Neural cell adhesion molecules and myelin-associated glycoprotein share a common carbohydrate moiety recognized by monoclonal antibodies L2 and HNK-1.

Cell surface molecules have been implicated in cell interactions which underlie formation of the nervous system. The analysis of the functional properties of such molecules has profited from the combined use of antibodies and cell culture systems. It has been suggested that the interplay between these molecules modulates cell-to-cell interaction at critical developmental stages. In the mouse, N-CAM and L1 antigen have been shown to mediate Ca2+-independent adhesion among neural cells. N-CAM plays a role in fasciculation of neurites and formation of neuromuscular junction. L1 is apparently not

Transient expression of a surface antigen on a small subset of neurones during embryonic development.

During development, neurones find and interconnect with their targets in a remarkably precise way. The unfolding of neuronal specificity involves a series of highly specific recognition events which are likely to be coordinated by the spatial and temporal expression of many different surface molecules. At early stages of development, neuronal recognition occurs most dramatically at the tips of growing axons, at growth cones and their filopodia. Previous studies on the grasshopper embryo suggest that specific filopodial contacts lead to the stereotyped patterns of selective axonal fasciculation

Cell recognition during neuronal development.

Insect embryos, with their relatively simple nervous systems, provide a model system with which to study the cellular and molecular mechanisms underlying cell recognition during neuronal development. Such an approach can take advantage of the accessible cells of the grasshopper embryo and the accessible genes of Drosophila. The growth cones of identified neurons express selective affinities for specific axonal surfaces; such specificities give rise to the stereotyped patterns of selective fasciculation common to both species. These and other results suggest that early in development cell linea

Diazepam does not prevent succinylcholine-induced fasciculations and myalgia. A comparative evaluation of the effect of diazepam and d-tubocurarine pretreatments.

To determine the effectiveness of diazepam pretreatment in preventing succinylcholine (SCh)-induced fasciculations and body pains, 587 patients were randomly allocated to six groups. Patients in Group I received no pretreatment and served as controls. Patients in Groups II and III were pretreated with 0.05 mg/kg of diazepam either 4-5 min (Group II) or 8-10 min (Group III) prior to SCh administration. Patients in Groups IV and V received 0.1 mg/kg of diazepam either 4-5 min (Group IV) or 8-10 min (Group V) prior to SCh administration, while patients in Group VI were pretreated with 0.05 mg/kg

Projection patterns of lateral-line afferents in anurans: a comparative HRP study.

Primary projections of the anterior and posterior lateral-line nerves were traced by means of transganglionic transport of horseradish peroxidase in species belonging to five of the six anuran superfamilies. Both anterior and posterior lateral-line nerve afferents each enter the brain via a single root which divides into two or more bundles. These bundles carry fibers from neuromasts only. No separate dorsal fascicle and no ampullary organs as in urodeles and gymnophions have been found. All bundles join in the neuropil of the nucleus intermedius to form ascending and descending fascicles. Two

Delayed neurotoxicity produced by an organophosphorus compound (Sumithion). A case report.

A case of late-onset manifestation of organophosphorous insecticide poisoning is described. The patient was a 70-year-old female who ingested 40 ml Sumithion emulsion (50% fenitrothion). At first, no toxic symptoms were apparent. However, 48 h after ingestion certain signs became apparent. An impediment in consciousness was observed. Fasciculation and muscular weakness were noted, while plasma and urinary 4-nitro-3-methylphenol (NMC), the degradation product of Sumithion, reached a maximum. Neither atropine sulfate nor PAM (pyridine 2-aldoxime methiodine) were effective. For 3 weeks the patien

In vivo actions of atraxin, a protein neurotoxin from the venom glands of the funnel-web spider (Atrax robustus).

The effects of atraxin, a neurotoxic protein from the venom glands of the funnel-web spider (Atrax robustus), have been studied in anaesthetized monkeys. At doses of 70 and 80 micrograms kg-1 i.v., atraxin caused respiratory disturbances (dyspnoea and apnoea), and profound alterations in heart rate and blood pressure. These doses also caused salivation, lachrymation, skeletal muscle fasciculation and an elevation in body temperature. Concurrent increases in firing were recorded from the phrenic nerve and from respiratory and other skeletal muscles. It is concluded that atraxin produces the sam

Isolation and characterization of a protein neurotoxin from the venom glands of the funnel-web spider (Atrax robustus).

