Multifaceted signaling of the cell surface receptor neuropilin-1 (NRP1): pathophysiological roles and therapeutic potential.

Neuropilin-1 (NRP1) is a developmentally conserved cell surface receptor that functionally acts to bind various ligands like class 3 semaphorins and several members of the VEGF family to engage in different signaling pathways that control cell migration and motility. Due to its widespread expression, NRP1 is implicated in immune function, axonal guidance, viral entry, tumor progression and angiogenesis. NRP1 is dysregulated across multiple cancer types and promotes tumor progression in a context-dependent manner in tumor cells and the tumor microenvironment, including endothelial, stromal, and

Taste and Smell Disturbances Associated With Secondary Adrenal Insufficiency: A Case Report.

BACKGROUND: Adrenal insufficiency often presents with nonspecific symptoms, impeding early diagnosis. Taste and smell disturbances are rarely recognized as presenting features. CASE PRESENTATION: We report the case of a 54-year-old man with a 3-month history of persistent taste and smell disturbances accompanied by fatigue, loss of appetite, weight loss, and diffuse body pain, including thigh stiffness. Endocrine evaluation with stimulation tests revealed markedly low levels of cortisol and adrenocorticotropic hormone, leading to a diagnosis of secondary adrenal insufficiency. After glucocort

Persistent Smell and Taste Disorders in the Post-COVID-19 Pandemic Era: A Cross-National Study of Prevalence and Quality of Life in the UK and the USA.

Anosmia/hyposmia affected 14.8%-19.6% of participants in the UK and the USA in the post-pandemic era. Since symptom onset, 37.1%-48.8% of affected participants reported no improvement, while 15.3%-17.6% reported worsening symptoms. Anosmia/hyposmia remains strongly associated with reduced quality of life across both countries. Among respondents reporting anosmia or hyposmia, a high proportion (88.8%-93.5%) also experienced concurrent parosmia.

Isolated Parosmia Caused by Vitamin B12 Deficiency.

Vitamin B12 deficiency classically presents with neurological or hematological abnormalities. Prior reports have shown that vitamin B12 deficiency also causes olfactory dysfunction when patients have concurrent neurological or hematologic findings. To our knowledge, this is the first case of isolated parosmia caused by vitamin B12 deficiency in the absence of neurological/hematological abnormalities or structural sinonasal disease. A 20-year-old man presented with a 3-month history of parosmic symptoms. Nasal endoscopy, brain MRI, and neurological examinations were normal. A laboratory evaluat

Olfactory ERP-based classification of anosmia and normosmia using machine learning.

OBJECTIVE: This study aimed to investigate whether electroencephalogram (EEG) data can be used to classify normosmia and anosmia using machine learning approaches and to examine how classification performance varies across single and combined chemical stimulation conditions. METHODS: EEG signals were recorded from 66 participants (26 normosmic and 40 anosmic) during olfactory stimulation with phenyl ethyl alcohol (PEA), hydrogen sulfide (H₂S), and carbon dioxide (CO₂), presented to the left and right nostrils. EEG data were averaged for each condition, resulting in six chemosensory event-rela

Navigating the efficacy, safety and outcome of intranasal platelet-rich plasma for persistent olfactory loss: a systematic review and meta-analysis.

BACKGROUND: Persistent olfactory dysfunction (OD) poses a significant clinical challenge with limited therapeutic options. Intranasal platelet-rich plasma (PRP), an autologous source of regenerative growth factors, presents a biologically plausible intervention, yet its efficacy across various OD etiologies remains synthetically underexplored. METHODS: This systematic review and meta-analysis adhered to PRISMA guidelines. A comprehensive search of PubMed/MEDLINE, Web of Science, and Cochrane Library (2010-2025) identified studies assessing intranasal PRP for persistent OD (> 6 months). Random

A Life Without Olfactory Function Has Limited Effects on Cerebral White Matter Morphology.

Absence of sensory input is associated with alterations in brain morphology; mainly in or near cerebral regions normally devoted to processing of the missing sense. However, multiple recent studies demonstrate that the only consistent morphological finding within the gray matter of individuals born without the sense of smell (isolated congenital anosmia [ICA]), are changes in or near the olfactory sulcus. In contrast, for the connecting tissue of the brain, the white matter (WM), previous studies have yielded inconsistent findings. Here, we show that individuals with ICA (n = 49) exhibit clear

The Unmet Need of Olfactory Testing in Inflammatory Disorders of the Upper Airways-An EAACI Position Paper.

The sense of smell, with its extensive evolutionary history, is highly prone to disorders that can have a profound impact on daily life. Anosmia affects approximately 5% of the population, with an additional 15% exhibiting reduced olfactory function. The prevalence of olfactory dysfunction (OD) varies by population and age group, and standardized testing reveals a broad range of impacts. OD includes various causes, most commonly aging, inflammation of the olfactory epithelium, upper respiratory tract infections (URTI), traumatic brain injury, and neurological conditions. The recent COVID-19 pa

Selective umami taste impairment in anosmic Parkinson's disease: associations with non-motor symptoms.

BACKGROUND AND PURPOSE: Olfactory dysfunction (OD) is a well-established prodromal feature of Parkinson's disease (PD). However, the association between olfactory loss and gustatory alterations - particularly umami perception - and their relationship with non-motor symptoms (NMS), affective status, reward-related eating behavior, and quality of life (QoL) remain incompletely characterized. The purpose of this study was to investigate the relationship between objectively defined anosmia and taste perception, NMS, mood disturbances, reward-driven eating behavior, and QoL in patients with PD. Exploring the Paradoxical Association Between Chest CT Pulmonary Involvement and Olfactory Dysfunction in COVID-19 Patients: A Prospective Cross-Sectional Study.

BACKGROUND AND AIMS: Olfactory dysfunction (OD) is common in COVID-19, yet its relationship with pulmonary involvement remains inconsistent. We evaluated the association between CT-based lung involvement and OD in laboratory-confirmed COVID-19. METHODS: In a prospective cross-sectional study (Yasuj University of Medical Sciences hospitals; December 26, 2021-May 22, 2022), OD was assessed using the CCCRC protocol and pulmonary involvement was categorized using a CT lung involvement score. Associations were examined using chi-square testing (categorical) and ANOVA where appropriate (α = 0.05).
