Predictive value of the neutrophil-to-lymphocyte ratio for 28-day mortality in elderly patients with sepsis: a cohort study based on the MIMIC-IV database.

BACKGROUND: The prognostic value of the neutrophil-to-lymphocyte ratio (NLR) in elderly patients with sepsis remains uncertain. This retrospective cohort study investigated the association between NLR at hospital admission and 28-day mortality in this population. METHODS: Data were extracted from the MIMIC-IV database (2008–2019), including 7,522 patients aged ≥65 years diagnosed with sepsis. The primary exposure was NLR measured upon intensive care unit (ICU) admission, and the outcome was 28-day all-cause mortality. Multivariable Cox proportional hazards models and Kaplan–Meier survival anal

Malignant neoplasm of skin with risk of sepsis: A bidirectional Mendelian randomization analysis.

Sepsis, a life-threatening condition characterized by dysregulated immune responses, shares pathophysiological features with malignant skin neoplasms, including immunosuppression and barrier disruption. Nonetheless, robust evidence for a causal relationship is lacking. To assess the causal association between malignant skin neoplasms and sepsis risk using bidirectional Mendelian randomization (MR). We performed a bidirectional 2-sample MR analysis under standard assumptions. Sepsis data came from the UK Biobank (1896 cases/484,488 controls; sepsis-3 criteria/International Classification of Dis

Ceralasertib plus durvalumab in Ras mutant and Ras wild type advanced non-small cell lung cancer: Two cohorts of the phase II platform National Lung Matrix Trial.

PURPOSE: Pre-clinical data suggests that ceralasertib, a potent ATR inhibitor, might reverse resistance to anti-PD-(L)1 in KRAS mutant lung adenocarcinoma (LUAD). Activity of ceralasertib/durvalumab was, therefore, assessed in two prospective, parallel immune checkpoint blockade (ICB)-resistant cohorts, one harbouring KRAS mutations (J1), the other without (NAJ), within the National Lung Matrix Trial. METHODS: Oral ceralasertib was administered (240 mg twice daily on days 15-28) with durvalumab (1500 mg IV, on day 1 of every 28-day cycle) until disease progression. Co-primary outcomes were ob

EA mitigates sepsis-induced liver injury by inhibiting the proinflammatory pathways IκBα/NF-κB and NLRP3-mediated pyroptosis.

The primary focus of sepsis therapy is to alleviate organ dysfunction. Sepsis-associated liver injury (SALI) occurs in about 40 % of sepsis patients, yet targeted therapeutic strategies remain unavailable. Electroacupuncture (EA) has emerged as a potential adjunctive therapy for sepsis, owing to its capacity to modulate the neuroendocrine-immune network. This study aims to investigate whether EA can alleviate SALI and its potential mechanisms. A murine model of sepsis was established via cecal ligation and puncture (CLP). The Zusanli (ST36) and Neiguan (PC6) acupoints, known for their anti-inf

Protocol for arbovirus infection of Aedes aegypti by intrathoracic microinjection or artificial blood feeding.

Here, we present a protocol for infecting adult female Aedes aegypti with arboviruses using intrathoracic microinjection or artificial blood feeding as alternative approaches. We describe steps for rearing mosquitos, virus preparation, intrathoracic microinjection, and artificial blood feeding. We then detail procedures for saliva collection, tissue dissection (midgut and carcass), and assessment of vector competence. The protocol provides practical guidance, critical experimental considerations, and validation steps to improve reproducibility and consistency across laboratory settings.