1. Ground venom glands from male and female funnel-web spiders (Atrax robustus) were extracted with acetic acid (0.35 M). 2. A protein constituent of these extracts, atraxin, which caused muscle fasciculation when applied to the phrenic nerve-hemidiaphragm of the mouse in vitro, was isolated by the sequential application of ultrafiltration, gel permeation and ion exchange chromatography. 3. When subjected to isoelectricfocusing, amino acid analysis, ultracentrifugation, ultraviolet and 1H nuclear magnetic resonance spectroscopy, it was shown that atraxin had an approximate molecular weight of

Pathfinding by neuronal growth cones in grasshopper embryos. II. Selective fasciculation onto specific axonal pathways.

In the previous paper (Raper, J.A., M. Bastiani, and C.S. Goodman (1983) J. Neurosci. 3:20-30) we showed that the growth cones of two sibling neurons, the G and C cells, follow the same route in the developing grasshopper neuropil until they reach a stereotypic choice point. Here their growth cones diverge from each other as G turns and extends anteriorly and C turns and extends posteriorly. In this paper we show that the G and C growth cones fasciculate and extend in opposite directions upon a specific bundle of four axons. This occurs even though many other axons are within filopodial reach

[Optochiasmatic diastasis in tumors with suprasellar extension and duplication of the chiasma].

The opto chiasmatic diastasis whose extreme form is a duplication of the chiasma, corresponds to a longitudinal cleavage, using the well known fasciculation of the optic fibres. It has been noticed four times in the sellar and para sellar expanding processes (two adenomas, one craniopharyngioma and one metastasis). Generally, this lesion of the chiasma goes with big tumors (grade II); it has been detected four times in frontal approaches (15%) while the rhinoseptal approach represents 85% of our cases. The diastasis of the optic fibers with tendency to form double chiasma, instead of being an

Monoclonal antibodies demonstrate the organization of axons in the leech.

Monoclonal antibodies have been generated that bind to subsets of neurons within the leech central nervous system (Zipser, B., and R. McKay (1981) Nature 289: 549-554). In this report we describe the binding patterns of monoclonal antibodies to subsets of axons in the leech using HRP-immunohistochemistry. Each antibody has a characteristic staining pattern in the connective, the large bundle of axons that runs the length of the nerve cord connecting each ganglion to its rostral and caudal neighbors. These staining patterns are consistent along the rostrocaudal axis of each animal, between anim

Development of order in the rat trigeminal system.

The trigeminal system of the rat is characterized by a high degree of order. The pattern of the distribution of vibrissae follicles on the face is replicated at each synaptic station between face and somatosensory cortex (Belford and Killackey, '80). The present study details the development of the trigeminal nerve, its intrinsic organization, and its relationship with its peripheral and central targets. We have observed that at early embryonic ages (E12 and E13) the trigeminal ganglion neurons grow out in straight lines without crossing, and the distance between these neurons and their periph

Cell adhesion molecules.

It has been proposed that cell-cell recognition occurs by means of local cell surface modulation of a small number of proteins rather than by expression of large numbers of different cell surface markers. Several different cell adhesion molecules (CAM's) have now been found in a number of vertebrate species in different tissues such as liver and striated muscle and even in a single complex structure such as the brain, where different molecules specific for neurons and glia have been identified. The neuron-specific molecule is involved in early embryonic events but also mediates neurite fascicu

Embryogenesis of peripheral nerve pathways in grasshopper legs. II. The major nerve routes.

In the preceding paper (H. Keshishian and D. Bentley, 1983a, Dev. Biol. 96, 89-102) the events leading to the morphogenesis of nerve 5B1 in the grasshopper embryonic metathoracic leg were presented. Here the role of later differentiating peripheral neurons in establishing the other major nerves of the leg is examined. In addition to the (tibial 1) (Ti1) pioneer neuron cell pairs that establish nerve 5B1 in the tibia femur, and coxa-trochanter, six later differentiating cells and/or cell pairs were identified and examined with respect to their role in peripheral nerve ontogeny. Nerve path pione

Isaacs' syndrome with muscle hypertrophy reversed by phenytoin therapy.

A 16-year-old boy was seen for severe episodic muscle cramps and generalized myokymia, consistent with Isaacs's syndrome. Bilateral calf hypertrophy (46-cm calf circumference) and ankle areflexia were noted. He was treated with phenytoin sodium, 300 mg/day. Within three months there was marked decrease in myokymia, total relief of cramps, return of ankle reflexes, and 6-cm reduction in calf circumference. We suggest that the excess muscle activity in Isaacs's syndrome may be responsible for the associated phenomena of muscle hypertrophy and areflexia.
