Quantitative Contrast-Enhanced Ultrasound Characterization of Perilesional Perfusion and Its Association with CD34-Positive Cell Density in a Rat Model of Hepatic Alveolar Echinococcosis.

Hepatic alveolar echinococcosis (HAE) is characterized by infiltrative growth and remodeling of perilesional host tissue, including microvascular changes. We investigated whether quantitative contrast-enhanced ultrasound (CEUS) could characterize perilesional perfusion and its animal-level association with CD34-positive cell density in a standardized rat model. Twenty female Sprague Dawley rats underwent ultrasound-guided intrahepatic HAE modeling and CEUS 10 weeks later. Peak enhancement (PE), rise time (RT), time to peak (TTP), wash-in rate (WiR), wash-in perfusion index (WiPI), and wash-in

Surgery and toceranib for renal cell carcinoma with caval thrombus in a dog.

A 14-year-old spayed female papillon dog was presented with hematuria and a right renal mass. Contrast-enhanced computed tomography revealed a tumor thrombus extending about 1.5 cm cranially from the right renal vein into the caudal vena cava (CVC), consistent with renal cell carcinoma and caval thrombus, with no signs of distant metastasis. The dog underwent open right-sided nephrectomy and CVC thrombectomy, using 2 tourniquets, with a total occlusion time of 12 min 13 s. Transient hypotension during surgery was successfully managed, and there were no major intra- or postoperative complicatio

First report of Taenia arctos (Cestoda: Taeniidae) from brown bears (Ursus arctos) in Romania.

BACKGROUND: The brown bear (Ursus arctos) is an apex predator with significant ecological importance and serves as a valuable indicator species for monitoring parasitic burdens in forest ecosystems. Owing to its complex ecology and varied diet, this species may play a key role in the life cycle of numerous pathogens, including cestodes of the genus Taenia, such as the recently described Taenia arctos. Among the potential intermediate hosts for T. arctos is the moose (Alces alces), a species sporadically present in northern Romania, where its habitat may overlap with that of U. arctos. In this

Diagnostic performance comparison of two ultrasound classification systems for hepatic alveolar echinococcosis: a single-center retrospective study.

BACKGROUND: Hepatic alveolar echinococcosis (HAE) is a severe zoonotic parasitic disease for which ultrasonography is the primary diagnostic tool. However, the heterogeneous imaging characteristics of HAE lesions present significant challenges to accurate diagnosis. To improve diagnostic reliability, this study compared the performance of two established ultrasound classification systems: the Echinococcus multilocularis National Health Commission Classification-Ultrasound (EMNHCC-US) and the E. multilocularis Ulm Classification-Ultrasound (EMUC-US). RESULTS: This study compared EMUC-US and EM

Enhancing the structural stability of P29-targeted monoclonal antibodies via β-hydroxybutyrylation modification improves their therapeutic performance in alveolar echinococcosis.

BACKGROUND: Alveolar echinococcosis (AE), a severe parasitic infection often likened to "parasitic cancer", still lacks effective treatments. Although our earlier ork on the P29 monoclonal antibody (P29 mAb) against the parasite-derived P29 antigen showed potential, its efficacy remained limited, prompting the need for improved biologic agents. METHODS: We applied β-hydroxybutyrylation (Kbhb) modification to engineer an enhanced antibody, P29 mAbKbhb, and comprehensively evaluated its properties using surface plasmon resonance, protease resistance assays, pharmacokinetic studies in C57BL/6 mi

IL-22-STAT3-CD155 axis in alveolar echinococcosis: a pivotal role in immune exhaustion and therapeutic potential.

BACKGROUND: Alveolar echinococcosis (AE), a lethal zoonosis caused by Echinococcus multilocularis (E.m) infection, is characterized by immune exhaustion that facilitates parasite evasion of host immunity and sustains chronic infection. The role and mechanisms of Interleukin-22 (IL-22), a key immunomodulatory cytokine, in E.m-induced immune responses remain unclear and warrant investigation. METHODS: Liver tissue samples from AE patients and E.m-infected mouse models were utilized to investigate IL-22 expression dynamics during AE progression and its correlation with disease progression. Analysis of Echinococcus multilocularis complete mitogenomes: evidence for the historical spread from Central Asia to the Northern Hemisphere.

Echinococcus multilocularis is a parasitic tapeworm responsible for alveolar echinococcosis (AE), a life-threatening zoonotic disease. Understanding its genetic diversity and historical spread is critical for developing effective control measures. Previous studies have suggested Central Asia as a potential origin of the parasite, but comprehensive global analyses encompassing multiple regions and time periods have been limited. This study aimed to analyze the genetic diversity, population structure, and phylogeography of E. multilocularis by sequencing the complete mitogenome from 384 isolates

Core-shell heterostructured nanozyme for alveolar echinococcosis treatment via targeted electron transfer and catalytic reactive oxygen species generation.

