Prostate Cancer Therapy via Inhibition of Negative Immune Regulation.

Prostate cancer exhibits complex pathophysiology, rendering its management arduous due to heterogeneity and recalcitrance toward conventional therapeutics. The increasing focus on immune checkpoint inhibitors for treatment stems from the limited existing therapeutic approaches and the impressive effectiveness of these inhibitors demonstrated in other malignancies. Denoted as immunologically "cold" with low tumor mutation burden (TMB), prostate cancer manifests diminished immune cell infiltration in tumoral tissue, potentially culminating in inadequate response to immune checkpoint blockade. Anti-GQ1b-Positive Bickerstaff Brainstem Encephalitis Complicated by Diffuse Hypertrophic Pachymeningitis: A Case Report.

A 36-year-old man developed acute progressive ataxia, an impaired consciousness, and ophthalmoplegia after an upper respiratory tract infection. A cerebrospinal fluid analysis revealed pleocytosis in the patient. Initial plain brain magnetic resonance imaging (MRI) showed no remarkable findings, whereas initial brain perfusion single-photon emission computed tomography demonstrated hyperperfusion in the brainstem. Follow-up contrast-enhanced MRI revealed diffuse dural thickening, and a histopathological examination supported the diagnosis of hypertrophic pachymeningitis. Anti-GQ1b antibodies w

Rational Design and Evaluation of Novel ATR Inhibitors for Osteosarcoma Therapy: An Integrated 3D-QSAR, Molecular Docking, and Molecular Dynamics Study.

BACKGROUND: Ataxia Telangiectasia and Rad3-related protein kinase (ATR) is a central regulator of the DNA damage response and represents an attractive therapeutic target in tumors with high replication stress, including osteosarcoma. This study aimed to characterize the structure- activity relationships of a distinct ATR-targeting scaffold and to guide the rational design of new inhibitors using integrated computational approaches. METHODS: A dataset of 23 previously reported derivatives was used to construct a Comparative Molecular Similarity Indices Analysis (CoMSIA) 3D-QSAR model, with 18

Clinical characteristics and treatment analysis of children with glucose transporter type 1 deficiency syndrome: a report of two cases.

BACKGROUND: Glucose transporter type 1 deficiency syndrome (GLUT1-DS) is a rare neurometabolic disorder caused by mutations in the SLC2A1 gene, leading to glucose transporter type 1 (GLUT1) dysfunction and cerebral energy failure. Clinically, it presents with intractable epilepsy, movement disorders, and developmental delay. Timely diagnosis remains challenging due to clinical heterogeneity. CASE DESCRIPTION: We report two pediatric cases that presented with early-onset drug-resistant epilepsy, ataxia, and developmental delay. Case 1 presented with refractory epilepsy that was poorly responsi

Stage-Specific Patterns of Imaging and Non-Motor Biomarkers in Spinocerebellar Ataxia Type 3.

OBJECTIVE: Advancements in disease-modifying therapies for spinocerebellar ataxia Type 3 (SCA3) underscore the need for robust biomarkers to monitor disease progression and evaluate treatment efficacy. Although motor phenotypes have been well characterized, less is known about non-motor and imaging-based features across different disease stages. This study aimed to characterize stage-specific patterns of non-motor and imaging biomarkers in SCA3. METHODS: This cross-observational study included 229 patients with SCA3 and 104 age- and sex-matched healthy controls from the Organization for South

Structural mechanism of TRPC3 inhibition by a potent antagonist.

The transient receptor channel canonical family member 3 (TRPC3) is a non-selective, calcium-permeable cation channel within the larger TRP superfamily. TRPC3 is expressed in both excitable and non-excitable cells, where it integrates multiple signaling pathways and is directly activated by binding of diacylglycerol following activation of G-protein coupled receptors. Increased TRPC3 signaling is implicated in several diseases, highlighting TRPC3 as a potential therapeutic target. However, the development of TRPC3 inhibitors with suitable potency and selectivity has been challenging due to the

[Ginsenoside Rg_1 attenuates OGD/R induced HT22 cell damage by inhibiting oxidative stress via ROS-ATM-CHK2 pathway].

