Tapeworms in an Apex Predator: First Molecular Identification of Taenia krabbei and Taenia hydatigena in Wolves (Canis lupus) from Romania.

The wolf (Canis lupus) is an apex predator with high mobility and trophic plasticity, serving as a valuable indicator of helminth transmission at the wildlife-livestock interface. Given the ecological overlap between wolves and both wild and domestic ungulates in Romania, we aimed to identify and molecularly characterize cestodes from wolves' small intestines. Between November 2022 and June 2025, small intestines from nine wolves were collected across four Romanian counties, frozen, and examined using classical parasitology (macroscopic and microscopic) and molecular methods (PCR amplification

Transplantation as a treatment option for hepatic echinococcosis past, present, and future - a pooled data analysis of 476 patients.

Liver transplantation (LT) serves as a critical therapeutic intervention for end-stage hepatic echinococcosis. Over time, the emergence of deceased donor LT (DDLT), living donor LT (LDLT), and ex vivo liver resection and autotransplantation (ELRA) has significantly influenced the clinical outcomes of end-stage echinococcosis. All English-language literature on echinococcosis-related LT published in the Web of Science, MEDLINE, and Embase databases from 1980 to 2023 was collected using relevant search terms. Patients' characteristics, surgical features, postoperative complications, mortality, a

Detection of Echinococcus spp. and other taeniid species in lettuces and berries: Two international multicenter studies from the MEmE project.

Cystic and alveolar echinococcosis are severe zoonotic diseases characterized by long asymptomatic periods lasting months or years. Viable Echinococcus spp. eggs released into the environment through the feces of canids can infect humans through accidental ingestion via hand-to-mouth contact or consumption of contaminated food or water. Both Echinococcus multilocularis and Echinococcus granulosus sensu lato are considered as foodborne parasites. However, when considering possible pathways of human infection, it appears that food and water-borne related variables do not significantly increase t

Zoonotic Echinococcus granulosus sensu lato genotypes G6 and G7: new insights from the global mitogenome analysis.

Cystic echinococcosis is a severe zoonotic disease caused by different species and genotypes belonging to the Echinococcus granulosus sensu lato (s.l.) complex. Among these, genotypes G6 and G7 are the second most common cause of human cystic echinococcosis. One of the very first steps towards understanding the epidemiology of G6 and G7 is to study their genetic and host diversity, population structure and phylogenetic relationships. For this, we sequenced near-complete mitochondrial genomes (12,850-12,856 bp) of 72 new G6 and G7 samples from eight countries and six host species, including hum

International multicentre study to validate the Segmental Sedimentation and Counting Technique (SSCT) for the surveillance of Echinococcus multilocularis in red fox.

The fox tapeworm, Echinococcus multilocularis, is the etiological agent of the severe zoonotic disease alveolar echinococcosis. The Sedimentation and Counting Technique (SCT) has long been considered the gold standard for detecting these worms in the intestines of dead foxes. Several alternative methods, including the Segmental Sedimentation and Counting Technique (SSCT), have been developed, aiming to reduce time of analysis while maintaining high sensitivity. In this study, the performance of the SSCT was first evaluated by comparison with the SCT in the context of E. multilocularis surveill

Unmasking the mimic: vertebral alveolar echinococcosis diagnosed by metagenomic next-generation sequencing.

A Siberian woman in her forties presented to a public hospital in northeastern Germany with chronic back pain and a paravertebral mass, initially misdiagnosed as spinal tuberculosis. Repeated biopsies and metagenomic next-generation sequencing (mNGS) ultimately confirmed vertebral alveolar echinococcosis. Haplotype analysis revealed a novel Asian-cluster variant, supporting the presumed origin of infection.

The Great Pretender: Echinococcal Infections Posing as Liver Cancer - Two Case Reports.

INTRODUCTION: Medical professionals are well aware that imaging techniques are not infallible in accurately diagnosing cancer or staging patients with histologically confirmed cancer. Pathologists often emerge as game changers, prompting a complete re-evaluation of the therapeutic approach. CASE PRESENTATIONS: Here we present 2 cases of patients initially diagnosed with suspicious cancer lesions that were subsequently identified as echinococcal hydatids. Beyond the clinical findings, the patients' perspectives vividly illustrate the psychological burden of misdiagnosis and the impact of treat

Developmental programming by prenatal sounds: insights into possible mechanisms.

In recent years, the impact of prenatal sound on development, notably for programming individual phenotypes for postnatal conditions, has increasingly been revealed. However, the mechanisms through which sound affects physiology and development remain mostly unexplored. Here, I gather evidence from neurobiology, developmental biology, cellular biology and bioacoustics to identify the most plausible modes of action of sound on developing embryos. First, revealing often-unsuspected plasticity, I discuss how prenatal sound may shape auditory system development and determine individuals' later cap

Immunoprotective effect and mechanism of rEg.P29 against CD4 + T cell-deficient mice with Echinococcus multilocularis infection.