Alveolar echinococcosis (AE), a severe parasitic disease, poses a significant therapeutic challenge due to the limitations of surgery and albendazole chemotherapy, particularly the difficulty in achieving targeted drug delivery and disrupting the robust structure of the protoscoleces (PSCs). This study introduces a novel therapeutic strategy of "targeted binding-electron transfer (ET)-osmotic disruption" to address these challenges. We designed a core-shell heterostructure nanoplatform (FZ) composed of a magnetite (Fe3O4) nanozyme core and a zinc oxide (ZnO) semiconductor shell. This architect

Exploring the Potential Role of Interleukin-33 in Inducing Eosinophil-Mediated Fibrosis in Hepatic Alveolar Echinococcosis.

BACKGROUND: Alveolar echinococcosis (AE) is a serious zoonotic parasitic disease. This study aimed to investigate the mechanisms underlying the formation of the dense fibrotic band surrounding hepatic alveolar echinococcosis (HAE) lesions, which impedes chemotherapeutic drug penetration. Additionally, the roles of Interleukin-33 (IL-33) and eosinophils in the progression of fibrosis within this band were examined. METHODS: IL-33/suppression of tumorigenicity 2 (ST-2) expression levels were compared between patients with HAE and healthy controls, as well as between close to lesion tissues (CLT

Macrophage-derived S100A9 drives liver fibrosis in Echinococcus multilocularis infection by up-regulating secreted protein acidic and rich in cysteine (SPARC) in hepatic stellate cells.

Alveolar echinococcosis (AE), caused by Echinococcus multilocularis(E. multilocularis) infection, is a severe parasitic disease characterized by progressive liver fibrosis. Although the damage-associated molecular pattern molecule S100A9 has been implicated in some inflammatory and fibrotic diseases, its role in AE induced liver fibrosis remains unclear. This study aimed to elucidate the function and mechanism of S100A9 in E. multilocularis induced liver fibrogenesis. Using proteomic analysis, we identified significant up-regulation of S100A9 in the livers of infected mice. In a stem cell-base

A fatal case of complex hepatic alveolar echinococcosis.

Hepatic alveolar echinococcosis (HAE), a life-threatening zoonosis, poses formidable surgical challenges when involving critical vasculature. Herein, we reported the periprocedural management dilemmas in radical resection for advanced HAE. A 58-year-old female visited the outpatient department presented with HAE. Imaging examination revealed extensive invasion of the hilum, bile duct, and several hepatic vessels, as well as left adrenal metastasis. The patient underwent right trisegmentectomy with left hepatic vein reconstruction, auto-transplantation, and adrenalectomy, with intraoperative Do

LAG3 constrains anti-parasitic response by effector CD4+ T-cell in early Echinococcus multilocularis-infected mice.

BACKGROUND: Immune checkpoint molecules such as lymphocyte activation gene-3 (LAG3) play a critical role in modulating host-pathogen interactions during chronic parasitic infections; however, their functions in early infection remain poorly defined. Using a murine model infected with Echinococcus multilocularis (E. multilocularis), a lethal helminth causing alveolar echinococcosis (AE), we elucidate the stage-specific regulatory functions of LAG3 in CD4+ T cell immunity during early infection. METHODS: Echinococcus multilocularis-infected mice were employed as the experimental model. Flow cyt

[Alveolar echinococcosis in a pregnant bitch - a case report].

Alveolar echinococcosis is a significant zoonotic disease characterized by infiltrative growth of metacestodes in intermediate hosts. It is primarily located in the liver and is associated with a high mortality rate if left untreated. To the authors' knowledge, the present report describes the first case of alveolar echinococcosis in a pregnant dog documented in the literature.A five-year-old, unneutered female Labrador Retriever was presented shortly after artificial insemination with abdominal distension, polydipsia, and polyuria. Further examinations led to the diagnosis of alveolar echinoc

Protoscolex Maturation in Echinococcus multilocularis Is Controlled by Host Genetic Factors Independent of Initial Development.

Alveolar echinococcosis, caused by Echinococcus multilocularis, poses a serious public health concern. This parasite requires rodents as intermediate hosts for protoscolex maturation in the liver and subsequent transmission to definitive hosts. We aimed to identify the host genetic factors controlling protoscolex development and maturation in congenic mice carrying susceptible DBA/2 genomic segments on a resistant C57BL/6N background. We assessed the hepatic protoscoleces and immune-related gene expression levels 16 weeks post-oral infection. All congenic strains developed protoscoleces; howev

CCR8 orchestrates an immunosuppressive niche in the liver to promote Echinococcus multilocularis infection.

Echinococcus multilocularis (E. m) infection causes alveolar echinococcosis (AE), a serious zoonotic disease characterized by invasive larval growth in the liver. The parasite establishes a chronic infection, suggesting effective modulation of host immunity. Here, we investigated the role of the CCR8/CCL1 chemokine axis in shaping the hepatic immune microenvironment during E.m infection. In infected wild-type (WT) mice, chronic infection specifically activated the hepatic CCR8/CCL1 axis, which was associated with a marked accumulation of FOXP3+ regulatory T cells (Tregs). Notably, although CCR
