Recent developments in Friedreich's ataxia: a state-of-the-art review.

Friedreich's Ataxia (FRDA) is a progressive disorder, which is autosomal recessive. It is one of the most common inherited ataxias in Europe, the Middle East, South Asia and North Africa. It is a life limiting condition, the time of onset until death is on average 36 years. A lack of certainty about specific phenotypic changes, has meant that, until recently, there have been no medications licensed for use in FRDA. Over the last 5 years, the development of omaveloxolone has stimulated increasing research into the mechanistic understanding of FRDA. We have conducted a critical review of all the

A phosphorylated variant of the mast/stem cell growth factor receptor KIT is upregulated in dorsal root ganglia of Friedreich ataxia.

Friedreich ataxia (FA) causes hypoplasia of nerve cells in dorsal root ganglia (DRG). Beyond hypoplasia, however, the lesion in DRG includes disorganization and proliferation of satellite cells, formation of residual nodules, and neuronophagia. Antibody microarray analyses of DRG lysates with 878 validated antibodies that target cell signaling proteins reveal prominent up-regulation of the mast/stem cell growth factor (SCF) receptor-tyrosine kinase KIT. In its bioactive form upon engagement with SCF, KIT undergoes dimerization and autophosphorylation at Tyr-936 amongst other sites, and is here

Interaction of FXTAS Family History and College Degree Attainment Predicts Trajectories of Cognitive and Motor Symptoms in FMR1 Premutation Carrier Women.

The present longitudinal study focuses on FMR1 premutation carrier women during midlife and early old age (n = 115). Bringing together the genetic risk factor of a family history of FXTAS and the environmental protective factor of higher education, the goal of the study was to determine how these factors potentially interact to predict self-reported FXTAS-type symptoms in women carrying the FMR1 premutation. We investigated age-related changes in symptoms, measured prospectively over a decade. Using an accelerated longitudinal design approach, we estimated trajectories of change extending from

Ancestral and divergent roles of the Marchantia polymorpha ATAXIA TELANGIECTASIA AND RAD3-RELATED kinase in maintaining genome integrity and clonal fitness.

The eukaryotic ATAXIA TELANGIECTASIA AND RAD3-RELATED (ATR) kinase is essential for ensuring genomic integrity under conditions that cause replication defects. It triggers a transient cell cycle arrest and DNA repair by activating the DNA damage response pathway. While ATR kinases appear to be functionally conserved, species-specific phenotypes can be observed across the plant kingdom. To investigate the ancestral roles of the plant ATR, we identified its homolog in the model liverwort Marchantia polymorpha, which we named MpATR. Similar to other plant species, MpATR preserves meristematic cel

A Rare Case of Spinocerebellar Ataxia 50: The First Reported Case from the Indian Subcontinent with a Review of the Literature.

Spinocerebellar ataxia (SCA)-50 is a recently reported rare subtype. We describe the first case of SCA-50 from the Indian subcontinent. A 22-year-old man presented with progressive cerebellar ataxia over 2 years, along with oculomotor abnormalities and imaging findings indicating mild cerebellar atrophy. Genetic testing revealed an NPTX1 gene mutation, which correlates with SCA-50. Its unique pathogenesis distinguishes it from other SCA subtypes.

DNA replication stress and translational repression converge to drive CDK1- and caspase-dependent apoptosis in Ewing sarcoma.

Despite aggressive multimodal therapy, including cytotoxic chemotherapy, surgery, and radiation, the prognosis for patients with Ewing sarcoma remains poor, particularly for those with metastatic or relapsed disease. Combining agents that increase DNA replication stress with ATR-CHK1-WEE1 pathway inhibitors, which disrupt the DNA damage response and cell cycle checkpoints, is a promising strategy under clinical investigation in Ewing sarcoma and other cancers. However, the mechanisms by which these drug combinations selectively kill cancer cells under replication stress remain incompletely und

Acute nitrate/nitrite poisoning in cattle caused by ingestion of Tithoniatubaeformis.

An outbreak of acute mortality affecting 13% of a herd of 450 beef cattle is described in the province of Salta, Argentina. The incident occurred after the introduction of the herd into a paddock that had previously been used for cattle confinement. Approximately 8 h after being introduced into the paddock, the affected animals developed hyperacute clinical signs, including weakness, ataxia, tachycardia and brown mucous membranes, followed by death within minutes. The autopsy findings included poorly coagulated dark chocolate-brown blood and generalized congestion, consistent with methemoglobi

Multimodal dynamics control activity of a glial glutamate transporter.

Membrane transporters move polar solutes across lipid bilayers to regulate cellular metabolism, signaling, and drug distribution. These proteins operate via an alternating-access mechanism, cycling between extracellular-, intermediate-, and intracellular-facing conformations. The human excitatory amino acid transporter 1 (EAAT1) protects neurons from excitotoxic damage by mediating the uptake of glutamate and aspartate into glial cells. Defects in EAAT1 function result in numerous pathologies, including epilepsy and ataxia, suggesting that positive modulation of these transporters might amelio

Loss of SARM1 Improves Phenotypes in a Mouse Model of Autosomal Recessive Spastic Ataxia of Charlevoix-Saguenay.

BACKGROUND AND OBJECTIVES: Autosomal Recessive Spastic Ataxia of Charlevoix-Saguenay (ARSACS) is a neurodegenerative disease caused by pathogenic variants in SACS. ARSACS is characterized by mitochondrial abnormalities and disruptions of the neurofilament cytoskeleton. In other conditions, these features have been linked to activation of Sterile Alpha and TIR Motif Containing 1 (SARM1), an enzyme that can trigger axon degeneration and neuronal death. Inhibition of SARM1 is an attractive therapeutic strategy because SARM1 is inactive in healthy cells and knockout of SARM1 has little or no delet

The role of FMR1 mRNA structure on the efficiency of non-canonical translation of toxic polyglycine protein.

Repeat-associated non-AUG (RAN) translation of mutant FMR1 messenger RNA (mRNA) containing CGG repeat expansions results in the production of a toxic polyglycine protein (FMRpolyG), which contributes to fragile X premutation-associated conditions (FXPAC), including fragile X-associated tremor/ataxia syndrome (FXTAS). The 5' untranslated region of FMR1 mRNA folds into a thermodynamically stable secondary structure at the region of excessively expanded CGG repeats and constitutes a template for RAN translation initiated from near-cognate start codons located upstream of the CGGs. Cis-regulatory

Immunotherapy-Induced Glutamic Acid Decarboxylase 65 Cerebellitis.

Immune checkpoint inhibitors (ICIs) are an important treatment modality for advanced cancer. However, they carry the risk of immune-related adverse events (irAEs) secondary to their mechanism of action. ICI medications function by inhibiting the checkpoints that suppress immune cells, which exist to prevent autoimmunity, and prime these cells to attack and eliminate cancer. While ICIs have substantially improved the prognosis for patients with advanced cancer, some patients experience adverse effects from this autoimmune state. One less common, but potentially life-threatening toxicity is cere

Cerebellar Ataxia and Chorea: Genetic Etiologies and Key Considerations for Diagnosis and Management.

BACKGROUND: The differential diagnosis of progressive ataxia is broad. Multiple degenerative, inherited, and acquired causes of ataxia must be considered. The presence of chorea as part of the clinical phenotype provides invaluable clinical information which may help to narrow the differential. OBJECTIVES: There are a limited number of conditions with both prominent chorea and cerebellar ataxia, and it is essential for clinicians to recognize these disorders to direct early disease-specific treatment. We aim to identify these conditions and propose a diagnostic approach. METHODS: We conducte