Alveolar echinococcosis (AE) is a zoonotic parasitic disease caused by infection with the larval stage of Echinococcus multilocularis and a major challenge to human public health. Vaccines are the most effective way to prevent and control infectious diseases. We previously revealed that the Echinocuccus granulosus recombinant protein P29 is a good vaccine candidate against E. granulosus. However, the protective and immunological mechanism of rEg.P29 against E. multilocularis remain unclear. In this study, CD4 + T cell-deficient mice are transferred with spleen CD4 + T cells isolated from wild-

Human alveolar echinococcosis in Slovakia: Epidemiology and genetic diversity of Echinococcus multilocularis, 2000-2023.

Human alveolar echinococcosis (AE) is a serious parasitic disease caused by larval stages of Echinococcus multilocularis. Between January 2000 and October 2023, 137 AE cases were confirmed in Slovakia. The average annual incidence increased from 0.031 per 100,000 inhabitants between 2000 and 2011, to an average of 0.187 since 2012, i.e. about six times. Among patients, 45.3% were men and 54.7% were women; the mean age at the time of diagnosis was 52.8 years. Most cases were diagnosed in the age groups 51-60 years and 61-70 years (33 cases each), and eight patients fell into the age category ≤

Confirmed Autochthonous Case of Human Alveolar Echinococcosis, Italy, 2023.

In September 2023, a patient in Italy who had never traveled abroad was referred for testing for suspected hepatic cystic echinococcosis. Lesions were incompatible with cystic echinococcosis; instead, autochthonous alveolar echinococcosis was confirmed. Alveolar echinococcosis can be fatal, and awareness must be raised of the infection's expanding distribution.

Molecular phylogenetic analysis of Echinococcus multilocularis from horses raised in Canada or Japan, using mitochondrial cytochrome b gene-targeted PCR.

Alveolar echinococcosis is a zoonotic disease caused by a larval-stage Echinococcus multilocularis infection. Geographical haplotyping targeting the parasite's mitochondrial cytochrome b (cob) gene has been reported for isolates from definitive and intermediate hosts (wild canids and rodents); however, there are limited reports on strain typing for the dead-end host, the horse, which could act as a sentinel for E. multilocularis. Accordingly, we investigated the diversity of E. multilocularis in isolates obtained from slaughtered Japanese and Canadian horses originating from the Iburi and Hida

A Bioengineered Nanovesicle Vaccine Boosts T-B cell Interaction for Immunotherapy of Echinococcus multilocularis.

Alveolar echinococcosis (AE) is a zoonotic parasitic disease, resulting from being infected with the metacestode larvae of the tapeworm Echinococcus multilocularis (E. multilocularis). Novel prophylactic and therapeutic interventions are urgently needed since the current chemotherapy displays limited efficiency in AE treatment. Bioengineered nano cellular membrane vesicles are widely used for displaying the native conformational epitope peptides because of their unique structure and biocompatibility. In this study, four T-cells and four B-cells dominant epitope peptides of E. multilocularis wi

Genome-wide transcriptome analysis of Echinococcus multilocularis larvae and germinative cell cultures reveals genes involved in parasite stem cell function.

The lethal zoonosis alveolar echinococcosis is caused by tumour-like growth of the metacestode stage of the tapeworm Echinococcus multilocularis within host organs. We previously demonstrated that metacestode proliferation is exclusively driven by somatic stem cells (germinative cells), which are the only mitotically active parasite cells that give rise to all differentiated cell types. The Echinococcus gene repertoire required for germinative cell maintenance and differentiation has not been characterised so far. We herein carried out Illumina sequencing on cDNA from Echinococcus metacestode

Risk factors for Echinococcus multilocularis intestinal infections in owned domestic dogs in a North American metropolis (Calgary, Alberta).

Human alveolar echinococcosis is increasingly documented in Alberta, Canada. Its causative agent, Echinococcus multilocularis (Em), can be transmitted to humans by infected dogs. We assessed the prevalence and associated risk factors for Em infections in domestic dogs in Calgary, Alberta, Canada. In this cross-sectional study that coupled collection and assessment of dog feces with a survey on potential risk factors, 13 of 696 (Bayesian true prevalence, 2.4%; 95% CrI: 1.3-4.0%) individual dogs' feces collected during August and September 2012 were qPCR positive for Em. Molecular analyses of exosome-derived miRNAs revealed reduced expression of miR-184-3p and decreased exosome concentration in patients with alveolar echinococcosis.

Both E. multilocularis and host-derived exosomes are involved in the pathogenic process of alveolar echinococcosis (AE). Exosomes secrete miRNAs that have regulatory roles in host-pathogen interactions in multiple ways. In the present study, we collected and purified supernatants of E. multilocularis cultures, as well as human plasma exosomes. High-throughput sequencing showed the identities of 45 exosomal miRNAs in E. multilocularis. The lengths of these miRNAs ranged from 19 to 25 nucleotides (nt), with the majority (n = 18) measuring 22 nt. Notably, emu-let-7-5p emerged as the most abundant
