Case report of anti-vimentin IgG-associated autoimmune encephalitis: diagnostic challenges and the role of 18F-DPA714 PET/MRI.

BACKGROUND: Anti-vimentin IgG-associated autoimmune encephalitis (AE) is a recently described autoimmune astrocytopathy characterised by relapsing-remitting cerebellar ataxia, brainstem involvement and pyramidal tract lesions. Nevertheless, several key aspects remain poorly elucidated, including the full clinical phenotypic spectrum (particularly peripheral nervous system (PNS) involvement), the interpretation of serological discrepancies and long-term management outcomes. Additionally, the imaging evidence for neuroinflammation and the evidence regarding evolution following treatment remain i

Targeting senescence-associated fibrotic signatures identifies syringin as an effective modulator of the ATR/CHK1-p53 axis in idiopathic pulmonary fibrosis.

Idiopathic pulmonary fibrosis (IPF) is a progressive and fatal interstitial lung disease closely associated with aging, yet the contribution of cellular senescence to fibrotic remodeling and its therapeutic implications remain incompletely understood. Syringin, a phenylpropanoid glycoside derived from Acanthopanax senticosus, has been reported to possess anti-inflammatory and antioxidant activities, but its role in IPF has not been elucidated. In this study, bulk lung transcriptomic data from the GTEx project and nine IPF cohorts were integrated with IPF scRNA-seq data to characterize senescen

Large-scale genetic profiling of hereditary ataxias in China: implications for a stepwise molecular diagnostic strategy.

BACKGROUND: Hereditary ataxias are clinically and genetically heterogeneous, which complicates clinical management, genetic counseling, and scientific research. Efficient molecular diagnosis is essential but requires molecular epidemiological considerations due to the variable genetic landscapes across regions and populations. In China, despite the established predominance of spinocerebellar ataxia type 3 (SCA3), comprehensive data on the genetic landscape of hereditary ataxias are limited and increasingly outdated, leaving insufficient evidence to support an epidemiology-informed molecular di

Improving Video-Based Prediction of Cerebellar Ataxia Severity Using a Pretrained Deep Learning Model.

BACKGROUND: Ataxia severity in degenerative cerebellar diseases (DCDs) is usually assessed with semi-quantitative clinical rating scales such as the Scale for the Assessment and Rating of Ataxia (SARA), which are subject to variability. Deep learning-based gait analysis offers an objective alternative, but labeled data in DCDs are limited. We investigated whether pretraining on gait videos from patients with Parkinson's disease (PD) could improve SARA score prediction in DCD. METHODS: Gait videos from patients with DCD were processed using pose estimation, and the extracted time-series keypoi

Circulating miR-181a-3p, ATR, and HMGB-1 as Integrated Predictors of Induction Blast Clearance in Adults with Acute Myeloid Leukemia.

METHODS: This study included 71 adult patients with newly diagnosed AML who received standard "7+3" induction chemotherapy. Baseline circulating miR-181a-3p and miR-181b-5p levels were quantified using quantitative real-time polymerase chain reaction, while ATM, ATR, and HMGB-1 protein levels were measured using enzyme-linked immunosorbent assay. Induction blast clearance was defined as a post-induction bone marrow blast count of <5%. Receiver operating characteristic (ROC) curve analysis was used to determine optimal cut-off values. Associations were evaluated using Spearman correlation and r

DNA damage response associated miRNA dysregulation in Ataxia-telangiectasia.

BACKGROUND: Ataxia-telangiectasia (A-T) is a rare autosomal recessive genome-instability disorder due to pathogenic variants in ATM, a central kinase of the DNA double-strand break signaling, checkpoint activation and stress-response pathways. Although microRNAs (miRNAs) regulate DNA damage response (DDR), apoptosis, senescence and cell-cycle control, the stress-responsive behavior of selected ATM/DDR-associated miRNAs remains incompletely characterized in A-T fibroblast models. METHODS AND RESULTS: Four predefined ATM/DDR-associated candidate miRNAs (hsa-miR-34a-5p, hsa-miR-26a-5p, hsa-miR-1

Anti-trapping scaffold protein: significance in cancer cell innate immune signaling and opportunities for immunotherapy.

