Oroxylum indicum (L.) Bark Ameliorates Anxiety and Depression: Evidence From Experimental and Computational Studies.

This study evaluated the antioxidant and neuropharmacological potentials of methanol extract of Oroxylum indicum bark (MOIB) to advocate the scientific basis of its traditional use in medical folklore. The preliminary phytochemical screening and GC-MS analysis identified twenty phytochemicals in MOIB. Besides, in EPM, MOIB exhibited an increase in time spent and the number of entries in open arms while an enhancement in head dipping was demonstrated by MOIB in HBT compared to control, indicating anxiolytic activities. Furthermore, a dose-dependent reduction in locomotor activities together wit

Acupuncture as a modulator of microglial polarization and pyroptosis in depression: evidence from a chronic unpredictable mild stress rat model.

BACKGROUND: Depression is a common psychiatric disorder, with stress-induced neuroinflammation playing a crucial role in its pathogenesis. Acupuncture has been demonstrated to be both effective and safe in the treatment of depression, but its precise mechanisms remain unclear. METHOD: This study investigated acupuncture's effects on neuroglia-associated neuroinflammation in chronic unpredictable mild stress (CUMS)-induced depressive rats. Rats underwent CUMS for 28 days, with the acupuncture group receiving treatment at Shangxing (GV23) and Fengfu (GV16) every other day. The fluoxetine group

Structure-activity Relationships of Triterpenoid Saponins Across Phylogenetically Diverse Organisms.

Saponins are structurally diverse bioactive metabolites found in more than 100 plant families and are synthesized by plants as protection against insects, fungi, and other organisms. The mode of action is related to their interference with membranes, and particularly the membrane sterols. As membrane sterol composition varies across kingdoms and species, species-specific differences in toxicity may therefore be expected. The aim of this study was to elucidate the structure-activity relationships of different saponins across four different organisms (Daphnia magna, Enchytraeus crypticus, Saccha

The contribution of additives to microplastic aquatic toxicity - A testing approach with model additives on selected aquatic organisms.

The challenge in assessing the overall toxicity of micro- and nanoplastics (MNP) lies in the complexity and variety of additives and their release, which is additionally affected by ageing processes of MNP in the environment. Our study aims to develop further the methodology to differentiate between the physical (particle-related) and chemical effects of MNP (contribution of additives) on the overall toxicity of MNPs. A testing methodology is presented that assesses the effects of well-defined, additive-loaded MNPs, pristine additive-free MNPs, and additives separately. This approach is comple

ICG-based fluorescence imaging to assess muscle perfusion for guiding surgical debridement after acute compartment syndrome.

BACKGROUND: Acute Compartment Syndrome (ACS) is a severe condition that can threaten limb viability, particularly in patients who experience prolonged immobility, such as in the found-down phenomenon. Inadequate removal of nonviable muscle after fasciotomy increases the risk of infection. While non-invasive imaging modalities have been explored for early ACS diagnosis, no tools currently exist to guide precise debridement intraoperatively. Existing technologies are limited by susceptibility to interference, lack of standardized thresholds and insufficient real-time surgical utility. This study

Postural and cardiorespiratory responses to emotionally-laden whole-body expressions.

Postural and autonomic adjustments are commonly described during the observation of emotional stimuli. Thus, this study investigated the physiological impact of emotional images viewing revealed by whole-body expressions. Body sway and cardiorespiratory signals were recorded in thirty-nine healthy young people who maintained an upright stance while observing 20 neutral, 20 happiness (positive) or 20 anger (negative) images from the Bodily Expressive Action Stimulus Test (BEAST) database. Participants rated each image considering their valence and arousal dimensions with the Self-Assessment Man

Dynamical modulation of hippocampal replay through firing rate adaptation.

During periods of immobility and sleep, the hippocampus generates diverse self-sustaining sequences of "replay" activity, which exhibit stationary, diffusive, and super-diffusive dynamical patterns. However, the neural mechanisms underlying this diversity in hippocampal sequential dynamics remain largely unknown. Here, we propose a unifying mechanism by showing that modulation of firing-rate adaptation strength within a continuous attractor model of place cells gives rise to these distinct forms of replay. Our model accounts for empirical data and yields several testable predictions. First, mo

Epidemiology and Natural History of Preclinical and Clinical Obesity: Insights From a UK Cohort.

OBJECTIVE: Obesity has been traditionally defined by BMI alone, but this metric has limitations in assessing body fat composition and adiposity complications. The Lancet Diabetes and Endocrinology Commission (LDEC) issued a new obesity definition to address these challenges, stratified by preclinical and clinical groups. We evaluated the epidemiology of preclinical and clinical obesity in the UK Biobank and associations with incident cardiovascular disease (CVD). METHODS: We performed retrospective cohort analyses of 502,233 adults enrolled in the UK Biobank. Obesity was categorized using the

Metabolomic insights into gut-brain axis dysregulation under unpredictable chronic stress in zebrafish.

