underwent curative surgery, a strategy to reduce the recommended 2-year postoperative albendazole (ABZ) regimen was evaluated. Guided by negative positron emission tomography computed tomography (PET-CT) and magnetic resonance imaging (MRI) results at 1 year, ABZ was safely discontinued earlier (median 14 months) in 10 patients, with no recurrence observed during the mean 47 months of follow-up, indicating that postoperative ABZ may be safely shortened with careful imaging follow-up. Interestingly, a retrospective ana

Mitochondrial Network Fragmentation Leads to Dysfunction of Macrophages During Echinococcus multilocularis Protoscoleces Infection.

Alveolar echinococcosis (AE) is a fatal foodborne parasitic disease caused by the larvae of Echinococcus multilocularis. The disease primarily affects the liver. Previous studies have found that Kupffer cells have an immune protective effect, but in the late stages of AE, they are associated with parasite immune escape. The present study analyzed the effects of Echinococcus multilocularis protoscoleces (PSCs) infection on the mitochondrial morphology and function of macrophages, as well as their phagocytic function and apoptosis. Infection with PSCs has been shown to result in the fragmentatio

Ultrasound-based population survey for abdominal cystic echinococcosis in Albania: Results from the NDTND project.

BACKGROUND: Albania is considered endemic for cystic echinococcosis (CE). Recent data on prevalence and incidence of CE in Europe suggests that the Balkan region is the epicentre of this neglected parasitic disease in Europe. In Albania, a retrospective study has estimated a mean incidence of 1.49 cases/100,000 people. We aimed to assess the prevalence of CE in eight municipalities across the country. METHODOLOGY/PRINCIPAL FINDINGS: We enrolled participants aged >5 years in an ultrasound (US)-based screening for CE in 23 villages in eight municipalities from three prefectures. Participants we

Prioritizing interventions for echinococcosis prevention in China: a population attributable fraction analysis.

BACKGROUND: Despite population-level control efforts, human echinococcosis continues to pose a public health challenge in China. Targeting modifiable risk factors at the individual level could be another recipe, but the priority targets for intervention remain unclear. Our aim was to quantify the potential health gains of hypothetical interventions on modifiable risk factors for human echinococcosis, informing effective control strategies. METHODS: We collected and aggregated data on the association between modifiable risk factors and human echinococcosis and national total echinococcosis (cy

[The infectious liver tumour].

Alveolar echinococcosis remains a rare parasitic disease, but the number of cases in Western Europe is increasing. Diagnosis is difficult due to a long incubation period and non-specific symptoms. Curative treatment can only be achieved through R0 resection. Suppressive therapy with albendazole can now achieve good long-term survival even in cases where surgery is not possible.

Transcriptomic Profiling of mRNAs, circRNAs, and lncRNAs in Mouse Liver at the Early Stage of Echinococcus multilocularis Infection.

Alveolar echinococcosis (AE) is a significant zoonotic disease caused by the larval form of Echinococcus multilocularis (E. multilocularis), posing substantial health risks. However, the roles of circular RNAs (circRNAs), long non-coding RNAs (lncRNAs), and messenger RNAs (mRNAs) in the early stages of E. multilocularis infection, particularly regarding liver involvement, remain unclear. This study aimed to investigate the molecular mechanisms of AE infection in mice during the early stage of secondary infection. The expression profiles of mRNAs, circRNAs and lncRNAs in mouse livers were analy

Macroscopic and Microscopic Aspects of Echinococcus multilocularis and Echinococcus granulosus in Humans: A Guideline for Pathologists.

BACKGROUND: Echinococcosis is a parasitic disease, caused by the larval stage of the genus Echinococcus. Echinococcus multilocularis and Echinococcus granulosus cause alveolar echinococcosis (AE) and cystic echinococcosis (CE) in humans. Pathologists may be confronted with the larval stage of both species. Diagnosis may be difficult due to the low incidence in daily routine. However, right diagnosis of the pathologist is still the gold standard and crucial for further treatment of the patient. SUMMARY: Macroscopically, specimens of AE show multiple small cysts and a bread-like appearance. Ins

Epidemiology and spatiotemporal patterns of human echinococcosis in China, 2006-2020.

China accounts for over 90% of global alveolar echinococcosis (AE) cases caused primarily by Echinococcus multilocularis. If left untreated, AE can have a 10-year mortality rate of 94%. Understanding its epidemiological patterns is essential for targeted control strategies. Surveillance data from 2006 to 2020 were obtained from the Public Health Scientific Data Center, and spatial and temporal trends were analyzed using spatial autocorrelation, hot spot analysis, and centroid migration techniques. A total of 51,403 echinococcosis cases were reported from 2006 to 2020, with an average annual in

TPEDCC-PEG-based photodynamic therapy exhibits potent therapeutic efficacy against echinococcosis by inducing oxidative stress and mitochondria-mediated apoptosis.

Alveolar echinococcosis (AE) is a fatal zoonosis caused by the larval stage of Echinococcus multilocularis, but there are no fully effective treatments for this disease to date. The aim of this study was to investigate the potential therapeutic implication of photodynamic therapy (PDT) for E. multilocularis. We synthesized a novel nanophotosensitizer, TPEDCC-PEG, with aggregation-induced emission (AIE) properties and investigated its in vitro and in vivo PDT effects against the larval stage of E. multilocularis, as well as the anti-Echinococcus mechanism. TPEDCC-PEG exhibited excellent physico