X-ray repair cross-complementing protein 1 (XRCC1) is a scaffold protein that functions as a molecular hub, serving as a platform for the recruitment and interaction of multiple DNA repair proteins to coordinate and facilitate multistep DNA repair processes. Furthermore, it acts as an antitrapping protein, ensuring the timely release of DNA repair proteins to prevent genotoxicity. XRCC1 participates in single-strand break repair (SSBR) pathways and has also been implicated in other DNA repair pathways. Preclinical studies have shown that XRCC1 deficiency delays SSB rejoining, induces mutations

DNA end-resection is stimulated by an interaction between BRCA1 exon 11 and TOPBP1.

Approximately 60% of the tumour suppressor protein BRCA1 is encoded by a single exon. Many tumours carry mutations in this exon, often resulting in exon skipping and thus a protein of a severely reduced size. Although none of the well-described protein domains of BRCA1 are encoded by this exon 11, the isoform lacking this part shows a hypomorphic activity in homologous recombination. To better understand the function of this large exon, we performed proteomic analyses to identify interaction partners via this part of the protein. Here, we report a DNA damage- and phospho-dependent interaction

Levofloxacin-induced Ataxia in a Child After Complicated Pneumonia: A Case Report.

A 6-year-old child with complicated necrotizing pneumonia developed acute ataxia, urticarial rash and urinary incontinence on the third day of levofloxacin therapy. No structural abnormalities were found on imaging. The antibiotic was discontinued, and corticosteroids with antihistamines were initiated, leading to complete symptom resolution without sequelae. This pediatric case underscores the need for vigilance regarding levofloxacin-induced movement disorders, even with short-term use.

Hearing shapes: the Bouba-Kiki effect in adult domestic dogs.

Iron-deficiency anemia is common worldwide and typically treated by oral iron supplementation. Excess enteral iron, however, may cause pathological outcomes. Developing new repletion approaches is thus warranted. Previous experimentation revealed that select amino acids (AAs) induce trafficking of transporters onto the enterocyte brush-border membrane (BBM) and enhance electrolyte absorption/secretion. Here, we hypothesized that certain AAs would increase the abundance of the main intestinal iron importer, divalent metal-ion transporter 1 (DMT1), on the BBM of duodenal enterocytes, thus stimul

Nutritional strategies in kidney transplantation: optimising outcomes beyond surgery.

Although kidney transplantation (KTx) remains the optimal therapy for end-stage kidney disease, long-term graft survival and patient outcomes continue to be limited by cardiometabolic complications and chronic allograft dysfunction. Nutritional management has evolved from supportive care to a core component of post-transplant follow-up. The early post-transplant phase is marked by catabolism, high-dose immunosuppression, and metabolic instability, leading to increased protein requirements but also potential susceptibility to hyperfiltration injury. In the stable post-transplant phase, a modera

Cells heal themselves: gene-edited hematopoietic stem cells restore frataxin deficiency in multiple organs.

BACKGROUND: Mortality rates increase due to iron deposition in the cardiac muscles of thalassemia major (TM) patients. Iron overload cardiomyopathy could be treated with a combination therapy of an iron chelator and an L-type calcium channel blocker. We designed a randomized controlled study to assess the potential of amlodipine, alongside chelation, in reducing myocardial iron concentration in TM patients compared with a placebo. OBJECTIVES: This study aims to estimate the change in myocardial iron concentration (MIC) determined by magnetic resonance imaging after 6 months of treatment with

Waist Circumference, Relative Fat Mass Index, and BMI as Classifiers of Diabetes Status: A Comparative, Cross-sectional Analysis in 82 Low- and Middle-Income Countries.

OBJECTIVE: Waist circumference-based measures can improve diabetes risk assessment beyond BMI. Yet, evidence to guide their integration into screening guidelines in low- and middle-income countries (LMICs) is limited, particularly to assess risk among individuals at intermediate BMI ranges. We evaluated the associations of waist circumference (WC), relative fat mass (RFM), and BMI with diabetes across 82 LMICs. RESEARCH DESIGN AND METHODS: Individual-level data were analyzed from nationally representative surveys across 82 LMICs, comprising 598,883 adults aged ≥25 years with a BMI of 18.5-29.
