Early-life stress alters adult social and coping behaviors in a sex-specific and domain-dependent manner.

Early Life Stress (ELS) increases the risk for mental health issues in humans, notably in major depression and anxiety disorders. ELS is frequently modeled in laboratory rodents by disrupting the early postnatal environment. Literature on ELS is expanding, yet studies on sex-specific differences remain mixed. We utilized a novel ELS protocol that subjected mouse pups of both sexes to maternal separation and removed pup-to-pup contact comfort during postnatal days 10 to 17. We hypothesized that this ELS protocol would induce depressive and anxiety-like phenotypes persisting into adulthood, with

An exploration of the relationships between the effects of psilocybin on behavior, 5-HT2A receptor occupancy, and neuroplastic effects in mice.

BACKGROUND: Psilocybin has shown rapid and sustained antidepressant effects in patients with major depressive disorder, yet the neurobiological mechanisms underlying these outcomes remain unclear. AIMS: This study aimed to bridge clinical and preclinical findings by investigating the relationships between 5-HT2A receptor occupancy (RO) achieved after administration of psilocybin and its effects on behavior and markers of neuroplasticity in mice. METHODS: In vivo 5-HT2A RO was determined via displacement of [3H]MDL-100,907 in the prefrontal cortex (PFC). To relate RO with behavioral outcomes

Deep Brain Stimulation of Lateral Habenula Ameliorates Depression-like Symptoms in Rats: Involvement of Mesolimbic Circuit.

Deep brain stimulation (DBS) is a promising approach for the treatment of psychiatric disorders including depression, and various targets have been tested for their usefulness. Among these lateral habenula (LHb), a critical site linked to depression, has attracted considerable attention in recent years. DBS at LHb produced antidepressant activity by modulating the monoamine levels. However, the precise circuitry that mediates the positive effects have not been clarified. Herein we employed chronic unpredictable mild stress (CUMS) protocol to generate rats with depression-like phenotype. These

Neuroprotective Effects of C-Phycocyanin in Chronic Unpredictable Mild Stress-Exposed Rats: Regulation of Neuroinflammation and CREB/BDNF Signaling.

Depression is a widespread neuropsychiatric disorder that significantly impacts emotional and cognitive function. Antidepressant medications are frequently accompanied by various adverse effects. C-phycocyanin has been previously shown to exert potent anti-inflammatory, and neuroprotective properties. Therefore, this study evaluated the therapeutic effects of C-phycocyanin against anxiety and depressive-like behaviors, and memory dysfunction in an animal model of chronic unpredictable mild stress (CUMS)-induced depression and explored the underlying mechanisms. Rats were daily exposed for six

Frailty and deconditioning on the acute take.

It is well known that older patients have a higher incidence of adverse hospital outcomes, such as inpatient falls, delirium and longer lengths of stay, and that older people are likely to become deconditioned during their time in hospital. It is often felt that deconditioning occurs as a consequence of a prolonged stay; however, the adverse effects of immobility can occur within hours of arrival into urgent care. Hospital-aquired deconditioning is in fact a multi-systemic syndrome that manifests in a number of ways outside muscle weakness, including delirium, healthcare-associated infection,

Conscious Sedation Versus General Anesthesia for Percutaneous Left Atrial Appendage Occlusion: A Systematic Review and Meta-Analysis.

BACKGROUND: Percutaneous left atrial appendage occlusion (LAAO) has traditionally been performed under general anesthesia (GA) to maintain patient comfort and immobility, especially in the setting of transesophageal echocardiography. AIMS: We aimed to compare the safety and efficacy of conscious sedation (CS) compared to GA for LAAO, concurrent with the expansion of intracardiac echocardiography guidance. METHODS: A systematic search of the PubMed, Embase, Cochrane Central, and Scopus databases was conducted through December 2024 for studies comparing CS versus GA for LAAO. For outcomes of p

Hampton's Hump as a Radiologic Clue to Pulmonary Embolism in a Young Patient.

Pulmonary embolism may occur even in young adults with subtle risk factors such as prolonged immobility from mobile gaming. Hampton's hump on chest CT, though classically associated with infarction, remains a critical clue for diagnosis. Early recognition enables prompt anticoagulation and favorable outcomes, even in low-risk individuals.

Pharmacological stimulation of hippocampal GPR35 impairs spatial navigation, learning, and memory.

GPR35 is an orphan receptor expressed in astrocytes and neurons of the central nervous system (CNS), including the hippocampus; however, its role in cognitive functions is largely unknown. This study administered intra-hippocampal injections of GPR35 compound-10 (GPR35C10; a GPR35-selective agonist), ML194 (a GPR35-selective antagonist), and the combination of both to elucidate their pharmacological actions in the Barnes maze. Compared to the vehicle (10 % DMSO) and ML194 groups, animals treated with GPR35C10 showed a higher latency and longer distance to solve the maze (p < 0.05) (an effect p

Assessing the Sperm Head DNA Damage in Frozen/Thawed Horse Spermatozoa via Xenogeneic ICSI.