This study focused on ginsenoside Rg_1(GRg_1) and aimed to explore whether it could alleviate oxygen-glucose deprivation/reoxygenation(OGD/R)-induced damage in mouse hippocampal neuronal(HT22) cells by regulating the reactive oxygen species(ROS)-ataxia telangiectasia mutated(ATM)-checkpoint kinase 2(CHK2) signaling pathway, thereby influencing apoptosis and oxidative stress responses. First, assessment of HT22 cell viability showed that GRg_1 significantly enhanced the viability of HT22 cells. Second, Western blot analysis demonstrated that GRg_1 markedly reduced the apoptosis rate and decreas

Frequency-dependent cerebellar circuits independently gate social vocalizations and movement.

Communication depends on precise coordination between motor execution and cognition. Here we reveal that the cerebellum exerts real-time control over social vocalizations in adult mice. Optogenetic activation of excitatory cerebellar output in the superior cerebellar peduncle suppressed ultrasonic vocalizations with frequency-dependent potency while inducing distinct motor phenotypes. Systematic functional mapping across cerebellar regions and cell types revealed that vocal suppression can occur in the absence of overt motor impairment, suggesting selective control of vocal output beyond gross

Juvenile Huntington Disease With Cerebellar-Predominant Atrophy Identified by Genome Sequencing.

Juvenile Huntington disease accounts for a small fraction of Huntington disease cases and is driven by the largest trinucleotide repeat expansions. We describe a 6-year-old boy with progressive global regression, generalized dystonia, and epilepsy, in whom serial imaging showed striking cerebellar-predominant atrophy with an initially normal-appearing striatum. Extensive metabolic testing, exome sequencing, and an ataxia repeat expansion panel were nondiagnostic. Research genome sequencing identified a large pathogenic huntingtin CAG repeat expansion that confirmatory testing sized beyond 180

What does it take to grasp a tool? A fine-grained multiple-case analysis of hand-tool and tool-object interaction errors in apraxia.

Apraxia of tool use is a neuropsychological syndrome characterized by difficulties in selecting or using familiar tools that cannot be better explained by primary motor, sensory, attentional, or language deficits. The neuropsychological literature indicates that tool use is a complex, multifaceted ability relying on heterogeneous cognitive processes and neural substrates. Patients may commit errors at different stages of the grasp-to-use sequence, including hand-tool interaction errors (e.g., holding tools upside-down), tool-object association errors (i.e., incorrect matching of the functional

Drug repurposing in hereditary ataxia.

Hereditary ataxias are rare disorders, and a context where small cohorts and high development costs make drug repurposing a pragmatic route. Technological advances are creating opportunities to complement pathway-guided plausibility with more target-supported and AI-nominated mechanistically grounded candidates. Also, digital twin and The Virtual Brain frameworks should be viewed in hereditary ataxias as future-oriented research tools to integrate multimodal data, support stratification, and generate testable system-level predictions in rare cohorts.

Ionizing radiation and Alzheimer's disease: epidemiology, mechanisms, and eCIRP-mediated tau pathology.

As human radiation exposure expands through medical imaging, nuclear industry growth, and the acceleration of crewed space exploration, understanding the molecular mechanisms linking radiation to Alzheimer's disease (AD) is becoming an increasingly urgent priority. Here, we review epidemiological, mechanistic, and therapeutic evidence for a relationship between ionizing radiation and AD, focusing on a neuroinflammatory pathway centered on extracellular cold-inducible RNA-binding protein (eCIRP). Epidemiological support comes from occupational cohorts of nuclear workers and studies linking resi

Ficd loss rescues motor impairments and reverses oligodendrocyte maturation deficits in a mouse model of spinocerebellar ataxia type 3.

Spinocerebellar ataxia type 3 (SCA3) is an inherited, fatal neurodegenerative disease caused by a pathological CAG repeat expansion in the ATXN3 gene, resulting in the selective degeneration of vulnerable neuronal populations. Recent work has identified impairments in oligodendrocyte maturation as a novel and robust feature of SCA3 pathogenesis. Oligodendrocytes synthesize myelin structural components through the endoplasmic reticulum (ER), rendering this organelle essential for white matter integrity. Despite this, the role of ER function in SCA3 remains unclear. In this study, we show that l