Growing evidence indicates that depression is closely associated with disturbances in the gut-brain axis. Although rodent models are currently widely utilised in depression pathophysiology research, the zebrafish (Danio rerio) has become a valuable and promising alternative model species. This study examined changes in neuroendocrine and neurochemical markers associated with behavioural modifications in a zebrafish model of unpredictable chronic stress (UCS). Following a 14-day UCS procedure, the open-field test (OFT), social interaction test (SIT), and light-dark test (LDT) were conducted. Wh

Discovery of the Underlying Mechanism of Ginger Juice Processed Ziziphi Spinosae Semen for Its Sedative-Hypnotic Effect on Insomnia Mice via Regulation of HPA Axis and cAMP/PKA Signaling Pathway.

Based on Traditional Chinese Medicine (TCM) theory, the efficacy and mechanism of Ginger juice processed Ziziphi Spinosae Semen (GJPZSS) for treating insomnia, particularly stress-related types, were investigated to provide empirical evidence. An insomnia model was induced in mice by DL-4-chlorophenylalanine (PCPA) and chronic tail clamping. The sedative effect was evaluated by behavioral tests. Serum components from GJPZSS were analyzed by UHPLC-Q-TOF-MS/MS, and 64 potential targets were identified. The cAMP signaling pathway was enriched as the core pathway by Kyoto Encyclopedia of genes and

The precision of CT-guided percutaneous sacral screw placement in 114 fragility fractures of the pelvis - a retrospective study.

BACKGROUND: Fragility fractures of the pelvis (FFP) are increasingly common, particularly in the ageing population. Traditionally, those low-energy pelvic ring fractures have been managed conservatively, but this approach can lead to immobility and therefore complications. Surgical intervention is indicated based on various factors including type of fracture, mobility, and pain. Minimal-invasive percutaneous sacroiliac and/or transsacral screw stabilisation offers an effective treatment, enabling immediate mobilisation. There are multiple technical methods for the implantation. This study aime

Mild fructose diet during periadolescent development increases emotionality, but not metabolic consequences of early life adversity in rats.

Early life stress (ELS) increases the risk of metabolic disorders such as obesity, as well as neuropsychiatric conditions including depression. Adolescence represents a critical window for metabolic programming, particularly under the influence of ELS and poor dietary habits. Animal models of ELS, such as maternal separation (MS180), induce long-term alterations in hypothalamic-pituitary-adrenal (HPA) axis function, glucose metabolism, and emotionality; however, it remains unclear whether this paradigm increases vulnerability to a chronic metabolic challenge. The aim of this study was to evalu

Combined administration of intracerebroventricular CB1 agonist ACEA and systemic TRPV1 agonist capsaicin induces synergistic antidepressant-like effects in rats.

Depression is a distressing mental disorder that affects millions of people worldwide. Current treatments include both psychological and pharmacological approaches. However, delays in clinical improvement and high percentage of patients unresponsive to conventional antidepressants highlight the need to identify novel compounds or strategies with optimal antidepressant actions. We have previously shown the antidepressant-like effects of the intraperitoneal (i.p.) administration of the TRPV1 agonist capsaicin (Cap). Moreover, accumulating evidence indicates that the endocannabinoid system plays

A paraventricular hypothalamic control of social stress susceptibility by targeting brown adipose tissue β3 adrenoceptor.

Adipose tissue disturbance underlies the pathogenesis of major depressive disorder (MDD); however, the underlying neural and molecular mechanisms remain elusive. Utilizing a social defeat stress mouse model of MDD, we identified a paraventricular hypothalamic (PVH) control of susceptibility to social stress via β3 adrenoceptor (β3AR) signaling in the brown adipose tissue (BAT). Susceptible mice exhibited a significantly elevated expression of BAT β3AR. Pharmacological activation of β3AR increased immobility time in the forced swim test in naïve mice, and decreased social interaction time in mi

Sex-specific difference on anxiety- and depressive-like behavior in neuronal growth regulator 1-knockout mice.

BACKGROUND: Sex differences are evident in anxiety and depression, and women more frequently present with comorbid anxiety and depression alongside gastrointestinal disturbances. This pattern suggests contributions from sex-specific biological mechanisms and gut-brain communication. Negr1, a molecule regulating neuronal growth and connectivity, has been linked to depression-relevant behaviors in animal models. However, its mechanisms and potential sex-specific effects remain unclear. METHODS: Behavioral tests were used to assess phenotypes related to depression, anxiety, and learning in male

Unraveling the role of IL-17 signaling pathway in breast cancer-related depression: insights from in vivo/in vitro models and transcriptomic analysis.

Breast cancer-related depression (BCRD) is a prevalent comorbidity that markedly reduces quality of life and can negatively influence treatment outcomes. The molecular basis of BCRD remains elusive, particularly the role of the IL-17 signaling pathway in the interaction between breast cancer and depression. We established a BCRD mouse model by inducing breast tumors and administering chronic corticosterone. Transcriptomic analysis was performed on brain and tumor tissues to identify differentially expressed genes (DEGs) associated with BCRD. Functional enrichment analyses were conducted to det