In the mouse, spermatozoa are highly resistant to DNA damage, even when frozen without cryoprotectants, and can produce offspring when subsequently used for ICSI (intracytoplasmic sperm injection). It is not known whether the same applies to other mammals as well. For example, in the horse, even conventional sperm freezing is still very problematic and frequently leads to sperm immobility. It has, however, never been tested whether sperm immobility also mirrors sperm head DNA damage, and if so, to what extent. In our study, we evaluated the damage to DNA in horse frozen and thawed motile and i

Targeting the NLRP3 inflammasome alleviates the comorbidity of chronic pain and depression via enhancing the autophagy.

The comorbidity of chronic pain and depression exhibits the bidirectional relationship and forms a vicious cycle in disease development, diagnosis and therapy. NLRP3 inflammasome-mediated neuroinflammation is a crucial factor for the pathogenesis of pain and depression. The underlying mechanisms of NLRP3 in the comorbidity of chronic pain and depression is unclear. In this study the spared nerve injury (SNI) mouse model was constructed, NLRP3 level was pharmacologically (MCC950) and genetically (Nlrp3−/−) regulated. As a result, SNI surgery induced pain hypersensitivity, depression-like, motor

Effects of UV-LED irradiation on decapod crustacean larval mortality, motility, and respiration: Implications for ballast water treatments and invasive species management.

Ballast water is a major vector for the transport of aquatic non-indigenous species among ecosystems worldwide. To comply with the International Convention for the Control and Management of Ships' Ballast Water and Sediments, vessels on international voyages commonly rely on Ballast Water Management Systems (BWMS) to meet the D-2 performance standard, often using ultraviolet (UV) irradiation as a key disinfection step. UV-light-emitting diodes (UV-LEDs) offer a mercury-free UV source with long lifespan and flexibility in wavelength selection; compared to conventional lamps, they are smaller in

General versus nongeneral anesthesia in transfemoral and transradial carotid artery stenting.

BACKGROUND: The optimal anesthetic strategy for carotid artery stenting (CAS) remains uncertain. General anesthesia (GA) ensures immobility and airway control but may increase hemodynamic instability, whereas monitored anesthesia care or local anesthesia permits real-time neurological assessment. This multicenter study evaluated the association between anesthetic modality and clinical outcomes after CAS. METHODS: Data were prospectively collected from 15 comprehensive stroke centers in the United States between January 2023 and December 2024. Adults undergoing CAS for atherosclerotic carotid

Next Generation Experimental Models of Venous Physiology and Pathology: Examining Human Relevance in Recapitulating Lower Extremity Flow and Valve Function in New Approach Methods (NAMs).

Venous return depends on compliant vessels, competent valves, and auxiliary pumps such as the calf and thoracic muscles. Disruption of these mechanisms, through immobility, valve dysfunction, or inflammation, creates disturbed flow and heterogeneous shear that drive venous dysfunction and disease progression. Traditional animal models, though informative, fail to replicate human venous geometry and hemodynamics. This review critically evaluates current in vitro and in silico venous models, highlighting their capabilities and limitations in reproducing physiological flow, endothelial responses,

Outcomes of neurosurgery and otolaryngology team surgery for revision vagus nerve stimulation implantation surgery.

PURPOSE: Vagus nerve stimulation (VNS) is a well-established FDA-approved option for drug-resistant epilepsy. Injury to the vagus nerve can result in vocal cord immobility, which can lead to significant dysphagia and communication deficits. We hypothesized that neurosurgery-otolaryngology collaboration for revision VNS implantation would reduce postoperative complications compared to neurosurgery-only revision. METHODS: A retrospective chart review was conducted on all consecutive patients who underwent VNS at a tertiary care children's hospital from 2005 to 2023. Details of the cases were co

Development and optimization of dothiepin hydrochloride-loaded nanostructured lipid carrier incorporated in situ nasal gel for enhanced brain-targeted antidepressant delivery.

This study aimed to develop dothiepin HCl-loaded nanostructured lipid carriers (NLCs) incorporated into an in situ nasal gel. NLCs were chosen to overcome the drug's poor solubility, extensive first-pass metabolism, and limited blood-brain barrier penetration, thereby enhancing brain-targeted delivery and antidepressant efficacy via the nasal route. DOTH.HCl suffers from poor solubility, extensive first-pass metabolism, and limited ability to cross the blood-brain barrier, which significantly restricts its therapeutic efficacy. Dothiepin HCl-loaded NLCs were prepared by melt emulsification and
